A Study on Changes in Antibacterial Activity of Pepsin-hydrolyzed Bovine Apo-lactoferrin at Various Method for Pasteurizations and pH Values (살균방법 및 pH 조건에 따른 Pepsin-hydrolyzed Bovine Apo-lactoferrin의 항균성 변화에 관한 연구)

  • This study was carried out to examine that pepsin-hydrolyzed bovine lactoferrin has applicabilities which are market milk and dairy products. The stability of pepsin-hydrolyzed bovine apo-lactoferrin and the change of its antibacterial character has been studied under various method for pasteurization (LTLT; 65$^{\circ}C$ / 30min., HTST ; 75$^{\circ}C$ / 15sec., UHT ; 135$^{\circ}C$ / 3sec.) and pH Values (pH 2.0, pH 4.0, pH 6.8). The ehated samples were assayed for minimal bacteriocidal concentrations (MBCs) and bacteriocidal effect against E. coli. The results obtained were summarized as follows: After fractionation of pepsin-hydrolyzed bovine lactofeerin by gel filtration. several peptide fractions were found that had strong antibacterial activity. SDS-PAGE showed that the one of these fractions with strong antibacterial activity, which had a molecular mass a range of 30∼33KDa. The MBCs for pepsin-hydrolyzed bovine lactoferrin fraction No. 2 against E. coli required to cause complete inhibition of growth varied within the range of 200∼400 $\mu\textrm{g}$/ml, depending on heat treatments and pH conditions. The peptide fraction No. 2 showed strong bacteriocidal activity against E. coli at LTLT and HTST treatments under acidic pH conditions. and was reduced activity at UHT treatment under pH 6.8 condition.

Effect of Low-Level Laser to Oral Microorganisms (저출력 레이저가 구강미생물에 미치는 영향)

  • This study is to observe the bacteriocidal effect of the Low Level Laser (LLL) against oral microorganisms which are related to the occurrence of periodontal diseases and oral malodors. The Porphyromonas gingivalis 2561 (P. gingivalis 2561) and Prevotella intermedia (Pr. intermedia) were treated with photosensitizing substance-toluidine blue O (TBO; C.I. 52040) and then radiated with the LLL which has 650nm wavelength for 1, 2, 3 and 5mins. continuously upon varying distances of 1, 2 and 3cm for each experimental groups. The results are as follows; 1. The P. gingivalis 2561 which was treated with TBO and then radiated with LLL at a distance of 3cm for 1min. showed 99.99% higher antibacterial effect in comparison to the experimental group treated only with TBO. 2. The Pr. intermedia which was treated with TBO and then radiated with LLL at a distance of 3cm for 1min. showed 99.8% higher antibacterial effect in comparison to the experimental group treated only with TBO. 3. The bacteriocidal effect of the P. gingivalis 2561 treated with TBO was found to gradually increase as the radiation time of LLL extended from 1min. to 3min. at 1min. intervals. 4. A slight decrease in bacteriocidal effect of the P. gingivalis 2561 was found as the radiation distance of LLL increased from 1cm to 3cm at 1cm intervals. 5. The bacteriocidal effect of the Pr. intermedia was found to slightly decrease as the radiation distance of LLL increased from 1cm to 3cm at 1cm. intervals. As the results shown above suggest, the bacteriocidal effect of LLL was found to increase as the radiation time extended and the distance shortened. Moreover, even the experimental group radiated with LLL at 3cm distance for 1min. which showed the lowest level of bacteriocidal effect, was found to have 99.8% higher bacteriocidal effect than the experimental group which was treated only with TBO and, therefore, this clearly shows the bacteriocidal effect of LLL against oral microorganisms. Thus, the use of LLL is thought to become very useful for suportive treatment for periodontitis and implantitis, and controlling oral malodors as long as it is used within the limits where there is no side effect.

Bacteriocidal Effect of Chamical Sanitizers on Natural Microflora and Pathogenic Bacteria in Fish (화학적 살균소독제를 활용한 생선중 존재하는 자연균총 및 주요 식중독균 제어)

  • In this study, we evaluated bacteriocidal effect of CaO (scallop shell powder) for the reduction of microorganism in Scomber japonicus and Pseudosciaena ciean manchurica, and compared with main chemical sanitizers such as chlorine, ethanol, hydrogen peroxide. As a result, the effectiveness of CaO showed dramatic reduction rate for total aerobic bacteria, Escherichia coli and Vibrio parahaemolyticus. The reduction rates of total aerobic bacteria for Scomber japonicus and Pseudosciaena ciean manchurica showed $2.6{\times}10^3\;and\;3.2{\times}10^4$ respectively; Those of Escherichia Coli were $6.3{\times}10^3\;and\;1.2{\times}10^4$, those of Vibrio parahaemolyticus were $5.4{\times}10^4\;and\;5.6{\times}\2.6{\times}10^310^4$, respectively. According to this result, bacteriocidal effect of CaO was about $10^4$ and was similar to bacteriocidal effects of other three chemical sanitizers. According to these results, CaO can alternate the currently used chemical sanitizers due to its natural origin as well as the effectiveness for sterilization.

