Objectives : To identify whether pattern identification using nasal endoscopy for rhinitis can be applied as a tool for evaluating rhinitis in routine care setting, we performed a inter-rater reliability study on this pattern identification. Methods : Two Korean medicine doctors assessed 290 left/right nasal endoscopy photograph cases of rhinitis patients with pattern identification using nasal endoscopy. This pattern identification consist of four assessment items, nasal membrane color(pale/hyperemia), nasal membrane humidity(dryness/dampness), rhinorrhea(watery/yellow), and turbinate membrane edema(atrophic/edematous). Cohen's kappa statistic and Percentage agreement were used to evaluate the inter-rater reliability. Results : Inter-rater percentage agreement and Kappa coefficient for left nasal endoscopy photograph cases was from 'slight' to 'moderate'(% agreement: 40.00-67.59%/Kappa: 0.06-0.407). Only the agreement of 'rhinorrhea (watery/yellow)' item was moderate(% agreement: 67.59%/Kappa: 0.407). Inter-rater percentage agreement and Kappa coefficient for right nasal endoscopy photograph cases was also from 'slight' to 'moderate'(% agreement: 42.41-68.97%/Kappa: 0.109-0.465). Only the agreement of 'rhinorrhea(watery/yellow)' item was moderate(% agreement: 68.97%/Kappa: 0.465). Conclusions : It is necessary to resolve problems such as cut-off value setting, bipolar evaluation values(pale/hyperemia, dryness/dampness, watery/yellow, atrophic/edematous) and weighting items. Further rigorous studies that overcome the limitations of the current research are warranted.

We have investigated efficiency of a recombinant subunit Pasteurella multocida toxin (PMT) that was mixed with a vaccine consisted of inactivated whole cells of Bordetella bronchiseptica, P. multocida (types A and D). For verification of the efficacy of the vaccine, all experimental pigs (suckling piglets, sow and gilts) in the three farms were vaccinated. Antibody titers against B. bronchiseptica and P. multocida type A of the vaccinated pigs by microplate agglutination were significantly higher than those of the control pigs (p < 0.05). Similar patterns were observed in the analysis of anti- PMT neutralizing antibody by serum neutralizing method using Vero cell (p < 0.05). Anti- P. multocida type D antibody titer of the vaccinated sows and gilts by ELISA showed significant differences with those of the non-vaccinated pigs (p < 0.05). Although antibody titers increased, it was unable to find out the difference in the clinical signs between the vaccinated and non-vaccinated pigs. However, the increase in body weight of the vaccinated piglets was observed in comparison with the non-vaccinated piglets on a farm. At slaughtering of the pigs, pathological lesions in the turbinate bones of the vaccinated pigs were significantly lower than those of the non-vaccinated pigs (p < 0.001). These results suggested that efficacy of the vaccine in pigs demonstrated to protect against atrophic rhinitis in Korea.
Pasteurella multocida는 돼지에서 위축성 비염, 폐렴을 비롯한 다양한 호흡기 질환을 일으키는 병원균이다. 본 연구에서는 돼지 위축성 비염에 대한 효과적인 순수정제 백신을 개발하고자 하는 기초 연구로서 P. multocida의 외막 단백질 H에 의해 유도되는 방어적 항체와 면역을 확인하였다. P. multocida의 외막 단백질을 포함하는 분획은 호흡기 질병 혼합 백신에 대한 항혈청과 불활화된 사균 세포에 대한 항혈청 모두에서 면역학적으로 검출 가능하였다. 선행 연구에서 분리한 외막 단백질 H 유전자는 재조합 발현 백터 제작에 이용되어 대장균으로부터 재조합 외막단백질 H를 정제하였다. 실험 동물 면역과 항혈청의 교차반응, ELISA를 통한 항체 역가의 측정 및 공격접종을 통하여, 재조합 외막 단백질 H는 높은항원성을 가지며, 지속적인 체액성 면역을 유도하는 것을 확인하였다. 외막 단백질 H는 순수정제 항원으로서 P. multocida에 의한 호흡기 질환에 대한 효과적인 방어를 유도할 수 있는 단위 백신 후보 단백질로 여겨진다.
