Occurrences of Major Diseases and Pests on 'Goldone', 'Redvita', 'Garmrok', New Cultivars of Kiwifruit (참다래 신품종 '골드원', '레드비타', '감록'의 주요 병해충 발생)

  • Kiwifruit has been cultivated in southern coastal regions of Korea since late 1970s. New cultivars have been successively released in recent years. In this study, we investigated major disease and pest incidences in new kiwifruit cultivars 'Goldone', 'Redvita' and 'Garmrok' at open field in Sacheon for 3 years and rain-proof field in Jeju for 2 years. For the bacterial canker, the 3 new cultivars showed more disease occurrence in Sacheon but rare in Jeju. For leaf spot disease, compared to disease incidence of 20% on 'Hayward' in Sacheon, cv. 'Garmrok' had high incidence about 60% but cv. 'Goldone' and cv. 'Redvita' had low incidence less than 20%. However, in Jeju, diseases incidences of all the new cultivars were lesser than 20%. In the case of Hemiptera, many Halyomorpha halys and Nezara antennata appeared in Sacheon but in Jeju Plautia stali was dominated. Isolated bacterial canker pathogen was identified as Pseudomonas syringae pv. actinidiae biovar 3. Leaf spots pathogens were Phomopsis sp., Phoma sp., Fusarium tricinctum and Alternaria alternata. This study shows the disease information on new kiwifruit cultivars and the adequate disease managements will be required.

Growth Promotion of Pavlova viridis by Bacteria Isolated from the Microalga (파블로바 비리디스로부터 분리한 세균에 의한 미세조류의 생장 촉진)

  • The marine microalga Pavlova viridis can grow fast and has the ability to accumulate essential nutrients for culturing marine animals, such as EPA and DHA, and it has been used as food for raring larval fish and prawn. The symbiotic relationship between the flagellate microalga Pavlova viridis and its associated bacteria was investigated. An axenic culture of P. viridis was obtained by repeated treatment of the microalga with an antibiotic cocktail. The axenic status was confirmed after sub-culturing three times in a sterile f/2 medium without an antibiotic. The axenic alga was then co-inoculated with five bacteria, arbitrarily designated as I1–I5, isolated from the alga to test the growth promotion of the algae. All bacterial strains promoted the growth of P. viridis, and bacterial isolate I3 was the most effective among the five bacteria tested. The cell number of P. viridis in the co-culture with I3 was significantly higher than that of the control culture. A sequence analysis of the 16S rRNA gene isolated from I3 revealed a 97% nucleotide sequence similarity to that of Citrobacter sp. The growth of strain I3 was also significantly enhanced by co-culturing with P. viridis, indicating a symbiotic relationship between the microalga and its associated bacterium. The association between the microalga and bacterium was confirmed by scanning electron microscopy.

Antifungal and Proteolytic Activity and Auxin Formation of Bacterial Strains Isolated from Highland Forest Soils of Halla Mountain (한라산 고지대 토양에서 분리한 미생물의 항균 및 단백질분해 활성, 오옥신 생산 특성)

  • Bacterial strains were isolated from forest soils of Halla mountain, Jeju island in Korea. The soil samples were collected at each altitude of 100m from 1,000 m above sea level. Total 398 strains were isolated and tested for their physiological characteristics of antagonistic and proteolytic activities, and auxin production. Among the isolates, 172 strains were selected as antifungal strains showing antagonistic activity against at least one of 8 plant fungal pathogens (Alternaria alternata, Botrytis cinerea, Collectotrichum acutatum, Fusarium oxysporum, Phytophthora capsici, Pythium ultimum and Sclerotinia sclerotiorum). In addition 203 strains for proteolytic activity and 26 strains for auxin production were characterized for further study. Je28-4 (Rhodococcus sp.) were showed 80% of control value against tomato gray mold in vivo. Thus, it is suggested that soil bacteria isolated from forest soils of Halla mountain can be important sources of bioactive compounds for improving plant growth or promising biocontrol agents.

Evaluation of Antimicrobial Activity and Disease Control Efficacy of Sodium Dichloroisocyanurate (NaDCC) Against Major Strawberry Diseases (딸기 주요 병원균에 대한 친환경제제 NaDCC의 항균활성 및 병 방제효과 평가)

  • Various diseases occur in fruits, leave and roots during strawberry cultivation and cause severe economical damage and huge amount of chemical fungicide use. Recently, as consumers' interest in safety of foods and organic agriculture produces have increased, control measures using alternatives for chemical fungicides have been newly developed in various ways. This study was conducted to test antifungal activity and control effect of sodium dichloroisocyanurate (NaDCC), using as disinfectants, against major disease pathogens of strawberry, Fusarium oxysporum (Fusarium wilt), Colletotrichum gloeosporioides (Anthracnose) and Phytophthora sp. (Phytophthora blight), and Xanthomonas fragariae (bacterial angular leaf spot) and evaluate availability as environment-friendly materials. When NaDCC was treated at the concentration range of 150 to 300 ppm, it suppressed significantly hyphal growth and reduced spore germination by more than 28%. In field condition, NaDCC showed excellent control effect (control value: 50%) against the bacterial angular leaf spot disease. Based on above-described results, we suggested that NaDCC can be used as alternative candidates to chemical pesticide alternatives of for controlling strawberry diseases.

