Optimal Temperature and Light Intensity for Improved Mixotrophic Metabolism of Chlorella sorokiniana Treating Livestock Wastewater

  • Mixotrophic microalgal growth gives a great premise for wastewater treatment based on photoautotrophic nutrient utilization and heterotrophic organic removal while producing renewable biomass. There remains a need for a control strategy to enrich them in a photobioreactor. This study performed a series of batch experiments using a mixotroph, Chlorella sorokiniana, to characterize optimal guidelines of mixotrophic growth based on a statistical design of the experiment. Using a central composite design, this study evaluated how temperature and light irradiance are associated with $CO_2$ capture and organic carbon respiration through biomass production and ammonia removal kinetics. By conducting regressions on the experimental data, response surfaces were created to suggest proper ranges of temperature and light irradiance that mixotrophs can beneficially use as two types of energy sources. The results identified that efficient mixotrophic metabolism of Chlorella sorokiniana for organics and inorganics occurs at the temperature of $30-40^{\circ}C$ and diurnal light condition of $150-200{\mu}mol\;E{\cdot}m^{-2}{\cdot}s^{-1}$. The optimal specific growth rate and ammonia removal rate were recorded as 0.51/d and 0.56/h on average, respectively, and the confirmation test verified that the organic removal rate was $105mg\;COD{\cdot}l^{-1}{\cdot}d^{-1}$. These results support the development of a viable option for sustainable treatment and effluent quality management of problematic livestock wastewater.

Characterization of Bottom Ash as an Adsorbent of Lead from Aqueous Solutions

  • This study investigated the potential of using bottom ash to be used as an adsorbent for the removal of lead (Pb) from aqueous solutions. The physical and chemical characteristics of bottom ash were determined, with a series of leaching and adsorption experiments performed to evaluate the suitability of bottom ash as an adsorbent material. Trace elements were present, such as silicon and aluminum, indicating that the material had a good adsorption capacity. All heavy metals leached during the Korea standard leaching test (KSLT) passed the regulatory limits for safe disposal, while batch adsorption experiments showed that bottom ash was capable of adsorbing Pb (experimental $q_e$ = 0.05 mg/g), wherein the adsorption rate increased with decreasing particle size. The adsorption data were then fitted to kinetic models, including Lagergren first-order and Pseudo-second order, as well as the Elovich equation, and isotherm models, including the Langmuir, Freundlich and Dubinin-Radushkevich isotherms. The results showed that pseudo-second order kinetics was the most suitable model for describing the kinetic adsorption, while the Freundlich isotherm best represented the equilibrium sorption onto bottom ash. The maximum sorption capacity and energy of adsorption of bottom ash were 0.315 mg/g and 7.01 KJ/mol, respectively.

Bacterial Community Structure in Activated Sludge Reactors Treating Free or Metal-Complexed Cyanides

  • The microbial activity and bacterial community structure of activated sludge reactors, which treated free cyanide (FC), zinc-complexed cyanide (ZC), or nickel-complexed cyanide (NC), were studied. The three reactors (designated as re-FC, re-ZC, and re-NC) were operated for 50 days with a stepwise decrease of hydraulic retention time. In the re-FC and re-ZC reactors, FC or ZC was almost completely removed, whereas approximately 80-87% of NC was removed in re-NC. This result might be attributed to the high toxicity of nickel released after degradation of NC. In the batch test, the sludges taken from re-FC and re-ZC completely degraded FC, ZC, and NC, whereas the sludge from re-NC degraded only NC. Although re-FC and re-ZC showed similar properties in regard to cyanide degradation, denaturing gradient gel electrophoresis (DGGE) analysis of the 16S rRNA gene of the bacterial communities in the three reactors showed that bacterial community was specifically acclimated to each reactor. We found several bacterial sequences in DGGE bands that showed high similarity to known cyanide-degrading bacteria such as Klebsiella spp., Acidovorax spp., and Achromobacter xylosoxidans. Flocforming microorganism might also be one of the major microorganisms, since many sequences related to Zoogloea, Microbacterium, and phylum TM7 were detected in all the reactors.

