The objective of this study was to investigate the relationship between body weight, body condition score (BCS), blood urea nitrogen (BUN), glucose, cholesterol and number of transferable embryos for the purpose of improving reproductive performance in Hanwoo donors. Seventy five cows, at random stages of the estrous cycle, received a CIDR together with injection of 1mg estradiol benzoate and 50 mg progesterone, and gonadotropin treatment begann. Four days later, the animals were superovulated with a total of 28AU FSH (Antorin, 2AU = 1 ml) administered twice daily in constant doses over 4 days. On the 3rd administration of FSH, CIDR was withdrawn and 25 mg $PGF_2{\alpha}$ was administered. Cows were artificially inseminated twice after estrous detection at 12 hr intervals. The cows received $100{\mu}g$ GnRH at the time of 1st insemination. Embryos were recovered 7 days after the 1st insemination. In conclusion, cows with body weight < 400, 400~450 and > 450kg had number of transferable embryos of $4.2{\pm}1.7$, $6.1{\pm}2.7$ and $4.8{\pm}2.6$, cows with BCS <2.25, 2.25~2.75 and ${\geq}2.75$ had number of transferable embryos of $4.6{\pm}1.6$, $5.7{\pm}2.4$ and $5.1{\pm}2.7$ respectively. These data indicate that a body weight and BCS for superovulation of CIDR-treated Korean native cows does not affect the embryo yield.
An important property of the intestine is the ability to secrete fluid. The intestinal secretion is regulated by a number of substances including vasoactive intestinal peptide (VIP), ATP and different inflammatory mediators. One of the most important secretagogues is adenosine during inflammation. However, the controversy concerning the underlying mechanism of adenosine-stimulated $Cl^-$ secretion in intestinal epithelial cells still continues. To investigate the effect of adenosine on $Cl^-$ secretion and its underlying mechanism in the rabbit colon mucosa, we measured short circuit current ($I_{SC}$) under automatic voltage clamp with DVC-1000 in a modified Ussing chamber. Adenosine, when added to the basolateral side of the muocsa, increased $I_{SC}$ in a dose-dependent manner. The adenosine-stimulated $I_{SC}$ response was abolished when $Cl^-$ in the bath solution was replaced completely with gluconate. In addition, the $I_{SC}$ response was inhibited by a basolateral Na-K-Cl cotransporter blocker, bumetanide, and by apical $Cl^-$ channel blockers, dephenylamine-2-carboxylate (DPC), 5-nitro-2-(3-phenyl-propylamino)-benzoate (NPPB), glibenclamide. Amiloride, an epithelial $Na^+$ channel blocker, and 4,4-diisothiocyanato-stilbene-2,2-disulphonate (DIDS), a $Ca^{2+}-activated$$Cl^-$ channel blocker, had no effect. In the mucosa pre-stimulated with forskolin, adenosine did not show any additive effect, whereas carbachol resulted in a synergistic potentiation of the $I_{SC}$ response. The adenosine response was inhibited by 10 ${\mu}M$ H-89, an inhibitor of protein kinase A. These results suggest that the adenosine-stimulated $I_{SC}$ response is mediated by basolateral to apical $Cl^-$ secretion through a cAMP-dependent $Cl^-$ channel. The rank order of potencies of adenosine receptor agonists was $5'-(N-ethylcarboxamino)adenosine(NECA)>N^6-(R-phenylisopropyl)adenosine(R-$ PIA)>2-[p-(2-carbonylethyl)-phenyl-ethylamino]-5'-N-ethylcarboxaminoadenosine(CGS21680). From the above results, it can be concluded that adenosine interacts with the $A_{2b}$ adenosine receptor in the rabbit colon mucosa and a cAMP-dependent signalling mechanism underlies the stimulation of $Cl^-$ secretion.
The experiment was crried out to evaluated the herbicidal properties of bensulfuron methyl [methyl 2-[[[[[(4, 6-dimethoxy pyrimidine-2yl) amino] carbonyl] amino] sulfonyl] methyl] benzoate]. No phytotoxicity was observed when bensulfuron methyl was applied at 3 and 6 g a.i./ 10a while the application rate 12 g a.i./10a slightly retared the growth of rice. The phytotoxicity decreased as the application time was delayed. The effect of application rate, leaching grade, transplanting depth, soil type and temperature on crop injury was little. Japonica variety (Dong-Jin) was more sensitive to bensulfuron methyl than indica X japonica variety (Sam-Kang). Bensulfuron methyl controlled effectively perennial weeds such as Sagittaria pygmaea Miq., Potamogeton diatinctus A. Benn., Cyperus serotinus Rottb., Sagittaria trifolia L., Eleocharis kuroguwai Ohwi. including most annual weeds except Echinochloa crus galli P. Beauv. The effect slightly decreased with lowering the temperature increasing the leaching grade. Application time and soil type employed did not affect the weeding effect.
