  • White birch bark is being called baekhwapi usually and is used urination, travail, removal of fever in Oriental medicine. Betulin had been known as main chemistry ingredient of white birch bark. So, in an experiment that see betulin and connection inorganic compounds separation and enforced to recognize antioxidative activity and anti HIV. Betulin's yield separated vast quantity for the first time in Korea by 6.9%, and 3,4-dihydrobenzoic acid in ethylacetate soluble as experiment result. Betulin, an Anti-Inflammatory Triterpenoid Compound, Regulates MUC5AC Mucin Gene Expression through NF-kB Signaling in Human Airway Epithelial Cells

  • Betulin is a triterpenoid natural product contained in several medicinal plants including Betulae Cortex. These medicinal plants have been used for controlling diverse inflammatory diseases in folk medicine and betulin showed anti-inflammatory, antioxidative, and anticancer activities. In this study, we tried to examine whether betulin exerts a regulative effect on the gene expression of MUC5AC mucin under the status simulating a pulmonary inflammation, in human airway epithelial cells. Confluent NCI-H292 cells were pretreated with betulin for 30 min and then stimulated with phorbol 12-myristate 13-acetate (PMA) for 24 h or the indicated periods. The MUC5AC mucin mRNA expression and mucin glycoprotein production were measured by reverse transcription - polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA), respectively. To elucidate the action mechanism of betulin, effect of betulin on PMA-induced nuclear factor kappa B (NF-kB) signaling pathway was also investigated by western blot analysis. The results were as follows: 1) Betulin significantly suppressed the production of MUC5AC mucin glycoprotein and down-regulated MUC5AC mRNA expression induced by PMA in NCI-H292 cells. 2) Betulin inhibited NF-κB activation stimulated by PMA. Suppression of inhibitory kappa B kinase (IKK) by betulin led to the inhibition of the phosphorylation and degradation of inhibitory kappa B alpha (IκBα), and the nuclear translocation of NF-κB p65. This, in turn, led to the down-regulation of MUC5AC glycoprotein production in NCI-H292 cells. These results suggest betulin inhibits the gene expression of mucin through regulation of NF-kB signaling pathway, in human airway epithelial cells.

Betulin suppressed interleukin-1β-induced gene expression, secretion and proteolytic activity of matrix metalloproteinase in cultured articular chondrocytes and production of matrix metalloproteinase in the knee joint of rat

  • We investigated whether betulin affects the gene expression, secretion and proteolytic activity of matrix metalloproteinase-3 (MMP-3) in primary cultured rabbit articular chondrocytes, as well as in vivo production of MMP-3 in the rat knee joint to evaluate the potential chondroprotective effect of betulin. Rabbit articular chondrocytes were cultured and reverse transcription-polymerase chain reaction (RT-PCR) was used to measure interleukin-$1{\beta}$ ($IL-1{\beta}$)-induced gene expression of MMP-3, MMP-1, MMP-13, a disintegrin and metalloproteinase with thrombospondin motifs-4 (ADAMTS-4), ADAMTS-5 and type II collagen. Effect of betulin on IL-$1{\beta}$-induced secretion and proteolytic activity of MMP-3 was investigated using western blot analysis and casein zymography, respectively. Effect of betulin on MMP-3 protein production was also examined in vivo. The results were as follows: (1) betulin inhibited the gene expression of MMP-3, MMP-1, MMP-13, ADAMTS-4, and ADAMTS-5, but increased the gene expression of type II collagen; (2) betulin inhibited the secretion and proteolytic activity of MMP-3; (3) betulin suppressed the production of MMP-3 protein in vivo. These results suggest that betulin can regulate the gene expression, secretion, and proteolytic activity of MMP-3, by directly acting on articular chondrocytes.

  • Betulin, a pentacyclic triterpenoid isolated from the bark of Betula papyrifera. Laboratory synthesized structural analogs were tested for antiviral activities against Epstein-Barr Virus (EBV) by immunofluorescent antiviral assay. Among the several analogs tested, betulin 3,28-diphosphate and uvaol exhibited significant antiviral activities against EBV. The $EC_{50}$ of betulin 3,28-diphosphate and uvaol was found to be $0.6\mu{M}$ and $0.7\mu{M}$ respectively.

