Fresh ginseng is prone to spoilage due to its high moisture content. For long-term storage, most fresh ginsengs are dried to white ginseng (WG) or steamed for hours at high temperature/pressure and dried to form Korean Red ginseng (KRG). They are further processed for ginseng products when subjected to hot water extraction/concentration under pressure. These WG or KRG preparation processes affect ginsenoside compositions and also other ginseng components, probably during treatments like steaming and drying, to form diverse bioactive phospholipids. It is known that ginseng contains high amounts of gintonin lysophosphatidic acids (LPAs). LPAs are simple lipid-derived growth factors in animals and humans and act as exogenous ligands of six GTP-binding-protein coupled LPA receptor subtypes. LPAs play diverse roles ranging from brain development to hair growth in animals and humans. LPA-mediated signaling pathways involve various GTP-binding proteins to regulate downstream pathways like [Ca2+]i transient induction. Recent studies have shown that gintonin exhibits anti-Alzheimer's disease and antiarthritis effects in vitro and in vivo mediated by gintonin LPAs, the active ingredients of gintonin, a ginseng-derived neurotrophin. However, little is known about how gintonin LPAs are formed in high amounts in ginseng compared to other herbs. This review introduces atypical or non-enzymatic pathways under the conversion of ginseng phospholipids into gintonin LPAs during steaming and extraction/concentration processes, which exert beneficial effects against degenerative diseases, including Alzheimer's disease and arthritis in animals and humans via LPA receptors.
약용식물인 천궁(Cnidium officinale Makino)의 미활용 부위의 생리활성 탐색을 위한 기초연구로서, 천궁 지상부(잎과 줄기)를 ethyl acetate, methanol, 물의 3가지 용매로 추출한 후, paper disc method와 microtiter broth dilution method를 이용하여 항균활성을 측정하였다. 천궁 지상부 ethyl acetate 추출물과 methanol 추출물은 비교적 높은 항균활성을 보였으며, Gram 양성균, Gram 음성균, 그리고 효모 등에 대해 broad한 항균범위를 지닌 것으로 나타났다. 특히 ethyl acetate 추출물은 0.4 mg/disc의 농도에서 Staphylococcus aureus, Salmonella typhimurium, Listeria monocytogenes등의 식중독균에 대해 각각 9.7 mm, 9.2 mm, 10.0 mm의 억제환을 보였다. Saccharomyces cerevisiae에 대한 항진균활성은 물 추출물과 유기용매 추출물에서 확인되었다. 그러나 물 추출물의 항균 및 항진균 활성은 유기용매 추출물에 비해 현저히 낮아 비교적 높은 농도에서 항균효과가 나타났다. 천궁 지상부 ethyl acetate 추출물의 항균활성은 사용 농도와 균의 종류에 따라 다른 것으로 나타났으며, 0.5 mg/mL의 농도에서 Staphylococcus aureus와 Pseudomonas aerogenes의 생육은 완전히 저해되었으나, 0.25 mg/mL 이하의 농도에서는 생육억제효과가 감소하는 것으로 나타났다. Enterobacter cloacae는 0.5 mg/mL의 추출물 처리에 의해 생육이 2 log cycle정도 억제된 반면, Escherichia cole는 비교적 저항성이 강한 것으로 나타났다. 천궁의 ethyl acetate 추출물은 열에 매우 안정하여 $100^{\circ}C$ 30분 또는 $121^{\circ}C$에서 15분간의 열처리(autoclave)에 의해서 항균활성이 감소되지않는 것으로 나타났다. 따라서 약용으로 이용되고 있는 천궁의 근경 이외의 미활용자원인 잎과 줄기 등의 지상부위를 이용한 천연 항균소재 개발이 가능할 것으로 판단된다.
