Heavy Metal Uptake by Balloon Flower Together with Investigating Soil Properties and Heavy Metal Concentrations in the Cultivated Soils

  • Soil properties and heavy metal (HM) concentrations in the field soils where balloon flowers (Platycodon grandiflorum, BF) were cultivated, were investigated together with HM (Cd, Cu, Pb, and Zn) accumulation by the BF roots. Basically, in most soils examined (51-97% among 65 samples), the chemical properties including soil pH, organic matter, available-P, and exchangeable cation contents appeared to be lower than the optimal ranges for balloon flower cultivation. There were no samples exceeding the standard limits for HM in soils. Instead, the total HM concentration levels in soils appeared to be maintained at around background levels for general soil in Korea. This implied that elevated HM accumulation in the soils caused by any possible input sources was unlikely. Even though the BF cultivated soils were not contaminated by HM, it was appeared that substantial amount of Cd was accumulated in BF roots with 1.5% and 35% roots samples exceeding the standard limits legislated for BF root ($0.81mg\;kg^{-1}DW$) and herbal plants ($0.3mg\;kg^{-1}DW$), respectively. This implied that the soil HM standard limits based on the total concentration does not reflect well the metal accumulation by plants and also it is likely that the Cd standard limits for BF and herbal plants is too restrict.

Membrane Filter와 Sucrose 농도가 도라지(Platycodon grandiflorum A. DC.) 기내 배양묘의 생장에 미치는 영향 (Effects of Membrane Filter and Sucrose Concentrations on the Growth of Balloon Flower (Platycodon grandiflorum A. DC.) Plantlets In Vitro)

  • 기내 배양환경을 조절하여 건전한 유식물체를 획득하기 위해 도라지를 무균발아시켜 shoot부분을 NAA 0.1 mg/L가 첨가된 MS 기본배지에 치상한 후 membrane filter 부착 유무와 sucrose 농도를 달리 처리하여 생장반응을 조사하였다. Membrane filter 미부착구에서는 배양용기 내의 $CO_2$$C_2$H$_4$ 농도가 membrane filter 부착구에 비해 높았으며 membrane filter가 부착되지 않았을 때 $CO_2$ 농도는 배양기간이 경과될수록 증가하였으나 $C_2$H$_4$ 농도는 감소하는 경향이었다. 식물체 생장은 membrane filter 부착구가 미부착구에 비해 양호하였고, 미부착구에서는 sucrose 농도가 증가할수록 생육이 양호하게 나타났다. 생체중과 건물중은 membrane filter를 부착하지 않은 sucrose 4.5% 처리구에 비해 membrane filter를 부착한 sucrose 무첨가구에서 비슷하거나 높았다. 엽록소 함량은 membrane filter 부착구에서 미부착구에 비해 높았으며 당 함량도 비슷한 경향이었는데 두 처리간 차이는 더 컸다. 기공은 membrane filter 부착구에서 미부착구에 비해 닫혀 있었으며 엽육조직은 membrane filter를 부착하거나 sucrose 농도가 높을수록 잘 발달하였다. 또한 상토에 이식한 후 13일째 생존율은 sucrose 처리 농도에 관계없이 membrane filter 미부착구는 0%인데 비해 부착구는 100%로 나타났다.

Endophytic Bacillus sp. CY22 from a Balloon Flower (Platycodon grandiflorum) Produces Surfactin Isoforms

  • Surfactin is a mixture of cyclic lipopeptides built from variants of a heptapeptide and a ${\beta}-hydroxy$ fatty acid produced by several strains of Bacillus sp. Surfactin isoforms produced by endophytic Bacillus sp. CY22 from a balloon flower were isolated and characterized. It was found that the purified surfactin had three isoforms with protonated masses of m/z 1,008, 1,022, and 1,036, and different structures in combination with Na, K, Ca ions using MALDI-TOF MS, ESI-MS/MS, and ICP MS, respectively. In the MS/MS analysis, the isolated surfactin had the identical amino acid sequence (LLVDLL) and hydroxy fatty acids (with 13 to 15 carbons in length), even though isolated from different Bacillus strains. The sfp22 gene, required for producing the surfactin, consisted of an open reading frame (ORF) of 675 bp encoding 224 amino acid residues with a signal peptide of 20 amino acids. The predicted amino acid sequence of sfp22 was very similar to that of Ipa-8.

