Bacteriological Examination of Retort Pouched Loach Soup and Soybean Paste Soup Containing Mud Snail (레토르트 파우치 추어탕과 우렁 된장국의 세균상 조사)

  • The present study is concerned with the bacteriological examination of retort pouched loach soup and soybean paste soup containing mud snail. It was found that before sterilization, viable cell counts (c.f.u/ml) in their media were for the loach soup 3.80${\times}$106 on TSA, 2.27${\times}$102 Endo agar, 5.20${\times}$102 on SS agar. With the culture media, SDA and TCBS, no microorgarnisms were isolated from the both soups. Sixteen species of microorgarnisms were identified for the unsterilized soups. In the loach soup on TSA, B. pantothenticus was the dominant species, followed by S. dysenteriae, C. sporogenes and some others were also identified, such on TSA, B. marinus was the dominant species, followed by S. aureus, S. saccharolytics and P. tetradius, S. adorifera, E. ictahuri, E. gergoviae, E. coli were also identified. On Endo agar, the two soups showed a similar bacteriological pattern, in which entrobacterium such as E. gergoviae and E. coli were identified. Particularly K. subsp. rhinoscleromatis for the loach soup. On SS agar, S. ficaria and P. prevotii in the loach soup, S. ficaria and P. tetradius in the soybean paste soup were identified respectively. Bacteriological examination was also carried out for the spoiled retort pouched soup in the market, in which thirteen microorgarnisms were isolated and its pattern almost similar to that before sterilzation. They were B. pantothenticus, S. dysenteriae, C. sporogenes, P. gergoviae, E. ictaluri, S. ficaria, K. subsp. rhinoscleromatis in the loach soup and B. marinus, S. aureus, S. saccharolytics, P. tetradius, E. ictaluri, S. ficaria, S. adorifera in the soybean soup.

Evaluation of the Bacteriological Safety for the Shellfish Growing Area in Jaranman.Satyangdo Area, Korea (자란만.사량도 패류생산해역의 위생학적 안전성 평가)

  • Bacteriological examination of a 12,700 ha area within Jaranman Saryangdo area, located in Jaran bay and Saryangdo island, was conducted with 35 fixed (designated areas and adjacent areas) seawater sampling stations between January 2006 and December 2008. According to results, the geometric mean range and the estimated 90th percentile range of fecal coliform counts in sea water samples collected in the designated area were <1.8-2.4 and <1.8-8.6 MPN/100mL, respectively. The estimated 90th percentile range of fecal coliform counts in sea water samples collected from 6 sampling stations in the adjacent areas were 6.1-34.6 MPN/100mL. Based on these results, bacteriological water quality of the designated areas in the Jaranman. Saryangdo area meet the NSSP (National Shellfish Sanitation Program) guidelines for approved area. The bacteriological sea water quality in Jaranman.Saryangdo area has been shown to be favorable at all investigated sampling stations except for July and August which coincided with heavy rainfall. In fact, the bacteriological water quality was not affected if rainfall was less than 30 mm rainfall. However, the degree of bacteriological contamination increased rapidly and the water quality exceeded approved NSSP guidelines set for rainfall above 60 mm.

  • Milk samples were collected from a total of 418 quarters of 214 Holstein cows. Of these, samples which were positive on California Mastitis Test (CMT) and above 200,000 cells/ml by somatic cell counts were subjected to bacteriological examination and antimicrobial susceptibility test. Major pathogens responsible for mastitis included coagulase-negative staphylococci (34.3%), coagulase-positive staphylococci (21.5%), gran-negative rod (coliforms and noncoliforms: 12.6%) and Streptococcus spp. (8.4%). These strains were tested with 13 antimicrobial agents by the Kirby and Bauer standardized disc diffusion method. The isolated pathogens were mostly susceptible to amoxicillin and cephalothin, but were resistant to erythromycin.

Usefulness of Sputum Induction with Hypertonic Saline in a Real Clinical Practice for Bacteriological Yields of Active Pulmonary Tuberculosis

  • Background: Mycobacterial identification in active pulmonary tuberculosis (APTB) is confirmative, even though successful rates using self-expectorated sputum are limited. Sputum specimens collected by hypertonic saline nebulization showed higher bacteriologic diagnostic sensitivities over those of self-expectoration, mostly studied in smear-negative or sputum-scarce patients. The efficacy of induced sputum was rarely assessed in real clinical settings. Methods: A prospective randomized case-control study was performed in one hospital. The subjects highly suspicious of APTB were asked to provide 3 pairs of sputum specimens in 3 consecutive days. The first pairs of the specimens were obtained either by self-expectoration (ES) from the next day of the visit or sputum induction with 7% saline nebulization in clinic (SI), and the other specimens were collected in the same way. The samples were tested in microscopy, culture, and polymerase chain reaction (PCR). The outcomes of the bacteriological diagnosis were compared. Results: Seventy six patients were assigned to either ES (38 subjects, median age of 51, 65.8% male) or SI (38 subjects, median age of 55, 52.6% male). APTB was clinically confirmed in 51 patients (70.8%), 27 in ES and 24 in SI. Among the APTB, more adequate specimens were collected from SI (41/65, 63.1%) than ES (34/80, 42.5%) (p=0.01). Bacteriological confirmation was achieved in 14 (58.3%) patients in SI, and 13 (48.1%) in ES (p=0.46). In the same-day bacteriological diagnosis with microscopy and PCR, there were positive results for 9 patients (37.5%) in SI and 7 patients (25.9%) in ES (p=0.37). Conclusion: Sputum induction improves sputum specimen adequacy. It may be useful for the same-day bacteriological diagnosis with microscopic examination and PCR.