  • Three lupane type triterpenoids were isolated from the methanol extract of Pyrus pyrifolia fruit peel through repeated silica gel column chromatography. Based on the spectroscopic methods, their structures were determined to be lupeol (1), betulin (2), and betulinic acid (3).

  • The simultaneous determination of platyphylloside, aceroside and betulin was established for the quality control of Betula platyphylla bark using a high performance liquid chromatography and diode-array UV/Vis detector (HPLC-DAD). Separation and quantification were successfully achieved with a INNO C18 column ($5{\mu}m$, 4.6 mm $I.D.{\times}150mm$) by gradient elution of a mixture of methanol and water at a flow rate of 1.0 ml/min. Validation of the developed method was performed by various factor such as linearity, specificity, precision, accuracy, system suitability and stability. This method was successfully applied to the determination of contents of platyphylloside, aceroside VIII and betulin in three batches of Betula platyphylla bark extract. These results suggest that the developed HPLC method is simple, effective and could be utilized as a quality control method for Betula platyphylla bark products.

  • Six compounds were isolated from the twigs of Ilex macropoda. Their structures were elucidated as betulinic acid, lupeol, betulone, betulin, erythrodiol and 11-oxo-erythrodiol by physicochemical and spectroscopic analysis. Among them, lupeol, betulone, erythrodiol and 11-oxo-erythrodiol were isolated for the first time from this plant.

천연물로부터의 항노화소재 개발: Triterpenoid계 식물성분인 Oleanolic acid의 항노화 효과

  • 전반적인 피부세포의 생리적 지능 자하로 인한 자연노화와 더불어, 여러 피부 스트레스 요인들이 다양하게 작용하여 발생하는 외인성노화를 방지하거나, 개선할 수 있는 항노화소재로서의 개발 가능성을 알아보기 위해, 예로부터 항염활성이 있다고 알려진 oleanolic acid(OA)를 포함한 ursolic acid(UA), betulin, betulinic acid(BA) 등의 triterpenoids가 어떻게 피부에서의 항노화 활성을 나타내는지를 알아보았다. 시험 결과, OA는 자외선에 의한 각질형성세포에서의 PGE$_2$ 생성과 섬유아세포(NHF)에 의한 matrix metalloproteinase-1(MMP-1) 분비를 억제하였다. 그리고, NHF의 procollagen 생성을 촉진하였으며, 이런 procollagen 생성촉진활성이 in vivo에서도 발현되는 것을 무모생쥐의 실험을 통해서 확인하였다. 또한 OA는 각질세포의 증식과 분화를 촉진하여 표피세포로 하여금 세라마이므와 필라그린 생성을 증가시키도록 하는 작용도 있음을 보여주었다. 더불어 실험한 UA, betulin, BA 들은 비록, betulin, BA의 경우 세포 독성이 다른 물질 들에 비해 높았고, UA가 각질세포의 분화를 오히려 억제하는 양상을 보이기는 했지만, 대부분의 기능은 OA와 유사하였다. 피부세포보호작용과 진피 기질물질에 대한 작용, 그리고, 표피의 장벽기능과 보습기능에 대해 시험한 본 연구는, 식물성분인 triterpenoids가 피부를 위한 항노화소재로서의 개발 가능성이 있음을 확인하는 계기가 되었고, 그 중에서도 OA가 보다 우수한 항노화 소재가 될 수 있음을 시사하고 있다.

Acyl-CoA: Cholesterol Acyltransferase Inhibitors from llex macropoda

  • Twigs from llex macropoda were extracted with MeOH, and the concentrated extracts were partitioned with $CH_2Cl_2$, EtOAc, n-BuOH, and $H_2O$. Repeated column chromatography of the $CH_2Cl_2$ fraction ultimately resulted in the isolation of two compounds, via activity-guided fractionation, using ACAT inhibitory activity measurements. According to the physico-chemical data, the chemical structures of these isolated compounds were identified as lupeol (1) and betulin (2). Compounds 1 and 2 were shown to inhibit the activity of hACAT-1 and hACAT-2 in a dose-dependent manner, and compounds 1 and 2 inhibited hACAT-1 with $IC_{50}$ values of 48 and $83{\mu}M$, respectively.