채소 식품의 건강 이미지가 기호와 섭취에 미치는 영향 (Influence of Healthy Image on Preference and Intake of Vegetables)

  • This study investigated the effects of image on the preference and intake frequency of 19 vegetables. A total of 359 usable surveys were collected using a convenient sampling method. The subjects included females (51.8%), university students (50.7%), home residents (66.9%) and subject's spending 20,000~40,000 won on meals/week (41.5%) and eat out 2~3 times/week (29.5%). The intake frequency of vegetables was 2~3 times per month. The healthy image of all vegetables was good overall and the average preference was 3.78 (out of 5 on the Likert). Tomatoes had the healthiest image, onions the highest preference, and Korean cabbage the highest intake frequency. For males, the vegetable with the healthiest image was sesame leaf, while the healthiest foods for females were broccoli and tomatoes. Elementary students had a healthier image of cucumber, bean sprouts, radish, sesame leaf, lettuce, radish leaf, and cabbage than university students and adults. Home residents had a healthier image of cabbage and burdock than other types of residents. Subject that st over 20,000 won per week on meals had a higher image of most vegetables. In terms of preference, males liked Korean cabbage, green pumpkin, balloon flower roots, radish leaf, and lotus root, but female liked tomatoes. In addition, elementary students, home residents, and subjects who eat out less often tended to prefer vegetables. In terms of intake, there was a high frequency of intake for all vegetables in adults. Home residents specifically had a higher intake of cucumber, carrot, bean sprouts, spinach, green pumpkin, balloon flower roots, lettuce, radish leaf, broccoli, burdock, lotus root, and tomato. Overall, the healthy image of vegetables had a positive influence on their preference and intake frequency. Therefore, to encourage the intake of vegetables, direct or indirect variables should be examined.

전처리 나물류(깐 도라지, 데친 고사리)의 제조공정 및 유통 중 저장조건의 표준화를 위한 연구 (Standardization of Manufacturing Process and Storage Condition for Pre-processed Foodstuffs (Pre-processed Namul: Peeled Balloon Flower Roots and Parboiled Bracken))

  • 깐 도라지와 데친 고사리는 단체급식에서 흔하게 사용되는 전처리 식재료이다. 이 연구는 이들 식재료의 제조공정과 시장에서 유통 중 최적 저장 조건을 확립하기 위해 수행하였다. 이들 식재료를 생산하는 제조업체에 대한 조사를 통해 가장 적합한 제조공정을 개발하였다. 조사한 제조업체 중 한 곳을 선정하여 시료를 채취하였으며 이 시료를 각각 다른 온도(4, 10, $20^{\circ}C$)에서 저장하면서 저장기간(1, 2, 3, 4, 5, 7, 10 days)에 따른 미생물학적, 이화학적, 관능적 품질 변화를 조사하였다. 저장 개시 시점의 도라지의 일반세균수는 5.26 log CFU/g이었고 $4^{\circ}C$ 저장군에서 일반세균수는 3일차까지 별다른 변화를 보이지 않았으나 4일차에 7.63 log CFU/g로 증가하였고 이후 7일차까지 완만하게 증가하였다. 그러나 10, $20^{\circ}C$ 저장군에서는 2일차에 각각 7.42 log CFU/g, 7.86 log CFU/g 수준까지 급격히 증가하였다. 이러한 양상은 데친 고사리에서도 유사하게 나타났다. 이화학적 품질변화의 경우 $20^{\circ}C$ 저장군에서 pH와 경도가 급속히 감소되는 결과를 보였다. 관능품질은 $4^{\circ}C$ 저장군에서 5일차까지 우호적으로 평가(9점 만점에 5~6점)되었으나 나머지 저장 온도군에서는 2일차부터 기호도가 급격히 감소하였다. 따라서 이들 전처리 나물의 최적 저장 조건은 $4^{\circ}C$에서 3일 이내인 것으로 확인되었다.

