실제 하수조건에서 조류-세균 복합군집의 생태적 상호작용 및 영양염류 제거 특성 규명 (Characterization of Algal-Bacterial Ecological Interaction and Nutrients Removal Under Municipal Wastewater Condition)

  • 하수를 이용해서 배양된 조류는 바이오디젤 생산에 유용한 자원이다. 그러나 실제 하수에서 조류의 영양염류 신진대사와 하수 세균과의 상호작용에 관한 연구는 미흡하다. 본 연구에서는 하수로 배양되는 대표적 조류균주인 Ankistrodesmus gracilis SAG 278-2에 의한 하수 내 질소, 인 제거 거동을 평가하였고, 조류와 상호작용하는 하수 내의 세균 군집을 분석하였다. 하수 슬러지 세균 군집과 비교하였을 때, 조류-세균 복합 군집은 하수 내보다 높은 영양염류 제거를 보였다. 16S rRNA 유전자 분석 결과, 조류-세균 군집에서 조류가 성장함에 따라 Unclassified Alcaligenaceae 세균이 선택적으로 우점됨을 알 수 있었고, 조류에 의해서 선택적으로 우점화된 하수세균은 자연 수질 환경에서 조류와 공생적으로 상호작용 하는 것으로 알려진 Alcaligenes faecalis subsp. 5659-H와 계통학적으로 가까운 것으로 밝혀졌다. 본 연구의 결과, 하수 내의 높은 영양염류 제거를 보이는 조류-세균 복합 군집에서의 조류의 성장 및 신진대사가 특정 세균의 분포에 영향을 주는 것을 알 수 있었다.

  • An in vitro experiment was carried out to investigate effects of solid associated (SAB) and liquid associated bacteria (LAB) and the type of incubation substrate on ruminal fermentation and gas production profiles. Bacterial fraction did not influence total numbers of bacteria. Gas production degradation parameters were significantly influenced by bacterial fraction and type of substrate (p<0.05). There was significant interaction between bacterial fraction and type of substrate in gas production (p<0.01). Total VFA concentration and acetic and propionic acid ratio were also influenced by bacterial fraction and type of substrate with little differences in individual VFA concentration.

Additive Main Effects and Multiplicative Interaction Analysis of Host-Pathogen Relationship in Rice-Bacterial Blight Pathosystem

  • Host-pathogen interaction in rice bacterial blight pathosystem was analyzed for a better understanding of their relationship and recognition of stable pathogenicity among the populations of Xanthomonas oryzae pv. oryzae. A total number of 52 bacterial strains isolated from diseased leaf samples collected from 12 rice growing states and one Union Territory of India, were inoculated on 16 rice varieties, each possessing known genes for resistance. Analysis of variance revealed that the host genotypes(G) accounted for largest(78.4%) proportion of the total sum of squares(SS), followed by 16.5% due to the pathogen isolates(I) and 5.1% due to the $I{\times}G$ interactions. Application of the Additive Main effects and Multiplicative Interaction(AMMI) model revealed that the first two interaction principal component axes(IPCA) accounted for 66.8% and 21.5% of the interaction SS, respectively. The biplot generated using the isolate and genotypic scores of the first two IPCAs revealed groups of host genotypes and pathogen isolates falling into four sectors. A group of five isolates with high virulence, high absolute IPCA-1 scores, moderate IPCA-2 scores, low AMMI stability index '$D_i$' values and minimal deviations from additive main effects displayed in AMMI biplot as well as response plot, were identified as possessing stable pathogenicity across 16 host genotypes. The largest group of 27 isolates with low virulence, small IPCA-1 as well as IPCA-2 scores, low $D_i$ values and minimal deviations from additive main effect predictions, possessed stable pathogenicity for low virulence. The AMMI analysis and biplot display facilitated in a better understanding of the host-pathogen interaction, adaptability of pathogen isolates to specific host genotypes, identification of isolates showing stable pathogenicity and most discriminating host genotypes, which could be useful in location specific breeding programs aiming at deployment of resistant host genotypes in bacterial blight disease control strategies.

  • Because of the importance of the type III protein-secretion system in bacteria-plant interaction, its function in bacterial pathogenesis of plants has been intensively studied. To identity bacterial proteins interacting with Xanthomonas hrp gene products that are involved in pathogenicity, we performed the glutathione-bead binding analysis of Xanthomonas lysates containing GST-tagged Hrp proteins. Analysis of glutathione-bead bound proteins by 1-DE and MALDI-TOF has demonstrated that Avr proteins, RecA, and several components of the type III secretion system interact with HrpB protein. This proteomic approach could provide a powerful tool in finding interaction partners of Hrp proteins whose roles in host-pathogen interaction need further studies.

In-Depth Characterization of Wastewater Bacterial Community in Response to Algal Growth Using Pyrosequencing

  • Microalgae have been regarded as a natural resource for sustainable materials and fuels, as well as for removal of nutrients and micropollutants from wastewater, and their interaction with bacteria in wastewater is a critical factor to consider because of the microbial diversity and complexity in a variety of wastewater conditions. Despite their importance, very little is known about the ecological interactions between algae and bacteria in a wastewater environment. In this study, we characterized the wastewater bacterial community in response to the growth of a Selenastrum gracile UTEX 325 population in a real municipal wastewater environment. The Roche 454 GS-FLX Titanium pyrosequencing technique was used for indepth analysis of amplicons of 16S rRNA genes from different conditions in each reactor, with and without the algal population. The algal growth reduced the bacterial diversity and affected the bacterial community structure in the wastewater. The following in-depth analysis of the deep-sequenced amplicons showed that the algal growth selectively stimulated Sphingobacteria class members, especially the Sediminibacterium genus population, in the municipal wastewater environment.

