The effects of the prevention of micelle formation and bile acid reabsorption, by using cholestyramine (CHOLN), a bile acid binding polymer, on the plasma lipid of Single Comb White Leghorn male chicks given diets containing medium chain triacylglycerol (MCT) and long chain triacylglycerol (LCT) were investigated. Corn oil and glyceryl tricaprylate were used as LCT and MCT sources, respectively. Plasma HDL cholesterol was reduced by CHOLN in all treatments. Plasma LDL cholesterol was reduced by CHOLN in chicks given LCT diet but not in MCT diet which could be accounted to the reduced plasma total cholesterol in LCT diet with CHOLN. It is concluded that bile acid binding does not alter the cholesteremic effect of MCT in the plasma of chicks.

In order to select probiotics with a high survival rate in gut and the growth inhibition of virulent pathogens to human beings or animals, we have examined a variety of microorganisms to assess the acid, bile, and pancreatic tolerance and the growth inhibition of E. colt O8 and Salmonella choleraesuis ATCC 8391. Lactobacillus acidophilus KCTC 3155 was shown to inhibit the growth of E. coli and Salmonella dramatically within 24 h of incubation, although it was vulnerable to the exposure of bile acids. Bacillus polyfermenticus showed a good growth inhibition against E. coli, with a moderate acid and bile tolerance, while Clostridium butyricum KCTC 1786 inhibited the growth of E. coli and Salmonella slightly with a good bile tolerance. However, Saccharomyces cerevisiae KCTC 7928 and Aspergillus oryzae KCTC 6075 did not inhibit the growth of E. coli and Salmonella, suggesting that these microorganisms can be used as the sources of nutritional suppliment rather than as probiotics itself.
We studied the effects of bile acids on the induction ofapoptosis in HepG2 human hepatocellular carcinoma cells. Treatment with either ursodeoxycholic acid (UDCA) or lithocholic acid (LCA) resulted in a dose- and time-dependent decrease in cell viability assessed by MTT assay. Both UDCA and LCA also induced genomic DNA fragmentation, a hallmark of apoptosis, indicating that the mechanism by which these bile acids induce cell death was through apoptosis. Cycloheximide, a protein synthesis inhibitor, blocked the apoptosis induced by these bile acids, implying that new protein synthesis may be required for the apoptosis. Intracellular $Ca^{2+}$ release blockers (dantrolene and 3,4,5-trimethoxybenzoic acid-8-(diethylamino)octyl ester) inhibited decreased cell viability and DNA fragmentation induced by these bile acids. Treatment of HepG2 cells with calcium ionophore A23l87 induced DNA fragmentation. These results suggest that UDCA and LCA induce apoptosis in the HepG2 cells and that the activation of intracellular $Ca^{2+}$ signals may play an important role in the apoptosis induced by these bile acids.
Survival and stability of 19 bifidobacterial strains included 13 isolates from Korean infants against acid, bile acid and oxygen exposure were examined. Acid resistance of selected strains at pH values of 4.0, 3.0 and 2.0 was tested. Among the bifidobacterial strains tested, B. bifidum B3, B. longum D6, and B. adolescentis F1 exhibited higher viable cell counts exposed to acid whereas other strains had various results. The abilities of the strains to grow in the MRS broth containing 0.2% thioglycolic acid and 0.2% oxgall were tested and the tolerance of B. bifidum B3 and B. longum D6 to bile acid were higher than that of others. Even though in same species, the tolerance of tested strains to bile acid were variable. Stabilities of tested strains to oxygen exposure were variable and B. bifidum and B. longum strains showed relatively higher viable cell counts after 48 hours exposure to aerobic incubation. These results demonstrated that the survival and stability of bifidobacterial strains to acid, bile acid, and oxygen exposure were variable and strain-dependent. Due to their tolerant ability to environmental factors like acid, bile acid, and oxygen, B. bifidum B3 and B. longum D6 had good potential properties as probiotic cultures and may be useful for industrial application.