피부의 피부 노화, 질환, 훼손은 내부적인 요인으로서 활성산소라는 사실이 밝혀진 이래로 합성 항산화제를 대체할 수 있는 천연물 항산화제에 대하여 많은 연구가 진행 중이다. 이들 중에 지실은 오래전부터 만성적인 염증, 알리지 완화, 황산화 기능을 하는 것으로 알려져 왔다. 미생물을 이용한 발효는 조직에 결합 되어 있는 생리활성 물질의 유리로 생체이용율(bioavailability) 훨씬 높아지는 것으로 알려 졌다. 따라서 본 연구는 지실 열수 추출물을 기질로 GRAS 알려진 균주를 이용하여 발효를 수행하고 발효가 항산화 능력에 미치는 영향에 대하여 알아보고자하였다. 지실 열수 추출물은 항산화 기능을 가지고 있는 hesperidine, naringine, luteoilin, imperatorin등의 플라보노이드 및 furocoumarin등이 동정이 되었고 함유량은 hesperidine naringine, imperatorin, luteolin 순이었다. 열수 추출물을 기질로 하여 분리된 Leuconostocs sp. strain JYK로 발효를 수행한 결과 지실 열수 추출물을 사용한 경우에는 $71.2{\pm}4.58(mg/g)$의 총 페놀 함량이 있는 것으로 측정이 되었고 반면에 발효 추출물에는 $89.2{\pm}13.47(mg/g)$으로 약 25.3% 증가된 것으로 나타났다. 총 플라보노이드 화합물은 지실 열수 추출물에는 $25.1{\pm}4.12(mg/g)$의 총 플라보노이드 함량이 있는 것으로 측정이 되었고 반면에 발효 추출물에는 $31.0{\pm}4.06(mg/g)$으로 약 23.5% 증가된 것으로 나타났다. DPPH 소거능은 지실 열수 추출물에는 70.9% 반면에 발효 추출물은 86.2% 측정이 되었다. 총 항산화 능력은 18.7% 발효추출은 26.6%로 발효를 통하여 약 40% 증가하였다. 따라서 미생물 발효를 통하여 천연물에 포함이 되어있는 기능성 물질에 대한 생체 이용률을 높인다면 산화적 스트레스에 의한 피부 노화 완화 및 개선 작용으로 기능성 화장품 및 식품의 소재로 활용 가치를 높여 줄 것으로 기대를 한다.
Silica is known as a promising osteoconductive material, and polycaprolactone is a bioactive and degradable material. The purpose of this study was to monitor the differentiation of MC3T3-E1 cells cultured on the layer-built silica/poly caprolactone non-woven fabric produced by electrospinning. Non-woven fabric (silica, polycaprolactone, PSP, SPS) was made by electrospinning and they were inserted in the 48 well cell culture plate. MC3T3-E1 cells were prepared by subculture. Cells were seeded to each well $1{\times}10^5$ concentration per well. Dulbecco's modified eagle medium with 10% FBS and 1% antibiotic-antimycotic solution was used. Confocal laser scanning microscope was taken 4 hours after incubation (95% air. 5% $CO_2$, $37^{\circ}C$). Cell proliferation was monitored by spectrophotometer on 1, 7, 14 days, and the morphology of the growing cells was observed by field emission scanning electron microscope. To monitor the differentiation of osteoblasts on the materials, MC3T3-E1 cells were incubated in 48 well culture plate after seeding with the density of $1{\times}10^5$ concentration. Then ELISA kit & EIA kit were used on to assess osteocalcin and osteopontin expression respectively. The other conditions were the same as above. MC3T3-E1 cells were proliferated well on all of the materials. There were no statistical differences among them. The osteopontin expression of silica, PSP, SPS was significantly higher than other groups on day 3 (p/0,05), but after that time, there were no statistically signigicant differences. The osteocalcin expression was significantly higher in silica and PSP than other groups on day 14. These findings show that PSP was as good as silica on the effect of osteoblast differentiation. The PSP non-woven fabric may have the possibility as bone graft materials.
타임, 바질, 로즈마리 등 20종의 허브 식물에 대한 80% 에탄올 추출물의 항산화 활성을 분석하였다. 탐색 대상 허브 식물 중 타임 추출물의 총 폴리페놀 함량은 6.90±0.02 mg CAE/100 g로 높은 값을 보였으며, 총 플라보노이드 함량은 1.71±0.19 mg NE/100 g으로 가장 높았다. 또한 DPPH radical 소거능은 93.14±0.23%로 매우 높은 수준의 항산화 활성을 나타냈다. 타임으로부터 플라보노이드를 추출하기 위하여 용매로서 물, 에탄올, 메탄올, 헥산 중 추출수율이 높은 헥산을 추출용매로 선정하였다. 반응표면분석법을 이용하여 타임 추출물의 최적화 추출조건으로 추출온도는 35.9 ℃, 농도는 1.63%, 시간은 192분이었다. 예측된 조건으로 제조한 타임 n-헥산 추출물의 총 플라보노이드 함량은 예측 값의 96.3%로 반응표면분석법에 의한 추출조건 최적화의 신뢰성이 검증되었다. 이상의 최적화된 추출 조건은 플라보노이드의 생산을 위한 타임의 용매추출 조건으로서 활용할 수 있을 것으로 기대된다.
