Comparison of the Genomic Structure of the Heat Shock Protein-88(Hsp88) Genes in the Four Entomopathogenic Fungal Strains, Paecilomyces tenuipes Jocheon-1, P. tenuipes, Cordyceps militaris, and C. pruinosa

  • Comparison on the genomic structure and phylogenetic relationship of the Hsp88 genes from P. tenuipes Jochoen-1, P. tenuipes, C. militaris and C. pruinosa was described. The Hsp88 genes from the three entomopathogenic strains, P. tenuipes Jocheon-1(strain), P. tenuipes(original species), and C. militaris contain the identical genomic structure, namely 5 introns and 6 exons with the length of 13, 62, 32, 1,438, 306, 288 nucleotides encoding 713 amino acid residues, whereas in case of C. pruinosa, it contains 4 introns and 5 exons with the length of 13, 62, 32, 1,744, 288 nucleotides encoding 713 amino acid residues. The genomic DNA length of the Hsp88 genes from P. tenuipes Jocheon-1 and P. tenuipes are both 2,600 nucleotides long in size. The Hsp88 genes from C. militaris and C. pruinosa are 2,582, 2,576 nucleotides long in size, respectively. Hsp88 genes of the P. tenuipes Jochoen-1, P. tenuipes, C. militaris and C. pruinosa also contain the conserved ATP-binding domain. Phylogenetic analysis of the Hsp genes of the four strains tested in this study showed that the fungal Hsp88 is divided into two separate clades, ascomycetes and deutromycete. Within the ascomycetes fungal clade, the P. tenuipes Jochoen-1 and P. tenuipes formed a subgroup, on the other hand, C. militaris and C. pruinosa formed another subgroup. Pair-wise comparison of P. tenuipes Jocheon-1 Hsp88 with those of P. tenuipes, C. militaris and C. pruinosa Hsp88s revealed significant identity in deduced amino acid sequence among these strains. The P. tenuipes Jocheon-1 Hsp88 showed 99% identity with the P. tenuipes, 97% identity with the C. militaris, and 98% identity with the C. pruinosa.

  • The genus Hypomyces contains fungi that grow on mushrooms, including agarics, boletes, and Aphyllophorales. While 53 Hypomyces species have been reported worldwide, only one was in Korea. In this study, two new Korean species were identified as H. luteovirens and H. tubariicola based on morphology and internal transcribed spacer sequencing.

Observation of Soft-Rot Wood Degradation Caused by Higher Ascomyceteous fungi

  • The capability of higher ascomyceteous fungi to cause typical soft-rot decay for wood under laboratory conditions is reviewed and discussed. Fungi tested were extremely active in the decomposition of timbers. Scanning electron micrographs illustrated typical soft-rot decay pattern of higher wood decay ascomycetes, with the exception of H. trugodes that caused white-rot decay. Most of the fungi tested could be grouped as soft-rot fungi that showed typical soft-rot type II. Hypha confined primarily to the resin canals in softwoods or vessel elements in hardwoods and spread tracheid to tracheid via pits of cell wall to cell wall with mechanical force.

Occurrence of Dumontinia tuberosa in Korea

  • Thirty-seven single spore isolates were obtained from specimens of ascomycetous fruiting bodies collected from Mt. Suri, Anyang in Korea. The fungal specimens and isolates were identified as Dumontinia tuberosa based on their morphological and cultural characteristics. This is the first record of this fungus occurring in Korea.

조계산일대의 고등 담자균류 (Fleshy Basidiomycetes in Mt. Jogye)

  • During a week from 6th August in 1976 to survey the fungal flora on higher fleshy fungi around Mt. Jogye 193 specimens were collected and then examined tentatively. As a result fo identification except several troublesome ones it was known that they lie within 66 genera, 110 species, 3 varieties, and 2 forms. Out of them five genera, Eutypa, Sarcosoma in Ascomycetes, and Corticium, Heimiella, Clitopilus in Basidiomycetes were confirmed as unrecorded genera to Korea. And therefrom 26 species and 1 form corresponding almost to the fourth of sum were identified as unrecorded species to Korea, among which Basidiomycetes are provided with descriptions and figures in this survey report.