Physiological changes in Eleocharis kuroguwai Ohwi as affected by bensulfuron-methyl {Methyl 2-[[[[[(4, 6-dimethoxy-2-pyrimidinyl)amino]carbonyl]amino] sulfonyl]methyl]benzoate} was determined to relate the characteristics with regrowth behavior. There were no changes in relative growth rate(RGR) during the period of growth cessation after application of bensulfuron-methyl. RGR's of the growth ceseased plants caused at 39 and 51 g/ha began to increase in between 25 and 30 days after application (DAA) and between 30 and 35 DAA, respectively. In untreated plant tuber carbohydrate rapidly decreased right after emergence and almost consumed within 40 days. There was no carbohydrate consumption during the period of growth cessation in bensulfuron-methyl-applied plant, but the content started to rapidly decrease with regrowth. Tuber viability lasted for 30 days in untreated plant, while tubers were viable for 60 and 70 days after application of bensulfuron-methyl at 39 and 51 g/ha, respectively. During the period of growth cessation the plants kept minimum respiration and photosynthesis, but with regrowth respiration and photosynthesis were resumed and rapidly increased.
Development of 3,700 folds resistance to etoxazole was found in the population of twospotted spider mite, Tetranychus urticae, collected from rose greenhouses in Buyo, Chungnam Provience in August 2000. This population was selected for 3yr with etoxazole to get 5,000,000 folds increase in resistance as compared to susceptible (S) strain. The etoxazole resistance was stablized for 16 months under the condition of no acaricide application. Inheritance and cross resistance in etoxazole to some acaricides of the etoxazole resistance strain (R) were investigated. There were differences of susceptibility in the etoxazole concentration-mortality relationships between $F_1$, $F_2$ progenies obtained from reciprocal cross with the S and R strains (R$_{♂}$${\times}$S$_{♂}$${\times}$R$_{♂}$). Degrees of dominance were 0.98 and 0.98 in $F_1$ and $F_2$ progenies of R$_{♂}$${\times}$S$_{♂}$, and -0.97 and -0.68 in $F_1$ and $F_2$ progenies of S$_{♀}$${\times}$R$_{♂}$ respectively. Inheritance in $F_1$ and $F_2$ progenies of R$_{♀}$${\times}$S$_{♂}$ were complete dominant. However $F_1$ and $F_2$ progenies of S$_{♀}$${\times}$R$_{♂}$ were incomplete recessive. These results suggest that inheritance of etoxazole resistance is controlled by a complete dominance. The R strain exhibited cross resistance to acequinocyl and emamectin benzoates in adult females, and milbemectin, amitraz and pyridaben in eggs. However they showed negatively correlated cross-resistance to bifenazate, a carbazate acaricide. These results may indicate bifenazate could be useful for the control of etoxazole resistant T. urticae population.

Acetolactate synthase (ALS) catalyzes the first common reaction in the biosynthesis of branched-chain amino acids, valine, leucine, and isoleucine. ALS is the common target of several classes of structurally diverse herbicides, the triazolopyrimidines, the imidazolinones, the sulfonylureas, and pyrimidyl-oxy-benzoates. We examined ihibitory activities of newly synthesized triazolopyrimidine sulfonamide derivatives using partially purified ALS from barley. $IC_{50}$ values for the active derivatives are 0.5nM∼8$\mu$M. Among them TP1 and TP2 are the most potent ALS inhibitors with $IC_{50}$ values of 0.5nM and 1.6nM, respectively. These inhibitors are more potent in the inhibition of barley ALS than commercial herbicides, Metosulam ($IC_{50}$;3.6 nM), Flumetsulam ($IC_{50}$;126 nM), and Cadre ($IC_{50};4 {\mu}M$). The progress curves for inhibition of ALS by TP2 showed that the amount of inhibition increases with time. The inhibition of ALS by TP2 was mixed-type inhibition with respect to pyruvate. Dual inhibition analyses of TP2 versus an imidazolinone, Cadre, and Leu showed parallel and intercepting kinetic pattern, respectively. The results suggest that TP2 binds to ALS competively with Cadre but not with Leu. Chemical modification of cysteinly residues in ALS decreased the sensitivity of ALS to Leu, while the modification did not affect the sensitivity of ALS to TP2 and Cadre.