The purpose of this study was to evaluate histologically the effect of LiF-maleic acid added calcium aluminate(LM-CA) bone cement & CA-PMMA composite bone cement on the healing of calvarial defect in Sprague-Dawley rats. The critical size defects were surgically produced in the calvarial bone using the 8mm trephine bur. The rats were divided in three groups : In the control group, nothing was applied into the defect of each rat. LM-CA bone cement was implanted in the experimental group 1 and CA-PMMA composite bone cement was implanted in the experimental group 2. Rats were sacrificed at 2, 8 weeks after surgical procedure. The specimens were examined by histologic analysis, especially about the bone-cement interface and the response of surrounding tissue. The results are as follows; 1. In the control group, inflammatory infiltration was observed at 2 weeks. At 8 weeks, periosteum and duramater were continuously joined together in the defect area. But the center of defect area was filled up with the loose connective tissue. 2. In the experimental group 1, the bonding between implanted bone cement and the existing bone was seen, which more increased in 8 weeks than 2 weeks. Inflammatory infiltration and the dispersion of implanted bone cement particles were seen in both 2 weeks and 8 weeks. 3. In the experimental group 2, implanted bone itself had a dimensional stability and no bonding between implanted bone cement and the existing bone was seen in both 2 weeks and 8 weeks. Implanted bone cement was encapsulated by fibrous connective tissue. In addition, inflammatory infiltration was seen around implanted bone cement. On the basis of these results, when LM-CA bone cement or CA-PMMA composite bone cement was implanted in rat calvarial defect, LM-CA bone cement can be used as a bioactive bone graft material due to ability of bonding to the existing bone and CA-PMMA can be used as a graft material for augmentation of bone-volume due to dimensional stability.
천연미백소재를 개발하기 위하여 대산(마늘)을 포함한 총10종의 약용식물 혼합물을 발효하여 얻은 발효물로부터 tyrosinase inhibition 활성을 확인하고, 이로부터 신규한 화합물 1종 외 3종의 활성 성분을 분리하였다. 마늘복합발효물의 영양성분 13항목을 분석하여 기능성 식품 소재로의 활용가능성을 확인하였다. 발효원액의 tyrosinase 저해효과는 88.6%로 나타났고, 활성성분의 분리를 위하여 EtOAc, n-BuOH 및 H2O로 분배, 추출하였다. 이중 수득률이 가장 높은 EtOAc 분획물(47g)로부터 silica gel 및 ODS column chromatography를 반복하면서 TLC와 효소저해 효과로부터 확인한 활성분획을 순차적으로 분리하였다. 화합물의 화학구조를 NMR 및 MS 등의 스펙트럼으로 분석한 결과 phenylpropanoid류의 신규 화합물인 2,4,5-trihydroxy-benzenepropanoic acid (1) (1.9 mg) 및 화합물 1과 동량으로 혼재하는 2,3,5-trihydroxy-benzenepropanoic acid (2)를 확인하였다. 이외에 2,4-dihydroxy-hydrocinnamic acid (3) (3.3 mg) 및 (+)sesamin (4) (6.1 mg)을 분리·동정하였다. 이 화합물들이 향후 본 조성물의 지표성분 및 기능성 성분으로 활용된다면 기업에서의 원천기술 확보를 통한 제품개발과 산업화에 매우 유용할 것으로 사료된다.
