육가공 작업장에서 발생되는 바이오에어로졸 저감을 위한 살균제 효능 실증 평가 (Efficacy Evaluation of Disinfectant for Reducing Bioaerosols Generated in a Meat Processing Workplace)

  • Objectives: This study aims to compare and evaluate the reduction efficiency of disinfectants used in the domestic meat processing industry to reduce bioaerosol exposure of meat industry workers and to use this as basic data for establishing work environment management measures. Methods: Thirteen disinfectants sold in South Korea were selected for evaluation and the bacterial reduction effect of the disinfectants was investigated. Bacterial suspension and surface disinfection tests were conducted to compare and analyze the antibacterial strength of the disinfectants. Pork carcasses, cutting boards, benches, and conveyor belts were selected for surface sterilization tests. Results: As a result of the bacterial suspension experiment test, all disinfectants had a bacterial reduction efficiency of more than 86%. Among them, the bacterial reduction efficiency of chlorine disinfectants was 99.93% on average. In the results of the pork carcass surface sterilization test, the rate of reduction of disinfectants made of quaternary ammonium compounds (QACs) was the highest. Tests of plastic cutting boards showed that chlorine disinfectants had the best sterilization effect. Experiments on stainless steel benches showed the best bacterial reduction efficiency for chlorine dioxide and QACs disinfectants. In the conveyor belt made of urethane, QACs disinfectants showed excellent sterilization effects. Conclusions: The study evaluated the disinfection power of disinfectants against bacteria occurring in domestic meat processing plants. All disinfectants were found to be effective in bacterial suspension experiments, and chlorine disinfectants were particularly effective. In surface sterilization experiments, sterilizing agents with QACs as the main ingredient were excellent.

  • 살균소독제의 살균소독력 평가를 위한 표준시험법을 확립하기 위하여 세균현탁액 시험법(bacterial suspension test)에 대해 연구하였다. 살균소독력은 G(-)인 E. coli ATCC 10536과 G(+)인 S. aureus ATCC 6538에 대하여 $20{\pm}1^{\circ}C$에서 $5min{\pm}10^{\circ}sec$간 반응하였을 때 초기근수를 5 log이상 감소시키고, 또한, 시험검증조건을 만족하는지를 검토하여 시험법을 확립하였다. 유통 중인 살균소독제 제품 36종에 대하여 확립된 시험법으로 살균소독력을 시험한 결과 모든 살균소독제는 제조자가 제시한 사용농도에서 살균소독력을 나타내었으며, 주요 식중독 균에 대한 살균효과도 관찰되었다. 확립된 시험법은 살균소독력을 측정하는 국가표준시험법으로서 살균소독제의 살균소독력 평가에 적합하였다.

수종의 역충전 재료의 치근단 밀폐력 비교 (COMPARISON OF THE SEALING ABILITY OF VARIOUS RETROGRADE FILLING MAIERIALS)

  • This study was performed to evaluate the sealing ability of various retrograde filling materials by using bacterial penetration and dye penetration test. One hundred and forty extracted human teeth with single, straight canals and mature apiece were collected and used for this study. All canals were instrumented using an engine driven Ni-Ti file (ProFile). After removing 3mm from the apex of tooth, a standardized 3mm root end cavity was prepared using an ultrasonic instrument. The 70 teeth were randomly divided into 7 groups : 6 groups for retrograde filling using Super-EBA, ZOE, Chelon-Silver, IRM, ZPC and amalgam. The 7th group was used as a negative control. Nail varnish was applied to all external root surfaces to the level of the reseated root ends to prevent lateral microleakages. The specimens were then sterilized in an ethylene oxide sterilizer for 24 hours. 2 mm of the reseated root was immersed in a culture chamber containing a Tripticase Soy Broth with a phenol red indicator. The coronal access of each specimen was inoculated every 72 hours with suspension of Proteus vulgaris. The culture media were observed every 24hours for color change indicating bacterial contamination. The specimens were observed for 4weeks. The remaining 70 teeth were submitted to a dye penetration test. The canals of all teeth were first sealed with AH26 and obturated using an Obtura II system. Root resection, root end preparation and retrograde filling was performed as above. All specimens were suspended in 2% methylene blue dye for 72 hours before being ion gitudinally split. The degree of dye penetration was then measured using a stereomicroscope at 10 magnification and evaluated. The results were as floows : 1. In the bacterial penetration, the degree of leakage was the lowest in the Super-EBA, followed by, in ascending order, ZOE, Chelon-Silver IRM and ZPC. The amalgam showed highest bacterial leakage of all(p<0.01). 2. In the dye penetration, the degree of microleakage was the lowest in the Chelon-Silver and Super-EBA, followed by, in ascending order, IRM, ZPC. The ZOE and amalgam showed the highest microleakage of all (p<0.05). These results suggested that the eugenol based cement, Super-EBA, have excellent sealing ability as a retrograde filling material.

