김치에 서식하는 Gram 양성세균의 분리 및 동정의 재평가 (Reevaluation of Isolation and Identification of Gram-positive Bacteria in Kimchi)

  • 김치발효 중 Gram 양성세균 또는 유산균의 분리와 동정을 시도하였다. 종의 다양성은 분리배지와 온도에 영양을 받았고, 다양성은 온도가 낮아질수록 감소하는 경향이었다. MRS는 세균의 분리에 KM(김치재료로 만든 자연배지)은 종의 수를 파악 하는데 각각 적합하였다. 분리균의 동정은 Bergey's manual of Systematic Bacterio]ogy (1986)를 기초로 하여 작성한 이 분농생표(dichotomous Identification scheme)에 의하여 선행하였다. 각 온도(5, 15, $25^{\circ}C$)에서 동정된 Gram 양성세균은 Leuconostoc 5종, Streptococcus 4종, Pediococcus 3종, Bacillus 2종 그리고 Ltobacillms 18종이였다. 각 온도에서 출현 빈도가 높은 종은 $25^{\circ}C$에서 LactobaIlus ptantanmz, Streptococcµs faffinolactis, Leuconostoc maιnteroid, subsp mlsentιroides 이었고, $15^{\circ}C$에서 L. mesenteroides Lactobacillus fructosus, L. maen teroid,l.I subsp. mesent,roid, 이였고, $5^{\circ}C$ 에서 Leuconostoc sp.(65.2 빈도)에 의하여 이루어졌다. 그리고 각 온도에 따른 김치발효 중 지금까지 알려진 Pediococcus cerevisiae 와 Streptococcus faecalis는 분리되지 않았다.

Analysis of Vaginal Lactic Acid Producing Bacteria in Healthy Women

  • Vaginal lactic acid-producing bacteria of 80 pre-menopausal women were studied by isolation on Blood and DeMan-Rogosa-Sharpe agar, PCR with group-specific primers for Lactobacillus-denaturing gradient gel electrophoresis (DGGE), and PCR with specific primers for V3 region in 16S rRNA-temporal temperature gel electrophoresis (TTGE). Conventional isolation method on media detected only one lactobacillus (Lactobacillus brevis) while TTGE detected only Lactobacillus sp. DGGE detected seven Lactobacillus species; L. coleohominis, L. crispatus, L. iners, L. reuteri, L. rhamnosus, L. vaginalis, and Leuconostoc lactis. L. acidophilus and L. gasseri, which are prevalent in Western women, were not detected in Korean women. Furthermore, L. rhamnosus, Leuc. lactis, L. coleohominis, and Weissella cibaria, which were not previously reported in the vaginal microbiota of Korean women, were detected. The five most prevalent LABs in vaginal microbiota in Korean women were L. iners, Enterococcus faecalis, L. crispatus, Leuc. lactis, and W. cibaria.

Isolation of Antibiotic-producing Bacteria Antagonistic to Fusarium oxysporum from Sesame-growing Soils and Evaluation of Their Antifungal Activity

  • For isolation of antibiotic-producing bacteria antagonistic to Fusarium oxysporum, a total of 327 microorganisms were screened from sesame-growing soils collected at various locations in Korea by the modified Herr's triple-agar-Iayer technique. Among the 36 bacterial isolates further screened by the dual culture test on tryptic soy agar, 10 were tested to show their antagonistic activity against 14 plant pathogenic fungi. Bacterial culture filtrates were shown either to inhibit some phytopathogenic fungal growth or to suppress F. oxysporum infection of sesame plants maintained in the green house. An isolate, B23, with the most prominent antagonistic activity was identified as Bacillus subtilis.