  • 본 연구는 혼합차수재 개발의 일부로 알루미늄 샛쉬 제작공정에서 산업 부산물로 생산되고 있는 gibbsite 와 자연 생성물인 점토광물이 흡착능을 조사한다. 연구결과는 다음과 같이 몇 가지로 요약할 수 있는데, 1) 구리, 카드뮴, 니켈, 페놀등의 흡착은 12 시간에서 48 시간 이내로 비교적 신속히 평형을 이루었다. 2) 흡착은 표면현상으로 흡착의 속도와 범위는 흡착제의 비표면적과 밀접한 관계가 있다. 3) Gibbsite 와 bentonite 표면에서 중금속의 흡착현상은 Freundich 등온흡착식 보다 Langmuir 등온흡착식의 적용이 보다 타당하였다. 4) 페놀의 경우 N 값이 1 에 가까운, 즉 선형 등온흡착식과 유사한 Freundich 식이 흡착해석에 유리하였다. 5) 중금속과 페놀은 다음 순서로 더 많은 흡착제거가 이루어 졌다. 흡착제 : $2{\mu}m-Al(OH)_3$ > 혼합시료 > $12{\mu}m-Al(OH)_3$ > Na-벤토나이트> $30{\mu}m-Al(OH)_3$

카올리나이트의 납 흡착 특성 (Adsorption Characteristics of Lead on Kaolinite)

  • 본 연구에서는 카올리나이트에 대한 납의 흡착 특성을 규명코자 실내에서 회분식 흡착시험을 수행하였다. 회분식 흡착시험에서는 흡착평형도달시간, 흡착능 및 흡착등온식을 연구하였고 pH와 혼합비에 따른 카올리나이트에 대한 납의 흡착특성에 미치는 영향을 분석하였다. 실험결과 카올리나이트에 대한 납의 흡착은 24시간 이내에 평형에 도달하였고 초기오염농도가 증가함에 따라 카올리나이트에 대한 납의 흡착량은 증가하나 초기농도 198mg/l 이상에서는 흡착율은 감소하였다 Freundlich 흡착등온식에 적용한 결과 흡착강도를 나타내는 계수 1/n은 0.9584이다. 그리고 pH 값이 증가함에 따라 초기오염농도에 관계없이 카올리나이트에 대한 납의 흡착량 및 흡착율은 증가되었으며 pH 8 이상에서는 일정한 값에 수렴하였다. 또한 혼합비가 증가함에 따라 흡착량은 증가하였다. 반면에 흡착율은 점점 증가하다가 일정 혼합비 8 이상에서는 감소하는 경향이 있음을 알 수 있었다.

산소용해수와 미생물제재를 이용한 호소 및 폐쇄수역의 정화기술 (Purification Technology in Closed Water like a Reservoir and Pond using Oxygen Solubilized Device and Standardized Microorganism Culture System)

  • The oxygen solubilized device(O.S.D) and standardized microorganism culture system is more efficient than physical and chemical purification techniques in closed water. This study was to determine how the O.S.D and standardized culture system is efficient in purification capacity in closed water based on the lab scale and pilot plant. In the batch test, inducing the quantitative results from pilot plant operation condition, removal efficiency of COD and TN were about 48.3% and 35% respectively, while SS and chlorophyll-a were 94.9% and 68.7%. The pilot plant results showed that suspended solid(SS) and chlorophyll-a removal efficiency were 60% and 59% respectively, due to coagulation characteristics by standardized culture. Total nitrogen(TN) and total phosphorus(TP)showed good effect for the purification of target pond water quality from field data. Additionally, released velocity was determined in control condition of $5.31mgPO{_4}^{3-}{\cdot}m^{-2}{\cdot}day^{-1}$ and $2486.8mgCOD{\cdot}m^{-2}{\cdot}day^{-1}$. Otherwise, phosphate and COD reflux in the aeration and microorganism condition was showed $-9.95mgPO{_4}^{3-}{\cdot}m^{-2}{\cdot}day^{-1}$ and $-397.88mgCOD{\cdot}m^{-2}{\cdot}day^{-1}$. This technology is the most effective not only removal of SS and chlorophyll-a but also control of phosphate and COD release which is very important phenomena in evaluating water quality in closed water like a reservoir and pond.

저장온도 및 종자 처리가 봄파종 인삼 출아와 생장에 미치는 영향 (Effects of Storage Temperature and Seed Treatment on Emergence and Growth Properties of Panax ginseng at Spring-sowing)

  • Background: In Korea, seeds of Panax ginseng C. A. Meyer need to be stored under cold temperature and high humidity condition for months to break physiological dormancy, making storage difficult until spring-sowing. This study was conducted to test the effects of seed storage conditions and seed treatment on the emergence of seedling after spring-sowing in a nursery greenhouse. Methods and Results: After dehiscence, endocarp dried seeds in mild or completely, and wet seeds were stored in $2^{\circ}C$ and $-3.5^{\circ}C$ during winter. Storage at $-3.5^{\circ}C$ resulted in a lower emergence rate (ER) than that at $2^{\circ}C$, and additional cold ($2^{\circ}C$) treatment before or after storage at $-3.5^{\circ}C$ increased the ER. Endocarp dehydration prevented pre-germination at $2^{\circ}C$ storage and increased the ER of seeds stored at $-3.5^{\circ}C$. ER was also dependent on the batch of seeds. However, seed treatments before sowing had only limited effects on ER. Root loss was the main reason for damping-off; prolonged cold storage of seeds increased damping-off, as the detection of pathogens was not high. Conclusions: This study showed that storage conditions such as temperature and moisture content of seeds, affect the ER after spring-sowing and vitality of seedlings, suggesting further attention on seed control for secure seedling stands after spring-sowing.