Retarding effects of the dietary fibers from tangerine peels on glucose, bile acid and cadmium transport were evaluated by dialysis method, and were compared with those of commercial dietary fibers(citrus pection, CM-cellulose, guar gum, $\alpha$-cellulose). Yields of total (TDF), insoluble(IDF) and soluble dietary fibers(SDF) from tangerine peels on the fresh matter basis were 2.84%, 1.95% and 0.39% respectively. The amount of insoluble fibers was 5.2 times higher than that of soluble fibers. Soluble fibers(guar gum, CM-cellulose, SDF, pectin) had the retarding effect on glucose transport, while IDF, TDF and $\alpha$-cellulose did not have. Guar gum showed the greatest effect, followed by CM-cellulose, SDF and pectin. Among the extracted fibers, only SDF had the effect on glucose transport retardation. Regarding bile acid dialysis, guar gum had the greatest retarding effect, and all dietary fibers from tangerine peels, especially SDF, showed the effect of bile acid retardation. On cadmium transport retardation, CM-cellulose had the greatest effect, followed by SDF, TDF, IDF, guar gum and pectin. Among the extracted fibers, SDF had the greatest effect on Cd trasport transport retardation. The extracted dietary fibers showed higher retarding effect on Cd transport than glucose and bile acid transport, and the effect of SDF was higher than IDF.

The purpose of the study was to find an effect of Medium Chain Triglycerids (MCT) diet on cholesterol metabolism in rat. Sprague-Dawley rats were fed two different diets containing MCT(trioctanoate) and corn oil respectively. After feeding to each group for four weeks, the levels of serum and liver cholesterol, the excretion rates of fecal and biliary steroids, and also bile acid composition were investigated. The results obtained from the study are as follows : (1) The average body weight gain in MCT group was almost same as that in the corn oil group. (2) The concentration of serum cholesterol in MCT group was lower than that in the corn oil group. Therefore it is confirmed that the cholesterol lowing action of MCT diet was practically high. (3) The concentrations of liver cholesterol and Triglyceride in MCT group were almost same as that in the corn oil group. Therefore it is thought that the level of liver lipids was not influenced by the difference of diet in this study. (4) The excretion rate of fecal neutral steroid in MCT group was significantly lower than that in the corn oil group, while the rate of fecal bile acid excretion was about same in both MCT and corn oil group. (5) The composition rates of fecal bile acid such as cholic acid, chenodeoxycholic acid and deoxycholic acid, a secondary acid of cholic acid, in MCT group were significantly lower than that in the corn oil group. (6) The excretion rates of biliary cholesterol and bile acid in MCT group were significantly higher than that in the corn oil group, while the composition rates of biliary bile acid such as chenoddeoxycholic acid and deoxycholic acid in MCT group were significantly higher than that in the corn oil group.
In the present study, the vacuolar apparatus were investigated in the hepatocytes of rats treated with DA by transmission electron microscopy of conventional and cytochemical thin sections. In the rats after 20 min of dehydrocholic acid treatment, the cis Golgj cisterns were sacculated in line. The saccule occasionally occured by elongation and attenuated neck. The lysosomes also showed protrudent saccule. The vesicles were observed near the cis Golgi cisterns, lysosome and bile canaliculi. Some of the vesicles appeared to be fused to bile canaliculi. The cis Golgi cisterns usually faced toward the bile canaliculi both in normal and experimental groups. The cis Golgi cisterns, protrudent saccule and vesicles were almost devoid of visible contents. The osmium deposits were heavy on the protrudent saccule as well as on the cis Golgi cisterns or on the vesicles isolated near by, but they were light or not observed on the vesicles in the immediate vicinity of bile canaliculi. The acid phosphatase activities appeared on the lysosome and vesicles located near by, but did not appear on the vesicles as approaching closer to the bile canaliculi. The evidence suggests that the vesicles are derived from the cis Gogi cistern and lysosomes and fuse to bile canaliculi for exocytosis, and that the activity in the vesicles is diminished as approaching closer to the bile canaliculi.