강원 동해안지역 도축돈의 전염성 관절염에 관한 조사 (A Survey on Prevalence & Incidence of the Infectious Arthritis on Slaughter Pigs in Eastern Kangweon-do)

  • This study was conducted to investigate the infectious arthritis on pigs slaughter in eastern Kangweon. During the period of between Feb. and Nov. 1992, 459 pigs were sampled at six abattoirs. Bacterial isolation and identification were perfomed from the arthritic lesions. In addition, drug susceptibility for the major isolated microoganisms were examined. The results obtained were summarized as follows : 1. From arthritic lesions bacteria were most frequently isolated, isolation rate being 21.1%. 2. The bacteria isolated from arthritic lesion, Staphylococcus spp. (33.9%), Coli form(39.3%) were most frequently isolated, while the genera of Mycoplasma were detected less frequently. 3. Antimicrobial drug susceptibility of the major organisms showed that all the isolated bacteria were susceptible to SXT(91.0%), cephalothin(82.5%), but resisted to penicillin and streptomycin.

  • 혐기성 광조건의 낮은 농도의 NH4+ 존재하에서 포도당으로부터 많은 양의 수소를 생성하는 홍색 비유황 광합성 세균을 수계혐기층으로부터 분리하였다. 이 세균은 형태적, 배양적, 생리적 특성에 따라 Rhodobacter sphaeroides KS 56으로 동정되었다.

Isolation and Characterization of Biofouling Bacteria in Ultra-high Purity Water Used in the Semiconductor Manufacturing Process

  • Bacteria were isolated and identified from an advanced high-purity water system that supplies ultra-high purity water (UHPW) for 16-megabyte DRAM semiconductor manufacturing. Scanning electron microscopic and microbiological observations revealed that the primary source of the bacteria isolated from the UHPW was detached cells from biofilms developed on the pipe wall through which the UHPW, a man-made and extremely nutrient poor environment, was passing. About 63-65% of the bacteria isolated from the UHPW and the pipe wall were Gram-positive, whereas only 10% of the bacteria isolated from the feed water were Gram-positive. The of Gram-positive bacteria and seven genera of Gram-negative bacteria. Strains of the UHPW bacteria effectively adhered to and formed a biofilm on the surface of polyvinyl chloride (PVC) pipe.

주요 식중독균 분리용 선택배지의 타당성 연구 (Evaluation of Selective Media for Isolation of Foodborne Bacteria)

  • 본 연구는 현행 식품공전상 제시되어 있는 식중독균 분리배지의 평가를 위해 Salmonella spp., Bacillus cereus, Listeria monocytogenes, Vibrio parahaemolyticus, Staphylococcus aureus의 5가지 식중독세균용 29가지 분리 선택배지를 Broth와 Food에서 시험하였다. Food는 닭고기, 쌀, 돼지고기, 고등어를 대상으로 5가지 식중독균을 접종후 균주의 recovery를 확인하였고. Broth는 5가지 식중독세균용 29가지 분리 선택배지를 사용하였다. Broth에서는 각 세정균에 대한 분리배지들 간에 유의적인 차이가 없었으나(P<0.05), Food에서는 그 차가 크지는 않았으나, 분리선택배지간의 분리능에 통계적 유의차가 존재하였다. 식품공전에 등재된 전통적인 분리 선택배지에서 분리된 균수가 Chromogenic 배지 등 새로운 선택배지에서 분리된 균수보다 많았다. 국제적으로 등재된 배지간의 선택성의 차이가 존재하였으며, 추후 다양한 배지가 식품공전에 등재되기 위해서는 추가 연구가 필요하다.

가축 소화기 병원성 세균을 저해하는 유산균의 분리 및 동정 (Isolation and Identification of Lactic Acid Bacteria Inhibiting Gastro-intestinal Pathogenic Bacteria of Domestic Animal.)

