생물고분자 생산 알칼리 내성 균주의 분리 및 특성 (Isolation and Characterization of Biopolymer Producing Alkali-Tolerant Bacterial Strain)

  • 미생물을 이용한 유용물질 생산연구의 일환으로 토양으로부터 생물고분자 생산균주를 분리하고, 이 분리균에 대한 균학적 성질 및 생성 생물고분자의 이화학적 성상을 검토하였다. 분리균주는 Bacillus속의 한 균종으로 동정 하였으며, 알칼리 내성균주의 특징을 나타내었다. 이 균주로부터 생성된 생물고분자는 성분분석, 정색반응 및 I.R. spectrum 분석결과 단백질 함량이 높고, 일부 amino sugar가 존재하나 uronic acid는 함유하지 않았다. 그러나 cetyl pyridinium chloride에는 침전되어 산성의 생물고분자인 것으로 추정되었다.

Bacterial Pathogens and Their Antimicrobial Susceptibility in Calves with Summer Pneumonia

  • Bovine respiratory disease (BRD) is one of the most important diseases in calves. It causes a huge economic loss in farms. BRD in calves is concentrated during winter because of the cold weather and lack of ventilation. However, BRD during summer in calves has continuously been a problem in farms. But there is no study about pathogens of summer pneumonia in calves and antimicrobial susceptibility in Korea. Therefore, aims of this study were to identify the pathogens and their antimicrobial susceptibility in calves with summer pneumonia. One hundred and one calves (2 weeks to 5 months after birth) with clinical sign of BRD from 5 farms were selected. After sampling by deep nasal swab, bacterial isolation and identification was conducted. Also, antimicrobial susceptibility test was performed. Pasteurella spp (49.4%), Staphylococcus spp (21.5%), Actinomyces spp (12.9%), E coli (10.7%), and Mannheimia haemolytica (5.3%) were isolated. The patterns of isolated pathogens from each farm were various. Also, the susceptibility of bacteria to antibiotics was showed a variety of patterns in each farm.

액비화 과정 중 인 이용 우수미생물 분리 및 특성 (Isolation and Characterization of Phosphorus Accumulating Microorganisms under Liquid Fertilization of Swine Slurry)

  • This study was conducted to investigate the bacterial capability to accumulate phosphorus during liquid composting process of pig slurry. Storage liquid compost and pig slurry were analyzed by using MALDI-TOF technique, which showed the colonies of Acinetobacter towneri and Bacillus licheniformis. In addition, bacterial colonies were isolated under high phosphoric acid conditions using X-phosphate MOPS medium with the addition of 2 mM $K_2HPO_4$. Microbial growth was observed in high and low phosphoric conditions due to the growth of bacterial diversities in the liquid fertilizer and slurry. The colonies isolated in the high phosphoric acid medium were uncultured bacterium clone and Acinetobacter sp. were identified by analysis of 16S rRNA gene sequences. Uncultured bacterium showed higher growth rate and excellent phosphorus ability then Acinetobacter sp.. In addition to Paenibacillus sp. AEY-1 isolated from pig slurry performed excellent phosphorus utilizing capability.

Eight unrecorded bacterial species isolated from soil and marine sediment in Korea

  • The Earth contains billions of microbial species, although the vast majority cannot be cultured in laboratories and are thus considered unidentified and uncharacterized. Extremophiles are microorganisms that thrive in extreme conditions, including temperature, salinity, and pH. Extremophilic microorganisms have provided important insights for biological, metabolic, and evolutionary studies. Between 2017 and 2019, as part of a comprehensive investigation to identify bacterial species in Korea, eight bacterial strains were isolated from marine and non-marine environments in Jeju Island. These strains were cultured under extreme salinity or pH conditions. Phylogenetic analysis using 16S ribosomal RNA(rRNA) gene sequencing indicated that all eight strains belonged to the phyla Gammaproteobacteria, Bacilli, and Alphaproteobacteria. Based on their high 16S rRNA gene sequence similarities(>98.7%) and the formation of strong monophyletic clades with their closest related species, all isolated strains were considered as an unrecorded strain, previously unidentified species. Gram stain reaction, culture conditions, colony and cell morphology, biochemical characteristics, isolation source, and National Institute of Biological Resources(NIBR) IDs are described in this article. The characterization of these unrecorded strains provides information on microorganisms living in Korea.

제주산 망아지의 Streptococcus equi subsp. zooepidemicus 감염에 의한 뇌막염 증례 (A case of meningitis caused by Streptococcus equi subsp. zooepidemicus infection in a foal in Jeju)

  • In November 2012, a 4-month-old foal with anorexia, neurologic signs with ataxia and death was submitted for necropsy to the diagnostic laboratory of Jeju Self-Governing Provincial Veterinary Research Institute. Grossly, yellowish-white fibrinous materials were seen on the ventral cerebrum, cerebellum, medulla oblongata and cervical spinal cord of the foal. Histologically, severe fibrino-purulent meningitis characterized by the infiltration of neutrophils and fibrionous exudates with bacterial colonies were presented in cerebrum, cerebellum, brain stem and cervical spinal cord. In the bacterial isolation using meningeal exudate, the small white colonies on blood agar plate with ${\beta}$-hemolysis were observed. Isolated bacteria were Gram-positive cocci and confirmed as Streptococcus (S.) equi subsp. zooepidemicus by VITEK 2 system. Based on the histopathological features and the bacterial test, this case was diagnosed as meningitis associated with S. equi subsp. zooepidemicus infection in a foal. In our best knowledge, this is the first report for fibrino-purulent meningitis caused by S. equi subsp. zooepidemicus infection in Jeju.

