Determination of Heavy Metal Concentration in Herbal Medicines by GF-AAS and Automated Mercury Analyzer

  • GF-AAS와 수은분석기를 이용하여 납, 비소, 카드뮴 및 수은의 회수율로 정확도를 측정한 결과 92.67-102.56% 범위에서 측정되었고, 정밀도를 측정한 결과 0.21-6.00 RSD%의 재현성을 보였으며, CODEX guideline에서 규정하는 범위에 적합하였다. 또한, FAPAS QC material을 검증결과, 회수율은 96.7-102.0%, 재현성은 0.33-4.93 RSD%로 우수한 결과를 나타냈다. 한약재 430건의 평균 Pb 함량은 254.9 ㎍/kg(N.D.-2,515.2)이었고, 평균 As 함량은 171.0 ㎍/kg (N.D.-2,465.2)이었으며, 평균 Cd 함량은 99.2 ㎍/kg (N.D.-797.1), 평균 Hg 함량은 6.0 ㎍/kg (N.D.-83.6)이었다. 분석 결과 우리나라에 유통되는 한약재 20종은 식품의약품안전처에서 규정하는 한약재의 중금속 함량에 대한 허용기준 이내의 결과로 모두 안전한 수준의 한약재로 나타났다. 이를 토대로 한약 규격품 제조업소를 품질관리의 기준으로 하여 유통되기 직전의 가공 포장 과정에서의 한약재 중 납, 비소, 카드뮴 및 수은의 함량을 조사하여 한약재 내 중금속 함량 실태를 파악하고, 국민에게 안전하고 우수한 한약재를 공급하기 위한 기초자료로 활용할 수 있을 것으로 사료된다.

수종 지혈 한약물이 Cytochrome P450 3A4 활성에 미치는 영향 (Influence of Five Herbal Medicines on Cytochrome P450 3A4 Drug-Metabolizing Enzymes in Human Liver Microsomes)

  • Objects : The aim of this study was to investigate the influence of five herbal medicines on cytochrome P450 3A4 drug-metabolizing enzymes in human liver microsomes. Methods : To use human liver microsomes, an extract of five herbal medicines, which are Artemisia princeps Pampan, Sophora jeponica Linne, Panax notoginseng F. H. Chen, Lithospermum Erythrorhizon Sieb., and Cirsium maackii Maxim, which together are called Jihyulyak(止血藥, drugs for arresting bleeding, hemostatics), was co-incubated and measured for relative enzyme activity in incubation condition compared to ketoconazole, a representative inhibitor of CYP 3A4. Results : We showed that all five of the traditional herbal medicines had no inhibition effect of CYP 3A4 at 10, 20, 30, 40, and $50{\mu}g/ml$ doses in human liver microsomes, although Sophora japonica Linne(SJL) showed a little inhibition at about 81% inhibition rate of control. However, this result is not enough to prove that SJL has a CYP 3A4 inhibition effect. Moreover, we can't make sure that those rates had significant induction effect on CYP 3A4. Conclusions : The result of this study could support that those herbal medicines are safer than chemical drugs, even if this is the basic step to prove that result.

청열약 수종의 Cytochrome P450 3A4 효소활성도에 미치는 영향 (The Effect to Drug Metabolizing Enzyme Cytochrome P450 3A4 by Chungyulyak)

  • In this study, the author experimented the influence of five herbal medicines, which are Lonicera japonica Thunb, Paeonia suffruticosa Andr., Fraxinus rhynchophylla Hance, Gardenia jasminoides Ellis, Scutellaria baicalensis George which are called 'Chungyulyak(淸熱藥)' on drug metabolizing enzyme cytochrome P450 3A4 in Human Liver Microsome. Above all, the reason for this study is that herbal medicines can be assumed that herbs might have interactions with drugs, other herbs, alcohol and chemicals whether those are much better synergy effects than expected effects when the medicine was treated alone or not. As a result, we showed that all of five traditional herbal medicines had no CYP 3A4 inhibition effect on 10, 20, 30, 40, $50{\mu}g/m{\ell}$ doses in Human Liver Microsome. However, this result are mostly not enough to prove that PMT has a CYP 3A4 inhibition effect. Moreover, it is not that those rates showed that those herbal medicines have CYP 3A4 induction effect. In conclusion, the result could support that those herbal medicines are more safe than chemical drugs even if this is the basic step to prove that result. Therefore, more specific studies to support this result, which are Kinetic study, cell and animal study then finally until clinical research, are required.

