Enterococcus faecium SA5의 기능적 특성과 인삼 ginsenoside Rb1의 전환 (Functional Characteristics of Enterococcus faecium SA5 and Its Potential in Conversion of Ginsenoside Rb1 in Ginseng)

  • 본 연구는 몽골 마유로부터 분리한 유산균 Enterococcus faecium SA5의 이화학 특성을 파악하고 유산균 E. faecium SA5의 ${\beta}$-glucosidase의 활성과 이를 통한 ginsenoside 전환을 확인하는 것을 목표로 진행되었다. E. faecium SA5는 내산성, 내담즙성을 나타내었으며 4종의 병원성 미생물(Salmonella typhimurium KCTC 3216, Listeria monocytogenes KCTC 3710, Bacillus cereus KCTC 1012, Staphylococcus aureus KCTC 1621)에 항균 활성을 가질 뿐만 아니라 항생물질 colistin, gentamycin, neomycin에 내성을 나타내었다. 또한, E. faecium SA5는 bile salt hydrolase 활성을 나타내어 혈액 내 콜레스테롤 수준 감소 효과가 있다고 사료되며 10% skim milk에서 배양하였을 때, pH가 감소하고 산도 및 생균수가 증가하는 것으로 보아 발효유 스타터로써의 활성을 갖는 것으로 판단되었다. 또한 E. faecium SA5의 ${\beta}$-glucosidase에 의해 ginsenoside $Rb_1$이 ginsenoside $Rg_3-s$$Rg_3-r$으로 전환되었음을 TLC 분석을 통해 확인하였다. 따라서 E. faecium SA5는 잠재적인 probiotics로 이를 이용하여 발효유 제조 및 ginsenoside 전환 관련 건강기능식품 개발에 응용할 수 있을 것으로 사료된다.

Screening of Bacteriocin-producing Enterococcus faecalis Strains for Antagonistic Activities against Clostridium perfringens

  • This study was conducted to isolate and characterize bacteriocin-producing bacteria against Clostridium perfringens (C. perfringens) from domestic animals to determine their usefulness as probiotics. Bacteriocin-producing bacteria were isolated from pig feces by the spot-on-lawn method. A total of 1,370 bacterial stains were isolated, and six were tentatively selected after identifying the inhibitory activity against the pathogenic indicator C. perfringens KCTC 3269 and KCTC 5100. The selected strains were identified as Enterococcus faecalis (E. faecalis) by 16s rRNA sequencing. Most of the isolated bacterial strains were resistant to 0.5% bile salts for 48 h and remained viable after 2 h at pH 3.0. Some E. faecalis also showed strong inhibitory activity against Listeria monocytogenes KCTC 3569, KCTC 3586 and KCTC 3710. In the present study, we finally selected E. faecalis AP 216 and AP 45 strain based on probiotic selection criteria such as antimicrobial activity against C. perfringens and tolerance to acid and bile salts. The bacteriocins of E. faecalis AP 216 and AP 45 strains were highly thermostable, showing anticlostridial activities even after incubation at $121^{\circ}C$ for 15 min. These bacteriocin-producing bacteria and/or bacteriocins could be used in feed manufacturing as probiotics as an alternative to antibiotics in the livestock industry.

돼지장에서 분리한 Lactobacillus salivarius의 생균제로서 특성 (Probiotic Properties of Lactobacillus salivarius Isolated from Piglet Intestines)

  • 돼지장에 존재하는 많은 균총들로부터 생균제를 분리하기 위하여 생균제로서 필수 조건인 내산성, 내담즙산, 유해균 억제능력들을 조사하였는데 그 결과 다른 균들에 비해 생균제로서 작용 능력이 뛰어난 한 균주인 PA10을 얻을 수 있었다. 생균제로서 중요한 기능인 설사 유발 대장균과 Salmonella를 억제하기 위하여 돼지의 장으로부터 내산, 내담즙의 특성을 가진 미생물틀을 분리 검색하였다. 이들 중, 위산과 담즙에 잘 견디며, 대장균 및 Salmonella에 대한 억제 능력이 뛰어난 한 미생물을 분리하였는데 Lactobacillus salivarious로서 동정되었다. 이미 생물은 pH 3의 위산과 0.3% 담즙산에서 각각 2시간, 24시간 동안 약 50~70%의 생존률을 나타냈으며, 대장균 또는 Salmonella를 MRS배지에서 혼합 배양시 24시간안에 이들 유해균을 완전히 제거할 수 있었다. 이 실험에서 L salivarius로 명명된 이 균은 돼지장에 서식하는 수 많은 미생물중 내산, 내담즙산, 유해균 억제능을 동시에 만족 시킬 수 있는 특성을 가지는 균주임을 알 수 있었다. 앞으로 균주 보존 조건을 확립하고, 동물 실험에서도 유사한 결과를 얻을 수 있다면 이 분리균은 생균제로서 유용하게 이용될 수 있을 것으로 생각된다.