Methane Production Potential of Food Waste and Food Waste Mixture with Swine Manure in Anaerobic Digestion

  • Purpose: Methane production potential in aerobic digestion was assessed according to feed to inoculum (F/I) ratio for food waste only, and mixing ratio of two materials for food waste and swine manure to give a basic data for the design of anaerobic digestion system. Methods: Anaerbic digestion test was performed using a lab scale batch reactor at $35^{\circ}C$ for six different feed to inoculum (F/I) ratios (0.50, 0.72, 1.14, 1.50, 2.14 and 3.41), three food waste to swine manure ratios (100:0, 60:40 and 40:60) with two different loading concentrations (10g VS/L and 30g VS/L). Results: For food waste only, the highest biogas yield of 1008 mL/gVS was obtained at 0.50 of F/I. For the co-digestion of food waste and swine manure mixture, the highest biogas yield of 1148 mL/gVS was obtained at a mixing ratio of 40:60 with loading concentration of 10g VS/L. Conclusions: F/I ratio for the food waste only, mixing ratio of food waste and swine manure, and co-substrate loading rate affected the biogas production rate. For the low loading rate, there was not so much difference according to the mixing ratio of food waste and swine manure, but for the high loading rate higher biogas yield was acquired for the co-digestion of food waste and swine manure than for the food waste alone (mixing ratio, 100:0).

활성탄(活性炭) 흡착(吸着)에 의한 취기유발물질(臭氣誘發物質) 제거(除去)에 관(關)한 연구(硏究) -GEOSMIN, 2-MIB를 중심으로- (A Study on the Removal of Taste and Odor Compounds by Activated Carbon Adsorption)

  • The occurrence of objectionable tastes and odors in drinking water is a common and widespread problem. The most troublesome odors are usually those described as muddy or earthy-musty. Two organic compounds which have been implicated as the cause of earthy-musty odor problems in water are geosmin and 2-Methylisoborneol. These earthy-musty organics have been shown to be metabolites of actinomycetes and blue green algae. The purpose of this paper is to describe adsorbability in removing these two oder causing compounds(geosmin and 2-MIB) upon various conditions like pH variation, adding humic acid and different activated carbon. The conclusion of this study are as followings. In batch test, carbon dosage is 10mg/100ml for geosmin and 15mg/100ml for 2-MIB. Both were in equilibrium state after 60 hours. In model simulation, F-P model described experiment data and modelling data appropriately in geosmin but F-S model not. In case of 2-MIB, models didn't describe relation between experiment and modelling data well. Two causative agents of earthy-musty odor compounds, geosmin and 2-MIB, are strongly adsorbed by activated carbon either coconut or brown. There appears to be no effect of pH (3,7,9) on adsorption of these two organics. Activated carbon proved to be more effective for removing geosmin than for removing 2-MIB. When activated carbon is. used in removing these two organics, the removal of these appeared to be adversely affected by back ground organic compounds, such as humic substances, due to competitive adsorption.

The Application of ParalluxTM System for Multi-Detection of (Fluoro)quinolone Class Antibiotics Residues in Raw Bovine Milk

  • This study aimed to apply the Parallux system to detect (fluoro)quinone antibiotics residues in raw bovine milk. The immunogen enabled the generation of a specific antiserum with a titer of 1/40,000. The $Parallax^{TM}$ kit using the antibody displayed $IC_{50}$ value of 10 to 150 ppb for (fluoro)quinolone antibiotics. $Parallax^{TM}$ kit was also sensitive for the detection of incurred (fluoro)quinolone at Korean Maximum Residual Levels in raw bovine milk as the result of dose response test. Cross reactivities of the antibody with the common (fluoro)quinolones were determined to be norfloxacin, 100%; enrofloxacin, 100%; ciprofloxacin, 100%; danofloxacin, 100%; nalidixic acid, 40%. Lower detection limit (LOD) values of the $Parallax^{TM}$ kit in raw bovine milk were determined to be norfloxacin, 4 ppb; enrofloxacin, 5 ppb; danofloxacin, 5 ppb; ciprofloxacin, 5 ppb and nalidixic acid, 10 ppb. The $Parallax^{TM}$ kit was run 8 times with five different concentrations of norfloxacin to determine the coefficient of variation (CV, %) of intra-assay, which was between 2.7% and 11.8%. To confirm the precision among kit batches for the inter-assay, five different batch kits were tested with 2 different concentration of norfloxacin. The CVs of the inter assay were 4.2% at 50 ppb, and 7.2% at 10 ppb norfloxacin, respectively.