Antimicrobial Effect of Phytochemicals to Listeria monocytogenes Isolated from Slaughterhouses

  • 본 연구에서는 도축장에서 분리된 Listeria monocytogenes를 저해할 수 있는 식물성 항균 물질을 탐색하고, 각 항균 물질의 최소살균농도(MBC)를 확인하였다. 잠재적인 항균 물질로 자몽종자 추출물, 감귤류 추출물, 생강 추출물, 배 추출물, 매실 농축액, 도라지 추출물, 대추 추출물, 오미자 추출물을 선정하였고, 대상 균주로는 국내 도축장에서 분리한 15 개의 L. monocytogenes 균주를 혼합하여 사용하였다. MBC 확인 결과, 최대 $4.0{\mu}g/mL$ 농도의 생강 추출물, 배 추출물, 매실 농축액, 도라지 추출물, 대추 추출물, 오미자 추출물은 고농도의 L. monocytogenes (7 Log CFU/mL)에 대해 균 저해 효과가 전혀 나타나지 않았다. 반면, 7 Log CFU/mL의 L. monocytogenes에 대한 자몽종자 추출물과 감귤류 추출물의 MBC는 $0.002{\mu}g/mL$인 것으로 확인되었다. 3 Log CFU/mL의 L. monocytogenes에 대한 MBC를 시험해 본 결과, 자몽종자 추출물과 감귤류 추출물의 MBC는 $0.001{\mu}g/mL$로 나타났다. 이상의 결과로 볼 때 자몽종자 추출물과 감귤류 추출물은 L. monocytogenes를 제어하기 위해 적용될 수 있을 것으로 판단된다.

조청의 이용실태 및 선호도 연구 (Study on Utilization and Perception of Jochung)

  • 본 연구는 조청의 이용실태 및 선호도와 대중화 방안을 조사하기 위하여 대구 경북에 거주하는 주부 445명을 대상으로 조사한 결과로 다음과 같다. 평소 가정에서 먹는 단맛의 정도는 적당하다가 52.4%로 가장 많았으며, 주로 사용하는 당류로는 올리고당 34.8%, 설탕 27.0%로 나타났다. 조청을 구입 유무로 72.8%가 구입한 경험이 있었으며, 이유로는 '가족, 친구, 이웃권유'와 '건강'이 높게 나타났다. 사용빈도로 일주일에 1~2회 정도 사용한다고 하였으며, 구매 시 고려사항으로는 '건강'이 49.7%, '맛'이 30.2% 순으로 높게 나타났다. 조청을 구입하지 않는 이유로는 '만들기 힘들어서'가 가장 높게 나타났으며, 개선 방안으로 '다양한 구매처'와 '조청의 다양한 기능성 제품 개발'이 높은 것으로 나타났다. 당 종류별 건강에 미치는 영향에 대한 인식정도에서는 화학감미료 1.56으로 가장 낮았으며, 꿀이 3.95로 가장 높게 나타났으며, 조청이 3.65로 다음 순으로 높게 나타났다. 60세 이상에서는 꿀과 조청 모두 가장 높게 나타났으며, 시판 조청에 대한 인식도와 선호도에서 대부분 인식도는 낮았으나 선호도는 높은 것으로 나타났다. 조청을 음식에 활용하여 접목하였을 때 선호도를 살펴본 결과, 한과류, 특히 다식에서 4.05로 가장 높게 나타났다. 이상의 결과, 시판 조청에 대한 인지도와 기능성 재료를 첨가한 조청에 대한 선호도가 낮았으며, 조청에 대한 인식 및 소비 증대를 위하여 위생 안전성을 확보하고, 소비자의 기호에 맞는 조청 개발과 품질 향상이 필요하리라 사료된다.