Potential Evaluation and Health Fostering Intrinsic Traits of Novel Probiotic Strain Enterococcus durans F3 Isolated from the Gut of Fresh Water Fish Catla catla

  • Over the last few years, marine environment was found to be a source of surplus natural products and microorganisms with new bioactive secondary metabolites of interest which can divulge nutritional and biological impact on the host. This study aims to assess the possible, inherent and functional probiotic properties of a novel probiotic strain Enterococcus durans F3 (E. durans F3) isolated from the gut of fresh water fish Catla catla. Parameters for evaluating and describing the probiotics described in FAD/WHO guidelines were followed. E. durans F3 demonstrated affirmative results including simulated bile, acid and gastric juice tolerance with exhibited significant bactericidal effect against pathogens Staphylococcus aureus, Salmonella Typhi, Escherichia coli and Pseudomonas aeruginosa. This can be due to the enterocin produced by E. durans F3 strain, which was resolute by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) gel with amplification of the anticipated fragment of a structural gene; enterocin A, followed by antibiotic susceptibility assessment. Effective antioxidant potentiality against ${\alpha}$-diphenyl-${\alpha}$-picrylhydrazyl free radicals including lipase, bile salt hydrolase activity with auto-aggregation and cell surface hydrophobicity was similarly observed. Results are proving the potentiality of E. durans F3, which can also be used as probiotic starter culture in dairy industries for manufacturing new products that imparts health benefits to the host. Finding the potent and novel probiotic strains will also satisfy the current developing market demand for probiotics.

음식물류폐기물의 자원화를 위한 항세균 활성 유산균의 개발 (From food wastes into useful probiotics: Development of Lactic acid bacteria as useful probiotics for animal feed without antibiotics using food waste)

  • 음식물류폐기물의 무항생제 액상사료화를 통한 고급자원화를 이루기 위하여 항생제 대체용 항세균 활성 유산균주 13 종을 돼지 장관 및 분변에서 분리하였다. 분리 유산균은 모두 내염성(Nacl 4%), 내산성 (pH4.0) 및 내담즙성 (oxgall 0.8%)을 보였으며, $37^{\circ}C$에서 생장이 가장 높게 나타났다. 분리 유산균주중 Lactobacillus plantarum CJY-22, Pediococcus sp. CJY-41, Lactobacillus brevis CJY-42 및 Lactobacillus arizonensis CJY-3 균주가 가장 높은 항세균 활성 및 광역의 항세균 활성 스펙트럼을 나타냈으며, Lactobacillus plantarum CJY-22 균주의 경우 $25^{\circ}C$에서도 생장이 원활하여 음식물류폐기물에 적용시 가온없이 발효공정을 수행할 수 있는 유용 균주로 적용할 수 있을 것으로 사료된다.

Screening and Characterization of Lactic Acid Bacteria Strains with Anti-inflammatory Activities through in vitro and Caenorhabditis elegans Model Testing

  • The present study was conducted to screen candidate probiotic strains for anti-inflammatory activity. Initially, a nitric oxide (NO) assay was used to test selected candidate probiotic strains for anti-inflammatory activity in cultures of the murine macrophage cell line, RAW 264.7. Then, the in vitro probiotic properties of the strains, including bile tolerance, acid resistance, and growth in skim milk media, were investigated. We also performed an in vitro hydrophobicity test and an intestinal adhesion assay using Caenorhabditis elegans as a surrogate in vivo model. From our screening, we obtained 4 probiotic candidate lactic acid bacteria (LAB) strains based on their anti-inflammatory activity in lipopolysaccharide (LPS)-stimulated RAW 264.7 cell cultures and the results of the in vitro and in vivo probiotic property assessments. Molecular characterization using 16S rDNA sequencing analysis identified the 4 LAB strains as Lactobacillus plantarum. The selected L. plantarum strains (CAU1054, CAU1055, CAU1064, and CAU1106) were found to possess desirable in vitro and in vivo probiotic properties, and these strains are good candidates for further investigations in animal models and human clinical studies to elucidate the mechanisms underlying their anti-inflammatory activities.

