Postharvest decay of onion bulbs was examined by inspecting the commercial packages in the market or in storage. Bulb rot incidence was unexpectedly high, and onion bulbs with 1st quality grade were rotten most severely by 51%, followed by 32% for 2nd and 21% for 3rd grades. This indicates that larger bulbs had higher incidences of bulb rots. Major pathogens associated with basal and neck rots were Fusarium oxysporum and Aspergillus sp. or Botrytis allii, respectively, of which basal rot was most prevalent and damaging during storage. Among the epiphytic microorgani는 from onion plants, several Bacillus and Paenibacillus spp. and previously selected Pseudomonas putida and Trichoderma harzianum had inhibitory efficacy against bulb rot pathogens. Among these B. amyloliquefaciens BL-3, Paenibacillus polymyxa BL-4, and P. putida Cha 94 were highly inhibitory to conidial germination of F. oxysporum and B. allii. P. putida Cha 94, B. amyloliquefaciens BL-3, P. polymyxa BL-4, and T. harzianum TM were applied in the rhizoplane of onion at transplanting. Initially antagonist populations decreased rapidly during the first one month. However, among these antagonists, rhizoplane population densities of BL-3, Cha 94, and TM were consistently high thereafter, maintaining about 10$^4$-10$^{5}$ cells or spores per gram of onion root up to harvest time. The other bacterial antagonist BL-4 survived only for two months. TM was the most effective biocontrol agent against basal rot, with the number of rotten bulbs recorded at 4%, while that of the control was 16%. Cha 94 was effective for the first 20 days, but basal rot increased thereafter and had about the same control efficacy as that of BL-3 and BL-4. When the antagonists were applied to the topping areas of onion bulbs at harvest, TM was the most effective in protecting the stored onion bulbs from neck rotting. The second effective antagonist was BL-3. TM and BL-3 completely suppressed the neck rot in another test, suggesting that biocontrol of postharvest decay of onion using these microorganisms either at the time of transplanting or at harvesting may be promising.
Soil actinomycetes of 703 strains were isolated from 25 sampling points in Cheju Island using 4 different media. Arginine glycerol salts agar containing soil extract was found to be the best medium for the isolation of soil actinomycetes. Soil samples from pasture land showed higher population and diversity of the actinomycetes than those from citrus field, forest, island, hill or valley. When the antibacterial activity of the 526 isolates was tested against three bacterial strains, Escherichia coli, Staphylococcus aureus and Pseudomonas solanacearum the frequency of the isolates with antibacterial activity varied much depending upon the media used for isolation and cultivation. BL106Ba, one of the 10 isolates that showed antibacterial activity against all the above 3 test strains, was chosen based upon the pH and heat stability of its antibacterial metabolites, and was identified as Streptomyces sp. based upon its cultural, morphological and physiological characteristics. The partially purified white crystalline substance obtained from the culture supematant of BL1063a through cation exchange chromatography(AG MP-50) and three times consecutive gel filtration(Sephadex G-10) showed high antimicrobial activity against gram positive and negative bacteria, but low activity against yeasts. The partially purified substance was found to contain at least four different compounds with antibacterial activity by both thin layer chromatography and high performance liquid chromatography.
This review described solvent-tolerant lipases and their potential industrial, biotechnological and environmental impacts. Although organic solvent-tolerant lipase was first reported in organic solvent-tolerant bacterium, many organic solvent-tolerant lipases are in not only solvent-tolerant bacteria but also solvent-intolerant bacterial and fungal strains, such as the well-known Bacillus, Pseudomonas, Streptomyces and Aspergillus strains. As these lipases are not easily inactivated in organic solvents, there is no need to immobilize them in order to prevent an enzyme inactivation by solvents. Therefore, the solvent-tolerant lipases have the potential to be used in many biotechnological and biotransformation processes. With the solvent-tolerant lipases, a large number insoluble substrates become soluble, various chemical reactions that are initially impossible in water systems become practical, synthesis reactions (instead of hydrolysis) are possible, side reactions caused by water are suppressed, and the possibility of chemoselective, regioselective and enantioselective transformations in solvent and non-aqueous systems is increased. Furthermore, the recovery and reuse of enzymes is possible without immobilization, and the stabilities of the lipases improve in solvent and non-aqueous systems. Therefore, lipases with organic-solvent tolerances have attracted much attention in regards to applying them as biocatalysts to biotransformation processes using solvent and non-aqueous systems.

