Mutagenic Characterization of a Conserved Functional Amino Acid in Fuculose-1-Phosphate Aldolase from Methanococcus jannaschii, a Hyperthermophic Archaea

  • To elucidate the putative role of the amido group in the metal binding of the fuculose-1-phosphate aldolase from Methanococcus jannaschii, we have examined a potential targen using site-directed mutagenesis. The replacement of asparagine 25 with leucine or threonine was shown to have a negative effect, not only on catlytic efficiency, but also on substrage recognition as well. The Hill coefficient values yeilded a value of =1. All metals used with the wild-type aldolases exhibited higher activity than that of the mutants. The spectra of the mutants were quite different from the wild-type aldolase. A highly conserved amino acid of asparagine 25 in a related family of aldolase odes not appear to provide sufficient evidence for evolution.

  • The studies could be summarized as follows: 1. Standard Rf Values were made as Table II for the identification of amino acids. 2. Amounts of free amino nitrogen in eggs were increased about 0.03% (Table IV) while the amounts of total nitrogen (Table III) were increased lower percentage during the ten days of development. 3. Three unknown substances were found newly during the development. 4. Eight ninhydrine positive amino acids were present, in the fresh embryonic eggs, but the numbers increased to eighteen after fifty days of development. 5. Through the development, there were large amounts of glutamic acid, aspartic acid and alanine. 6. After fifty days development, the amount of amides, such as asparagine, glutamine and alanine were increased very large which were very small amount before it and the amount of asparagine were more than that of glutamine. 7. Tryptophane, Cystine , Histidine were detected after 50 days of development. This is very remarkable fact.

  • 인공해수에서 일어나는 구리의 부식에 대하여 알라닌, 아스파라긴, 아스파틱산, 글루타민 그리고 메티오닌의 부식억제 효과를 조사하였다. 아미노산들의 흡착은 흡착 분자들의 상호작용 때문에 Temkin의 logarithmic isotherm이 잘 적용되었으며, 부식억제 효율은 아미노산의 농도에 의존하였다.

  • Paper partition chromatography has been applied to estimate the amino acids in the liver of Bombina orientalis Boulenger. The results obtained are as follows : 1) The presence of cystine, serine, glycine, threonine, alanine, $\beta$-alanine, tyrosine, histidine, asparagine, tryptophane, methionine, valine, phenylalanine, leucine and an unknown substance has been demonstrated in the alcoholic extracts of the material. 2) The presence of glutamine, cystine, serine , glycine, arginine, threonine, histidine, alanine, methionine, proline, valine ,phenylalanine, leucine, and two unknown substances has been demonstrated in the acid-hydrolyzed liver. 3) Tyrosine, asparagine, and trypotphane have been demonstrated only in the alcoholic extract. 4) Proline, glutamine and arginine have been demonstrated only in the acid-jydrolyzed liver.

  • AF-001A is a novel antifungal cyclic glyco-peptise isolated from Pseudomonas cepacia AF6008. AF-001A is a mixture of AF-011A1 and AF-001A2. Each compound contains glycine(1), serine(2), asparagine(1), 2,4-diaminobutyric acid(1), beta-hydroxytyrosine(1), xylose(1) and a methylene chain amino acid(1). Additionally, A1 contains one beta-hydroxyasparagine that is replaced with as asparagine in A2. AF-011A showed high in vitro antifungal activity against various animal and plant pathogenic fungi and caused no irritation on the skin of rabbits.

Asparagine Residue at Position 71 is Responsible for Alkali-Tolerance of the Xylanase from Bacillus Pumilus A-30

  • The xynA gene encoding an alikali-tolerant endo-1,4-${\beta}$-xylanase (XYN) was cloned from the alkalophilic Bacillus pumilus A-30. The nucleotide sequence of a 974-bp DNA fragment containing the xynA was determined. An ORF of 684 nucleotides that encoded a protein of 228 amino aicds was detected. Asparagine-71 of XYN from B. Pumilus A-30 showed to be highly conservative in alkaline xylanases of family G/11, upon comparing the amino acid sequences of 17 family G/11 xylanases. Site-directed mutation of N71D of the xynA gene resulted in a decrease of 12.4% in the specific acitivity and a significant decline in the enzyme activity in the alkaline pH range.

  • Major components affecting nonenzymatic browning in stored ginger paste were investigated using five synthetic model solutions. The model systems were stored at $40^{\circ}C$ for 30 days and analyzed for browning, in addition the contents of sugars, organic acids, ascorbic acids, amino acids and gingerols were determined. Among the chemical components, fructose, asparagine and ascorbic acid were the main contributors to the browning development of ginger paste, while gingerol compounds were browning inhibitors.

Effect of Supplement nutrition on the Mycelial Growth of Lentinus edodes

  • Mycelial growth of L. edodes by supplement nutrition of softwood was studied on a sawdust medium. The sawdust used was from the following softwood species : Larix leptolepis, Pinus densiflora and Pinus koraiensis. The added nutritions consisted of carbon nutritions(sucrose, active carbon, xylose, glucose, paper pellet), nitrogen nutritions(potassium nitrate, ammonium chloride, asparagine, glutamic acid) and vegetable oil(rice bran oil). The sawdust medium was a mixture of 76% sawdust, 20% rice bran, 3% carbon nutrition, 0.4% nitrogen nutrition and 0.6% calcium carbonate. Following addition of carbon and nitrogen nutritions on the sawdust medium proved most suitable : L. leptolepis (glucose, glutamic acid), P. densiflora (active carbon, asparagine) and P. koraiensis (xylose, glutamic acid). The highest mycelial growth was obtained from sawdust medium of optimum condition with 97% of L. leptolepis, 110% of P. densiflora and 98% of P. koraiensis. This study has provided useful preliminary information for the cultivation of L. edodes.

Monascus anka로부터 황색소 생성 변이주의 분리 및 특성 (Isolation and characteristics of yellow-pigment producing mutants of Monascus anka.)

  • 천연 황색소 생산을 위하여 Monascus anka Nakazawa et Sato IFO 4478(KCCM l1832 strain) 균주를 모균주로 하여 UV 조사 및 NTG(N-methyl-N'-nitro-N-nitrosoguanidine)처리 및 이들 방법을 병용하는 방법을 사용하여 모균주와는 색소생성 특성이 다르게 황색소를 단일성분으로 생성하는 변이주를 분리하고 그 특성을 조사하였다. leucine, cystein, aspartic acid, glutamic acid 와 asparagine 등 5종의 아미노산에 대한 영양요구주를 분리하였고 이중 asparagine 영양요구주 Y7은 colony 형태와 균사의 직경에서 약간의 차이를 나타내지만 전형적인 Monascus 균주의 특성을 지니고 있었으며 특히 유성생식에 의한 번식 특성이 강하여 색소 생성에 유리한 조건을 지니고 있었다. 그리고 Y7은 밝은 색조의 황색소를 단일 성분으로 생성하며 10대 이상의 계대배양에서도 색소 생성 특성이 안정하였고 색소 생성량이 모균주보다 약 2.2배 높은 값을 나타내고 있었다. 이때 생성된 황색소는 373 nm에서 단일 흡수 peak를 나타내고 물, 에탄올 등 극성용매에 대한 용해성이 높은 특성을 나타내며 생성된 색소의 대부분이 균체 밖으로 배출되는 특성을 지니고 있었다.