Inhibition of Streptococcus mutans biofilm formation on composite resins containing ursolic acid

  • Objectives: To evaluate the inhibitory effect of ursolic acid (UA)-containing composites on Streptococcus mutans (S. mutans) biofilm. Materials and Methods: Composite resins with five different concentrations (0.04, 0.1, 0.2, 0.5, and 1.0 wt%) of UA (U6753, Sigma Aldrich) were prepared, and their flexural strengths were measured according to ISO 4049. To evaluate the effect of carbohydrate source on biofilm formation, either glucose or sucrose was used as a nutrient source, and to investigate the effect of saliva treatment, the specimen were treated with either unstimulated whole saliva or phosphate-buffered saline (PBS). For biofilm assay, composite disks were transferred to S. mutans suspension and incubated for 24 hr. Afterwards, the specimens were rinsed with PBS and sonicated. The colony forming units (CFU) of the disrupted biofilm cultures were enumerated. For growth inhibition test, the composites were placed on a polystyrene well cluster, and S. mutans suspension was inoculated. The optical density at 600 nm ($OD_{600}$) was recorded by Infinite F200 pro apparatus (TECAN). One-way ANOVA and two-way ANOVA followed by Bonferroni correction were used for the data analyses. Results: The flexural strength values did not show significant difference at any concentration (p > 0.01). In biofilm assay, the CFU score decreased as the concentration of UA increased. The influence of saliva pretreatment was conflicting. The sucrose groups exhibited higher CFU score than glucose group (p < 0.05). In bacterial growth inhibition test, all experimental groups containing UA resulted in complete inhibition. Conclusions: Within the limitations of the experiments, UA included in the composite showed inhibitory effect on S. mutans biofilm formation and growth.

배추 무름병에 대한 저항성 품종 검정 (Screening of Disease Resistance of Chinese Cabbage Cultivars and Lines to Bacterial Soft Rot)

  • Erwinia carotovora subsp. carotovora에 의한 배추 무름병은 배추에서 가장 큰 문제가 되고 있는 병중 하나이다. 본 연구는 배추 품종별 무름병 저항성을 조기에 검정하고자 실시하였다. 선발된 균주를 이용하여 세균현탁액과 mineral oil을 4:1로 혼합하여 배추의 중앙 기부에 10m1 관주 접종하는 mineral oil 접종법으로 접종하였다. 국내의 3개 종묘회사와 대학으로부터 분양 받은 43개의 배추품종 및 계통의 저항성 정도를 조사하였다. 그 결과 C3-26, C3-28, C3-29, C29-51-51-52는 저항성, 강타배추, 금촌얼갈이, 미니배추, 금빛, DB50, 지부, 평층, 산촌배추는 감수성인 것으로 나타났으며, 나머지는 중간정도의 저항성을 나타냈다.

  • The purpose of this study was to evaluate the usefulness of Ga-67 scintigram in predicting the disappearance of inflammatory activity in mandibular lesions of dogs. Inflammatory bony lesions were produced artificially by the injection of sclerosing solution and bacterial suspension through the distal root canals of the lower 4th premolars in 10 dogs. The serial Ga-67 scintigrams and periapical radiograms were obtained. After 3 to 6 weeks duration of antibiotic therapy, the experimental animals were killed, and the mandibles were dissected. Bacteriologic culture and antibiotic sensitivity test were done. 1. The following results were obtained. The negative Ga-67 scan has 100% predictive value, and is a strong evidence of the resolution of inflammatory activity. In 2 animals of positive Ga-67 scan, I animal had negative culture. In 14 cases of positive Ga-67 scan, radiographically there was increase in the size of lesion in 9 cases, no change in 5 cases. In 8 cases of negative Ga-67 scan, radiographically there was increase in the size of lesion in I case, no change in 6 cases and decrease in 1 case.

The role and characterization of .betha.-1, 3-glucanase in biocontrol of fusarium solani by pseudomonas stutzeri YPL-1

  • An antifungal Pseudomonas stutzeri YPL-1 produced extracellular chitinase and .betha.-1, 3-glucanase that were key enzymes in the decomposition of fungal hyphal walls. These lytic extracellular enzymes markedly inhibited mycelial growth of the phytopathogenic fungus Fusarium solani. A chitinase from P. stutzeri YPL-1 inhibited fungal mycelial growth by 87%, whereas a .betha.-1, 3-glucanase from the bacterium inhibited growth by 53%. Furthermore, co-operative action of the enzymes synergistically inhibited 95% of the fungal growth. The lytic enzymes caused absnormal swelling and retreating on the fungal hyphal walls in a dual cultures. Scanning electron microscopy clearly showed hyphal degradation of F. solani in the regions interacting with P. stutzeri YPL-1. In an in vivo pot test, P. stutzeri YPL-1 proved to have biocontrol ability as a powerful agent in controlling plant disease. Planting of kidney bean (Phaseolus vulgaris L.) seedlings with the bacterial suspension in F. solani-infested soil significantly suppressed the development of fusarial root-rot. The characteristics of a crude preparation of .betha.-1, 3-glucanase produced from P. stutzeri YPL-1 were investigated. The bacterium detected after 2 hr of incubation. The enzyme had optimum temperature and pH of 40.deg.C and pH 5.5, respectively. The enzyme was stable in the pH range of 4.5 to 7.0 and at temperatures below 40.deg.C, with a half-life of 40 min at 60.deg.C.