A comparative study was performed in gnotobiotic and greenhouse conditions to evaluate the effect of exogenous application of indole-3-acetic acid (IAA) and inoculation of Methylobacterium spp. possessing 1-aminocyclopropane-1-carboxylate deaminase (ACCD) and IAA activity on red pepperand tomato seedling growth and development. Application of 1.0 ${\mu}g\;mL^{-1}$ IAA positively influenced root growth while high concentrations (>10.0 ${\mu}g\;mL^{-1}$) suppressed root growth of red pepper and tomato under gnotobiotic condition. On the other hand, inoculation of Methylobacterium strains with ACCD activity and IAA or without IAA enhanced root growth in both plants. Similarly, under greenhouse condition the inoculation of Methylobacterium sp. with ACCD activity and IAA enhanced plant fitness recorded as average nodal length and specific leaf weight (SLW) but the effect is comparable with the application of low concentrations of IAA. Seedling length was significantly increased by Methylobacterium strains while total biomass was enhanced by Methylobacterium spp. and exogenous applications of < 10.0 ${\mu}g\;mL^{-1}$ IAA. High concentrations of IAA retard biomass accumulation in red pepper and tomato. These results confirm that bacterial strains with plant growth promoting characters such as IAA and ACCD have characteristic effects on different aspects of growth of red pepper and tomato seedlings which is comparable or better than exogenous applications of synthetic IAA.
Kimchi was fermented in permeability-controlled polyethylene containers, in glazed onggi (Korean ethnic earthenware) or glass bottles at $5^{\circ}C$ for 8 weeks. During 4 weeks of storage, kimchi fermented in the permeability-controlled container showed a stable fermentation pattern, in terms of changes in pH and acidity, compared with kimchi fermented in the other containers. With respect to changes in bacterial counts, kimchi fermented in polyethylene containers showed vigorous multiplication of lactic acid bacteria, especially Lactobacillus sp., but slow growth of total aerobic bacteria. The springiness of kimchi fermented in the polyethylene containers was optimal (about 10% more than that of glass bottle-fermented kimchi), and the overall acceptability and hardness of container-fermented kimchi were excellent upon sensory evaluation. The DPPH radical-scavenging activity of kimchi fermented in polyethylene containers was also greater (91%) than that of kimchi fermented in glazed onggi (73%) or glass bottles (63%). The $O_2$ and $CO_2$ permeabilities of the polyethylene containers were higher (458 and $357\;mmol\;h^{-1}\;m^{-2}\;atm^{-1}$, respectively) than were those of the other containers; the permeability ratio was 0.8. Glass bottles showed no permeance. The results indicate that permeability-controlled polyethylene containers may be used for kimchi fermentation.

The complex ecosystem of intestinal micro flora is estimated to harbor approximately 400 different microbial species, mostly bacteria. However, studies on bacterial colonization have mostly been based on culturing methods, which only detect a small fraction of the whole microbiotic ecosystem of the gut. To clarify the initial acquisition and subsequent colonization of bacteria in an infant within the few days after birth, phylogenetic analysis was performed using 16S rDNA sequences from the DNA iso-lated from feces on the 1st, 3rd, and 6th day. 16S rDNA libraries were constructed with the amplicons of PCR conditions at 30 cycles and $50^{\circ}C$ annealing temperature. Nine independent libraries were produced by the application of three sets of primers (set A, set B, and set C) combined with three fecal samples for day 1, day 3, and day 6 of life. Approximately 220 clones ($76.7\%$) of all 325 isolated clones were characterized as known species, while other 105 clones ($32.3\%$) were characterized as unknown species. The library clone with set A universal primers amplifying 350 bp displayed increased diversity by days. Thus, set A primers were better suited for this type of molecular ecological analysis. On the first day of the life of the infant, Enterobacter, Lactococcus lactis, Leuconostoc citreum, and Streptococcus mitis were present. The largest taxonomic group was L. lactis. On the third day of the life of the infant, Enterobacter, Enterococcus faecalis, Escherichia coli, S. mitis, and Streptococcus salivarius were present. On the sixth day of the life of the infant, Citrobacter, Clostridium difficile, Enterobacter sp., Enterobacter cloacae, and E. coli were present. The largest taxonomic group was E. coli. These results showed that microbiotic diversity changes very rapidly in the few days after birth, and the acquisition of unculturable bacteria expanded rapidly after the third day.

In this paper, we systematically investigated the influence of some selected ligands on the U-phosphate precipitation induced by soil bacteria. These organics are widely ranging from acetate, lactate, salicylate and citrate to oxalate. The results revealed that uranium could be biomineralized on bacteria as $UO_2HPO_4{\cdot}4H_2O$ or $(UO_2)_3(PO_4)_2{\cdot}4H_2O$. The influence of organic ligands on the biomineralization had clear-cut correlations with its complexation abilities to uranyl. It was clearly found that the U-phosphate biomineralization was affected noticeably by the strong ligands (oxalate and citrate). Further study discovered that when the organic ligands were uncompetitive with biotic $PO_4^{3-}$ for uranyl, the transformation of uranyl species from ${\beta}-UO_2(OH)_2$ colloidal particles to free $UO_2^{2+}$-ligands ions could facilitate the U-phosphate biomineralization. However, when the organic ligands competed with biotic $PO_4^{3-}$ for uranyl, the U-phosphate biomineralization were inhibited. Our results highlight the importance of complex interactions of strong organic ligands with uranyl during the bacterial precipitation of U-P compounds and thus for the mobilization and immobilization of radio-nuclides in the nature.
