A bacterial strain, designated T116, degrading terephthalic acid (TPA) was isolated from the soil around Taegu industrial area into which dye works wastewater flow. The isolate was identified as pseudomonas sp. based on its morphological and physiological characteristics. Degradation of TPA by the strain T116 was confirmed with UV scanning and HPLC. About 90% and 98% of TPA were degraded after 36 and 60 hours, respectively, during the culture in a liquid medium containing 0.1% TPA. Addition of KH2PO4 at a final concentration of 100ppm enhanced the chemical oxygen demand (COD) removal rate about 50% from dye works wastewater by Pseudomonas sp. T116. Optimum pH and temperature for COD reduction from wastewater were 7.0 and 3$0^{\circ}C$, respectively. The bacterium was applied to the continuous culture for the treatment of dye works wastewater whose TPA concentration and CODMn were 2,200ppm and 1,620ppm, respectively. It was observed that 90-95% of COD was eliminated after 4 days culture in the continuous culture with a retention time of 37 or 47 hours.
Two bacterial strains capable of degrading trichloroethylene (TCE), isolated form soils contaminated with various chlorinated alkenes, were identified as Alcaligenes odorous N6 and Nocardia sp. Hl7. In addition, four KCTC strains, including three strains of Pseudomonas putida and one strain of Sphingomonas chlorophenolica, exhibited an ability to degrade toluene. A. odorans N6 and Nocardia sp. H17 degraded 84% of the initial amount of TCE in a basal salts medium (BSM), containing 0.2 mM TCE as the sole source of carbon and energy, in a day. The optimal pH for growth was within a range of 7.0-8.0. A mixed culture of the four toluene-degrading isolates degraded 95% of 0.2 mM TCE with 1.5 mM toluene as an inducer, whereas no TCE was degraded by the same mixture without an inducer. When a mixed culture of all 6 isolates was used, the degradation efficiency of 0.2 mM TCE was 72% without an inducer, in a day, and 82% with toluene as an inducer. In a continuous treatment, 1,000 mg/1 of TCE in an artificial wastewater was completely removed within 18 h when an activated sludge was used along with the microbial mixture, which was 27 h laster than when only an activated sludge was used. Accordingly, it would appear that such a microbial mixture could be effectively applied to the biological treatment of wastewater containing TCE with or without an inducer.

In order to compare bacteria] community structure and diversity in activated sludges, terminal restriction fragment length polymorphism (T-RFLP) of PCR-amplified 16s rDNAs was analyzed for 31 domestic and industrial wastewater treatment plants (WTPs). Regardless of the characteristics of the wastewaters, the bacteria] community structures of activated sludges appeared diverse and complex. In particular, activated sludges in domestic WTPs contained higher bacterial diversity than those in industrial WTPs. It was also found that terminal restriction fragment (T-RF) profiles derived from domestic WTPs were very similar with each other, although activated sludges were collected from different plants at different locations. Interestingly, activated sludges of a WTP where restaurant and toilet sewages of a company were managed showed a bacterial community structure similar to that of domestic WTPs. Activated sludges in leather industria] WTPs also showed a high similarity. However, other wastewaters possessed different bacterial communities, so that overall similarity was as low as about $30\%$. Since activated sludges from WTPs for domestic wastewaters and a company sewage appeared to hold similar bacterial communities, it was necessary to confirm if similar wastewaters induce a similar bacterial community. To answer this question, analysis of T-RFs for activated sludges, taken from another 12 domestic WTPs, was conducted by using a 6FAM$^{TM}$-Iabeled primer and an automated DNA sequencer for higher sensitivity. Among 12 samples, it was again found that T-RF profiles of activated sludges from Yongin, Sungnam, Suwon, and Tancheon domestic WTPs in Kyonggi-do were very similar with each other. On the other hand, T-RF profiles of activated sludges from Shihwa and Ansan WTPs were quite different from each other. It was thought that this deviation was caused by wastewaters, since Ansan and Shihwa WTPs receive both domestic and industrial wastewaters. From these results, it was tentatively concluded that similar bacterial communities might be developed in activated sludges, if WTPs treat similar wastewaters.