Evaluation of the Bacteriological Safety of the Shellfish-growing Area in Gangjinman, Korea (강진만 패류생산해역의 위생학적 안전성 평가연구)

  • The sanitary quality of a shellfish-growing area in Gangjinman, Korea, was assessed through a bacteriological examination of seawater form January 2007 to December 2009. The range of the geometric mean (GM) and the estimated 90th percentile for most-probable-number (MPN) values for fecal coliforms in samples collected in the designated area were 2.0-2.4 and 6.1-11.5 MPN/100 mL, respectively. Therefore, the bacteriological quality of seawater in the designated area in the Gangjinman area met the National Shellfish Sanitation Program (NSSP, USA) guidelines for approved areas. Also, the sanitary status of shellfish harvested from the designated area met the Korean Shellfish Sanitation Program (KSSP) fecal coliform criterion (<230 MPN/100 g). However, the degree of bacteriological contamination increased rapidly, and water quality exceeded the criteria for approved areas when large amounts of rain fell before the sampling date. Fortunately, fecal pollutants did not affect the bacteriological quality of a buffer zone established between the shoreline and the designated area. These results indicate that the current buffer zone established around the designated area protects the designated area properly.

  • To test flavonoids for antibacterial activity against oral micraorganisms, flavonoids, quercetrin and naringenin, were incorporated into two pharmaceutical preparations in the form of tooth paste. Samplees of dental plaque, the msot accused dental deposit which initiates the gingival and periodental diseases, were collected from the teeth surface of ten dental students at one week interval before and after using placebo, followed by two formulae of tooth paste containing 0.1% of quercetrin and naringenin (formulas I and II, respectively). The amount of dental plaque was assessed by the quigley and Hens index. Then plaque samples were subjected to bacteriological examination of Gram stain and plate counts of microorganisms. The results revealed that most of Gram negative cocci and bacilli were highly affected by the two formulae : the number of actinomycetes were decreased after using formula I and disappeared completely by the sue of formula II, while the number of Gram positive streptococci was highly decreased after the treatment with the two formulae. These results indicate a possible use of flavonoids to inhibit dental plaque formation.

Evaluation of the Bacteriological Quality of a Shellfish-growing Area in Kamak Bay, Korea

  • The sanitary quality of a shellfish-growing area in Kamak Bay, Korea, was assessed through a bacteriological examination of seawater and oysters from January 2004 to December 2006. From seawater samples collected at 28 stations, the range of geometric mean (GM) and the estimated 90th percentile for most-probable-number (MPN) values of fecal coliforms were <1.8-2.4 MPN/100 mL and 1.8-6.0 MPN/100 mL, respectively. The observed fecal coliform GM and the estimated 90th percentile did not exceed the fecal coliform water quality standards of 14 MPN/100 mL and 43 MPN/100 mL. Therefore, the bacteriological quality of seawater at this shellfish-growing area met the Korean Shellfish Sanitation Program (KSSP) criteria for a growing area used for export. The range of the fecal coliform GM and the estimated 90th percentile MPN values of oyster samples were 19.2-160 MPN/100 g and 20.2-166.9 MPN/100 g, respectively, and the range of the viable cell count was 30-1900 CFU/g. Thus, the fecal coliform value for the oysters and the viable cell count were less than the current shellfish quality standards of 230 MPN/100 g and 50,000 CFU/g, respectively. The bacteriological quality of the oysters complied with the criteria for domestic use and export of shellfish.