Asian dust storms originating in the arid desert of China and Mongolia usually occur from late winter through spring, and more than one million tons of dust per year is transported to the Korean Peninsula by the prevalent westerly winds. We supposed that these dust particles could include bioaerosols and act as carriers of microorganisms. In order to clarify the dynamics of microorganisms moving with these particles, the concentration and composition of microorganisms associated with settled particles were compared between samples collected during Asian dust events and those under non-dust periods. From February to April 2008, settled dust particles were collected at one location in Ulsan using rainfall meter of 200 mm diameter. During this period, there was one Asian dust event in Ulsan. The bacterial concentrations were higher in samples collected during Asian dust event than those under non-dust period, whereas fungal concentrations were rather similar regardless of the Asian dust event. We analyzed 16S rRNA gene sequences of 45 bacterial isolates obtained from the settled particle samples. These isolates belonged to either genus Bacillus or genus Streptococcus and were tentatively identified as B. amyloliquefaciens, B. aryabhattai, B. atrophaeus, B. licheniformis, B. megaterium, B. methylotrophicus, B. pumilus, B. sonorensis, B. subtlis, B. vallismortis, S. epidermidis, and S. succinus. In cases of fungal isolates, genera such as Mucor, Alternaria, Cladosporium, and Aspergillus were tentatively identified from samples collected at both Asian dust and non-Asian dust periods. It appears that endospore-forming bacteria such as Bacillus sp. rather than fungal spores are more likely to be associated with Asian dust particles.
Bioaerosols generated from composting facilities and landfills may create health risks for workers and nearby residents. To determine the levels of culturable airborne bacteria and fungi in bioaerosols, samples were seasonally collected at a composting facility and a landfill in Ulsan, Korea with an impaction-type sampler. Concentrations of heterotrophic bacteria averaged (in $MPN/m^3$) $6.5{\times}10^3$ (range $1.5{\times}10^2-1.5{\times}10^4$) in the composting facility and $3.9{\times}10^3$ (range $6.0{\times}10^1-9.3{\times}10^3$) at the entrance of the facility. These concentrations were 460 and 280 times higher than those of reference sites. Coliform bacteria were detected both inside and entrance of the facility. On the landfill, heterotrophic bacterial concentrations averaged (in $MPN/m^3$) $4.9{\times}10^2$ (range $1.7{\times}10^2-1.0{\times}10^3$), while they averaged $3.7{\times}10^2$ (range $4.8{\times}10^1-1.3{\times}10^3$) at the parking lot of the landfill. These concentrations were 35 and 26 times higher than those of reference sites. When we isolated and tentatively identified heterotrophic bacteria, Pseudomonas luteola was the most dominant species in bioaerosols from the composting facility, whereas the most abundant one in reference samples was Micrococcus sp. Average concentrations of airborne fungi were measured between $4.8{\times}10^2$ and $7.9{\times}10^2\;MPN/m^3$ depending on sites, which were 2.1-3.4 times higher compared to those of reference sites. While Cladosporium, Alternaria, and Penicillium were commonly identified fungal genera, genus Aspergillus was identified only in bioaerosols from the composting facility.
Vegetables including Chinese cabbage and radish have been grown in alpine areas such as Muju, Namweon, Jinan and Jangsu during the shortage period of vegetables. The incidence of various diseases, environmental factors such as temperatures and rainfalls, and aphid populations as virus vector were observed at 10-day intervals from July to September in those areas. Disease incidence showed no significant difference among locations. Major diseases in Chin ese cabbage were bacterial soft rot, white spot, downy mildew, mosaic virus, Alternaria leaf spot and Fusarium seedling blight. Major diseases in radish were virus, white rust, black rot, root rot, leaf spot and Fusarium seedling blight. Disease incidence reached peak on Aug. 20 with $27\%$ infection in radish and with $20\%$ infection in Chinese cabbage during the growing season, and declined thereafter. Percentage of infection in each growth stage showed $25\%$ at root thickening stage and $26\%$ at the harvest time in radish; and the head formation stage, $24\%$ at the harvest time. The data indicate that disease incidence in radish increased rapidly at late growing stage and progressively increased in Chinese cabbage Seedling blight caused by Fusarium sp. and root rot caused by Aphanomyces sp. were also observed in those areas. Cool and wet weather appeared to be favorable for disease incidences during the rainy period of growing season although average temperature was about $25^{\circ}C$. Populations of aphids were lower in the alpine vegetable growing area than that of flat areas. Aphids as virus vectors from total aphids collected were $73.5\%$ or 289 virus vectors /993 total aphids in Namweon and $18.1\%$ or 31 virus vectors/171 total aphids in Muju. The most prerevalent species of aphids was Myzus persicae Sulz.
