Antioxidant Status and the Extent of Health Risks in Obese Korean Children

  • Objective: The purpose of this study was to determine serum antioxidant nutrients and the relationship between serum antioxidants and risks of chronic diseases in obese Korean children Methods: Normal weight Korean school children (0=170), mean age of 11.5$\pm$1.5, and obese (body fat mass > $28\%$) children (0=176), mean age of 11.0$\pm$1.8, were recruited Fat mass ($\%$) was determnined by Bioelectrical Impedance (BEI), and body mass index (BMI) was calculated Fasting blood was collected to measure serum antioxidant nutrients, vitamin A, vitamin E and zinc. Serum lipid profiles including total cholesterol (TC), high density cholesterol (HDL) and triglyceride (TG), and blood glucose, glutamic oxaloacetic transaminase (GOT), and glutamic pyruvic transaminase (GPI) were also determined. Differences in serum blood measurements between obese and normal children were assessed by independent t test. Pearson's correlation analysis was used to determine the relationship between variables. Results: Blood glucose, GPT, total cholesterol, and triglycerides concentrations were significantly higher among obese boys, compared to normal boys (p<005). Significantly lower concentrations of serum vitamin E, after adjustment for TG and TC, was shown in obese boys (0.26 mg/mg) and obese girls (0.31 mg/mg), compared to normal boys (0.36 mg/mg) and girls (0.38 mg/mg) (p<0.05). Fat mass ($\%$) was negatively con-elated with serum vitamin A and vitamin E. Conclusion: Obese Korean children showed insufficient serum vitamin E concentration and increased risk for diabetes, atherosclerosis, and liver disease. Since lower vitamin E concentration was negatively con-elated with atherogenic index, improved vitamin E status in children may decrease the risk of atherosclerosis later in life.

Effects of Sodium Intake on the Association between the Salt-Sensitive Gene, Alpha-Adducin 1 (ADD1), and Inflammatory Cytokines in the Prevalence of Children Obesity

  • Objective: To examine the effects of sodium intake on the correlations between the saltsensitive gene ${\alpha}$-adducin 1 (ADD1) and inflammatory cytokines in Korean childhood obesity. Methods: A total of 2,070 students aged 8-9 years old participated in this study. The anthropometrics, serum biochemistry profile, inflammatory cytokines, and three-day dietary assessment were analyzed according to sex, obesity degree, and ADD1 polymorphism. Results: The obesity prevalence was higher in boys (15.6%) than in girls (11.9%). Boys also showed higher values in anthropometrics; lipid, glucose, and insulin profiles; total calorie intakes, as well as those of sodium and calcium compared with those of the girls. The more obese were boys and girls, the higher were the anthropometrics and the blood levels (total cholesterol, triglyceride, low-density lipoprotein cholesterol, fasting blood sugar, and insulin), but the lower was high-density lipoprotein cholesterol. The obese boys had significantly higher sodium and Na/K intakes, while the obese girls had higher visfatin level and Na/K intake. In addition, an increase in the risk factors for blood pressure and obesity in ADD1 variants was identified. Serum tumor necrosis $factor-{\alpha}$($TNF-{\alpha}$) significantly increased with increasing sodium intake in the ADD1 W allele carriers, regardless of sex. The presence of obesity with the ADD1 W allele induced inflammatory accelerators such as $TNF-{\alpha}$ or C-reactive protein(CRP) with higher sodium intake. Conclusion: Obese children with an ADD1w allele can experience a more complex form of obesity than non-obese when exposed to an obesity-inducing environment and need to be controlled sodium intake in the diet.

랫트의 실험적 동맥경화증에 대한 $\beta$-Glucan의 예방 효과 (Preventive Effect of $\beta$-Glucan on the Experimental Atherosclerosis in Rats)

  • The present studies were undertaken in attempt to investigate the preventive effect of $\beta$-glucan from barley ad diltiazem on cholesterol and vitamin D2 induced-atherosclerotic rat. The results obtained were summerized as follows. 1.The group, fed only the mixture of cholesterol and vitamin D2, showed significant increase of calcium, inorganic phosphorus, total cholesterol, lipid LDL-cholesterol and phospholipid in serum, and total lipid in the liver (p<0.05) as comparing with normal group. The aorta showed severe damage of disorganization, necrosis and lipid deposition in the elastic membrane. 2. The group fed mixture of cholesterol and vitamin D2 plus diltiazem simultaneously, showed significant increase of total cholesterol, total lipid and phospholipid in serum, and total lipid and triglyceride n the liver (p<0.05) as comparing with normal group, but the significant decrease of calcium and inorganic phosphorus in serum(p<0.05) as comparing with the atherogenic control group. The aorta showed slight damage of elastic membrane and lipid deposition as comparing with the atherogenic control group. 3.The group, fed mixture of cholesterol and vitamin D2 puls $\beta$-glucan simultaneously, showed significant decrease of total cholesterol, LDL and VLDL-cholesterol, total lipid, phospholipid and triglyceride in serum, and total lipid in the liver as comparing with the atherogenic control group(p<0.05), but the significant increase of calcium in serum as comparing with the normal group(p<0.05). The aorta showed no changes in elastic fiber and no lipid deposition in comparing with the atherogenic control group.

