Optimization of Extraction of Marker Compounds from Red Ginsengs by Accelerated Solvent Extraction Using Response Surface Methodology (반응표면분석법을 사용한 가속 용매 추출에 의한 홍삼 지표성분의 추출 최적화)

  • A new method based on accelerated solvent extraction (ASE) combined with response surface methodology (RSM) has been developed for optimization of the extraction of ginsenoside [Rb1, Rg1, and Rg3(20S)], total phenolics, and benzopyrene in red ginseng. The RSM method, based on a five level and two variable central composite design, was employed to obtain the optimal combination of extraction conditions. In brief, ginsenosides Rb1, Rg1, and Rg3(20S) and total phenolics with undetectable benzopyrene were optimally extracted with 50% ethanol as an extraction solvent, extraction temperature of $158^{\circ}C$, extraction time of 20 min, extraction pressure of 2,500 psi, flush volume of 60%, and one extraction cycle. The contents of ginsenosides and total phenolics in red ginseng extracted by ASE under optimum conditions were significantly higher than those extracted by sonication and reflux extraction.

Atmospheric Concentration and Size Distribution of Airborne Particulates, Benzopyrene and Heavy Metals (대기(大氣) 중 Benzopyrene 및 중금속(重金屬)의 농도(濃度)와 입경분포(粒徑分布))

  • Total suspended particulate (TSP) in the atmosphere was collected and size-fractionated by Andersen high volume air sampler for the past two years (Mar. 1987-Feb. 1989) in Seoul. The concentrations of several polycyclic aromatic hydrocarbons and heavy metals were determined to investigate the atmospheric concentrations, seasonal variations and its relationship with the size distribution of suspended particulate matter. The arithmetic mean concentration of total suspended particulate was $229.48\;{\mu}g/m^3$. The concentrations of heavy metals were $2971.94\;ng/m^3$ for Fe, $767.75\;ng/m^3$ for Zn, $765.80\;ng/m^3$ for Pb, $218.40\;ng/m^3$ for Cu, $129.91\;ng/m^3$ for Mn, respectively. And the concentration of PAHs were $3.23\;ng/m^3$ for benzo(a)pyrene, $2.71\;ng/m^3$ for benzo(k)fluoranthene, $4.53\;ng/m^3$ for benzo(ghi)perylene, respectively. The mass-size distribution of TSP was lowest in the particle size range $1.1-3.3\;{\mu}m$ increased as the particle size increased or decreased. But PAHs, Pb and Zn abounded in particles below $2.0\;{\mu}m$, while Fe and Mn abouned in particles above $2.0\;{\mu}m$. TSP and its chemical compositions showed the seasonal variations. The concentrations of anthrophogenic origin like TSP, PAH and heavy metals in the fine particles were highest in winter and lowest in summer. PAH and Ph analyzed showed significant correlations with each other and with TSP concentration in fine particles, indicating that the particles in which they are contained have a similar behavior in the atmosphere.

  • An analysis of changes in herbal tea composition according to the difference in processing conditions showed slightly reduced crude protein content and increased, moisture, crude fat and solid elution rate after treatment using the ash puffing process compared to roasting. Benzopyrene content was significantly reduced to 0.18 ppb from 0.51 ppb. This result indicated that, the $B({\alpha})P$ content differed depending on the processing condition and raw materials. Generation of food $B({\alpha})P$ is mainly include the thermal decomposition of food cooking, when the processing which is a main component of food carbohydrate, protein, fat reason despite severe heat treatment as a whole is to be detected even though the $B({\alpha})P$ in this way is considered to be. The taste, aroma and color did not show a large difference, but the strong bitters taste decreased.

  • We investigated effects of several chemical carcinogens, i.e., $benzo({\alpha})pyrene$ (BP),7,12-dimethylbenz(a)anthracene (DMBA), nitrosomethyl urea (NMU), and nicotine on the replication, cytolyticity, DNA synthesis, and protein synthesis of type 1 herpes simplex virus (HSV-1) in viral infected Vero cell monolayers. We observed that the BP and DMBA did not show such activity. All chemical carcinogens did not inhibit the synthesis of viral DNA, but the expression of gamma viral proteins that are expressed from the newly synthesized progeny viral DNA was somewhat notably inhibited by BP and DMBA. However, the synthesis of alpha and beta viral proteins was not altered by the chemical carcinogens. These data indicate that the gamma viral proteins expressed from the newly synthesized DNA in the presence of chemical carcinogens in the culture medium may be defective. This is further supported by the fact that the virus fail to replicate in the presence of these chemical carcinogens, in spite of viral DNA and proteins are somewhat normally synthesized.

  • The following study is the result of herbal teas puffed at different temperatures between $140{\sim}220^{\circ}C$. There was change of single breadth that some carbonization occurs according to rise of processing temperature and crude ash content rises relatively, and crude protein and crude fat content had hardly changed and moisture content decreased. The solid elution rate of the herbal teas appeared by 0.18~0.27% (w/w), it increased as puffing temperature rises. The reason for the increase in solid elution rates is due to the breakage of cross bridges between the raw materials in the herbal tea which are carbohydrates, proteins, lipids and etc. after treatments of physical changes rather than chemical ones. Benzopyrene content happened difference in B(${\alpha}$)P content according to processing temperature, raw material by 0.18~0.24 ppbs.