Screening and Identification of Alkaline Protease Inhibitor-Producing Marine-derived Actinomyces. (Alkaline protease inhibitor를 생산하는 해양유래 방선균의 탐색 및 동정)

  • In this study, we screened and identified the bacterial strain showing high alkaline pretense inhibitor activity from marine environment. Nine bacterial strains with alkaline pretense inhibitor activity were isolated from marine sediments. Among them, strain C12 had the highest alkaline pretense inhibitor activity and was selected for further taxonomical study. On the basis of morphological and physiological characteristics, strain C12 was identified as the genus Streptomyces. A phylogenetic analysis of the 165 rDNA showed that the isolated strain was actually a member of the genus Streptomyces, because the determined sequence exhibited a higher homology with Streptomyces thermocarboxydus. Morphological characteristics showed cylindrical spore chain and smooth spore surface by scanning electron microscope. Strain C12 was grown on all media except for ISP 9 agar. This strain could be grown in the medium containing up to 9% NaCl.

Bacteriological Quality of Sea Water in Deukryang Bay, Korea (득량만 해수의 세균학적 수질)

  • A bacteriological study of sea water in Deukryang Bay was conducted to evaluate sanitary conditions of the bay and compliance of waters with the recommended bacteriological criteria for the designated area of shellfish cultivation. Sea water samples were collected at the established sampling stations (fig. 1) from May 1995 to November 1996. During the study period, coliform group, fecal coliform, classification of coliform group with IMViC reactions and pathogenic vibrios were analyzed. Coliform group and feral coliform MPN's were ranged from $<\;3.0\~4600/100m{\ell}\;and\;<\;30\~1,100/100m{\ell}$, respectively. The bacteriological criteria of sea water in shellfish growing area should be less than 70 per l00ml of sea water for median value of coliform MPN, and below $10\%$ of the samples which contain over than 230 for coliform MPN or over than 43 for fecal coliform MPN, Most of the waters from 26 sampling stations were complied water coliform criteria recommended for designated shellfish growing area. Then, the ratios of the samples with move than 230/10ml of coliform group MPN and more than 43/100ml of fecal coliform MPN were $7.4\%$ and $8.5\%$, respectively. The bacterial density of the sea water was deeply affected by rainfall amount. For example, coliform bacterial counts of sea watery after 48 hours from 93 mm rainfall were $6\~7$ times higher than those of without rainfall. During the study period, infectious bacteria such as Vibrio cholerae, Salmonella sp. an d Shigella sp. were not detected from the samples, but detection ratios of Vibrio parahaemolyticus and Vibrio vulnificus were $15\~20\%$ in summer months.

Involvement of Multiple Signaling Molecules in Peptidoglycan-induced Expression of Interleukin-1α in THP-1 Monocytes/Macrophages (THP-1 단핵구의 펩티도글리칸 유래 인터루킨-1 알파 발현에서 TLR2, PI3K/Akt/mTOR, MAPKs의 역할)

  • The expression of interleukin-1α (IL-1α) is elevated in monocytic cells, such as monocytes and macro-phages, within atherosclerotic arteries, yet the cellular molecules involved in cytokine upregulation remain unclear. Because peptidoglycan (PG), a major component of gram-positive bacterial cell walls, is detected within the inflammatory cell-rich regions of atheromatous plaques, it was investigated if PG contributes to IL-1α expression in monocytes/macrophages. Exposure of THP-1 monocytic cells to PG resulted in elevated levels of IL-1α gene transcripts and increased secretion of IL-1α protein. The transcription and secretion of IL-1α were abrogated by OxPAPC, an inhibitor of TLR2/4, but not by polymyxin B that inhibits lipopolysaccharide-induced TLR4 activation. To understand the molecular mechanisms of the inflammatory responses due to bacterial pathogen-associated molecular patterns (PAMPs) in diseased arteries, we attempted to determine the cellular factors involved in the PG-induced upregulation of IL-1α expression. Pharmacological inhibition of cell signaling pathways with LY294002 (a PI3K inhibitor), Akti IV (an inhibitor of Akt activation), rapamycin (an mTOR inhibitor), U0126 (a MEK inhibitor), SB202190 (a p38 MAPK inhibitor), SP6001250 (a JNK inhibitor), and DPI (a NOX inhibitor) also significantly attenuated the PG-mediated expression of IL-1α. These results suggest that PG induces the monocytic or macrophagic expression of IL-1α, thereby contributing to vascular inflammation, via multiple signaling molecules, including TLR2, PI3K/Akt/mTOR, and MAPKs.