박과 작물에 과일썩음병을 일으키는 Acidovorax citrulli 검출을 위한 nested-PCR 검사법 개발 (Development of Nested-PCR Assay to Detect Acidovorax citrulli, a Causal Agent of Bacterial Fruit Blotch at Cucurbitaceae)

  • 박과 작물에서 과일썩음병(bacterial fruit blotch)을 일으키는 Acidovorax citrulli를 종자로부터 검출하기 위한 특이적이고 민감한 nested-PCR 방법을 개발하였다. 본 연구에서는 Next Generation Sequencing을 이용하여 draft genome sequencing을 얻은 후 이를 분석하여 PCR 프라이머를 디자인하였고, 이들 프라이머의 A. citrulli에 대한 특이성을 확인하여 Ac-ORF 21F/Ac-ORF 21R의 nested PCR 프라이머를 최종 선발하였다. Ac-ORF 21F/Ac-ORF 21R는 오직 A. citrulli에서만 특이적으로 140bp 크기의 DNA를 증폭하였으며, 그 검출민감도는 1차 PCR 검출한계(10 ng genomic DNA/PCR)보다 검출한계를 10,000배 증가시켰다. 개발된 nested-PCR 방법을 통해 병원균을 인공접종한 수박 종자의 외부검사에서 $10^1cfu/ml$까지 인공 접종 한 모든 종자 시료에서 병원균을 검출하였고, 병원균을 인공접종 한 수박 종자의 내부검사에서는 병원균이 검출되지 않았다. 자연 감염 수박 종자의 외부검사에서는 10개의 반복 시료 중 2개에서, 그리고 종자 내부검사에서는 10개의 반복 시료 중 5개에서 A. citrulli를 검출하였다. 본 연구에서 개발한 nested-PCR은 특이성과 민감도가 높고 인공접종과 자연감염 수박 종자에서도 병원균의 검출이 가능하여 박과 작물의 종자로부터 A. citrulli를 검출하는데 효과적으로 사용될 수 있을 것으로 생각된다.

Antimicrobial Efficacy of the Disinfectant Solution Nanoxil® Against Fish Pathogenic Bacteria

  • Fish pathogenic bacteria are a considerable danger of farmed fish and a source of economic loss in the fish farming industry. In this study, $Nanoxil^{(R)}$ was compared to hydrogen peroxide and a silver colloid in terms of disinfection efficacy against E. tarda, V. anguillarum and S. iniae. A bactericidal efficacy test conducted by a broth dilution method was used to determine the lowest effective dilution of the disinfectant following exposure to test bacteria for 30 min at $4^{\circ}C$. $Nanoxil^{(R)}$ and test bacteria were diluted with distilled water (DW), hard water (HW) or an organic matter suspension (OM) according to the treatment condition. Under the OM condition, the bactericidal activity of $Nanoxil^{(R)}$ against E. tarda exhibited a lowered efficacy compared to that under the DW and HW conditions. $Nanoxil^{(R)}$ at 500 fold (dilutions on) under all of the conditions demonstrated a high bactericidal efficacy against S. iniae. As $Nanoxil^{(R)}$ possess bactericidal efficacy against fish pathogenic bacteria such as E. tarda, V. anguillarum and S. iniae, this disinfectant solution can be used to limit the spread of fish bacterial diseases.

Antimicrobial Efficacies of Citra-Kill®, Disinfectant Solution against Salmonella Typhimurium and Brucella Ovis

  • Salmonellosis and brucellosis have caused a considerable danger of farmed animals and economic loss in animal farming industry. In this study, the disinfection efficacy of Citra-Kill$^{(R)}$, a commercial disinfectant, composed to quaternary ammonium chloride and citric acid was evaluated against S. typhimurium and Brucella ovis. A bactericidal efficacy test by broth dilution method was used to determine the lowest effective dilution of the disinfectant following exposure to test bacteria for 30 min at $4^{\circ}C$. Citra-Kill$^{(R)}$ and test bacteria were diluted with distilled water (DW), hard water (HW) or organic matter suspension (OM) according to treatment condition. On OM condition, the bactericidal activity of Citra-Kill$^{(R)}$ against S. typhimurium and Brucella ovis was lowered compared to that on HW condition. As Citra-Kill$^{(R)}$ possesses bactericidal efficacy against animal pathogenic bacteria such as S. typhimurium and Brucella ovis, this disinfectant solution can be used to control the spread of animal bacterial diseases.