  • In this study, we evaluated bacteriocidal effect of CaO (scallop shell powder) for the reduction of microorganism in lettuce, and compared with main chemical sanitizers such as chlorine, ethanol, hydrogen peroxide. As a result, the effectiveness of CaO showed dramatic reduction rate for total aerobic bacteria, Escherichia coli, Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, and Salmonella Typhimurium and were $5.9{\times}10^3,\;1.3{\times}10^5,\;5.9{\times}10^3,\;2.7{\times}10^6,\;3.6{\times}10^3,\;4.5{\times}10^3\;and\;2.6{\times}10^4$, respectively. CaO did not show better disinfecting efficiency than chlorine or hydrogen peroxide which were used as sanitizer. In Bacillus cereus case, it showed $10^6$ reduction rate, and were $10^2{\sim}10^5$ times better reduction than ethanol sanitizer. According to these results, CaO can alternate the currently used chemical sanitizers due to its natural origin as well as the effectiveness for sterilization.

Effects of Phytic Acid on Viability of Vibrio vulnificus and on Septicemia-Induced Mice (피틴산이 비브리오균의 생존과 마우스의 패혈증에 미치는 영향)

  • EDTA is known to have bacteriocidal effect on Vibrio vulnificus, pathogen of septicemia by osmotic shock in seafoods. Attempts were made to elucidate the bacteriocidal effect of phytic acid (PA) as a substitute for EDTA against V. vulnificus and its inhibition effect on the septicemia, which induces liver damage of the mice by the pathogen. Viable cells of V. vulnificus with the initial titre of $1.7{\times}10^6$ c.f.u. $ml^{-1}$ decreased by 90.6% after 1 min and 99.6% after 5 min in distilled water. The titre decreased by 65.9% and 94.5% in 2 mM solution of $Mg^{2+}$. In 0.1 mM solution of PA, the rate of decrease in titre was 97.4% after 1 min of incubation and 99.8% after 5 min, compared to 95.7% and 99.8% in 0.1 mM solution of EDTA. The bacteriocidal effect of PA solution at a concentration of 1 mM was marked: the rate of decrease in titre was 99.9% after 1 min. In relation to the bacteriocidal effect, PA was evaluated as a potential therapeutic agent for V. vulnificus septicemia in mouse. When the survival periods of mice were investigated by PA and EDTA treatment after the pathogen injection, the group of mice which infected by a low concentration of the strain survived longer than that inoculated at high concentration; also, the ratio of survival was 1.3 times higher in PA than in EDTA, showing that the fatal rate depended on the inoculation concentration. Although survival periods of mice induced with liver damage by carbon tetrachloride and then inoculated with the strain showed a similar trend, the fatal rate of mice was 2 times faster than those inoculated with only pathogen into normal liver, These results indicate that the infection by V. vulnificus was more fatal to those with liver disease. Also, symptoms of hemorrhage and inflammation on the mice with induced liver damage were reduced in case there was phytic acid treatment at each concentration.

  • Bacteriocidal effect of calcium oxide (CaO, scallop shell powder) on reduction of microorganism in sesame leaf was evaluated through comparison with chemical sanitizers such as chlorine, ethanol, and hydrogen peroxide. CaO showed 0.55-1.49, 0.85-2.56, 0.16-1.08, 0.30-1.14, and $0.19-1.07\;log_{10}$ CFU/g reductions of total aerobic bacteria, total coliforms, Escherichia coli, Listeria monocytogenes, and Salmonella typhimurium respectively, indicating possible use of CaO as effective natural sanitizer.