한국산 잣기름이 정상토끼 및 동맥경화증 유발 토기의 적혈구막 지방산조성에 미치는 영향 (Effect on Fatty Acid Composition of red Blood Cell Membranes of supplement of Korean Pinenut Oil, rich in 5-Olefinic acids, in Normocholesterolemic and Atherosclerotic New Zealand White Rabbits)

  • The present study was carried out in normal and atherosclerotic New Zealand white(NZW) rabbits, to evaluate the effect of dietary supplementation with Korean pinenut oil, on the total fatty acid composition of red blood cell membranes. Erythrocyte fatty acids of chain lengths ranging fro 12 : 0 to 22 : 6$\omega$3 were identified. In study I, palmitic acid(16 : 0) was found to be the main erythrocyte membranes fatty acid, followed in turn by stearic(18 : 0), oleic(18 : 1), and linoleic acids(18 : 2$\omega$6). There was a large increase in the proportion of 20 : 4$\omega$6 in 10% pinenut oil group on days 40 and 80(compared with control), although the increase was not statistically significant. Most erythrocyte fatty acids were less affected by the types or levels of oils supplemented for periods fo 40 and 80days. In study II,at the end of the dietary treatment, the levels of 12 : 0, 14 : 0, 15 : 0 and 18 : 0 were significantly decreased in the 5% soybean oil/cholesterol group, whereas those of 12 : 0, 14 : 0 and 18 : 0 were significantly decreased, but that of 18 : 2$\omega$6 was significantly increased in the 10% soybean oil/cholesterol group, compared to the cholesterol group. No significant differences in fatty acid composition were seen between the pinenut oil/cholesterol group, whereas the 10% soybean oil/cholesterol group had significantly elevated 18 : 2$\omega$6, compared to the cholesterol group. The proportion of 18 : 4$\omega$6 was significantly raised in the 5% soybean oil/cholesterol and 5% pinenut oil/cholesterol group, and the content of 16 : 1$\omega$7 was significantly lowered in the 5% and 10% soybean/cholesterol groups, compared to the cholesterol group at 10 weeks supplementation. In studies I,II and III, the total proportions of saturated fatty acids were not significantly affected by the types of levels of oils supplemented throughout the study period.

제2형 당뇨 마우스 모델에서 청혈플러스의 항고지혈 및 항산화효과 (Hypolipidemic and Anti-oxidant Effects of Chunghyl Plus in Type II Diabetic Mice Model)

  • This study was perfomed to investigate the effects of Chunghyul-plus(CHP) on oxidative damage and hyperlipidemia in db/db mouse. After treatment with CHP, safety in cytotoxicity, heavy metal toxicity, production of reactive oxygen species(ROS), nitric oxide (N0) and proinflammatory cytokine IL-Ib, TNF-a, IL-6 in RAW 264.7 cells. Serum total cholesterol, LDL cholesterol, HDL cholesterol, triglyceride, insulin, GLP-1, glucose, food intake, body weight, organ weight, AST, ALT, ALP, BUN, creatine and histologic change of liver and aorta were measured in db/db mouse after oral administration of CHP. CHP showed safety in cytotoxicity and toxicity of liver and kidney for logn time administration. CHP increased the DPPH and ABTS radical scavenging activity. CHP showed significant inhibitory effect on reactive oxygen species (ROS), and showed inhibitory effect on nitiric oxide(NO) compared to control group. CHP decreased cytokine IL-6 production significantly, and decreased IL-1β and TNF-α compared to control group. CHP decreased body and organ weitht, intake food, and glucose levels compared to control group. CHP decreased total cholesterol and triglyceride significantly, and decreased LDL-cholesterol levels and increased HDL-cholesterol levels compared to control group. CHP decreased atherogenic index and cardiac risk factor significantly. CHP increased serum insulin and GLP-1 compared to control group. In histologic examination, lipophagy in the liver and aorta decreased in CHP treated mice and the cell was regular and boundary of vessel wall was clear compared to control group. These results suggest that CHP is effective in antioxidation activity and treatment and prevention of hyperlipidemia, atherosclerosis, diabetes, ischemic heart disease, stroke and other cardiocerebrovascular disease.