항진균성 물질을 생산하는 지의류 내생 곰팡이의 선별 및 특성 (Isolation and Characteristics of Endolichenic Fungi Producing Antifungal Compound)

  • 새로운 항진균성물질을 분리할 목적으로 지의류은행에서 107종의 지의류 내생 곰팡이를 분양받아 이들의 항진균성 활성을 조사하였다. Candida albicans를 이용한 항진균 활성검사에서 2종류의 지의류 내생 곰팡이 EL123과 EL156을 최종적으로 선별하였다. 이들은 MYA 배지와 EMM 배지에서 공통적으로 높은 곰팡이 성장저해능을 보였다. 지의류 내생 곰팡이의 5.8S rRNA를 포함하고 있는 internal transcribed spacer 부분을 PCR 증폭 후 nucleotide sequence 분석 및 NCBI Blast 분석 결과 EL123 (JF714250)은 Thielavia microspora와 염기서열이 95%의 유사도를 보였고, EL156 (JF714251)은 Cryptosporiopsis diversispora와 99%의 유사도를 보였으며, 2종 모두 자낭균류(ascomycetes)에 속하였다. 형태적 특성으로는 EL123은 가지 친 형태의 균사가 관찰되었고, EL156은 직선형의 균사가 관찰되었으며, EL156이 EL123보다 액체배양 중에 많은 양의 항진균성 물질을 생성하였다.

Notes on Lichen Genus Buellia De Not. (lichenized Ascomycetes) from South Korea

  • Based on a literature survey and assessment of the important features of lichen genus Buellia (spore shape and size, anatomy of the exciple as well as analysis of the lichen substances), the present paper describes four new records of B. maritima, B. polyspora, B. spuria and B. stellulata from South Korea. Among them, B. maritima and B. polyspora are firstly reported in East Asia including in China, the Korean Peninsula and Japan. Brief description of all the new records along with earlier described species placed under genus Amandinea and Hafellia are provided with their distribution and chemistry. A key to all the Buellia species reported so far from South Korea is also provided.

  • A. oryzae에 있어서 protoplast를 이용한 형질전환이 아닌 세포벽이 부분적으로 분해된 cell을 이용하여 electroporation으로 형질전환시켰고, novozyme234, hemicellulase와 celluclast를 사용하여 형질전환 효율이 어떻게 다른지를 비교 분석 하였다. Hemicellulase를 $^{\sim}10^8$ cell에 처리하여 A. oryzae에서 83 transformants/10ug of DNA를 얻었고, novozyme234와 celluclast를 사용하였을 때는 4.3 transformants/10ug of DNA를 얻었다.

Molecular and Morphological Characterization of Two Novel Species Collected from Soil in Korea

  • Two fungal species of ascomycetes were discovered during the screening of soil microflora from the Gangwon Province in Korea: Didymella chlamydospora sp. nov. (YW23-14) and Microdochium salmonicolor sp. nov. (NC14-294). Morphologically, YW23-14 produces smaller chlamydospores (8.0-17.0 × 7.0-15.0 ㎛) than D. glomerata and D. musae. The strain NC14-294 was characterized by smaller conidiogenous cells (4.9-8.8 × 2.0-3.2 ㎛) compared with the closest strains M. fisheri and M. phragmitis. The detailed descriptions, illustrations, and discussions regarding the morphological and phylogenetical analyses of the closely related species are provided to support the novelty of each species. Thus, YW23-14 and NC14-294 are described here as newly discovered species.

  • 자낭균류에 속하는 100여 개체의 버섯표본을 관찰한 결과 3속 9종의 미기록 속과 종이 동정되었기에 보고하는 바이다. 미기록 속은 땅주발버섯 속(Tarzetta,), 살갗버섯 속(Mollisia), 점버섯속(Hypocrea) 등이다. 미기록 종은 노란긴대주발버섯(Cyathipodia cupuliformis), 솜털긴대주발버섯(Cyathipodia villosa), 갈색땅주발버섯(Tarzetta catinus), 오목땅주발버섯(Tarzetta cupularis), 갈색투구버섯(Cudonia confusa), 작은잎맥섯(Rutsroemia petiolorum), 노란상수리술잔고무버섯(Hymenoscyphus pileatus), 연한살갗버섯(Mollisia ventosa), 점버섯(Hypocrea rufa) 등이다.
