Estimation and Association of Genetic Diversity and Heterosis in Basmati Rice

  • A representative group of 38 improved basmati lines including maintainers of sterile lines were studied for genetic diversity utilizing Mahalanobis $D^2$ statistics. A wide diversity was observed having ten clusters with high intra- and inter-cluster distance. Heterosis was estimated utilizing the cytoplasmic male sterile lines from the clusters having high intra- and inter-cluster distance. Highly heterotic hybrids were obtained from the hybridization programme. Cross combinations IR68281A/Pusa 1235-95-73-1-1, IR68281A/RP 3644-41-9-5, Pusa 3A/UPR 2268-4-1, IR 68281A/Pusa Basmati-1, IR68281A/BTCE 10-98, and IR58025A/HKR 97-401 were found to be highly heterotic for grain yield/plant with other agronomic and quality traits. Additionally, a positive association of intra-cluster distance with heterosis was observed, which could be utilized as a guideline for predicting heterosis in basmati hybrid rice breeding program. Also, a positive association between inter-cluster distance and heterosis was observed.

Evaluation of Bacterial Blight Resistance Using SNP and STS Marker-assisted Selection in Aromatic Rice Germplasm

  • A molecular survey was conducted to identify the presence of the bacterial blight resistance genes (Xa1, Xa4, xa5, xa13 and Xa21) in 86 accessions of aromatic rice obtained from germplasm. The results revealed that the resistance gene Xa4 (32.5%), Xa21 (17%), and xa5 (16%) were widely observed in tested rice germplasm. Among tested rice germplasm, 49 accessions showed the presence of more than one of five R genes, and 37 accessions possessed none of the R gene. TALLi and 05-IRRi-M-46 showed the presence of Xa4, xa5, xa13 and Xa21. Rice race $415{\times}Ir352$ exhibited positive amplicon for the Xa1, Xa4, xa5 and Xa21. Hyangmibyeo1hos, Ir841-85-1-1-2 and Jasmine85 showed the positive amplicon for the Xa1, Xa4 and xa5 genes. Yekywin Yinkya Hmwe and Khao Dawk Mali105 showed the presence of Xa1, Xa4 and Xa21 gene. Masino Basmati showed the presence of xa5, xa13, Xa21 genes. Xa1 and Xa21 genes were noticed in Mihayngbyeo, Tarana Deshi, Mayataung and AZUCENA. Hyangmibyeo2ho, Basmati 6311 and Basmati405 possessed only two R genes such as Xa4 and xa5, and xa5 and xa13, respectively. The evaluation results of bacterial blight resistance genes in aromatic rice germplasm will help in breeding of multi disease resistant varieties.

Screening of Rice Blast Resistance Genes from Aromatic Rice Germplasms with SNP Markers

  • Rice blast is one of the serious devastating diseases. This study was carried out to determine the genetic diversities of blast resistance (R) genes form 86 accessions of aromatic rice with eight SNP markers, z4792, zt4792, z60510, zt6057, k6415, k6411, k39575 and t256, which showed the close-set linkage to 6 major genes, Piz, Piz-t, Pik, Pik-m, Pik-p, and Pit. Four accessions of indica type, Mayataung, Yekywin Yinkya Hmwe, Basmati9-93, and Basmati5854, showed the positive amplicons of six major genes. Among 86 accessions, 83 accessions were detected both or one of Piz and Piz-t genes. Seventy three accessions contained the Piz gene with z4792 marker. In addition, 30 and 71 accessions possessed Piz-t gene with zt4792 and zt6057 markers, respectively. Ten accessions showed the positive bands for the Piz-t gene with both zt4792 and zt6057 markers. Only one accession, Khau Nua Keo, was not amplified for both Piz and Piz-t gene. But japonica type, Gerdeh, possessed only Piz gene between Piz and Piz-t. Fifty two accessions showed the three of Pik multiple genes and Pit gene. Four accessions, Iari7447, Daebunhyangdo2, Shiyayuuine, and Basmati 6129 possessed a Pik-p gene. Especially, Pit gene on chromosome 1 was detected with t256 marker in all of 83 accessions, exception of A-2, one accession of japonica type.

