A report of 26 unrecorded bacterial species in Korea, belonging to the Bacteroidetes and Firmicutes

  • An outcome of the study to discover indigenous prokaryotic species in Korea, a total of 26 bacterial species assigned to the classes Bacteroidetes and Firmicutes were isolated from diverse environmental samples collected from soil, tidal flat, freshwater, seawater, wetland, plant roots, and fermented foods. From the high 16S rRNA gene sequence similarity (>99.0%) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that these 26 species have been described in Korea; therefore 14 strains for the order Flavobacteriales and two strains for the order Cytophagales were assigned to the class Bacteroidetes, and 8 strains for the order Bacillales and 4 strains for the order Lactobacillales were assigned to the class Firmicutes are reported for new bacterial species found in Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs are also described in the species description section.

  • Since Anton van Leeuwen­hoek first observed a surface-associated multicellular structure of bacterial cells in the 17th century, it has been shown to exhibit an ability to form a biofilm by numerous bacterial species. The biofilm formation is composed of distinct developmental stages, which include an attachment/adhesion of a single cell, a proliferation toward monolayered coverage, a propagation to aggregated microcolony, a maturation to 3-dimensional structure, and subsequently a local degradation. Investigation to identify the essential factors for bacterial biofilm formation has been performed via classical genetic approaches as well as recently developed technologies. The initial stage requires bacterial motility provided by a flagellum, and outermembrane components for surface signal interaction. Type IV-pilus and autoaggregation factors, e.g., type I-fimbriae or Ag43, are necessary to reach the stages of monolayer and micro colony. The mature biofilm is equipped with extracellular polymeric matrix and internal water-filled channels. This complex architecture can be achieved by differential expressions of several hundred genes, among which the most studied are the genes encoding exopolysaccharide biosyntheses and quorum-sensing regulatory components. The status of our knowledge for the biofilms found in humans and natural ecosystems is discussed in this minireview.

Description of 39 unrecorded bacterial species in Korea, belonging to the class Alphaproteobacteria

  • During an investigation of the biodiversity of bacterial species in Korea, we discovered many indigenous prokaryotic species. A total of 39 bacterial strains in the class Alphaproteobacteria were isolated from various environmental samples collected from marine organisms, sea water, fresh water, tap water, mud flats, activated sludge, mineral water, tidal flats, soil and decayed plants. From the high 16S rRNA gene sequence similarity (>98.7%) and formation of robust phylogenetic clades with the most closely related species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that any of these 39 Alphaproteobacteria species have been described in Korea. Specifically, 18 species in 11 genera in the order Sphingomonadales, 11 species in 10 genera in the order Rhizobiales, two species in two genera in the order Caulobacterales, six species in six genera in the order Rhodobacterales and two species in two genera in the order Rhodospirillales were found in Korea.   • Within a comprehensive, widescale investigation of indigenous prokaryotic species in Korea, 29 bacterial strains in the phylum Bacteroidetes were isolated from diverse environmental habitats that included soil, plant roots, natural caves, tidal flats, freshwater from lakes, and seawater. Based on their high 16S rRNA gene sequence similarities (>99.1%) and the formation of robust phylogenetic clades with the closest type species, each strain likely belonged to an independent and predefined bacterial species. There are no publications or official reports of the isolation of these 29 species in Korea. Our study provides strong evidence that seven species in three genera in the order Cytophagales, 15 species in 13 genera in the order Flavobacteriales and seven species in five genera in the order Sphingobacteriales, all within the phylum Bacteriodetes, are new reports of bacterial species in Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs are described in the species description section.