유산소성 운동이 콜레스테롤식이 흰쥐의 혈청지질에 미치는 효과 (Effects of Aerobic Exercise on Serum Lipid in Cholesterol-Dietary Rats)

  • 본 연구는 생후 4주령의 Sprague Dawley 흰쥐 47마리를 대상으로 2% 콜레스테롤 식이와 정상식이 그리고 여러가지 강도의 유산소운동(트래드밀 런닝)과 비운동을 실시하여 혈청지질과 adipose tissue를 분석해서 다음과 같은 경과를 얻었다. 1. 콜레스테롤식이에 의해 형중T-C치가 유의하게 증가되었으나 유산소성운동에 의해 감소하는 경향을 나타내었다. 2. 콜레스테롤식이에 의해 혈중 HDL-C와 LDL이 약간 증가야였으나, 유산소성운동에 의해 모두 감소하는 경향을 나타내었다. LDL/HDL-C는 간한 운동을 한 CRA군에서만 감소하는 경향을 나타내었다. 3. 콜레스테롤식이에 의해 혈액중의 phospholipid 및 triglyceride는 감소하는 경향을 나태내었으나, 유산소성운동에 의해 약간 증가하는 경향을 나태내었다. 4. 정상식이군에서 유산소성운동에 의해 adipose tissue가 유의하게 감소되었으며, 콜레스테롤 식이군에서도 유산소성운동에 의해 adipdse tissue가 감소하는 경향을 나타내었다. 이상과 같은 연구결과를 통하여 유산소성운동이 콜레스테롤식이에 의해 증가도니 혈중 총콜레스테롤을 저하시키며, 동백경과 위험인자인 LDL은 낮추어 동맥경화성 질병을 예방할 것으로 사료된다.

김치 젖산균의 In Vitro에서 콜레스테롤 저하 효과 (In Vitro Evaluation of Cholesterol Reduction by Lactic Acid Bacteria Extracted from Kimchi)

  • 본 연구는 김치에 분포하는 것으로 알려져 있는 대표적인 젖산균에 관한 콜레스테롤 저하 효과에 관하여 연구하였다. Lactobacillus 1종, Leuconostoc 2종과 Weissella 2종을 대상으로 콜레스테롤 저하 효과를 측정하였다. 김치 초기 발효균인 Leu. citreum은 55.46% 콜레스테롤 감소율을 보였고, 대표적인 산패균인 Lac. plantarum은 55.91%, Leu. mesenteroides은 56.37%였으며 W. kimchii,, W. confusa 등에서도 55% 이상의 콜레스테롤 저하 효과가 나타났다. 김치의 발효 숙성 과정에 나타나는 젖산균들이 시험관 내에서 콜레스테롤 저하 효과가 확인되어, 향후 in vivo 조건하에서 콜레스테롤 저하에 어느 정도 영향을 미치는지 검토해볼 필요가 있다고 사료된다.

랫트의 실험적 동맥경화증에 대한 칠성장어유 및 옥수수유의 예방적 효과 (Preventive Effect of Lamprey Oil and Corn Oil on the Experimental Atherosclerosis in Rats)