Human Liver Microsome을 이용한 수종 이기약의 간대사효소에 미치는 영향 (The Effect to the Hepatic Metabolic Enzume that Uses Human Liver Microsome Caused by a Bariety of Yigiyak)

  • In this study, we experimented the influence of three herbal medicines, which are Saussurea lappa Clarke, Poncirus trifoliata Rafin, Citrus aurantium Linne, which are called 'Yigiyak(理氣藥)' on drug metabolizing enzyme cytochrome P450 3A4 in Human Liver Microsome. Above all, the reason for this study is that herbal medicines can be assumed that herbs might have interactions with drugs, other herbs, alcohol and chemicals whether those are much better synergy effects than expected effects when the medicine was treated alone or not. As a result, we showed that all of five traditional herbal medicines had no CYP 3A4 inhibition effect on 10, 20, 30, 40, $50{\mu}g/m{\ell}$ doses in Human Liver Microsome even Saussurea lappa Clarke showed a little inhibition as about 93% and 79% inhibition rate of control. However, this result are mostly not enough to prove that SLC has a CYP 3A4 inhibition effect. Moreover, it is not that those rates showed that those herbal medicines have CYP 3A4 induction effect. In conclusion, the result could support that those herbal medicines are more safe than chemical drugs even if this is the basic step to prove that result. Therefore, more specific studies to support this result, which are Kinetic study, cell and animal study then finally until clinical research, are required.

유통 한약재의 잔류농약 및 잔류이산화황의 함량 분석 (The analysis of Residual Pesticides and Sulfur Dioxide in Commercial Medicinal Plants)

  • Objectives : This study was investigated to determine the contents of pesticide residues and sulfur dioxide residues in commercial herbal medicines in Korea. Methods : Chromatographic test was performed on 100 samples consisted with 10 kinds of medicinal plants including improted and domestic products. To establish 19 pesticide residues (DDE, DDD, DDT, Dieldrin, Methoxychlor, BHC isomers, Aldrin, Endosulfan isomers, Endrin, Captan, Procymidone, Chlorpyrifos and Imidacloprid) in commercial herbal medicines, chromatographic equipments were used with the gas chromatography-mass detector and gas chromatography-electron capture detector for qualitative analysis. The imidacloprid analysis was performed by high performance liquid chromatograpgy-ultraviolet detector at 270 nm UV wavelength. The contents of sulfur dioxides were analyzed by modified Monnier-Williams method. All methods were based on notification procedure of Korea Food & Drug Administration (KFDA). Results : The residual pesticides were not founded in improted and domestic samples. Among 100 samples, the residues of sulfur dioxide in 73 samples were not detected and 25 samples showed contents in the range of 0~21.90 mg/kg. The excess samples of MRLs were 2 samples (30 mg/kg to medicinal herbs), Asiasari Radix et Rhizoma (Imported product) and the average amount of sulfur dioxide in 2 unsuitable samples were 14.83 mg/kg. These samples were found to transgress KFDA regulatory guidance of residual sulfur dioxide. Conclusion : These results are able to use as basic data to improve the reliability and value of commercial medicinal herbs.

활혈거어약의 Cytochrome P450 3A4 효소활성에 미치는 영향 (Influence of Five Herbal Medicines on Cytochrome P450 3A4 Drug-Metabolizing Enzyme Activity)

Methods: By using of human liver microsomes, we extracted Cnidium officinale Makino, Rhus verniciflua Stokes, Prunus persica Batsch, Corydalis remota Fisch, Carthamus tinctorius Linne, which are called Hwalhyulgeoouhyak(活血祛瘀藥). Then they were incubated and measured for relative enzyme activity under incubation conditions compared to ketoconazole, which is known as a representative inhibitor of CYP 3A4. Results: We showed that all of five traditional herbal medicines had no inhibition effect of CYP 3A4 at 10, 20, 30, 40, and 50${\mu}g/m{\ell}$ doses in human liver microsomes, although Rhus verniciflua Stokes (RVS) showed a little inhibition as about 95% enzyme activity of control. However, this result was not enough to prove that RVS has a CYP 3A4 inhibition effect. Moreover, we can't confirm that those rates have significant induction effect on CYP 3A4. Conclusions: The result of this study could support that those herbal medicines are more reliable than chemical drugs, even if this is a basic step to prove that result.

