Genetic Analysis on Floury Endosperm Characteristics of 'Namil(SA)-flo1', a Japonica Rice Mutant Line (남일벼 돌연변이 후대 계통 'Namil(SA)-flo1'의 분질배유 특성에 대한 유전분석)

  • Rice varieties with suitable flour-making quality are required to promote rice processed-food industry and boost rice consumption in Korea. 'Namil (SA)-flo1' is an advanced mutant line with floury endosperm which shows good flour-making quality under dry-milling process. Genetic analysis was carried out to localize the chromosomal region responsible for the floury endosperm of 'Namil (SA)- flo1'. By using 94 F2 progenies, which were derived from 'Namil (SA)-flo1' ${\times}$ 'Milyang 23', floury grains percentage was investigated as phenotypic data, and genotyping was conducted with 54 SSR markers. Association analysis showed that the target genetic region for floury endosperm is on middle-low region of chromosome 5. Through further association analysis with increased number of SSR markers on chromosome 5, we found that genotypic variation in RM164 explains 79.7% of the variation in floury grains percentage of F2:3 seeds. The floury endosperm locus was localized on 17.7-20.7 Mbp region of chromosome 5 and will be further analyzed for fine mapping and gene identification.

Fine mapping of rice bacterial leaf blight resistance loci on K1 and K2 of Korean races of Xoo (Xanthomonas oryzae) using GWAS analysis

  • Bacterial leaf blight(BLB), caused by X. oryzae pv. oryzae(Xoo), is one of the most destructive diseases of rice due to its high epidemic potential. Understanding BLB resistance at a genetic level is important to further improve the rice breeding that provides one of the best approaches to control BLB disease. In the present investigation, a collection of 192 accessions was used in the genome-wide association study (GWAS) for BLB resistance loci against four Korean races of Xoo that were represented by the prevailing BLB isolates under Xoo differential system. A total of 192 accessions of rice germplasm were selected on the basis of the bioassay using four isolated races of Xoo such as K1 and K2. The selected accessions was used to prepare 384-plex genotyping by sequencing (GBS) libraries and Illumina HiSeq 2000 pairedend read was used for GBS sequencing. GWAS was conducted using TASSEL 5.0. The TASSEL program uses a mixed linear model (MLM). The results of the bioassay using a selected set of 192 accessions showed that a large number of accessions (93.75%) were resistant to K1 race and K2 resistant germplasm proportion remained between 66.67. The genotypic data produced SNP matrix for a total of 293,379 SNPs. After imputation the missing data was removed, which exhibited 34,724 SNPs for association analysis. GWAS results showed strong signals of association at a threshold of [-log10(P-value)] more than 5 (K1 and K2) for nine of the 39 SNPs, which are plausible candidate loci of resistance genes. These SNP loci were positioned on rice chromosome 2, 9, and 11 for K1 and K2 races. The significant loci detected have also been illustrated and make the CPAS markers for NBS-LRR type disease resistance protein, SNARE domain containing protein, Histone deacetylase 19, NADP-dependent oxidoreductase, and other expressed and unknown proteins. Our results provide a better understanding of the distribution of genetic variation of BLB resistance to Korean pathogen races and breeding of resistant rice.

  • Recently days, soybean production in paddy field is increasing, from 4,422 ha in 2016 to 10,658 ha in 2021 in Korea. It is easy for Phytophthora stem and root rot (PSR) occurring in paddy field condition, when it is poorly drained soils with a high clay content, and temporary flooding and ponding. Therefore PSR resistant soybean cultivar is required. The objective of this study is to identify QTL region and candidate genes relating to PSR resistance of the race in main soybean cultivation area in Korea. 210 soybean materials including cultivars and germplasm were used for inoculation and genome-wide association study (GWAS). Inoculation was conducted using stem-scar method with 2 replications in 2-year for the race 3053 from Kimje and 3617 from Andong. 210 materials were genotyped with Soya SNP 180K chip, and structure analysis and association mapping were conducted with QTLMAX V2. The results of inoculation showed that survival ratio ranged from 0% to 96.7% and mean 9.7% for 3053 and ranged from 0% to 100% and mean 7.6% for 3617. Structure analysis showed linkage disequillibrium (LD) was decayed below r2=0.5 at 335kb of SNP distance. Significant SNPs (LOD>7.0) were identified in Chr 1, 2, 3, 4, 5, 11, 14, 15 for 3053 and Chr 1, 2, 3, 7, 10, 14 for 3617. Especially, LD blocks (AX-90455181;15,056,628bp~AX-90475572;15,298,872bp) in Chr 2 for 3053 and 3067 were duplicated. 29 genes were identified on these genetic regions including Glyma.02gl47000 relating to ribosome recycling factor and defense response to fungus in Soybase.