The purpose of this study was to determine the effect of different feeding ratios of whole crop barley silage on the embryo production in Hanwoo donors. All donors were basically fed 2.5 kg concentrate daily. Donors were divided into three groups according to the different feeding of forage; hay 70% and rice straw 30% (control, n = 21), whole crop barley silage 80% and rice straw 20% (T1, n = 25), and whole crop barley silage 60% and rice straw 40% (T2, n = 23) fed based on TDN 6.70/ BW 500 kg. All Hanwoo donors received a CIDR together with injections of 1 mg estradiol benzoate and 50 mg progesterone ($P_4$, Day 0). Four days later, they were superovulated with 28 mg FSH twice daily IM in decreasing doses over 4 days. Then donors received 2 doses of $PGF_2{\alpha}$ (25 and 15 mg) with the 5th and 6th injections of FSH on Day 6. CIDR were withdrawn at the 6th FSH injection and the donors received $100\;{\mu}g$ GnRH 36 h after the second $PGF_2{\alpha}$ injection. The donors were artificially inseminated twice, at 8 and 24 h after GnRH, and embryos were recovered 7 or 8 days after the 1st insemination. The flush rate of the donors following positive superovulation responses did not differ among groups (76.2~96.0%, p>0.05). The number of corpus luteum (CL) at embryo recovery also did not differ among groups (10.6~14.0, p>0.05). Furthermore, the mean numbers of total ova (9.4, 10.5 and 12.0) and transferable embryos (5.3, 12.0 and 6.5) did not significantly differ among the control, T1 and T2 groups, respectively (p>0.05). However, mean concentrations of serum $P_4$ of the T1 (64.2 ng/ml) and T2 groups (55.7 ng/ml) were higher than that of control group (43.3 ng/ml, p<0.01), while serum cholesterol concentrations in the control (105.8 mg/dl) and T2 groups ($96.9\;{\pm}\;mg/dl$) were significantly lower than in the T1 group (121.1 mg/dl, p<0.05). Conclusively, whole crop barley silage can be fed a good substitute for hay forage for Hanwoo donors. Furthermore the ratios of whole crop barley silage 60% and rice straw 40% might be more worthful for embryo production.
Bioreporter bacteria, such as recombinant bioluminescent bacteria, have been used for the detection of specific compounds in complex environmental media. In this study, optimum conditions for the preparation and application of deep-freezed and Iyophilized recombinant bioluminescent strain KG1206 were investigated for the future application on contaminated environmental sites. Genetically engineered microorganism, Pseudomonas putida mt-2 KG1206, contains TOL plasmid and the plasmid inserted $P_{m}$, promoter on the upper part of lux gone in vector pUCD615, and m-toluate and benzoate are considered direct inducers for bioluminescence. Optimum conditions determined for the preparation and application of the deep-freezed and lyophilized strain were followings: cryoprotective agent (24% sucrose), lyophilization time (12 hrs), strain concentration ($OD_{600}=0.6$), reconstitution for freezed strain (quick reconstitution at $35^{\circ}C$), reconstitution for lyophilized strain ($3{\sim}6$ hrs exposure on LB medium), carrying conditions (keep at $20^{\circ}C$ after reconstitution). These results demonstrate the feasibility of deep-freezed or lyophilized state of genetically engineered bioluminescent strain for environmental usage.
This study was undertaken to elucidate the behavior of herbicide bensulfuron-methyl[methyl-2-[[[[[(4,6-dimethoxy pyrimidine-2yl)amino]carbonyl]amino]sulfonyl]methyl]benzoate]in soils. Adsorption of the herbicide in soils was mainly correlated with content of organic matter and clay, and canon exchange capacity. Adsorption distribution coefficient(Kd value) in clay loam soil was greater than those in loam and sandy loam soils. The Kd value decreased in the order of zeolite, bentonite, halloysite and laziolite clay minerals. Bensulfuron-methyl moved to 3cm deep in clay loam soil and 4cm deep in sandy loam and herbicide treated layer was 0 to 2cm profile in the two soils. The decomposition rate of bensulfuron methyl varied with the soil properties. The rate was slower in sterilized soil than in nonsterilized. Addition of organic matters to the soils accelerated the decomposotion. The degradation was more rapid in 30$^{\circ}C$ soil temperature than in 20$^{\circ}C$.
Difference in sensitivity of Eleocharis kuroguwai tubers to bensulfuron- methyl{methyl 2-[[[[[(4, 6-dimethoxy-2-pyrimidinyl) amino] carbonyl] amino] sulfonyl] methyl] benzoate} at different burial depths were determined with respect to tuber emergence, sprouting of lateral buds, carbohydrate consumption of the tuber, and growth and new tuber production of the regrown plants. Days required to regrowth from the growth cessation due to bensulfuron-methyl were shorter in shallow-buried tubers than in deep-buried tubers. With application of bensulfuron-methyl shallow-buried tubers consumed less carbohydrate in the tuber than deep-buried tubers as compared with in deep-buried tubers during the period of growth cessation and greater regrowth also occurred in the former. Fast and great regrowth in shallow-buried tubers resulted in great production of dry matter and new tubers. However, the differences obtained were not due to bensulfuron-methyl, but due mainly to ecological emergence and growth characteristics of tubers buried at different depths.