In this study, characteristics of wastewater treatment of sewage intermittently mixed with industrial wastewater is examined by investigating the operational status of each unit operation and measuring water quality. The bioreactor operating condition was measured for MLSS concentration 2,000~3,000 mg/L, HRT 5.3~16.3 hour, SRT 2.8~66.6 day, and SVI frequently showed the value above 200 which was higher than the optimal range of 50~150. It is thought that the sludge is not in suitable condition for sedimentation caused by the incoming industrial wastewater. When industrial wastewater is come into the system, MLDO inside of bioreactor rapidly increased, rate of nitrification is steeply decreased, and Pin floc. is spilled in the secondary clarifier. In the observance of microorganism showed that various bacterial floc. and ciliata were found as well as actinomycetes and filamentous bacteria(Sphaeotilus) which is known to cause bulking. Efficiency of each unit operation was fairly good in average. However, efficiency of the bioreactor treatment showed high fluctuation by unstable operating condition by intermittently incoming industrial wastewater.
The membrane-aerated biofilm reactor (MABR) is a promising municipal wastewater treatment process. In this study, two cross-flow MABRs were constructed to explore the carbon and nitrogen removal performance and bacterial succession, along with changes of influent loading shock comprising flow velocity, COD, and NH4-N concentrations. Redundancy analysis revealed that the function of high flow velocity was mainly embodied in facilitating contaminants diffusion and biosorption rather than the success of overall bacterial populations (p > 0.05). In contrast, the influent NH4-N concentration contributed most to the variance of reactor efficiency and community structure (p < 0.05). Pyrosequencing results showed that Anaerolineae, and Beta- and Alphaproteobacteria were the dominant groups in biofilms for COD and NH4-N removal. Among the identified genera, Nitrosomonas and Nitrospira were the main nitrifiers, and Hyphomicrobium, Hydrogenophaga, and Rhodobacter were the key denitrifiers. Meanwhile, principal component analysis indicated that bacterial shift in MABR was probably the combination of stochastic and deterministic processes.
To remove nitrate from wastewater, a novel bioelectrochemical denitrification system is introduced. In this proposed system, biological reactions are coupled with reactions on the electrode, whereby the electrons are transferred to the bacterial enzymes via a mediator as an electron carrier. The denitrification reaction was achieved with permeabilized Ochrobactrum anthropi SY509 containing denitrifying enzymes, such as nitrate reductase, nitrite reductase, and nitrous oxide reductase, and methyl viologen was used as the mediator. The electron transfer from the electrode to the enzymes in the bacterial cells was confirmed using cyclic voltammetry. A high removal efficiency of nitrate was achieved when the bioelectrochemical system was used with the permeabilized cells. Furthermore, when the permeabilized cells were immobilized to a graphite felt electrode using a calcium alginate matrix containing graphite powder, a high removal efficiency was achieved (4.38 nmol/min mg cell) that was comparable to the result when using the free permeabilized cells.
Ammonia-oxidizing bacteria (AOB) are chemolithoautotrophs that play a key role in nitrogen removal from advanced wastewater treatment processes. Various AOB species inhabit and their community compositions vary over time in the wastewater treatment bioreactors. In this study, a hypothesis that operational and environmental conditions affect both the community compositions and the diversity of AOB in the bioreactors was proposed. To verify the hypothesis, the clone libraries based on ammonia monooxygenase subunit A were constructed using activated sludge samples from aerobic bioreactors at the Pohang, the Palo Alto, the Nine Springs, and the Marshall wastewater treatment plants (WWTPs). In those bioreactors, AOB within the Nitrosomonas europaea, N. oligotropha, N.-like, and Nitrosospira lineages were commonly found, while AOB within the N. communis, N. marina, and N. cryotolerans lineages were rarely detected in the samples. The AOB community structures were different in the bioreactors: AOB within the N. oligotropha lineage were the major microorganisms in the Pohang, the Palo Alto, and the Marshall WWTPs, while AOB within the N. europaea lineage were dominant in the Nine Springs WWTP. The correlations between the AOB community compositions of the wastewater treatment bioreactors and their operational (HRT, SRT, and MLSS) and environmental conditions (temperature, pH, COD, $NH_3$, and $NO_3{^-}$) were evaluated using a multivariate statistical analysis called the Redundancy Analysis (RDA). As a result, COD and $NO_3{^-}$ concentrations in the bioreactors were the statistically significant variables influencing the AOB community structures in the wastewater treatment bioreactors.