Systemic Induction of the Small Antibacterial Compound in the Leaf Exudate During Benzothiadiazole-elicited Systemic Acquired Resistance in Pepper

  • Plants protect themselves from diverse potential pathogens by induction of the immune systems such as systemic acquired resistance (SAR). Most bacterial plant pathogens thrive in the intercellular space (apoplast) of plant tissues and cause symptoms. The apoplastic leaf exudate (LE) is believed to contain nutrients to provide food resource for phytopathogenic bacteria to survive and to bring harmful phytocompounds to protect plants against bacterial pathogens. In this study, we employed the pepper-Xanthomonas axonopodis system to assess whether apoplastic fluid from LE in pepper affects the fitness of X. axonopodis during the induction of SAR. The LE was extracted from pepper leaves 7 days after soil drench-application of a chemical trigger, benzothiadiazole (BTH). Elicitation of plant immunity was confirmed by significant up-regulation of four genes, CaPR1, CaPR4, CaPR9, and CaCHI2, by BTH treatment. Bacterial fitness was evaluated by measuring growth rate during cultivation with LE from BTH- or water-treated leaves. LE from BTH-treatment significantly inhibited bacterial growth when compared to that from the water-treated control. The antibacterial activity of LE from BTH-treated samples was not affected by heating at $100^{\circ}C$ for 30 min. Although the antibacterial molecules were not precisely identified, the data suggest that small (less than 5 kDa), heat-stable compound(s) that are present in BTH-induced LE directly attenuate bacterial growth during the elicitation of plant immunity.

점안용 소염약침액의 안전성 및 유효성 평가 (The Experimental study of safety and efficacy in using Soyeom Pharmacopuncture solution as eyedrops)

  • Objective : This experimental study was performed to investigate the safety of Soyeom Pharmacopunture solution manufactured by extraction of alcohol and water. To identify the use of it as eyedrops, the eye irritation test of rabbits and antibacterial test of Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, and Candida albicans was performed. Methods : 1. The eye irritation test of this material was performed according to the Regulation of Korea Food & Drug Administration(2005. 10. 21, KFDA 2005-60). After Soyeom pharmacopuncture solution was administered in the left eye of the rabbits, eye irritation of the cornea, iris and conjunctiva was observed at 1, 2, 3, 4 & 7day. 2. After administering Soyeom Pharmacopuncture solution on bacterial species (Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis, MIC(Minimum Inhibition Concentration) and the size of inhibition zone were measured. Anti-bacterial potency was also measured using the size of inhibition zone. Results : 1. After Soyeom pharmacopuncture solution was administered in the left eye of the rabbits, it was found that none of nine rabbits have abnormal signs and weight changes. 2. After Soyeom pharmacopuncture solution was medicated in the left eye of the rabbits, no eye irritation of the cornea, iris and conjunctiva was observed at 1, 2, 3, 4 & 7day. 3. There was no response to MIC on bacterial species (Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) after Soyeom pharmacopuncture solution was medicated. Conclusions : The present study suggests that Soyeom pharmacopuncture solution is a non-toxic and non-irritant medicine, which does not cause eye irritation in rabbits, but dosen't have anti-bacterial effects on bacterial species which cause Keratitis. These study result recommends that more research on other herbal medicines of eye drop for Keratitis are required.

Effect of Gamma Irradiation on Shelf-life Extension and Sensory Characteristics of Dak-galbi (Marinated Diced Chicken) during Accelerated Storage

  • This study examined the effect of gamma irradiation on shelf-life extension and sensory characteristics of dak-galbi. Commercial dak-galbi sauce was gamma-irradiated at 0, 5, 10, 15, 20, 25, and 30 kGy. The dak-galbi sauce (200 g) was then added to diced chicken (800 g) for cooking, and the cooked dak-galbi samples in vacuum bags were stored at $35^{\circ}C$ for 5 d. Dak-galbi samples were analyzed on d 0, 1, 2, 3, 4, and 5 for microbial analysis (plate count agar), thiobarbituric acid (TBARS) and volatile basic nitrogen (VBN) measurements, and on d 0 for sensory evaluation. On d 0, total bacterial populations were below detection limit after dak-galbi marinated with the gamma-irradiated (${\geq}15\;kGy$) sauces were cooked, and the samples marinated with higher dose irradiated dak-galbi sauce had lower (p<0.05) bacterial populations during storage. TBARS values of the dak-galbi samples marinated with non-irradiated sauce were not different ($p{\geq}0.05$) with those marinated with irradiated sauces on d 0, and the TBARS values increased (p<0.05) during storage at $35^{\circ}C$, regardless of irradiation dose. In the VBN analysis, there was no difference ($p{\geq}0.05$) in VBN values among irradiation doses on d 0, but VBN values decreased (p<0.05) as irradiation dose increased during storage. Moreover, there were no significant differences ($p{\geq}0.05$) in sensory characteristics among irradiation doses. These results indicate that use of gamma irradiation on dak-galbi sauce may be useful in shelf-life extension without compromising the sensory characteristics of dak-galbi.