Optimal Growth Conditions for the Two Euryhaline Cyanobacterial Clones, Anabaena sp. CB-MAL21 and CB-MAL22 Isolated from Mankyeong Estuary, Korea

  • As a result of the 2-year monthly monitoring of the phytoplankton community at 3 stations in Mankyeong Estuary, Korea, we learned that cyan bacterial species of the genus Anabaena occurred at most sampling points with huge salinity differences (0.1-32.5 psu). We isolated several clones of Anabaena spp. from the monitoring stations, and screen out two euryhaline and nitrogen-fixing Anabaena clones, CB-MAL21 and CB-MAL22. The two clones were grown under various environmental gradients such as temperature (20, 30, 35 and 40$^{\circ}C$), salinity (0, 2, 5, 15 and 30psu), and $PO_4^{3-}$-P concentration (0, 1.6, 8.0, 40 and 200 ${\mu}M$M). Growth of CB-MAL21 and CB-MAL22 was measured by daily monitoring of chlorophyll fluorescence from each experimental culture for more than three serial transfers. Both the two experimental clones did not grow at 0psu. Maximal growth rates of the two clones were markedly reduced at lower $PO_4^{3-}$-P concentrations showing negligible growth at 0 and 1.6 ${\mu}M$M. However, growth of CB-MAL21 was not affected by low $NO_3^--$ concentration in culture media, showing the nitrogen-fixing ability. Maximum biomass yields of the two clones decreased dramatically at 35 and 40$^{\circ}C$. Optimal growth conditions for the two experimental clones were determined to be 20-30$^{\circ}C$, 40 ${\mu}M$M $PO_4^{3-}$-P, and wide salinity range from 5.0 to over 30psu. Best growth of CB-MAL21 was shown at (20$^{\circ}C$-15psu), which is less saline and cooler condition than those (i.e., 30$^{\circ}C$-30psu) for the best growth of CB-MAL22. The euryhaline and nitrogen-fixing CB-MAL21 strain thus can be a candidate laboratory culture for the future cyan bacterial marine biotechnology in temperate coastal waters.

Assessment of Mycobacterial Viability by Fluorospectrophotometry (형광분광측정법에 의한 항산균의 생명력 평가)

  • Viable potential of Mycobacterium smegmatis, a slow grower in vitro cultivation and of M. leprae, an obligate intracellular parasitic bacterium, which can not be cultured yet in vitro was assessed by fluorospectrophotometry. Bacterial cells in different numbers and under various physiological status were incubater with fluorescein diacetate(FDA). After an incubation of the bacterial preparations with FDA at specified conditions, amount of fluorescein inside bacteria was measured by a fluorospectrophotometer at 470nm and 510nm of excitation and emission wavelengths, respectively. Fluorounit given by such bacteria showed a correlation with assessment of viability of the same preparations made by other methods, such as optical density and colony forming units of M. smegmatis and intracellular ATP content of M. leprae. The possible use of fluorospectrophotometry in assessing viability or physiological potential of bacteria, particularly intracellular parasites and fastidious organisms to culture in vitro is discussed in relation to other methods.

Antimicrobial Activity and Coloration of Environment-Friendly Biopolymer, Bacterial Cellulose (환경친화적 바이오폴리머인 세균 섬유소의 항균활성과 염색성)

  • In order to develop bacterial cellulose (BC) with antimicrobial activity against pathogenic microorganisms, silver and chitosan were incorporated into BC, respectively. Experiment results showed that antimicrobial activity against pathogenic microorganisms was improved with increasing silver concentration. Chitosan also showed a direct proportion between its concentration and antimicrobial activity. These results suggest that antimicrobial effects of BC using silver and chitosan are well proven to be effective. We also tested the stainability of BC with natural colorant for the application of food industry. Stainability of BC was enhanced with increasing natural colorant concentration. Decolorization of BC stained was observed by dipping it into distilled water with one hour-intervals. As a result, there was no significant difference. Combination of natural colorant-stainability and antibiosis of BC is expected to be useful in making colored antibiotic BC in various industrial application areas, considering its antimicrobial activity, high stainability and low decolorization tendency.