Gas Chromatography-Mass Spectrometry에 의한 향미 유전자원의 방향성 화합물 비교분석 (Comparison Analysis of Aromatic Compounds in the Aromatic Rice Germplasm by Gas Chromatography and Mass Spectrometry)

  • 국내 육성향미자원과 도입 향미 유전자원의 2AP 함량을 비교분석하여, 2AP 함량이 높고 농업적 형질이 우수한 자원을 30품종의 향미 유전자원에서 방향성 화합물을 SPME 추출법으로 추출하고 GC-MS로 각 자원이 가지는 방향성 화합물을 비교분석 및 동정하였다. 1. 3종류의 유기용매의 2AP 추출효율은 ethanol > acetonitl > methanol 순으로 확인되었으며, ethnaol의 경우 $30{\sim}90^{\circ}C$의 온도범위에서 3가지 유기용매 중 ethanol이 2AP 추출효과가 가장 뛰어났고, 특히 $90^{\circ}C$에서 30분간 추출하였을 때 2AP 추출함량이 가장 높았고 추출 시간이 경과할수록 2AP의 함량이 감소하였다. 현미의 분쇄정도에 따른 2AP 추출함량은 5초간 분쇄하였을 경우 가장 높았으며 분쇄 시간이 길어질수록 낮아지는 경향이었다. 2. 선발자원의 향에 대한 관능검사는 국내 향미자원 5품종 중 Hyangmibyeo1ho, Arnaghyangchalbyeo가 중간 정도의 향을 띄었고 인디카형 도입자원 19품종 중에서 11품종과 자포니카형 도입자원 6품종 중 2품종이 중간 이상의 향을 나타났다. 3. 30품종의 평균 2AP함량은 국내 육성 향미자원이 도입 향미자원에 비해 낮았으며 도입자원에서는 인디카형이 자포니카형 보다 높은 2AP 함량을 나타내었다. 4. 156종의 방향성 화합물 기능기별로 분류하면 alcohol류 33종, acid류 23종, ketone류 24종, hydrocarbon류 20종, ester류 16종, aldehyde류 16종, ether류 4종, amine류 5종 phenol류 3종, base류 2종, 및 기타 10종으로 분류되었다. 5. 156종의 방향성 화합물 중 생태형별 출현빈도가 50% 이상인 방향성 화합물을 살펴보면, 국내육성 향미자원에서는 2-acetyl-1-pyrroline 외 28종, Japonica type 도입 향미자원에서는 9,12-octadecadienoic acid 외 31종, Indica type 도입 향미자원에서는 naphthalene 외 23종이었다. 6. GC-MS 분석결과 검출된 국내 육성 향미자원의 방향성 화합물의 수는 평균 38개($36{\sim}41$개), 평균 total peak area는 61.1%($51.6{\sim}79.3%$)이었다. 도입 자포니카형 향미자원의 방향성 화합물의 수는 평균 39개($26{\sim}45$개), 평균 total peak area는 63.2%($40.9{\sim}81.9%$)이었다. 도입 인디카형 향미자원의 방향성 화합물의 수는 평균 39.6개($26{\sim}53$개), 평균 total peak area는 72.8%($33.8{\sim}98.8%$)이었다. 7. 30 품종으로부터 검출된 156종의 방향성 화합물의 출현 유무(0/1)를 기본 matrix로 하여, UPGMA/Neighbor-join tree 분석을 통해 분류한 결과, 7개의 그룹(Group $I{\sim}VII$)으로 나눠어졌다. 각 그룹의 66% 이상의 자원에서 나타난 향(aroma/odor)성분을 살펴보면, alcohol, aldehyde, acid, ether류 등의 휘발성 성분들의 비율이 품종별로 높았다. 따라서 본 실험의 결과는 대량의 향미 유전자원의 방향성 화합물에 대한 GC/GC-MS분석결과로, 향후 우수한 향미 품종 육성에 필요한 자료로 활용될 수 있다고 사료된다.