Optimizing the fermentation condition of low salted squid jeotgal by lactic acid bacteria with enhanced antioxidant activity

  • Lactic acid bacteria (LAB) are widely used as starter culture in food fermentation due to their harmless entity and health beneficial properties along with the ability to change texture, aroma, flavor and acidity of food products. In this study, five different LAB (FB003, FB058, FB077, FB081, and FB111) isolated from different Korean traditional fermented foods, assigned to Lactobacillus plantarum, Pediococcus pentosaceus, Weissella viridescens, Lactobacillus sakei, and Leuconostoc mesenteroides, respectively, on the basis of their physiological properties and 16S rRNA sequence analysis, to use as fermentation starter and check their ability to fasten the ripening time as well as the overall optimization in the fermentation condition. To check their suitability as starters, their safety, acid and bile tolerance, NaCl and temperature resistance, susceptibility to common antibiotics, and antimicrobial activities were determined. Squid jeotgal samples were prepared by adding $10^8CFU/g$ of each strain in different samples, which were then kept for fermentation at $4^{\circ}C$ and checked for their antioxidant activities at 0, 7, 15, and 21-day intervals. The samples fermented with FB003 and FB077 displayed the highest antioxidant activity. This study revealed two effective starter cultures (FB003, FB077) for squid jeotgal fermentation, which presented increased functionalities. The results of this study will lead to the development of novel industrial-scale production avenues for jeotgal preparation, and offer new insights into the prevention and control of chronic diseases.

Identification of Potential Bacillus subtilis Probiotics from Korean Soybean Paste and Their Antimicrobial and Immune Activities

  • The potential probiotic of a total of 15 Bacillus species isolated from Korean soybean paste (doenjang) was evaluated. Among those tested, the CSY191 and CSY388 strains were selected as probiotic bacteria due to their acid and bile tolerance, respectively. These strains were classified as Bacillus subtilis based on morphological, physiological, and chemotaxonomic features as well as on phylogenetic analysis based on their 16S rDNA sequences. These strains CSY191 and CSY388 showed a significant survival with rate range of 30.0 to 58.3% and of 31.0% to 58.1%, respectively, under artificial gastric acidic conditions at pH 3.0. These CSY191 and CSY388 strains appeared to have high antimicrobial activity against Salmonella Typhimurium, Bacillus cereus and Listeria monocytogenes. Also, methanol extractions (surfactin-like compounds) of strain CSY191 and strain CSY388 activated RAW264.7 microphages and induced the production of nitric oxide (NO) in a concentration-dependent manner, respectively. Therefore, strain CSY191 and strain CSY388 can be used as potential probiotics.

한국 여성의 질에서 분리한 유산균의 Gardnerella vaginalis에 대한 항균효과 및 특성 규명 (Characterization and Antimicrobial Activity against Gardnerella vaginalis of Vaginal Lactobacillus spp. Isolated from Korean Women)

  • Bacterial vaginosis (BV) is caused by microbial imbalance of the vaginal ecosystem and overgrowth of anaerobic bacteria. The antibiotic treatment often results in very high recurrence of BV because it disturbs the vaginal ecosystem. The high recurrence rates suggest a need for alternative therapeutic methods and probiotics are being recognized as alternative or additional treatment method for BV. The purpose of this study was to investigate how human vaginal isolates of Lactobacillus spp. inhibit the BV-associated pathogen Gardnerella vaginalis. Results show that selected strains significantly reduced the viability of G. vaginalis. Among these selected strains KLB410 and KLB416 were further selected based on acid/bile tolerance and identified through 16S rRNA gene sequencing being Lactobacillus plantarum. Further studies are underway to demonstrate that the selected strain can be applied as potential probiotics for recovering vaginal ecosystem.

Complete genome sequence of Latilactobacillus curvatus CACC879 and its functional probiotic properties

  • Latilactobacillus curvatus CACC879 originated from swine feces in Korea, and its probiotic properties have been analyzed. The complete genome of strain CACC879 contained one chromosome 1,398,247 bp in length and three circular plasmids, namely, pCACC879-1 (591,981 bp), pCACC879-2 (14,542 base pairs [bp]), and pCACC879-3 (45,393 bp). The complete genome encodes a total of 2,077 genes, including 25 rRNA genes and 90 tRNA genes. In addition, probiotic stability- genes acid/bile related to salts tolerance, the biosynthesis of cobalamin (vitamin B12), riboflavin (vitamin B2), and CRISPR/Cas9 were found in the whole genomes. Remarkably, L. curvatus CACC879 contained the antioxidant-related (peroxiredoxin) and bacteriocin-related genes (lysM and blpA). Overall, these results demonstrate that L. curvatus CACC879 is a functional probiotic candidate for animal industry applications.