  • 미생물을 이용한 생분해성 고분자 생산연구의 일환으로 토양으로부터 고점성의 생물고분자 생산균주를 분리하고, 이 분리균에 대한 분류학적 성질 및 생성 생물고분자의 이화학적 성상을 검토, 규명하였다. 분리균주는 Alcaligenes속의 한 균종으로 동정하였으며, 호알카리성 균주의 특성을 나타내었다. CPC(cetylpridinium chloride)침전 및 acetone처리하여 정제한 산성 고분자 물질은 carboxyl기를 갖고 자외 흡수가 강한 glutamic acid만을 구성성분으로 하였고, glutamic acid는 $\gamma$-peptide 결합의 $\gamma$-polyglutamic acid로 존재하였다. 중화당량은 약 350으로 $\gamma$-PGA 2.7잔가당 1개의 음이온이 존재하였으며, 분자량은 약 $6.5{\times}10^5$Daltons이었다.

Plant-growth promoting traits of bacterial strains isolated from button mushroom (Agaricus bisporus) media

  • A diverse group of plant-growth promoting bacteria were isolated in button mushroom (Agaricus bisporus) media to investigate the plant-growth promoting traits of compounds including indole acetic acid (IAA), ammonia, 1-aminocyclopropane-1-carboxylic acid deaminase, siderophore, and hydrogen cyanide. Twenty-one bacterial strains showing positive effects for all the test traits were selected and classified to confirm bacterial diversity in the media habitat. Plant-growth promoting traits of the isolates were also assessed. All strains produced IAA ranging from 20 ㎍/mL to 250 ㎍/mL. Most of the isolates produced more than 80% siderophore. Four strains (Pantoea sp., PSB-08, Bacillus sp., PSB-13, Pseudomonas sp., PSB-17, and Enterobacter sp., PSB-21) showed outstanding performances for all the tested traits. In a bioassay of these four strains using mung bean plant, the best growth performances (23.16 cm, 22.98 cm, 2.27 g/plant, and 1.83 g/plant for shoot length, root length, shoot dry weight, and root dry weight, respectively) were obtained from the plants co-inoculated with Bacillus sp., PSB-13. The resultant data indicate that button mushroom media have got a diverse group of bacteria with plant growth promoting abilities. Thus, the media could be a good recycling resource for using to an effective bio-fertilizer.

A report on 38 unrecorded bacterial species in Korea in the class Gammaproteobacteria

  • During an investigation of indigenous prokaryotic species in the Republic of Korea, a total of 38 bacterial strains belonging to the class Gammaproteobacteria were isolated from diverse environments. Samples were collected from soil, seawater, sand, sedimentary soil, rabbit feces, rat intestines, marine wetland, and tidal flats. The strains were identified to the species level using the high 16S rRNA gene sequences and showed high similarity (>98.7%) with the closest bacterial species and formed a robust clade in the neighbor-joining phylogenetic tree; it was determined that each strain belonged to independent, predefined bacteria species within the class Gammaproteobacteria. The 38 strains of Gammaproteobacteria analyzed in this study have not been reported in the Republic of Korea. Therefore, this study describes 20 genera of 13 families in 8 orders: Aeromonadales, Alteromonadales, Cellvibrionales, Enterobacterales, Lysobacterales, Oceanospirillales, Pseudomonadales, and Vibrionales. For each species, we describe Gram reaction, strain ID, isolation source, colony and cell morphology, cultural, physiological, and basic biochemical characteristics.

Isolation of a Lipolytic and Proteolytic Bacillus licheniformis from Refinery Oily Sludge and Optimization of Culture Conditions for Production of the Enzymes

  • With the increasing demand for enzymes in industrial applications there is a growing need to easily produce industrially important microbial enzymes. This study was carried out to screen the indigenous refinery bacterial isolates for their production of two industrially important enzymes i.e. lipase and protease. A total of 15 bacterial strains were isolated using Soil Extract Agar media from the oil-contaminated environment and one was shown to produce high quality lipase and protease enzymes. The culture conditions (culture duration, temperature, source of nitrogen, carbon, and pH) were optimized to produce the optimum amount of both the lipase (37.6 ± 0.2 Uml-1) and the protease (41 ± 0.4 Uml-1) from this isolate. Productivity of both enzymes was shown to be maximized at pH 7.5 in a medium containing yeast extract and peptone as nitrogen sources and sucrose and galactose as carbon sources when incubated at 35 ± 1℃ for 48 h. Bacterial strain SAB06 was identified as Bacillus licheniformis (MT250345) based on biochemical, morphological, and molecular characteristics. Further studies are required to evaluate and optimize the purification and characterization of these enzymes before they can be recommended for industrial or environmental applications.

Isolation of Nine Bacteriophages Shown Effective against Erwinia amylovora in Korea

  • Erwinia amylovora is a devastating bacterial plant pathogen that infects Rosaceae including apple and pear and causes fire blight. Bacteriophages have been considered as a biological control agent for preventing bacterial infections of plants. In this study, nine bacteriophages (ΦFifi011, ΦFifi044, ΦFifi051, ΦFifi067, ΦFifi106, ΦFifi287, ΦFifi318, ΦFifi450, and ΦFifi451) were isolated from soil and water samples in seven orchards with fire blight in Korea. The genetic diversity of bacteriophage isolates was confirmed through restriction fragment length polymorphism pattern analysis. Host range of the nine phages was tested against 45 E. amylovora strains and 14 E. pyrifoliae strains and nine other bacterial strains. Among the nine phages, ΦFifi044 and ΦFifi451 infected and lysed E. amylovora only. And the remaining seven phages infected both E. amylovora and E. pyrifoliae. The results suggest that the isolated phages were different from each other and effective to control E. amylovora, providing a basis to develop biological agents and utilizing phage cocktails.