Screening of Resistance Genes Linked to Brown Planthopper Using STS Marker in Aromatic Rice Germplasm

  • Brown planthopper (BPH) is a serious insect pest of rice crop throughout rice growing countries, and yield loss due to its infection can be up to 60%. This study aimed to evaluate efficiency of molecular markers for screening BPH resistance accessions among 86 aromatic rice germplasm Eighty-six accessions of aromatic rice germplasm included two accessions of Tongil type (bred in Korea), 28 accessions of japonica type and 56 accessions of indica type. We applied eight STS markers (pBPH9, pBPH19, pBPH20, pBPH21, AJ09-b, RG457L, RG457B, and 7312.T4A) which were linked to four of BPH resistance genes, Bph1, Bph13(t), Bph10, and Bph18(t) respectively. One japonica type accession, 415XIr352, and six indica type accessions possessed one or four positive bands when tested with four STS markers linked to Bph1 gene. One indica type aromatic rice, Basmati9-93, showed the target bands linked to the Bph10 gene. The other accessions did not show same fragments as the respective resistant lines. Bph13(t) is the most widely introduced resistance gene and only one accession showed positive bands implying that this accession might harbor Bph10 and Bph18(t) genes. Three aromatic accessions, Domsiah, Khao Dawk Mali 105 and 415XIr352 showed gene pyramiding of Bph1 and Bph13(t). Two indica aromatic rice, Ds 20 and Basmati 9-93, possessed at least two BPH resistance genes, Bph1, Bph18(t) and Bph13(t), Bph18(t), respectively. These results indicates that aromatic rice germplasm have narrow diversities of BPR resistance genes.

Screening of the Dominant Rice Blast Resistance Genes with PCR-based SNP and CAPS Marker in Aromatic Rice Germplasm

  • The objective of this study was to determine the genetic diversities of major rice blast resistance genes among 84 accessions of aromatic rice germplasm. Eighty four accessions were characterized by a dominant 11 set of PCR-based SNP and CAPS marker, which showed the broad spectrum resistance and closest linkage to seven major rice blast resistance (R) genes, Pia, Pib, Pii, Pi5 (Pi3), Pita (Pita-2), and Pi9 (t). The allele specific PCR markers assay genotype of SCAR and STS markers was applied to estimate the presence or absence of PCR amplicons detected with a pair of PCR markers. One indica accession, Basmati (IT211194), showed the positive amplicons of five major rice blast resistance genes, Pia, Pi5 (Pi3), Pib, Pi-ta (Pi-ta2), and Pik-5 (Pish). Among 48 accessions of the PCR amplicons detected with yca72 marker, only five accessions were identified to Pia gene on chromosome 11. The Pib gene was estimated with the NSb marker and was detected in 65 of 84 accessions. This study showed that nine of 84 accessions contained the Pii gene and owned Pi5 (Pi3) in 42 of 84 accessions by JJ817 and JJ113-T markers, which is coclosest with Pii on chromosome 9. Only six accessions were detected two alleles of the Pita or Pita-2 genes. Three of accessions were identified as the Pi9 (t) gene locus.

아종특이적 STS 마커를 이용한 벼 품종의 유전다양성 분석 (Genetic Diversity of Rice Collections using Subspecies-specific STS Markers)

  • 전 세계에서 광범위하게 수집한 벼 유전자원 320개를 63개의 아종특이적마커로 분석하여 유전자원의 다양성, 유연관계 및 유전집단의 구조분석을 하여 아종특이적마커의 아종판별 효율을 검정하고 아종의 게놈 구성을 검토하고자 본 시험을 수행하였다. 1. 본 연구에서 사용한 63개의 아종특이적 마커는 벼 품종을 인디카와 자포니카 두 아종으로 구분하는데 효과적으로 이용할 수 있었다. 2. 실험에 사용한 320개의 벼 유전자원들은 자포니카군(128개)과 인디카군(178개)으로 나눌 수 있었고 Basmati370, Shennong27 등 14개 품종은 중간형 admixture형태의 품종으로 구분할 수 있었다.