  • 토종닭 소장으로부터 가축 소화기 병원성 유해세균의 분리를 위하여 무작위 선별법과 중층-도말법을 사용하여 총130주의 유산균을 1차 분리하였다. 1차 분리한 130주를 paper disc법을 사용하여 최종적으로 살모넬라 균, 포도상구균, 대장균에 대하여 저해능이 우수한 7주를 분리하였다. 분리 균주의 동정을 위하여 1차로 API CHL kit를 사용하였고, 동정의 정확성을 높이기 위하여 2차로 16S rRNA sequencing 방법을 사용하였다. 그 결과 BD14 균주는 Pediococcus pentosaceus로 동정되었으며 나머지 6주는 Lactobacillus sp.로 동정되어 모두 안전성 있는 유산균임을 확인하였다. 여러 병원균에 가장 항균력이 큰 유산균은 L. pentosus K34이었으며, 이 유산균은 염산과 담즙산에 대한 내성에서도 가장 우수한 균주로 나타났다. 10종의 항생제에 대한 감수성을 조사한 결과 ciprofloxacin에 대하여 모든 균주가 내성을 보였고, BL 균주를 제외하고 모든 균주가 colistin, streptomycin에 대하여 내성을 보였다. 또한 BD14, BD16, K34 균주가 gentamicin에 대한 내성을 보였다.

Composition and Diversity of Gut Bacteria Associated with the Eri Silk Moth, Samia ricini, (Lepidoptera: Saturniidae) as Revealed by Culture-Dependent and Metagenomics Analysis

  • The polyphagous eri silk moth, Samia ricini, is associated with various symbiotic gut bacteria believed to provide several benefits to the host. The larvae of S. ricini were subjected to isolation of gut bacteria using culture-dependent 16S rRNA generic characterization, metagenomics analysis and qualitative enzymatic assays. Sixty culturable aerobic gut bacterial isolates comprising Firmicutes (54%) and Proteobacteria (46%); and twelve culturable facultative anaerobic bacteria comprising Proteobacteria (92%) and Firmicutes (8%) were identified inhabiting the gut of S. ricini. The results of metagenomics analysis revealed the presence of a diverse community of both culturable and un-culturable gut bacteria belonging to Proteobacteria (60%) and Firmicutes (20%) associated with seven orders. An analysis of the results of culturable isolation indicates that these bacterial isolates inhabited all the three compartments of the gut. Investigation on persistence of bacteria coupled with metagenomics analysis of the fifth instar suggested that bacteria persist in the gut across the different instar stages. In addition, enzymatic assays indicated that 48 and 75% of culturable aerobic, and 75% of anaerobic gut bacterial isolates had cellulolytic, lipolytic and nitrate reductase activities, thus suggesting that they may be involved in food digestion and nutritional provision to the host. These bacterial isolates may be good sources for profiling novel genes and biomolecules for biotechnological application.

구취를 유발하는 혐기성 세균의 증식을 억제하는 유산 간균의 분리 및 동정 (Isolation and identification of Lactobacillus inhibiting the production of halitosis by anaerobic bacteria)

  • There are normal inhabitants doing medically useful functions in the body. There are many kinds of bacteria performing specific functions in the oral cavity. Two strains of lactic acid bacteria were isolated from normal inhabitants of children 's oral cavity, which inhibited the the production of halitosis by anaerobic bacteria. The authors identified the isolates by the lest using API 50 CHL medium kit. 1. Two isolates were Gram-positive bacilli and produced hydrogen peroxide. 2. The optical density was 1.286 in the supernatant of Fusobacterium nucleatum after vortexing for 30 minutes, whereas in the supernatant of combined Fusobacterium nucleatum and each isolate, they were reduced to 0.628 and 0.497, which the percentages of coaggregation between them were 29.4% and 57.8%, respectively. 3. The optical density of Fusobacterium nucleatum precipitate was 1.794 in the culture media containing cysteine and $FeSO_4$, being reduced to 1.144 and 0.915 in the coaggregated precipitates of Fusobacterium nucleatum and each isolate. 4. The optical density of Porphyromonas gingivalis precipitate was 1.932 in the culture media, being reduced to 1.170 and 1.266 in the coaggregated precipitates of Porphyromonas gingivalis and each isolate. 5. When two isolates were tested with API 50 CHL medium kit, those were identified as Lactobaciallius salivarius and Lactobacillus delbrueckii subsp. lactis.