A Follow-up Survey on the Socio-medical Status of the Drop-out Cases of Tuberculosis in Jeonju Health Center (보건소등록결핵환자(保健所登錄結核患者)에 대(對)한 사회의학적(社會醫學的) 조사연구(調査硏究))

  • A socio-medical survey was carried out on 1,108 cases of pulmonary tuberculosis who had registered and 220 cases unregistered at Jeonju health center in 1973, during from June 1 to July 31, 1974. As the results of this survey, the following conclusions were obtained. 1. Of the total 1,108 cases of pulmonary tuberculosis there were 708 new cases, while remaining 400 were old cases and rate of registration was 4.0 per thousand person. 2. The highest rate of registration of the newly diagnosed pulmonary tuberculosis could be found in bacteriological examination while 54.4 per cent in X-ray examination. 3. As for the educational status of the tuberculosis patients, primary school graduates contituted the greatest proportion or 64.6 per cent, middle school graduates 13.6 per cent, high school graduates 6.8 per cent and collge graduates only 0.7 per cent. 4. By age group of the cases, at age of 20 to 29 years occupied 23.6 per cent (262 out of 1,108 cases), 17.9 per cent at age of 40 to 49 years. 5. The greatest proportion or 38.4 per cent of the cases had no occupation. 6. The living standard of the patient's household, low class constituted 60.6 per cent of the total households. 7. By distribution of residential area, farming area was 5.0 per thousand person, 4.0 in downtwon and suburban areas. 8. The greatest proportion or 70.0 per cent of positive cases in X-ray examination was unregistered in August, 63.6 per cent in July 1973. 9. 220 out of 1,108 cases(19.9%) were unregistered pulmonary tuberculosis in X-ray and bacteriological examinations. 10. For age groups of unregistered caes, most prevalent age group was 30 to 39. 11. Regarding on the cases of unregistration, 'indifference for disease' occupied highest rate with 31.9 percent' and 'private secret' with 15.7 per cent. 12. Of the total 457 cases drop-outed, there were 78 complete heald cases while remaining 207 inactive.

SSOP Program Development for HACCP Application in Fresh Raw Fish Manufacturing (싱싱회류 생산업체의 HACCP 적용을 지원하기 위한 SSOP Program 개발과 성과)

  • This study aims at developing an easily-applicable Sanitation Standard Operating Procedure(SSOP) program for Fresh raw-fish manufacturing. It compares the sanitation inspection results of SSOP-program-applied procedure and non-applied procedure. The samplings for bacteriological examination were collected from the touching surface of food and from the personal workers. The sanitation status was recorded in the check-up list. The results were statistically analyzed to ascertain the efficiency of the SSOP program. The present official KFDA check-up list for sanitation inspection is composed of 51 items. Each item is judged to be fit or unfit to the standards. When the unfit items are more than 6, the work-place is judged to be insanitary. But in this study, the results of sanitation inspection are scaled in points in the 70% allotment, and the results of bacteriological examination are scaled in points in 30%, totaling 100%. Below 60 scores are judged to be insanitary and administratively punished; 61-75 scores are judged to be average; 76-85 good; above 86 excellent. The sanitation inspection scores by the improved check-up list went up by two grades from 71.8 (before SSOP program) to 88.6 (after SSOP program). The results of bacteriological examination also improved from 57.5 to 98.2. The total scores of both results converted in the allotment of 70% and 30% also showed an improvement by two grades from 66.4 (before SSOP program) to 93.4 (after SSOP program). The above results show that the manufacturers participated in this study are following the 8 essential sanitary procedures which are needed to anchor the HACCP system.

The Study on Safety of Scolopendrid Aqua-acupuncture (오공약침(蜈蚣藥鍼)의 안전성(安全性)에 관한 연구(硏究))

  • Objective : Recently scolopendrid aqua-acupuncture has been a good effect on pain control but it has not been known about clinical safety. The purpose of this study was to investigate acute toxicity of scolopendrid aqua-acupuncture. Method : In order to prove the clinical safety of scolopendrid aqua-acupuncture, We have observed a bacteriological examination and clinical pathology test after scolopendrid aqua-acupuncture treatment. Balb/c mice were injected intravenous with Scolopendrid aquaacupuncture treatment for $LD_{50}$ and acute toxicity test. We analyzed physical reaction(side effect)and clinical pathology test before and after Scolopendrid aqua-acupuncture treatment of mice and 20 patients suffering from pain, who admitted department of Acupunture and Moxibustion, College of Oriental Medicine, Won-Kwang University Kwangju hospital. Results : In the Blood agar plate and Nutrient agar plate, a bacteriological examination did not show a bacillus. In acute $LD_{50}$ toxicity test, there was no mortality thus unable to attain the value. Examining the toxic response in the acute toxicity test, there was no sign of toxication. In acute toxic test, running biochemical serum test couldn't yield any differences between the control and experiment groups. In the 20 patients treated with Scolopendrid aqua-acupuncture, hematologic test did not show remarkable change. In the 20 patients treated with Scolopendrid aqua-acupuncture, Liver function test(AST, ALT, ALP) showed a slight decrease on the contrary, and abnormal rate showed a decrease of 5.0% compared with previous study. Reanl function test(BUN, Cr) and abnormal rate showed a decrease of 5.0% compared with previous study. In the 20 patients treated with Scolopendrid aqua-acupuncture, Electrolyte were normal range before and after treatment. In the Urine analysis of 20 patients, Leukocyte, Protein, Glucose, Keton, Bilirubin, U-bilinogen were not detected before and after Scolopendrid aqua-acupuncture treatment, and the rest almost made no difference.
