Culture Conditions for Mycelial Growth of Coriolus versicolor

  • Coriolus versicolor, is one of the most popular medicinal mushrooms due its various biologically active components. This study was conducted to obtain basic information regarding the mycelial culture conditions of C. versicolor. Based on the culture, and MCM media were suitable for the mycelial growth of the mushroom. The optimum carbon and nitrogen sources were dextrin and yeast extract, respectively, and the optimum C/N ratio was 10 to 2 when 2% glucose was used. Other minor components required for optimal growth included thiamine-HCl and biotin as vitamins, succinic acid, lactic acid and citric acid as organic acids, as well as $MgSO_4{\cdot}7H_2O$ as mineral salts.

  • The purpose of the study was to identify the needs assessment for urban agricultural program. The findings were as follows; Citizens at urban cities said that educational program of 'shortage of farming technology education' should be developed, and wanted to be given 'farming technology' of urban agriculture education area more than other education areas. The citizens asked for urban agricultural program with both theories and practice education than existing urban agricultural program that relied upon basic theory education. And, they wanted urban agricultural program that was based on 'farming technology' to consist of practice education and theory education at the ratio of 80 and 20 and to be professional and differentiated. The citizens preferred 3 to 6 months of education period the most, and more than 95 percent of them wanted to join the program when it is developed.

  • This study was carried out to determine the basic mycelial culture conditions for Poria cocos growth. According to colony diameter and mycelial density, suitable media for mycelial growth were Malt yeast extract, Potato dextrose agar, Yeast extract agar, and Yeast malt agar. The optimum temperature for mycelial growth was between 25 and $35^{\circ}C$, and the optimum pH value was between 4 and 7. Carbon and nitrogen sources were fructose and yeast extract. The optimum C/N ratio was about 10 to 1 with 2% glucose. Other minor components for optimal growth were thiamine-HCl and nicotinamide as vitamins, acetic and lactic acid as organic acids, and $MgSO_4{\cdot}7H_2O$ and $FeSO_4{\cdot}7H_2O$ as mineral salts.

  • Meat derived from skeletal muscles of animals is a highly nutritious type of food, and different meat types differ in nutritional, sensory, and quality properties. This study was conducted to compare the results of previous studies on the muscle fiber characteristics of major porcine skeletal muscles to the end of providing basic data for understanding differences in physicochemical and nutritional properties between different porcine muscle types (or meat cuts). Specifically, the muscle fiber characteristics between 19 major porcine skeletal muscles were compared. The muscle fibers that constitute porcine skeletal muscle can be classified into several types based on their contractile and metabolic characteristics. In addition, the muscle fiber characteristics, including size, composition, and density, of each muscle type were investigated and a technology based on these muscle fiber characteristics for improving meat quality or preventing quality deterioration was briefly discussed. This comparative review revealed that differences in muscle fiber characteristics are primarily responsible for the differences in quality between pork cuts (muscle types) and also suggested that data on muscle fiber characteristics can be used to develop optimal meat storage and packaging technologies for each meat cut (or muscle type).

농업분야 탄소인지예산제도 도입을 위한 국가연구개발사업의 탄소저감 기여도 평가 방안 (Evaluation Methods of Carbon Reduction Contribution for Green Budget of National R&D Projects in Agricultural Sector)

  • Carbon neutrality is emerging as a new paradigm for the international society by transiting from climate change to climate risk. This study proposes evaluation methods for the carbon reduction contribution of climate-related national R&D projects in order to introduce a green budget system in the agricultural sector. We considered the domestic and foreign green budget systems and classified national R&D projects into positive, negative, and neutral from the perspective of carbon reduction. The results of this study propose three methods to estimate the monetary costs and carbon benefits by adopting the framework for the economic evaluation of national R&D projects conducted by the Rural Development Administration. These methods support to evaluate the potential contribution to carbon reduction of national R&D projects in the agricultural sector. Finally, the proposed methods were tested and verified for the efficiency and validity of evaluating carbon reduction contribution. These evaluation methods of the carbon reduction contribution can be used as a basic methodology for the pre-budget calculations of national R&D projects and the contribution for the greenhouse gas reduction budget.