For the purpose of development of non-toxic and biodegradable flocculant, chitosan complex was isolated from Ganoderma lucidum wastes. The isolated complex was identified as the expected chitosan-glucan complex by IR specta. The complex was extracted by treatment of 50% NaOH solution at 120$\circ$C for 5 hrs, namely optimal condition and solubilized with 2% acetic acid for fur-ther use as flocculant. Preliminary experiments showed that the solubilized complex had higher flocculation activity of 1.3 fold than commercial chitosan at 400 mg/l concentration in soybean curd wastewater. Also the solubilized complex removed 83% of MLSS and 60% of COD in the soybean curd wastewater treated by photosynthetic bacteria, 50% of turbidity and 21% of MLSS in sugar industry wastewater, and 90% of turbidity and 89% of MLSS in alcohol fermentation wastewater. Bacterial cell flocculation activities of the solubilized chitosan-glucan complex were 89% in Bacillus subtilis broth, 81% in Streptococcus lactis broth, and more than 90% in Escherichia coli broth after standing for 2 days. The results reveal that chitosan-glucan complex from Ganoderma lucidum wastes can substitute for commercial chitosan as non-toxic and biodegradable flocculant.
Ammonia-oxidizing bacteria (AOB) were enriched by repeating fed-batch cultivations in an AOB-selective medium of activated sludges from a domestic wastewater treatment plant. Enriched culture showed strong capabilities of ammonia oxidation [0.810 mg $NH_4^+$-N/mg mixed liquor suspended solids (MLSS)$\cdot$day] as well as $NO_x^-$-N production (0.617 mg $NO_x^-$-N/ mg MLSS$\cdot$day). Degree of enrichment was examined through fluorescent in situ hybridization (FISH) analyses using an AOB-specific Cy3-labeled oligonucleotide probe (NSOl90) and terminal-restriction fragment length polymorphism (T-RFLP) analyses. FISH analyses confirmed that the fraction of AOB among 4',6-diamidino-2-phenylindole (DAPI)-stained cells increased from about less than $0.001\%$ to approximately $42\%$ after enrichment of AOB, and T-RFLP analyses showed that bacterial community became simpler as enrichment was continued. When the enriched culture of AOB was added (150 mg/l as dry suspended solid) to the normal activated sludge (3,000 mg/l as dry suspended solid), nitrification efficiencies were improved from 0.020 mg $NO_x^-$-N/mg MLSS$\cdot$day to 0.041 mg $NO_x^-$-N/mg MLSS$\cdot$day in a synthetic wastewater and also from 0.0007 mg $NO_x^-$-N/mg MLSS$\cdot$day to 0.0918 mg $NO_x^-$-N/mg MLSS$\cdot$day in a real domestic wastewater. Therefore, it is expected that this enrichment method could be used for improving efficiency of nitrification in wastewater treatment plants.

The effect of recycling ratio and fixed media on nitrate and phosphate removal was investigated in a pilot-scale wastewater treatment unit using synthetic wastewater. Addition of fixed media increased nitrate removal from 45 to 58% while no noticeable change was observed for Chemical Oxygen Demand (COD) and phosphate removal (<5%). Nitrate removal efficiency also enhanced (Ca 7%) when the influent wastewater flow was doubled (2Q), however phosphate removal was decreased from 40.9 to 26.6% with the increasing recycling rate. The attached biomass analysis showed the presence of bacteria (73.4 $mg/cm^2$) on the surface of added media in anoxic reactor. Pseudomonas aeruginosa a common denitrifying bacterium dominated the bacterial growth (58%) in the anoxic reactor which was determined using Fluorescence In Situ Hybridization (FISH) analysis.