Mercury Contents of Medicinal Plants and the Cultivated Soils in Korea

  • This study was conducted to identify transition characteristics of mercury in several selected medicinal plants and to find the appropriate management for production of safety food. Cultivated soils and medicinal plants were collected at 29 sites for Angelica gigas (Korean angelica root), 68 sites for Platycodon grandiflorum (Balloon flower), 35 sites for codonopsis lanceolata (Deoduck), 36 sites for Dioscorea batatas (Chinese yam), 32 sites for Rehmannia glutinosa (Foxglove), 16 sites for Cnidium officinale makino (cnidium), and 26 sites for Astragalus membranaceus (milk vetch root) during the harvest season of 2013. Mercury in the soils and medicinal roots were analyzed with a Direct Mercury Analyzer. Average content of mercury in soils cultivated medicinal plants was $0.023mg\;kg^{-1}$ (range: from 0.003 to $0.074mg\;kg^{-1}$) and average content of mercury in medicinal plants was $0.003mg\;kg^{-1}$ (range: from 0.001 to $0.011mg\;kg^{-1}$), indicating that mercury in the surveyed soils and medicinal plants were not exceeded the Korean regulation.

태음조위탕(太陰調胃湯)의 기원(基源), 변천과정(變遷過程) 및 구성원리(構成原理) 재고(再考) (Reconsideration on the Origins, Changes, and Constructive Principle of Taeeumjowi-tang)

  • Objectives This paper was aimed to review the preceding research on the origins, changes and constructive principle of Taeeumjowi-tang.Methods We reviewed Jema Lee's original works including Dongyisusebowon and relevant books to analyze Taeeumjowi-tang-related items. To compensate insufficient evidence of literature, we also refocued physiological and pathological pecularities of Taeeumin and relevant symptomatology.Results and Conclusions 1) The origins of Taeeumjowi-tang should be Mahwang-tang, Saengmaek-san, Seokchangpowonji-san, and Gondam-san. 2) Taeeumjowi-tang went through Saengmaek-san, Sanyakhwawi-jeon, Bopyesaengmaek-tang, Gilgyungsaengmaek-san, and Bopyewon-tang before it was built as the final version. 3) Taeeumjowi-tang consists of a) big blue lilyturf (麥門冬) and Omija (五味子) which are directly of help to exhale and disperse qi-fluid of Lung, b) balloon-flower (桔梗) which helps circulation of qi-fluid from Head to Lung, c) adlay (薏苡仁) and dry nut (乾栗) which are directly of help to raise up qi-fluid of Wiwan, d) ephedra (麻黃) which helps circulation of qi-fluid from Wiwan to skin, e) calamus (石菖蒲) which directly helps the psychological function of Ear, and f) radish seed (蘿葍子) which slightly eliminate the excessive function of Small intestine, treating Wiwanhan symptomatology in Taeeumin.

한국 약용작물의 기술 동향 및 특허 전망 (Technology Trends and Patenting Prospects of Medicinal Plants in Korea)

  • Background: Medicinal plants are widely used in Asia. They have proven to be an invaluable asset in modern drug discovery and their demand has been steadily increasing across various industries. Methods and Results: Using 4,867 valid patents related to 12 oriental medicinal plants of 10 country groups, the growth and development potential of patents was evaluated. The cites per patent (CPP) and patent family size (PFS) indices were used to evaluate the market capability and technological level of the collected patents. Meanwhile, the patent impact index (PII) and technology strength (TS) were used to compare the technological competitiveness of patents among various technology types and markets. Both CPP and PFS indices showed that magnolia-vine and balloon flower have numerous core or original patents. Furthermore, an increase in both PII and TS indices was observed. A newly designed intellectual property multi-layer (IPM) model predicted that the medicine, genome and cosmetic categories have a high possibility of patent application growth. Conclusions: The IPM model can be used to provide the scope of particular technology fields for patent development. In addition, this study can assist patents to advance in the international market and guide the development of a national industrial strategy.