  • 주요 신재생에너지인 바이오에너지의 일환으로 조류를 이용한 바이오에너지 및 자원화 기술에 대한 관심이 높아지고 있다. 조류는 영양염류 제거 능력을 활용해서 하수와 같은 오폐수 내 난분해성오염물질과 영양염류 제거의 고도처리도 가능하다. 조류와 박테리아 간의 생태적인 상호작용이 조류를 활용한 하수처리 및 하수자원화에 중요한 역할을 함에도 불구하고, 실지 하수 조건에서 조류와 박테리아간의 생태학적인 상호작용에 관한 과학적인 정보가 부족하다. 본 연구에서는 하수에서 배양이 잘 되고, 지질함량이 높다고 알려진 국내 조류 종인 Ankistrodesmus gracilis SAG 278-2의 하수오염물질 제거 특성과 조류 주입에 따른 하수 박테리아 군집의 반응을 실지 하수 조건에서 연구하였다. 하수 박테리아의 수가 증가는 조류의 성장 속도를 감소시켰으나, 반면 조류의 성장은 박테리아의 생존 및 내성호흡 생분해 속도에는 영향을 주지 않았다. 조류가 주입된 하수에서 난분해성 유기물질 및 총질소의 제거 향상이 관찰되었다. 박테리아 16S rRNA 유전자 T-RFLP 분석에 따르면 조류의 주입은 시간에 따라 박테리아 군집에 영향을 주었다. 박테리아 16S rRNA 유전자 PCR 증폭, clone 및 염기서열 분석 결과, 하수 내 조류의 성장은 박테리아 군집 구성을 변화시키며, 조류와 함께 공동 성장 가능한 박테리아는 Sediminibacterium, Sphingobacterium, Mucilaginibacter 속에 속하는 개체로 판명되었다.

키토산의 양성자화가 세균막과의 상호작용에 미치는 효과 연구 (Molecular Dynamics Simulation Studies of the Effects of the Protonation State of Chitosan in Interactions with Bacterial Membranes)

  • The cationic biopolymer chitosan has several applications in medicine. Chitosan is the deacetylated derivative of chitin, the second most abundant naturally occurring polymer. Recent studies have investigated the relationship between chitosan and antibacterial activity. However, the molecular interactions and mechanisms have not been detailed. This study used molecular dynamics simulations to study interactions between chitosan and anionic bacterial membranes (POPE-POPG) and electrically neutral non-bacterial membranes (POPC). We calculated the free energy using umbrella sampling to compare the interactions between membranes and chitosan in different protonation states. Fully protonated chitosan interacted most strongly with the bacterial membranes, but weakly with non-bacterial membranes. These results suggest that electrostatic interactions are the main mechanism of the antibacterial activity of chitosan, and they provide insights into the design of novel antibacterial and antimicrobial agents.

Changes in Structural and Functional Responses of Bacterial Communities under Different Levels of Long-Term Compost Application in Paddy Soils

  • Soils amended for long-term with high levels of compost demonstrated greater abundance of bacterial members of the phylum Bacteroidetes whereas a decreasing trend in the relative abundance of phylum Acidobacteria was noted with increasing levels of compost. Metabolic profiles predicted by PICRUSt demonstrated differences in functional responses of the bacterial community according to the treatments. Soils amended with lower compost levels were characterized by abundance of genes encoding enzymes contributing to membrane transport and cell growth whereas genes encoding enzymes related to protein folding and transcription were enriched in soils amended with high levels of compost. Thus, the results of the current study provide extensive evidence of the influence of different compost levels on bacterial diversity and community structure in paddy soils.

마우스에 있어서 Dermatophilus 양 균에 의한 농양의 형태 병리 발생 (Morphopathogenesis o the abscess induced by Dermatophilus-like microorganism in mice)

  • NC strain mice were inoculated intraperitoneally with Dermatophoilus-like microorganism strain W254 isolated from the sulfur granules of the swine tonsils. The development of the resulting abscesses in NC strain mice was investigated at regular time intervals by means of histological, histochemical and electron microscopical methods. The results obtained were as follows ; 1. The inoculated bacteria have settled in the subserous site of the peritoneal organ and provoked cellular response to form neutrophil, macrophage and connective tissue layers at the time of one week after inoculation, and thereafter were found to have formed typical actinomycotic abscess two weeks after inoculation. 2. The persistant abscesses were appeared to reveal close interaction between higher activity of organism and tissue reaction and contained hyaloid bodies within or outside bacterial clumps. 3. The hyaline materials were appeared to be composed of protein rich in hydroxyphenyl group by histochemical examination. The materials were presumed to be produced by the interaction between bacterial activity and host cell reaction. 4. The bacteria in the abscesses showed the various developmental stages of coccoid body, mulberry-like body tuber-shaped body and filamentous body, and it was confirmed that the mesosomal development was related to septation of the bacterial cells.