Development of Hazardous Materials Management Standard for Decoction Type of Personalized Herbal Medicine

  • Objectives: This study was conducted to development of hazardous materials management standards for the decoction type of personalized herbal medicines (PHMs). Methods: This study was conducted in two stages. We searched documents about criteria to use words such as 'Herb', 'Herbal medicine', and 'Botanical Drug' and summarized the results. We organized the committee consisted of seven experts, and held two meetings to reach an agreement on hazardous management standards of the decoction type of PHMs. Results: The seven documents were presented in the literature review and six items related to hazardous management standards of decoction were identified. The second expert meeting brought that a total of six items, including heavy metal, pesticide residues, sulfur dioxide, benzopyrene, mycotoxin, and micro-organism limits, were selected for safety management of decoction type of PHMs. Also, the criteria and test methods for each standard were suggested for monitoring the decoction type of PHMs. Conclusion: The study suggested hazardous material management standards and criteria for the decoction types of PHMs. In the future, it would be necessary to conduct a pilot test to ensure the validity and credibility of the safety management standard and criteria. Furthermore, the government level safety management system should be introduced to verify the safety of decoction medicines.

  • This survey was conducted to monitor the safety of red ginseng products in Gwangju, in 2010. Among 100 samples, 37 were beverages, 5 were functional health foods on the market, and 58 were beverages from a tailor-made shop. All samples were negative for coliform bacteria. Aerobic plate counts were detected from 13 samples in the 58 tailor-made products but not detected in the other types of products. Benzoic acid was detected in 5 samples (range, 19.2-543.0 mg/kg). Among heavy metals, lead was detected, ranging from 28.8-62.3 ${\mu}g/kg$, cadmium, from 1.15-4.18 ${\mu}g/kg$, and mercury from 0.10-0.18 ${\mu}g/kg$. Benzopyrene was not detected in any samples. Ginsenoside Rg1 and Rb1 were detected in 0.1-23.4 mg/90 mL of beverages and 12.1-66.8 mg/90 mL of functional health foods. These results indicate that red ginseng products are safe in terms of microbial contaminants and hazardous chemical compounds such as heavy metals and benzopyrene.

  • DNA damage accumulates in cells as a result of exposure to exogenous agents such as benzopyrene, cigarette smoke, ultraviolet light, X-ray, and endogenous chemicals including reactive oxygen species produced from normal metabolic byproducts. DNA damage can also occur during aberrant DNA processing reactions such as DNA replication, recombination, and repair. The major of DNA damage affects the primary structure of the double helix; that is, the bases are chemically modified. These modification can disrupt the molecules'regular helical structure by introducing non-native chemical bonds or bulky adducts that do not fit in the standard double helix. DNA repair genes and proteins scan the global genome to detect and remove DNA damage and damage to single nucleotides. Direct reversal of DNA damage, base excision repair, double strand break. DNA repair are known relevant DNA repair mechanisms. Four different mechanisms are distinguished within excision repair: direct reversal, base excision repair, nucleotide excision repair, and mismatch repair. Genetic variation in DNA repair genes can modulate DNA repair capacity and alter cancer risk. The instability of a cell to properly regulate its proliferation in the presence of DNA damage increase risk of gene mutation and carcinogenesis. This article aimed to review mechanism of excision repair and to understand the relationship between genetic variation of excision repair genes and head and neck cancer.

The Changes of Benzo[${\alpha}$]pyrene Content in Herbal Tea Containing Schizandra chinensis, Astragalus membranaceus, Platycodon grandiflorum and Liriope platyphylla Affected by Roasting Temperature (Roasting 온도에 따른 오미자, 황기, 길경 및 맥문동을 첨가한 한방차의 벤조피렌 함량 변화)

  • The following is the study result of herbal tea roasted at different temperatures between $80{\sim}140^{\circ}C$. Depending on treatment temperature the water content decreased, some carbonization occurred and crude ash content relatively increased. Also crude protein and crude fat changed little. Benzopyrene content (0.17~0.35ppb) showed a tendency to increase with higher treating temperature. From this result, the $B({\alpha})P$ content differed depending on the treatment temperature and raw materials. In case of roasting, the actual inside temperature is around $200^{\circ}C$ but since the surface temperature of the roaster reaches around $2,000^{\circ}C$ some portion of $B({\alpha})P$ content was presumed to be produced from the area that came in contact with this surface. Solid elution rate of herbal tea showed 0.18~0.35%(w/w) and the rate of solid elution decreased with higher roasting temperature. There was no big change in $80{\sim}110^{\circ}C$ treatment section but the solid elution decreased rapidly in $110{\sim}140^{\circ}C$ section. The reason for decreasing solid elution rate at higher treatment temperature is because the compact inner tissue makes elution difficult.