The detection of Bordetella bronchiseptica which is supposed to be an agent of the infectious atrophic rhinitis of swine, is likely to receive more attention in the future as the pork industry comes to realize that eradication of this infection from breeding herds is a practical possibility. Experiments described here were carried out to establish the rapid plate agglutination test for the detection of the infectious atrophic rhinitis of swine in the field using the criteria of antigen preparation, effects on the antigenecity after storing of the antigen and reaction appearing time. Also, the agglutinabilities between the plate and tube method were compared and the degree of pathological lesions were recorded in relation to tube agglutination titers. Obtained results were as follows: 1. No differences were noted in the agglutinabilities on the plate agglutination test between the treatments in antigen preparation-formolized, merthiolate-killed and living organism. 2. The agglutinability of the antigens did not show any significant changes until 10 weeks of storage at 4 C; however, after 10 weeks of storage, non-specific reaction was observed with the HPCD control sera. 3. The results of the plate and tube agglutination tests were not comparable but the effective use of the plate method in Bordetella bronchiseptica eradication programs in pigs especially in the sow is stressed as a screening test.
Bordetella (B.) bronchiseptica is a causative agent of swine atrophic rhinitis that promotes colonization of the mucous membrane of the swine nasal cavity by Pasteurella (P.) multocida. Mixed infection with B. bronchiseptica and P. multocida leads to growth inhibition of pigs, resulting in significant economic loss. There are many commercial vaccines for atrophic rhinitis, including B. bronchiseptica as a killed vaccine antigen (Ag). However, the immunogenicity of killed B. bronchiseptica Ag has not yet been elucidated; therefore, this study was conducted to investigate the immunogenicity of killed B. bronchiseptica Ag and the type of immune response it induces. In vitro assays using mouse spleen cells and flow cytometry revealed that B. bronchiseptica Ag induced high proliferation capability of lymphocytes, especially B lymphocytes, and the proliferating cells showed a significant response to interleukin (IL)-2. B. bronchiseptica Ag also enhanced the production of IL-12, a representative cytokine for cell-mediated immunity. In vivo experiments using mice showed that the injection of B. bronchiseptica Ag markedly induced Ag-specific antibody. Taken together, these results indicate that B. bronchiseptica Ag has high immunogenicity by itself.
The capsular serogroupes and drug susceptibility of 111 isolates of Pasteurella multocida from pigs with atrophic rhinitis and pneumonic lesions were investigated. Of the 111 P multocida isolates, 42 were from lung lesions, 47 from nasal turbinate lesions and the remaining 22 from the nasal swabs. P multocida isolates were typed for capsular serogroupes A by hyaluronidase inhibition of capsule and D by acriflavine auto-agglutination. Most isolates(64.9%) were type A, 23.4% were type D and the remaining 11.7% were untypable. Resistance to triple sulfa(97.3%) was most frequent, followed by resistance to tiamulin(71.2%), tylosin(56.8%), streptomycin(36.9%), and neomycin(36.0%). The majority of the organisms were susceptible in order of prevalence to baytril(100%), ampicillin(98.2%), linsmycin(97.3%), colistin(97.3%), cephalothin(94.6%), gentamicin(93.7%), amikacin(92.3%), tetracycline(91.9%), trimethoprim/sulfamethoxazole(91.0%), and kanamycin(90.1%). No differences in drug resistance in relation to capsular serogroupes of P multocida and the origin of lesions were noted. A high prevalence of multiple drug resistance was observed and the most common resistant patterns were Sss, Tm, Ty(12.6%) and Sm, Sss, Tm, Ty(8.1%) patterns.
The present study was conducted to investigate the incidence of Pasteurella multocida infection in Youngnam swine herds during the period from March 1988 to February 1989 and some properties of the isolated organisms. P multocida was isolated from 22(43.1%) of 51 growing pigs of 4 to 12 weeks of age and from 8(80.0%) of 10 herds. From nasal turbinates of 102 slaughtered pigs, 47(46.1%) pigs were culture positive and pigs from 8(88.9%) of 9 herds were found to be infected with P multocida. From lungs of 101 slaughtered pigs, 42(41.6%) pigs were culture positive and the pigs from 11(91.7%) of 12 herds were found to be infected with P multocida. The majority of biochemical and cultural properties of the P multocida isolates were identical to those of the standard strains. The isolation frequencies of P multocida in relation to pig snout lesion grades of 0 to 5 were 28.6%, 41.6%, 48.0%, 50.0%, 85.7%, and 100%, respectively.