한약재 수치에 관한 연구 (III) -후박의 수치전.후 Magnolol의 함량분석 및 시험관내에서 최종당화산물 생성억제 효능- (Studies on the Processing of Herbal Medicines (III) -HPLC Analysis of Magnolol and Inhibitory Effects on the Formation of Advanced Glycation Endproducts(AGEs) in Vitro of Unprocessed-and Processed Magnolia Bark-)

  • Advanced glycation end products(AGEs) are largly involved in the pathogenesis of diabetic nephropathy. It is obvious that inhibition of AGEs formation is important in preventing the occurrence and progression of diabetic nephropathy. In diabetes, this reaction is greatly accerated and is important in the pathogenesis of diabetic complications, especially diabetic nephropathy. Therefore, to seek possible AGEs inhibitors in herbal medicines, unprocessed - and processed Magnolia Bark were examined in vitro as basic data for aniaml experiment. The content of magnolol in unprocessed Magnolia Bark was $0.796{\pm}0.072%$, and after processing was decreased to $0.586{\pm}0.101%(p<0.01)$. The content of AGEs was measured by their intrinsic fluorescence. The $IC_{50}({\mu}g/ml)$ values of aminoguanidine, unprocessed- and procesled Magnolia Bark are $38.845{\pm}8.36{\mu}g/ml$, $54.264{\pm}3.153{\mu}g/ml$ and $27.882{\pm}1.836{\mu}g/ml$, respectively. This result means that prcessed Magnolia Bark was more effective than aminoguanidine, as positive control.

유통 한약재의 중금속 함량 조사 (Determination of Heavy Metal Contents in Medicinal Herb)

  • 국내에서 유통되고 있는 한약재의 안전성을 검토하고자 국내산 한약재 28종, 33건과 수입산 37종 56건 등 총 65종 89건을 구입하여 중금속 함량을 조사하였다. Hg의 경우 평균 함유량이 0.010 mg/kg (domestic : 0.010, imported : 0.010)이었으며, Pb의 경우에는 0.380 mg/kg (domestic : 0.311, imported 0.449), Cd은 0.080 mg/kg (domestic : 0.101, imported : 0.059), As를 분석한 결과 2.085 mg/kg (domestic : 1.845, imported : 2.324)으로 나타났다. 또한 Mn은 31.564 mg/kg (domestic 33.844, imported : 29.283), Zn은 15.436 mg/kg (domestic : 18.703, imported : 12.168), Cu가 3.406 mg/kg (domestic : 3.374, imported : 3.437), Fe의 경우 134.944 mg/kg (domestic : 108.327, imported : 161.561)으로 분석되었다. Hg, Pb, Cd,와 As에 대한 전반적인 경향은 대부분의 검사항목에서 안전한 것으로 분석되었으며, 수입산 한약재와 국내산 한약재간의 특별한 차이를 보이지 않았다. 그러나 극히 일부 시료에서 비록 적은 양이지만 기준치를 약간 초과하는 결과를 보여 향후 안전한 한약재를 국민들에게 공급하기 위해서는 보다 더 세심한 관리가 필요할 것으로 생각된다. 이러한 결과로 보아 추후 개별 중금속기준치 설정 및 국내 유통 생약의 개별 중금속 함유량실태 파악하는데 있어서 기초 자료로 활용이 가능할 것으로 판단된다.

Comparison of Relative Antioxidant Capacity and Antiperoxidation Activity of Traditional Medicines in Vitro

  • To investigate the antioxidant capacity of traditional Korean medicines, water extracts from 57 species were tested on their antioxidant activity using radical scavenging effects against ABTSㆍ+. Of which, Rhus javanica, Caesalpinia sappan, Rosa rugosa, Spatholobus suberectus, and Magnolia obovata showed strong antioxidant capacities at 10㎍ concentration. Therefore, antioxidant capacities of 5 traditional medicine extracts in the different concentration (1㎍, 5㎍, and 10㎍) were determined. The 5 traditional medicine extracts was detected in antioxidant capacity dose dependently. R. javanica was showed the highest antioxidant capacity, the antioxidant activity at 1 ㎍ of herbal extract being 0.85 mM TE. At the same time, the antiperoxidation effects of these 5 medicines were determined. Lipid peroxidation in brain homogenates induced by NADPH and ADP-Fe/sup 2+/ was strong inhibited by S, suberectus, R. rugosa, and M. obovata. Extract of C. sappan was showed the highest inhibition against lipid peroxidation compared with 4 herbal extracts in vitro. These traditional medicines are a potent antioxidant capacity and antiperoxidation activity, further investigation into the in vivo antioxidant therapeutic potential for treatment of human disorders such as Alzheimer's disease.

Herbal pathies (Unani, Ayurveda) need to review their way of research

  • World health organization has recently published a strategic plan for the development and promotion of traditional system of medicine. Herbal pathies especially Unani and Ayurvedic systems of medicines have great scope in this aspect. But, there are several problems with these pathies in the current era, as the way of research and identification is still on classical system. The correct identification of medicinal plant is one of the major problems in both the system. This should be corrected with the modern tools and techniques. The various types of data including recent discoveries, economical growth, ethnobotanical literature and extremely rapid increase in herbal journals and books have emerged great scope for these pathies. At the same time several challenges and threats are present including herb-drug interaction, false reports, toxicity studies etc. In this review paper, opportunities, threats, and researches to be focused will be discussed.