Comparative Analyses for Aroma and Agronomic Traits of Native Rice Cultivars from Central Asia

  • Aromatic rice has become popular owing to its aroma. Growing demand for aromatic rice has spurred interest in the development of domestic cultivars that offer similar combinations of grain attributes such as texture, cooking characteristics, aroma, and taste. In this study, the most important agronomic attributes and aroma of 26 cultivars from Afghanistan, Iran, and Uzbekistan, and controls from Japan, Thailand, and India were characterized. Also $F_2$ populations derived from the cross between(Jasmine 85 aromatic$\times$Nipponbare non-aromatic) and(Jasmine 85$\times$Basmati 370 aromatic) were obtained. Tasting individual grains, cooking test, 1.7% KOH sensory test, and molecular marker analysis have been applied to distinguish between aromatic and non-aromatic rice. Diversity for some traits of agronomic importance, such as plant height was detected among countries, e.g. Afghan cultivars classified as tall, and Iranian and Uzbek intermediate and short, respectively. Differentiations of panicle, grain, leaf, basal internode, and culm dimension among rice cultivars, indicating the source of rice diversity in Central Asia. According to the results, 6 of 10, 2 of 7, and 0 of 6 of Afghan, Iranian, and Uzbek rice cultivars were scored as aromatic, respectively. Therefore, Afghan cultivars are a good source of aromatic rice germplasm for Central Asia. The expression between aromatic and non-aromatic, and aromatic and aromatic combinations has been evaluated. The observed segregation ratio of these crosses in the $F_2$ populations was tested by $x^2$ analysis against the expected ratio for a single gene. A segregation ratio of 3:1 between non-aromatic and aromatic combination has been detected, while segregation has not been detected between the aromatic and aromatic combinations. Also, parallel results were obtained from the tested aromatic rice cultivars. Thus, our results suggest that a single recessive gene controls aroma in all aromatic rice cultivars.

Global Rice Production, Consumption and Trade: Trends and Future Directions

  • The objectives of this paper are (i) to analyze past trends and future directions of rice production, consumption and trade across the world and (ii) to discuss emerging challenges and future directions in the global rice industry. Rice is a staple food of over half of the world's 7.7 billion people. It is an important economic, social, political, and cultural commodity in most Asian countries. Rice is the $1^{st}$ most widely consumed, $2^{nd}$ largely produced, and $3^{rd}$ most widely grown food crop in the world. It was cultivated by 144 million farms in over 100 countries with harvested area of over 163 million ha producing about 745 million tons paddy in 2018. About 90% of the total rice is produced in Asia. China and India, the biggest rice producers, account for over half of the world's rice production. Between 1960 and 2018, world rice production increased over threefold from 221 to 745 million tons (2.1% per year) due to area expansion from 120 to 163 million ha (0.5% per year) and paddy yield increase from 1.8 to 4.6 t/ha (1.6% per year). The Green Revolution led massive increase in rice production prevented famines, provided food for millions of people, reduced poverty and hunger, and improved livelihoods of millions of Asians. The future increase in rice production must come from yield increase as the scope for area expansion is limited. Rice is the most widely consumed food crop. The world's average per capita milled rice consumption is 64 kilograms providing 19% of daily calories. Asia accounted for 84% of global consumption followed by Africa (7%), South America (3%), and the Middle East (2%). Asia's per capita rice consumption is 100 kilograms per year providing 28% of daily calories. The global and Asian per capita consumption increased from the 1960s to the 1990s but stable afterward. The per capita rice consumption is expected to decline in Asia but increase outside Asia especially in Africa in the future. The total milled rice consumption was about 490 million tons in 2018 and projected to reach 550 million tons by 2030 and 590 million tons by 2040. Rice is thinly traded in international market because it is a highly protected commodity. Only about 9% of the total production is traded in global rice market. However, the volume of global rice trade has increased over six-fold from 7.5 to 46.5 million tons between the 1960s and 2018. A relatively small number of exporting countries interact with a large number of importing countries. The top five rice exporting countries are India, Thailand, Vietnam, Pakistan, and China accounting for 74% of the global rice export. The top five rice importing countries are China, Philippines, Nigeria, European Union and Saudi Arabia accounting for 26% of the global rice import. Within rice varieties, Japonica rice accounts for the highest share of the global rice trade (about 12%) followed by Basmati rice (about 10%). The high concentration of exports to a few countries makes international rice market vulnerable to supply disruptions in exporting countries, leading to higher world prices of rice. The export price of Thai 5% broken rice increased from 198 US$/ton in 2000 to 421 US$/ton in 2018. The volumes of trade and rice prices in the global market are expected to increase in the future. The major future challenges of the rice industry are increasing demand due to population growth, rising demand in Africa, economic growth and diet diversification, competition for natural resources (land and water), labor scarcity, climate change and natural hazards, poverty and inequality, hunger and malnutrition, urbanization, low income in rice farming, yield saturation, aging of farmers, feminization of agriculture, health and environmental concerns, improving value chains, and shifting donor priorities away from agriculture. At the same time, new opportunities are available due to access to new technologies, increased investment by the private sector, and increased global partnership. More investment in rice research and development is needed to develop and disseminate innovative technologies and practices to overcome problems and ensure food and nutrition security of the future population.