A total of 98 strains of Bordetella bronckiseptica isolated from pigs affected the infectious atrophic rhinitis(AR) during 1978 were surveyed for drug sensitivity to 26 chemotherapeutic agents, and minimum inhibitory concentration(MIC), incidence rate of resistant strain and resistant patern from the strains which were obtained from the different pig farm in Jeonnam province were examined. The results obtained are summarized as follows. 1. Most of the strains tested were resistant to Ampicillin (AB, PC), spiramycin(SPO, sulfa drugs (SD) (MIC:$400.0{{\mu}g/ml}$) and streptomycin(SM) (MIC:$200.0{{\mu}g/ml}$). Of the 75.0% of strains were also resistant to penicillin(PC) (MIC:$200.0{{\mu}g/ml}$) and of the 14.3 of strains were inhibited to grow to tetracycline(TC), chlortetracycline(CTC), oxytetracyc-line(OTC), erythromycin(EM), tylosin(TS), leucomycin (LM) and chloramphenicol (CP) (MIC:$6.25{{\mu}g/ml}$). On the other hand, most of the strains tested were inhibited to grow to kanamycin(KM), gentamycin(GM) neomycin(NM) (MIC:$25.0{{\mu}g/ml}$) and to colistin(CL) (MIC:$12.5{{\mu}g/ml}$). 2. Incidence rate of resistant strains to main chemotherapeutic agents was 100.0% of sulfa drugs, 96.4% of streptomycin, 85.7% of penicillin, tetracycline, chloramphenicol and erythromycin, 46.4% of gentamycin, 17.9% of colistin and 0.0% of kanamycin and nalidixic acid.
Atrophic rhinitis (AR) is the one of important respiratory diseases and causes severe economic losses in pig industry. Severe attempts have been made to reduce the economic losses by preventing the disease. One of the methods is the spraying of antibiotics into nasal cavity of piglets. Recently, the efficacy of the spraying with kanamycin and gentamicin was reduced in the Korean swine industry. Therefore, the preventive methods have been required to be changed based on the antimicrobial susceptibility profiles of causative agents of swine AR. Based on the current situations of this disease, Bordetella (B.) bronchiseptica and Pasteurella (P.) multocida 4D were isolated from pigs with clinical signs of AR. The isolation rates of B. bronchiseptica and P. multocida 4D were 58.5% and 32.9%, respectively. In the antimicrobial susceptibility test, the bacteria were resistant to kanamycin and gentamicin which have been used as the spraying agents, but they were susceptible to amikacin. A new spraying agent was developed using amikacin using ${\beta}$-glucan and yakbaltag as supplementary agents. Field efficacy of the agent was carried out with different schedule. The results from this study suggested that the newly developed spraying agents might be helpful to prevent AR in swine.
The immune-stimulating activities of Bordetella bronchiseptica antigens containing dermonecrotoxin (BBD) loaded in chitosan microspheres (CMs) have already been reported in vitro and in vivo with a mouse alveolar macrophage cell line (RAW264.7) and mice. Therefore, this study attempted to demonstrate the successful induction of mucosal immune responses after the intranasal administration of BBD loaded in CMs (BBD-CMs) in colostrum-deprived pigs. The BBD was introduced to the CMs using an ionic gelation process involving tripolyphosphate (TPP). Colostrum-deprived pigs were then directly immunized through intranasal administration of the BBD-CMs. A challenge with a field isolate of B. bronchiseptica was performed ten days following the final immunization. The BBD-specific IgG and IgA titers, evident in the nasal wash and serum from the vaccinated pigs, increased with time (p<0.05). Following the challenge, the clinical signs of infection were about 6-fold lower in the vaccinated pigs compared with the nonvaccinated pigs. The grades for gross morphological changes in the turbinate bones from the vaccinated pigs were also significantly lower than the grades recorded for the nonvaccinated pigs (p<0.001). Therefore, the mucosal and systemic immune responses induced in the current study would seem to indicate that the intranasal administration of BBD-CMs may be an effective vaccine against atrophic rhinitis in pigs.
