Lectin Activity and Chemical Characteristics of Escherichia coli, Lactobacillus spp. and Bifidobacterium spp. from Gastrointestinal Mucosa of Growing Pigs

  • Lectin activities and chemical characteristics of Escherichia coli, Lactobacillus spp. and Bifidobacterium spp. originating from the porcine cecal mucosal layer were studied based on hemagglutination assay (HA) and hemagglutination inhibition assay (HIA). Although all the bacterial strains were able to agglutinate erythrocytes of porcine or rabbit origin, much higher HA titers were consistently observed for Lactobacillus spp. than for E. coli or for Bifidobacterium spp. A remarkable reduction in HA titers occurred by the treatment of E. coli and Lactobacillus spp. with protease or trypsin and of Bifidobacterium spp. with protease, trypsin or periodate. There were no significant effects on the HA titers of the three groups of bacteria after the treatment with lipase. Hemagglutination of E. coli was strongly inhibited by D (+)-mannose and D (+)-galactose; Lactobacillus spp. by $\alpha$-L-rhamnose and methyl-$\beta$-galactopyranoside; Bifidobacterium spp. by D (+)-alactose, $\alpha$-L-rhamnose, $\alpha$-L-fucose, L (+)-arabinose, D (+)-mannose, D (-)-fructose at a relatively low concentration (1.43 to 3.75 mg/ml). These results, combined with the enhanced HA activities of the three bacterial strains by modification of rabbit erythrocytes with neuraminidase and abolished HA activity of E. coli after treatment with $\beta$-galactosidase, indicate that it might be the glycoproteinous substances surrounding the surface of the bacterial cells that are responsible for the adhesions of these microorganisms by recognizing the specific receptors on the red blood cell.

  • Human milk is frequently the only food source for a newborn during the initial stage of life after birth. Milk provides not only the nutrients necessary for the infant's growth, but also ingredients that may enable the infant to thrive. Human milk oligosaccharides (HMO) are considered to be these beneficial ingredients for the health of infant. It has been reported that around 5 to 10 g unbound oligosaccharides and around 20 to over 130 different HMO are present in 1L of human milk. The suggested health mechanisms of HMO's roles in host defense are 1) blocking bacterial adhesions, 2) binding to a toxin receptor on the extracellular domain, and 3) postbiotic effect resulting from the increase of probiotics such as Bifidobacteria and Lactobacilli. Among the prebiotic oligosaccharides, mixtures of long chain fuetooligosaccharides (10%) and galactooligosaccharides (90%) in infant formula are demonstrated to increase the number of Bifidobacteria and Lactobacilli to the levels seen in human milk fed infants.

  • Effect of the roasted barley tea in commercial markets on the adherence to the saliva-coated hydroxyapatite(HA) beads and the cell surface hydrophobicity of Streptococcus mutans and Streptococcus sobrinus as cariogenic microorganism was examined in vitro. Adherence activity and hydrophobicity in bacteria tested in all the barley tea samples decreased and the values were different according to the type of tea and the type of treatment. The inhibition of bacterial adsorption to HA beads suggest that barley tea active molecules as catechins and melanoidins may adsorb to a host surface, preventing the tooth receptor from interacting with any bacterial adhesions. The obtained results showed that the barley tea may inhibit bacterial adherence, the first step of the pathogenesis of dental caries in which these microorganism are involved.

Necrotic Proctitis and Escherichia coli Septicemia in a Bottlenose Dolphin Tursiops truncatus (큰돌고래(Tursiops truncatus)에서 괴사성 직장항문염과 대장균 패혈증)

  • We report a case of Escherichia coli septicemia in a 6-year-old male bottlenose dolphin (Tursiops truncatus). Gross lesions included turbid reddish yellow ascites, fibrous adhesions of rectum and peritoneum, multifocal mucosal ulcers of rectum, and systemic petechiae. Multifocal necrosis with bacterial colonies was observed histologically in mucosal membrane of rectum and anus, and also in caudal mesenteric lymph node, inguinal lymph node, tracheobronchial lymph node, tonsil, spleen, liver, and lung. E. coli was isolated in pure culture from multiple organs including blood, spleen, mesenteric lymph node, liver, lung, and ascites. The E. coli was serotype O25. This case was diagnosed as a septicemia caused by E. coli serotype O25 associated with proctitis.

Generation of a monoclonal antibody against AgI/II, a cellular surface protein of Streptococcus mutans GS5 (Streptococcus mutans GS5의 세포막 단백질 Ag I/II에 대한 단항체의 생산)

  • Most of oral streptococci express the Antigen I/II (AgI/II) proteins, cell wall anchored adhesions. AgI/II protein binds to salivary agglutinin glycoprotein, a component of tooth pellicle and to ligands in other bacteria. These associations play important roles in bacterial colonization. Recently, it was reported that diverse host molecules also interact with AgI/II protein and that these interactions induce inflammatory responses from host cells. Among mutans streptococci containing -type hemolytic activity, Streptococcus mutans is a causative agent for dental caries. Compared with many other strains of S. mutans, GS-5 strain is unique in that this bacterium expresses truncated secretory AgI/II protein due to the nonsense mutation in the agI/II gene. This indicates that S. mutans GS-5 has a different clinical role and a recent report supported this idea based on the results from clinically isolated S. mutans strains. Previously, we had cloned agI/II gene from S. mutans GS-5 and generated recombinant N-terminal AgI/II protein. In this study, we further produced a hybridoma line expressing anti-AgI/II monoclonal antibodies named as 1C11A. This antibody showed high sensitivity to AgI/II protein in Western blot and ELISA. This new reagent will provide a basis for investigating the mechanisms of AgI/II-related diseases.

Effect of fabrication method and surface polishing on the surface roughness and microbial adhesion of provisional restoration (임시 수복물의 제작 방식과 표면 연마가 표면 거칠기와 세균 부착에 미치는 영향)

  • Purpose: This study aims to investigate the effects of provisional restoration fabrication methods and surface polishing on surface roughness and microbial adhesion through in vitro experiments. Materials and Methods: 120 cylindrical provisional restoration resin blocks (10 × 10 × 2.5 mm) were manufactured according to four fabrication methods, and 30 specimens were assigned to each group. Afterwards, they were divided into non-polishing group, #400 grit SiC polishing group, and #800 grit SiC polishing group and polished to a 10 × 10 × 2 mm specimen size (n = 10). The surface roughness Ra and Ry of the specimen was measured using a Surface Roughness Tester. Three specimens were extracted from each group and were coated with artificial saliva, and then Streptococcus mutans were cultured on the specimens at 37℃ for 4 hours. The cultured specimens were fixed to fixatives and photographed using a scanning electron microscope. For statistical analysis, the two way of ANOVA was performed for surface roughness Ra and Ry, respectively, and the surface roughness was tested post-mortem with a Scheffe test. Results: The fabrication method and the degree of surface polishing of the provisional restorations had a significant effect on both surface roughness Ra and Ry, and had an interaction effect. There was no significant difference in Ra and Ry values in all polishing groups in DLP and LCD groups. Conclusion: The fabrication method and surface polishing of the provisional restoration had a significant effect on surface roughness and showed different adhesion patterns for S. mutans adhesion.