Gas chromatography를 이용한 향미 유전자원의 2-acetyl-1-pyrroline 정량분석 (Quantification of 2-Acetyl-1-pyrroline from the Aroma Rice Germplasm by Gas Chromatography)

  • 본 실험은 국내 육성향미자원과 유전적 배경이 서로 다른 도입향미 유전자원의 2AP 함량을 비교분석하여, 2AP 함량이 높고 농업적 형질이 우수한 자원을 선발하여 우수한 향미자원의 육성에 필요한 기초 자료를 제공하고자 실시하였다. 3종류의 유기용매에 대한 추출효율은 ethanol>acetonitle>methanol 순으로 확인되었으며, ethnanol의 경우 $30-90^{\circ}C$의 온도범위에서 3가지 유기용매 중 2AP 추출효과가 가장 뛰어났고, 특히 $90^{\circ}C$에서 30분간 추출하였을 때 2AP 추출함량이 가장 높았고 추출 시간이 경과할수록 2AP의 함량이 낮았다. 현미의 분쇄정도에 따른 2AP 추출함량은 5초간 분쇄하였을 경우 가장 높았으며 분쇄 시간이 길어질수록 낮아졌다. 선발한 30품종 중 국내 향미자원과 자포니카형 도입자원의 평균 출수일은 각각 130.3일과 131.8일로 비슷하였으며 인디카형 도입자원은 약 137.1일로 가장 늦었다. 평균 1,000립중은 인디카형 도입자원이 19.9 g으로 가장 낮았으며, 국내 향미자원이 21.65 g이었고 자포니카형 도입자원은 25.5 g으로 높았다. 선발자원의 향에 대한 관능검사는 국내 향미자원 5품종 중향남벼 2호, 아랑향찰벼가 중간 정도의 향을 띄었고 인디카형 도입자원 19품종 중에서 11품종(57.9%)과 자포니카형 도입자원 6품종 중 2품종(33.3%)이 중간 이상의 향을 나타났다. 선발한 30품종의 평균 2AP 함량은 국내 육성 향미자원이 도입 향미자원에 비해 낮았으며 도입자원에서는 인디카형이 자포니카형보다 높은 2AP 함량을 나타내었다. 국내 육성 향미자원은 아랑향찰벼외 4품종과 도입 향미자원 중 인디카형은 Basmati6313 외 18품종 및 자포니카형은 자향나 861 외 5품종을 선발하였다.