Bacteriocidal Effects of Ultraviolet Irradiation for Reducing Bovine Mastitis Derived from Environmental Contamination (우분뇨 유래 젖소 유방염 저감을 위한 자외선 조사 살균의 효과 규명)

  • Bovine mastitis is an important disease causing serious economic loss in dairy production and food poison in public health. The major causative agents of bovine mastitis include Escherichia coli (E. coli), Streptococcus agalactiae (S. agalactiae), Staphylococcus aureus (S. aureus). These bacteria were found in milk and environmental condition such as feces, water, soil and so on. Recently, many cases of mastitis are derived from environmental contamination of micro-organisms, which important factors for the spread of this disease in farm. Ultraviolet irradiation (UV) has been used as disinfection for waste and water in clinical and industrial facilities. Moreover the UV irradiation has been used as useful bactericidal agents to remove bacterial biofilms in environmental condition. In this study, we determined the bacterial replication in different percentage of water content (PWC) in sterilized saw dust and feces complexes from farm, and results showed that slightly decreased growth pattern of E. coli and S. agalactiae but increased growth pattern of S. aureus in various PWC (200, 400 and 600%) until 144 h incubation. In the bacteriocidal effect of UV irradiation to bacteria in saw dust and feces complex, the results showed that bacteriocidal effect was depended on the UV irradiation time, irradiation distance and PWC. Especially the antibacterial activity of UV irratiation is stronger in low PWC (50%), long time irradiation (50 sec), and short distance (5 cm) than other condition of this study. Furthermore UV irradiation with stirring showed increased the bactericidal effect compared without stirring. These results suggested that bovine mastitis causing agents may survive long time in environmental condition especially saw dust and feces complexes in farm and can cause a various disease including mastitis. Moreover, these data can be used as basis for application and development of UV disinfection to control of bovine mastitis from environmental contaminated bacteria in dairy farm.

The Antibacterial Activity of Garlic Juice Against Pathogenic Bacteria and Lactic Acid Bacteria. (병원성 세균과 젖산균에 대한 마늘의 항균작용)

  • This study was carried out to determine the inhibitory effect of garlic juice against Escherichia coli, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella typhimurium, Shigella flexneri. Staphylococcus aureus, Streptococcus mutans, Virio. parahaemolyticus which are food pathogenic bacteria and Lactobacillus acidophilus, Lactobacillus brevis, Lactobacillus casei, Lactobacillus plantarum, Lactococcus. lactis, Leuconostoc mesenteroides which are lactic acid bacteria. An aqueous extract of garlic was bacteriocidal against Gram-positive and Gram-negative bacteria in all concentrations (0.1∼2.5(w/v)%) tested in this experiment. Especially 0.5(w/v)% garlic juice inactivated completely E. coli, S. typhimurium, S. flexineri, V. parahaemolyticus and 1.0(w/v)% garlic juice perfectly reduced P. aeruginosa, S. mutans. Generally, the experiment result indicate that garlic juice restrains the growth of the pathogenic bacteria better than the lactic acid bacteria. Therefore, garlic has potential for the preservation of processed foods.

Antimicrobial effects of curcumin against pathogenic bacteria in fish (어류의 병원성 세균에 대한 curcumin의 항균효과)

  • The present study was to investigate anti-microbial effects of curcumin on major bacterial pathogens for farmed fish, such as Aeromonas hydrophila, A. salmonicida subsp. masoucida, A. salmonicida subsp. salmonicida, Edwardsiella tarda, Vibrio vulnificus, V. paraheamolyticus using disk diffusion, minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) tests. In disc diffusion test, curcumin exhibited concentration-dependent antimicrobial activities to all bacteria pathogens used in the study. Antimicrobial effects of curcumin was found differently depending on bacterial species when determined by MIC or MBC tests. For examples, E. tarda and A. hydrophila was respectively the most sensitive bacterium for bacteriostatic and bacteriocidal effect of curcumin. Collectively, curcumin could be a potential natural drug for controlling pathogenic bacteria in the aquaculture industry.

Ozone Treatment for Circulation of Fresh Poultry Meat (신선계육의 유통을 위한 Ozone 처리 효과)

  • The utilization of ozone as a disinfectant for removing poultry meat microorganisms and then cleaning the poultry rinse water was investigated. When microbial suspensions were treated with ozone at 2, 500ppm/min for 40min, microorganisms were not detectable perfectly. The bacteriocidal effect of ozone by temperature was enhanced greater at 7$^{\circ}C$ than $25^{\circ}C$. All poultry meat microorganisms were killed by ozone treatment at 1, 530ppm for 50min. The pathogenic bacteria such as Salmonella sp. were more vulnerable and not detected by ozone treatment for 20min. Ozonation of the suspension for 20min and 50min increased light transmission at 500nm to 58% and 145%, respectively. The order of COD removal was ozone treatment(21%), coagulant((Al)2SO4) treatment(41%), ozone treatment after coagulant treatment(54%).