  • 콜레스테롤과 Vitamin D2로 유발시킨 동맥경화증 랫트에 칠성장어유와 옥수수유의 급여에 따른 예방적 효과를 조사할 목적으로 다음 네 개의 식이군으로 나누어 실험한 바, 첫째군은 정상대조군, 둘째군은 콜레스테롤(40/kg) 과 Vitamin D2(300,000 IU/kg)를 시험개시 1일부터 연속적으로 4일간 투여한 동맥 경화증 대조군, 셋째군은 동맥경화증 유발 처치와 함께 15% 칠성 장어유 식이(w/w)를 8일간 급여한 칠성장어유 식이군, 넷째군은 동맥경화증 유발처치와 함께 15% 옥수수유 식이 (w/w)를 8일간 급여한 옥수수유 식이군으로 하여 다음과 같은 결과를 얻었다. 동맥경화증 대조군은 정상대조군에 비하여 혈청 중의 총지질, 인지질. 총콜레스테롤, HDL-cholesterol, LDL-chlesterol 치의 현저한 증가를 보였고 (p<0.01), 또한 심장의 중성지방 및 콜레스테롤치의 증가(p<0.01), 대동맥의 중성지방(p<0.01) 및 콜레스테롤치 (p<0.05)를 보였다. 한편 대동맥은 심한 손상으로 내피세포의 탈락 및 내막과 중막에 석회화, 괴사, 지방 침착을 보였고, 관상동맥은 석회화, 괴사, 백혈구의 침윤이 현저하게 관찰되었다. 칠성장어유 식이군은 정상대조군에 비하여 혈청 중성지방치의 r마소 (p<0.01), 혈청 HDL-cholesterol 치의 증가(p<0.01), 심장콜레스테롤치 (p<0.05),의 증가를 보였고, 동맥경화증 대조군에 비하여는 혈청 중의 총지질, 중성지방, 인지질, 총콜레스테롤, LDL-cholesterol , VLDL-cholesterol 치 (p<0.01), 및 LDL치의 감소(p<0.05), 또한 간장 콜레스테롤치의 증가(p<0.05), 심장의 중성지방 (p<0.01) 및 콜레스테롤치 (p<0.05)의 r마소, 대동맥 중성지방 (p<0.01) 및 콜레스테롤치 (p<0.05) 의 감소를 보였다. 또한 옥수수유 식이군에 비하여 혈청 총지질(p<0.05)의 감소, 총콜레스테롤 및 HDL-cholesterol 치의 감소 (p<0.01), 심장중성지방치의 감소 , 대동맥 중성지방치의 감소 (p<0.01)를 보였다. 한편 대동맥은 동맥경화증 대조군에 비하여 미약한 석회화와 괴사된 thru을 보였으나 칠성장어유 식이군에서와 같이 지방침착은 관찰되지 않았고 관상동맥에서도 동맥경화증 대조군에 비해 미약한 석회화와 괴사된 소견을 보였다. 이상의 결과에서 불포화지방산을 다량으로 함유한 칠성장어유 및 옥수수유는 동맥경화증 랫트에 있어 생체내 지질대사에 영향을 미침으로써 혈청지질치를 저하시키고 동맥벽내에 지방침착을 억제시켰으며, 이러한 효과는 n-6계 다불포화지방산을 많이 함유하고 있는 옥수수유보다 n-3rP 다불포화지방산을 다량으로 함유한 칠성장어유에 더욱 크게 나타났다

  • Acyl-CoA:cholesterol acyltransferase (ACAT, EC 2.3.1.26) plays an important role in the control of intracellular free cholesterol content via its cholesterol esterifying activity. ACAT inhibitors are expected to be effective for treatment of atherosclerosis and hypercholesterolemia. In the course of a screening program for ACAT inhibitors from microbial sources, GERI-BP001 M, A, and B were isolated from the fermentation broth of a fungal strain. GERI-BP001 compounds were isolated from a culture broth of Aspergillus fumigatus F37 by acetone extraction, EtOAc extraction, SiO$_2$ column chromatography, and reverse phase HPLC. The structure of GERI-BP001 coumpounds were determined by $^1$H-NMR, $\^$l3/C-NMR, 2D-NMR, NOESY, and long range C-H COSY experiments. GERI-BP001 M, A, and B inhibit ACAT activity in an enzyme assay system using rat liver microsomes by 50% at concentrations of 75, 147, and 71 ${\mu}$M, respectively.

달래(Allium monanthum)가 콜레스테롤을 투여한 흰쥐의 혈청 성분에 미치는 영향 (Effects of Wild Garlic on Serum Component of Cholesterol Fed Rats)

  • To study effects of wild garlic on components of lipid and protein, blood glucose level, and cholinesterase activity in rat's serum, Sprague-Dawley strain of 40 male rats are divided into 5 groups which are Normal, Control, A, B, and C group. Normal group is fed only Basal diet, and Control group is fed Basal diet and 0.5ml/day of 2.5% cholesterol solution, In addition to a diet of control group, A group is fed 25% wild garlic flour, B group 0.5ml/2day of concentratded wild garlic juice, C group 0.5ml/2day of concentrated ethanol extract of wild garlic, After 8 weesk the rats were fasted for 12 hours, and then decapitated to collect blood. The results of analysis of the rat's serum were summarized as follows, 1. Wild garlic diet, specially ethanol extract has an influence on decreasing the level of total cholesterol and blood glucose in rat's serum, 2. HDL-cholesterol and phospholipid content of rat's serum are increased by wild garlic diet. Therefore I think that wild garlic is good food for preventing development of atherosclerosis and diabetes.