샐러드와 김밥의 Bacillus cereus 분석에 의한 통계적 검체채취 계획 수립 (Determination of Statistical Sampling Plans for Bacillus cereus in Salad and Kimbab)

  • 시중에 유통 중인 샐러드와 김밥을 각각 100종 수거하여 B. cereus 함량을 정량분석하였다. 샐러드는 불검출이 54종이었으며 10 CFU/g 이하가 8종, 100 CFU/g 이하가 25종, 1,000 CFU/g 이하가 11종이었으며 1,000 CFU/g 이상인 것이 2종이었다. 샐러드의 대한 모니터링 결과 평균은 1.18 log CFU/g 이었으며 표준편차는 0.71 log CFU/g이었다. 김밥에서는 100개의 검체중 20개의 검체에서 B. cereus가 검출되었으며 평균은 1.01 log CFU/g 이었으며 표준편차는 0.71 log CFU/g이었다. 모니터링 결과를 이용하여 통계적 검체채취 방법을 수립하고자 하였다. 검체채취 방법은 국제미생물위원회에서 제공하는 검체채취프로그램인 NEW sampleplan program을 이용하였으며 검체갯수를 고정한 후 미생물 기준인 m과 M, 그리고 허용한계값인 c 값을 변화하면서 산출되는 합격률이 0.95에 가장 근사한 값을 선정하였다. 그 결과 샐러드, 김밥에서의 가장 적합한 검체채취 기준은 3분법을 사용하는 것으로 n = 5, c = 0, m = 1,000, M = 10,000과 n = 5, c = 2, m = 100, M = 1,000의 기준이 설정되었다. 특히 후자에 서술된 기준은 현재 뉴질랜드에서 즉석섭취식품에서의 B. cereus에 대한 규격으로 사용하고 있어 국제적인 경향에도 부합함을 알 수 있었다.

Model for Estimating CO2 Concentration in Package Headspace of Microbiologically Perishable Food

  • Levels of carbon dioxide gas, a metabolite of microbial growth, have been reported to parallel the onset of microbial spoilage and may be used as a convenient index for a packaged food's shelf life. This study aimed to establish a kinetic model of $CO_2$ production from perishable food for the potential use for shelf life control in the food supply chain. Aerobic bacterial count and package $CO_2$ concentration were measured during the storage of seasoned pork meat at four temperatures (0, 5, 10 and $15^{\circ}C$), and their interrelationship was investigated to establish a mathematical model. The microbial growth at constant temperature was described by using model of Baranyi and Roberts. $CO_2$ production from the stored food could be explained by taking care of its yield and maintenance factors linked to the microbial growth. By establishing the temperature dependence of the microbial growth and $CO_2$ yield factor, $CO_2$ partial pressure or concentration in package headspace could be estimated to a limited extent, which is helpful for controlling the shelf life under constant and dynamic temperature conditions. Application and efficacy of the model needs to be improved with further refinement in the model.

천연소독제 Clean Natural의 Salmonella typhimurium에 대한 복귀돌연변이시험 (Bacterial Reverse Mutation Test of Clean Natural using Salmonella typhimurium)

  • Clean Watural은 주성분이 참나무로부터 추출정제한 목초액과 프로폴리스로 구성된 새로운 천연 살균소독제이다. Salmonella typhimurium TA98, TA100, TA102, TA1535 및 TA1537 등의 5종의 균주를 이용한 복귀돌연변이성 시험을 수행한 결과, 모든 균주에서 대조군과 비교하여 시험용량군 사이에 돌연변이 유발성이 관찰되지 않았다. 그러나, 앙성대조물질에서는 대조군과 비교하여 통계학적으로 유의한 균주의 증가가 관찰되었다. 따라서, 시험물질인 천연 살균소독제, Clean Natulat은 돌연변이 유발 가능성이 없는 것으로 평가되었다.