  • The purpose of this study is development of the management and guide system for convenient use of new address guide system for systematic management and use of spatial information gained by mapping of numerical map of building and road to large basic new address, survey and input of main gate, decision of the road session, naming and input of road name, grant of numbering of all houses and buildings, new address guide, real life geographical information and added information service on web site in Jecheon-si city area. In this study, the development and establishment of new address management and guide system in local cities are designed by making full use of GIS function of client server based system. A guide system is designed the same database as management system, and it is transformed into Web GIS environment and developed in order to add the various functions of life GIS as well as new address guide. In the result, the effectiveness of new address system and guide of Jecheon-si city could be highly increased owing to not only citizens' easy access but also easy availability of various informations necessary in life by developed its system.

Structures and Policies of British Geographic Information Dissemination for Korea National GIS Project (국가지리정보사업 추진을 위한 영국지리정보 유통구조 및 정책 연구 - 영국지리정보원의 역할을 중심으로-)

  • The objective of this paper is to discuss geographic information policies and strategies of British government and suggest effective policies for Korea National GIS project GIS that has been implemented geographic information infrastructures into public sectors and private markets for the last ten years. To obtain the research aim, this paper reviews the main factors of the Britain GIS project such as distribution structure and process of GI markets, government policies and strategies that are led by Ordnance Survey, most leading mapping agency in the UK. In conclusion, some issues have been explored with reference to the experiences of the Britain GIS projects. The first of these is the nature of geographic information and the second concerns the notion of the circulation policies of spatial data, and the last proposes Korea GIS policies and strategies for successful geographic information and spatial data implementation. The findings of the analysis of the Britain GIS development indicate that a shift began to take place from central government coordinate toward more extensive utilization of private and commercial sectors. This reflects both the increasing importance of geospatial data circulation in all levels of GIS stakeholders. Finally these discussions are particularly to be the issues where multi-agency collaboration of Korea government is concerned and can take the form of joint ventures by consortiums of both involving data producers and data users in order to increase commercial participation for value-added geospatial items, and to encourage both research and development sectors with low or free price policies.

Mapping Technique for Heavy Snowfall Distribution Using Terra MODIS Images and Ground Measured Snowfall Data (Terra MODIS 영상과 지상 적설심 자료를 이용한 적설분포도 구축기법 연구)

  • This study is to make snowfall distribution map for the 4 heavy snowfall events of January 2001, March of 2004, December of 2005 and January of 2010, and compare the results for three cases of construction methods. The cases are to generate the map by applying IDW(Inverse Distance Weighting) interpolation to 76 ground measured snowfall point data (Snow Depth Map; SDM), mask out the SDM with the MODIS snow cover area (MODIS SCA) of Terra MODIS (MODerate resolution Imaging Spectroradiometer) (SDM+MODIS SCA; SDM_M), and consider the snowdepth lapse rate of snowfall by elevation (Digital Elevation Model; DEM) to the second case (SDM_M+DEM; SDM_MD). By applying the MODIS SCA, the SCA of 4 events was 62.9%, 44.1%, 52.0%, and 69.0% for the area of South Korea. For the average snow depth, the SDM_M decreased 0.9cm, 1.9cm, 0.8cm, and 1.5cm compared to SDM and the SDM_MD increased 1.3cm, 0.9cm, 0.4cm, and 1.2cm respectively.