Bile salt hydrolase (BSH, EC 3.5.1.24) activity, which cleaves amide bond between carboxyl group (bile acid) and amino group (glycine or taurine), is commonly detected in gut-associated species of human and animal. During the screening of BSH active strains from the fecal samples of elderly human volunteers, strain CU30-2 was isolated on the basis of the highly active BSH producing activity. A bsh gene of the isolate was cloned into the pET22b expression vector and overexpressed in Escherichia coli BL21 (DE3) Gold by induction with 1mM IPTG. The overexpressed BSH enzyme with 6x His-tag was purified with apparent homogeneity using a $Ni^+$-NTA agarose column and characterized. The BSH enzyme of E. faecalis CU30-2 exhibited approximately 50 times higher activity against glycol-conjugated bile salts than tauro-conjugated bile salts having the highest activity against glycocholic acid. Considering the prevalence of E. faecalis strains in the human GI tract and glycol-conjugates dominated bile acid composition of human bile, further study is needed to investigate the impact of the BSH activity exerted by E. faecalis strains to the host as well as to the BSH producing strains.
Methylene bis (3, 4, 6-trichlorophenoxy acetic acid) 'MTPA' has been developed for the purpose of treatment of clonorchiasis. It has been rpeorted that, in patients treated with MTPA, the flukes in the liver were killed, elevated serum bilirubin returned to normal and the patients´ general condition was improved. However it took $1{\sim}4 $ weeks to obtain sufficient therapeutic effects. In rabbits, excretion of bilirubin in the bile was increased by the MTPA, and this action was enhanced by a combination of deoxycholic acid with MTPA. This study was designed as a part of a series to increase the therapeutic effect of MTPA, by observing the relation of the blood level of MTPA with the excretion of MTPA in the bile, and the excretion of MTPA with bilirubin excretion in the bile caused by the injection of MTPA alone or in combination with deoxycholic acid. $^{14}C-labeled$ MTPA alone or with deoxycholic acid were injected into the ear veins of rabbits. The amout of bile, MTPA and bilirubin in the bile and the blood level of MTPA were measured. The results obtained were as follows: 1. The amount of excreted bile was decreased gradually as the time elapsed in both groups, that is groups injected with MTPA alone and with deoxycholic acid, without any significant difference between either group. 2. The largest amount of MTPA excretion in the early stage of the MTPA excretion in both groups, but deoxycholic acid caused an increase in blood level of MTPA whereas biliary excretion of MTPA decreased, especially in the early stage after drug injection. 3. The significant increment of bilirubin excretion began within an hour and it reached peak level in $2{\sim}2\frac{1}{2}$ hours after drug injection in both groups, but the amount of excreted bilirubin was larger in the combined group. The above results suggest that deoxycholic acid interferes with the biliary excretion of MTPA, and that there is no close relation between the increased excretion of MTPA and bilirubin excretion. But there is a close relation between blood level or tissue concentration of MTPA and bilirubin excretion. Concerning the influence of deoxycholic acid on the therapeutic effect of MTPA, deoxycholic acid would enhance the effect of MTPA, if the parasites take the drug from the blood, but diminish its effectiveness if they take the drug from the bile.
The purpose of this study was to examine the effect of resistant starch(RS) in hyperchlesterolemia and colon cancer. The subjects of this study was eight college women participating in the general starch diet(GSD) period for 5 days and resistant starch diet(RSD) period for 7 days. RSD contains 30g or the RS. On the last day of each program blood were collected. And for the last 3 days of each diet period, the amount of all the food consumed by the subjects and feces were collected. Food was measured to determine and compared the energy, protein and fat intakes. The amount of total cholesterol, HDL-cholesterol, LDL-cholesterol and volatile fatty acids in plasma and the amounts of bile acids in feces were measured by gas chromatography. The results obtained were as follows, Daily energy intake was higher in the RSD compared with the GSD, Protein and fat intakes were lower in the RSD compared with the compared with the GSD. Volatile fatty acid contents in plasma, the amounts of acetic acid, propionic acid and valeric acid were higher in the RSD compared with the GSD. The amounts of bile acids in feces, cholic acid, chenodeoxycholic acid and lithocholic acid were higher in the RSD compared with the GSD, But the amount of deoxycholic acid n the RSD period was significantly low. Secondary/primary ratios of bile acids was lower in the RSD compared with GSD, respectively. We speculate that , RS consumption decreases colonic mucosal proliferation as a result of the decreased formation of cytotoxic secondary bile acids.