Purpose: Various bone graft materials are being used for periodontal tissue regeneration. Th materials are being developed continuously for ideal clinical effects. Therefore, it is necessary to identify the clinical characteristics of each bone graft material through comparing the various bone graft materials statistically and in doing so, proposing a more efficient bone graft material. In this study, the following results were attained through comparing the clinical effects among the bone graft materials, using the statistical method based on the clinical studies published at the department of periodontology of Yonsei hospital. Materials and Method: 6 selected studies of department of Periodontology at Yonsei University Hospital were based on clinical study of bone grafting in intrabony defects. It was compared the clinical parameters among the 6 clinical studies, using the statistical META analysis. Result: When comparing the probing depth reduction, there was a relatively great amount of decease when using the xenograft, Anorganic Bovine Derived Hydroxapatite Bone Matrix/Cell Binding Peptide(ABM/P-15: PepGen $P-15^{(R)}$) and the autogenous bone and absorbable membrane, d, 1-alctide/glycolide copolymer(GC: $Biomesh^{(R)}$). The allogfrafts showed a relatively low decrease in the probing depth and clinical attachment change. It also showed a slight decrease in the bone probing depth. The allografts showed various results according to different bone graft materials. When comparing the ABM/P-15 and bovine bone $powder(BBP^{(R)})$, ABM/P-15 showed a relatively high clinical attachment level and the bovine bone powder showed a relatively high clinical attachment level. The probing depth change and gingival recession change showed a lower value than the mean value between the two bone graft materials. The synthetic bone showed a relatively high decrease in clinical attachment level and periodontal probing depth change. There was a relatively larger amount of gingival recession when using Bioactive Glass(BG) but a relatively low bone regeneration effect was seen. Conclusion: Good restorative results of the periodontal tissue can be attained by applying the various bone graft materials being used today after identifying the accurate clinical effects.
치과용 임플란트 재료로 주로 사용되는 지르코니아 및 티타늄 합금은 생체불활성 특징으로 인하여 골유착 및 골형성 능력이 떨어진다. 이러한 문제를 쉽고 간단하게 해결하기 위한 방법으로는 생체활성 물질을 표면에 코팅하여 생체 활성을 높이는 방법이 있다. 본 연구에서는 우수한 골결합 능력을 가진 실리케이트계 세라믹인 아커마나이트(Ca2MgSi2O7)를 고상반응법으로 합성하고, SBF 용액 내 침적실험을 통하여 합성 아커마나이트 분말의 생체활성을 분석하였다. 고상반응 출발원료로는 탄산칼슘(CaCO3), 탄산마그네슘(MgCO3), 이산화규소(SiO2) 분말을 사용하였다. 분말을 혼합 및 건조한 후, 가압 성형하여 디스크 형태로 만든 후, 고상반응 온도를 변화시키며 아커마나이트 상의 합성을 유도하였다. 합성된 아커마나이트 펠릿의 용해 및 생체활성 분석을 위하여 SBF 용액 내 침적 시키고, 침적시간에 따라 아커마나이트의 표면 용해 및 하이드록시아파타이트 석출을 분석하였다. 합성반응 온도가 높아질수록 아커마나이트 상이 뚜렷하게 나타난 반면에, SBF 용액 내 용해는 천천히 진행되었다. 합성된 아커마나이트 분말의 생체활성도는 대체적으로 우수하였으나, 그 중에서도 1100℃에서 고상반응 하여 합성한 분말에서 적절한 용해 및 하이드록시아파타이트 입자의 석출이 잘 일어나는 것으로 분석되었다.
본 연구에서는 복분자 열매의 성숙도에 따른 미백 활성의 차이를 확인하기 위해 B16F10 cell에서 melanin 생성량을 측정하고, 기능성분인 ellagic acid의 함량 변화를 확인하였다. 또한 ellagic acid의 함량에 따른 tyrosinase 저해 활성도 측정하였다. B16F10 cell에서 MRF 및 IRF 추출물은 200 ㎍/mL 농도까지 세포 독성을 나타내지 않았고, Melanin 생성량은MRF 추출물에 비해 IRF 추출물에서 더 좋은 억제 활성을 보여주었다. 또한 복분자 성숙도에 따른 ellagic acid의 함량 변화를 확인한 결과, MRF 추출물에 비해 IRF 추출물에서 ellagic acid의 함량이 높게 측정되었고, tyrosinase 저해 활성 평가에서도 MRF 추출물에 비해 IRF 추출물에서 더 좋은 억제 활성을 보였다. 결과를 종합해보면, MRF 추출물에 비해 IRF 추출물에서 ellagic acid의 함량이 더 높았으며, 이로 인해 tyrosinase의 저해 및 melanin 생성 억제 또한 IRF 추출물에서 더 좋은 것으로 판단된다. 따라서, 복분자 미성숙과는 천연 미백 소재로써 우수한 기능성 화장품 원료가 될 것이라 생각한다.

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