Design of ICT-based Agricultural Safety Monitoring System Models

  • Objective: This study carried out base research to build an agricultural safety monitoring system through ICT convergence to reduce safety accidents and enhance welfare in life in the agricultural field. Background: The functions and values of rural villages as the space of living are recognized anew, but occupational accident rate due to farm work accidents is on the rise each year. Therefore, the seriousness of such a problem emerges. The convergence technology combining ICT is recently applied to industries overall, and therefore better services are offered. However, studies on ICT convergence has not yet been applied to the agricultural safety field. Method: This study identified ICT convergence service technology trends and representative serious accident types mainly occurring in agricultural activities. This study defined the major factors of farm work accidents and ICT to solve those accident factors including the sensor technology, wired/wireless communication technology and location information service, and applied them to prototype PCB for the development of an agricultural safety monitoring system. Results: This study proposed an emergency monitoring system for farmers and a harmful environment monitoring system. The ICT technology to prevent farm work accidents can be summarized as sensing technology, ICT and network technology and user interface technology. This study also designed PCB module configuration and situation judgment algorithm as basic research for proposed monitoring system development. Conclusion: The ICT-based agricultural safety monitoring research proposed in this study is expected to become the basis to build a future real time monitoring system, and also is expected to contribute to social safety and welfare service improvement for farmers. Application: The ICT convergence farmer accident prevention system will make contributions to the prevention of serious farm work accidents.

붉은자루동충하초의 균사생육에 관한 연구 (Cultural Characteristics of Mycelial Growth by an Entomogenous Fungus, Cordyceps pruinosa Petch)

  • This study was carried out to obtain basic data on physiological characteristics for an artificial cultivation of fruiting body of C. pruinosa. C. pruinosa showed the most favorable growth on the MCM medium. The optimal condition for the mycelial growth was obtained at 25$^{\circ}C$ and pH 5.0, respectively. Carbon sources such as arabinose, mannose, xylose were favorable for stimulating a mycelial growth and fruiting bodies of C. pruinosa. Ammonium nitrate, ammonium citrate and ammonium tartrate of nitrogen sources also appeared to be good in the mycelial growth and fruiting body formation.

  • Phellinus genus belonged to Hymenochaetaceae of Basidiomycetes and has been well known as one of the most popular medicinal mushrooms due to high antitumor activity. This study was carried out to obtain the basic information for mycelial culture conditions of Phellinus linteus, P. baumii, and P. gilvus. According to colony diameter and mycelial density, the media for suitable mycelial growth of them were shown in MEA, glucose peptone, and MCM. The optimum temperature for mycelial growth was $30^{\circ}C$. Carbon and nitrogen sources were man nose and malt extract, respectively. The optimum C/N ratio was 10:1 to 5:1 with 2% glucose concentration, vitamin was thiamine-HCl, organic acid was succinic acid, and mineral salt was $MgSO_{4}{\cdot}7H_{2}O$.

  • Ganoderma applanatum is one of the most popular medicinal mushrooms due to the various biologically active components it produces. Based on the colony diameter and mycelial density, PDA, YMA and MCM media were suitable for the mycelial growth of the mushroom. The optimum temperature for mycelial growth was found to be $25{\sim}30^{\circ}C$. The optimum carbon and nitrogen sources were mannose and dextrin, respectively, and the optimum C/N ratio was 2 to 10 when 2% glucose was used. Other minor components required for the optimal growth included thiamine-HCl and biotin as vitamins, succinic acid and lactic acid as organic acids, and $MgSO_4$ $7H_2C$, $KH_2PO_4$ and NaCl as mineral salts.

Silk Fibroin Microsphere and Its Characterization

  • Using gel filtration chromatography, high molecular silk fibroin with high purity was obtained and silk flbroin microsphere particles (SFMP) could be simply made by spray dryer method. Also, some of the physicochemical properties of SFMP and morphology were investigated. The average molecular weight of pure silk fibroin protein dissolved in calcium chloride is about 61,500g/㏖ as measured by gel permeation chromatography. SFMP was spherical in shape, and particles, sized average of 2 ${\pm}$ 10 ${\mu}$, were observed by SEM and particle analyzer, respectively. Obtaining microspheres particles by spray dryer method accelerated the transition from the random coil to the $\beta$-sheet structure during spray dryer treatment. It was identified by the basic fourier transform infrared spectroscopy of SFMP. The swelling ratio of SFMP is majorly dependent on the pH of the solution, not on the occurred gelation. The characteristic structure, which might be applicable to immobilization of drugs is superior to other matrix materials for the use of biomaterials with skin affinity.