A Study on Object-Based Image Analysis Methods for Land Cover Classification in Agricultural Areas (농촌지역 토지피복분류를 위한 객체기반 영상분석기법 연구)

  • It is necessary to manage, forecast and prepare agricultural production based on accurate and up-to-date information in order to cope with the climate change and its impacts such as global warming, floods and droughts. This study examined the applicability as well as challenges of the object-based image analysis method for developing a land cover image classification algorithm, which can support the fast thematic mapping of wide agricultural areas on a regional scale. In order to test the applicability of RapidEye's multi-temporal spectral information for differentiating agricultural land cover types, the integration of other GIS data was minimized. Under this circumstance, the land cover classification accuracy at the study area of Kimje ($1300km^2$) was 80.3%. The geometric resolution of RapidEye, 6.5m showed the possibility to derive the spatial features of agricultural land use generally cultivated on a small scale in Korea. The object-based image analysis method can realize the expert knowledge in various ways during the classification process, so that the application of spectral image information can be optimized. An additional advantage is that the already developed classification algorithm can be stored, edited with variables in detail with regard to analytical purpose, and may be applied to other images as well as other regions. However, the segmentation process, which is fundamental for the object-based image classification, often cannot be explained quantitatively. Therefore, it is necessary to draw the best results based on expert's empirical and scientific knowledge.

Spatial Distribution Characteristics of Vertical Temperature Profile in the South Sea of Jeju, Korea (제주 남부해역 수온 수직구조의 공간분포 특성 파악)

  • To visualize the characteristics of vertical seawater temperature data, in the ocean having 3D spatial characteristics, 2D thematic maps like horizontal seawater temperature distribution map at each depth layer and 3D volume model using 3D spatial interpolation are used. Although these methods are useful to understand oceanographic phenomena visually, there is a limit to analyze the spatial pattern of vertical temperature distribution or the relationship between vertical temperature characteristics and other oceanic factors (seawater chemistry, marine organism, climate change, etc). Therefore, this study aims to determine the spatial distribution characteristics of vertical temperature profiles in the South Sea of Jeju by quantifying the characteristics of vertical temperature profiles by using an algorithm that can extract the thermocline parameters, such as mixed layer depth, maximum temperature gradient and thermocline thickness. For this purpose spatial autocorrelation index (Moran's I) was calculated including mapping of spatial distribution for three parameters representing the vertical temperature profiles. Also, after grouping study area as four regions by using cluster analysis with three parameters, the characteristics of vertical temperature profiles were defined for each region.

Assessment of the Inundation Area and Volume of Tonle Sap Lake using Remote Sensing and GIS (원격탐사와 GIS를 이용한 Tonle Sap호의 홍수량 평가)

  • The ability of remote sensing and GIS technique, which used to provide valuable informations in the time and space domain, has been known to be very useful in providing permanent records by mapping and monitoring flooded area. In 2000, floods were at the worst stage of devastation in Tonle Sap Lake, Mekong River Basin, for the second time in records during July and October. In this study, Landsat ETM+ and RADARSAT imagery were used to obtain the basic information on computation of the inundation area and volume using ISODATA classifier and segmentation technique. However, the extracted inundatton area showed only a small fraction than the actually inundated area because of clouds in the imagery and complex ground conditions. To overcome these limitations, the cost-distance method of GIS was used to estimate the inundated area at the peak level by integrating the inundated area from satellite imagery in corporation with digital elevation model (DEM). The estimated inundation area was simply converted with the inundation volume using GIS. The inundation volume was compared with the volume based on hydraulic modeling with MIKE 11. which is the most poppular among the dynamic river modeling system. The method is suitable for estimating inundation volume even when Landsat ETM+ has many clouds in the imagery.

Vegetation Mapping and Fodder Value of Plant Communities at the natural Grassland (자연초지 식생군락의 사료가치와 식생도 작성)

  • This trial was carried out to find out the degree of fodder value of different plant communities and to make the plant sociological vegetation map for all plant communities at the natural grassland of the Rehberg-Alm in the Bavarian Alps, southern part of Germany during 1982-1983. 1. Allogenic succession of the plant communities at natural grassland was much more influenced by the change of soil moisture or/and surface water than sheep grazing. 2. The plant communities at the Rehberg-Alm were Nardetum alpigenum, Poo-Prunelletum, Cirsium arvense Cirsium Vulgare-Association, Caricetum davallianae, Rumicetum alphini, Caricetum paniculatae and Disturbed lowland bog-Stand. 3. By the sheep grazing will be improved the inferior plant community of Nardetum alpigenum to the most desirable Poo-Prunelletum plant community at the mountainous grassland gradually. 4. General fodder value in this area depended heavily on the composition of vegetation of the plant communities. The highest fodder value was the Poo-Prunelletum with 4.4 and the next was the Nardetum alpigenum with 2.5. The others were not suitable for grazing pasture due to less fodder value.
