Toxic effects of arsenic on growth, hematological parameters, and plasma components of starry flounder, Platichthys stellatus, at two water temperature conditions

  • The purpose of this study is to investigate the changes in growth, hematological parameters, and plasma components of juvenile starry flounder, Platichthys stellatus, following exposure to varying arsenic concentrations present at different water temperatures. P. stellatus (total length, $15.9{\pm}0.4cm$; body weight, $62.2{\pm}4.2g$) were exposed for 4 weeks to waterborne arsenic (sodium arsenite, As) at 0, 150, 300, and $600{\mu}g/L$ at temperatures of $12^{\circ}C$ and $18^{\circ}C$. Toxic effects of As exposure on P. stellatus were higher at the higher temperature and the growth and hematological parameters measured decreased with increasing arsenic concentration, while the concentration of plasma components measured increased. This indicates that waterborne As exposure and water temperature can cause toxic effects on growth, hematological parameters, and plasma components in Platichthys stellatus.

Investigation of As(III) Sorption by Sand and Alumina under Anoxic Conditions

  • Under anoxic conditions, this study investigated removal of dissolved As(III) by Si and Al oxides including natural sand, chemically washed sand (silica), alumina, and activated alumina. Despite the similar surface area, natural sand showed greater extents of As(III) sorption than chemically washed sand. This was likely due to the high reactivity of Fe(oxyhydr)oxide impurities on the surface of natural sand. For both sands, As(III) sorption was the greatest at pH 7.1, in agreement with the weakly dissociating tendency of arsenous acid. Also, the least sorption was observed at pH 9.6. At basic pH, elevated silicate, which originated from the dissolution of silica in sands, would compete with As(III) for sorption. Due to the highest surface area, activated alumina was found to quantitatively immobilize the initially added As(III) (6.0×10-7-2.0×10-5 M). Alumina showed As(III) sorption compared to or greater than chemically washed sand, although the former had less than 6% of the surface of area the latter. The greater reactivity of alumina than chemically washed sand can be explained by using the shared charge of oxygen.

Arsenic Detoxification by As(III)-Oxidizing Bacteria: A Proposition for Sustainable Environmental Management

  • Arsenic (As), which is ubiquitous throughout the environment, represents a major environmental threat at higher concentration and poses a global public health concern in certain geographic areas. Most of the conventional arsenic remediation techniques that are currently in use have certain limitations. This situation necessitates a potential remediation strategy, and in this regard bioremediation technology is increasingly important. Being the oldest representativse of life on Earth, microbes have developed various strategies to cope with hostile environments containing different toxic metals or metalloids including As. Such conditions prompted the evolution of numerous genetic systems that have enabled many microbes to utilize this metalloid in their metabolic activities. Therefore, within a certain scope bacterial isolates could be helpful for sustainable management of As-contamination. Research interest in microbial As(III) oxidation has increased recently, as oxidation of As(III) to less hazardous As(V) is viewed as a strategy to ameliorate its adverse impact. In this review, the novelty of As(III) oxidation is highlighted and the implication of As(III)-oxidizing microbes in environmental management and their prospects are also discussed. Moreover, future exploitation of As(III)-oxidizing bacteria, as potential plant growth-promoting bacteria, may add agronomic importance to their widespread utilization in managing soil quality and yield output of major field crops, in addition to reducing As accumulation and toxicity in crops.

Stress Granules Inhibit Coxsackievirus B3-Mediated Cell Death via Reduction of Mitochondrial Reactive Oxygen Species and Viral Extracellular Release

  • Stress granules (SGs) are cytoplasmic aggregates of RNA-protein complexes that form in response to various cellular stresses and are known to restrict viral access to host translational machinery. However, the underlying molecular mechanisms of SGs during viral infections require further exploration. In this study, we evaluated the effect of SG formation on cellular responses to coxsackievirus B3 (CVB3) infection. Sodium arsenite (AS)-mediated SG formation suppressed cell death induced by tumor necrosis factor-alpha (TNF-a)/cycloheximide (CHX) treatment in HeLa cells, during which G3BP1, an essential SG component, contributed to the modulation of apoptosis pathways. SG formation in response to AS treatment blocked CVB3-mediated cell death, possibly via the reduction of mitochondrial reactive oxygen species. Furthermore, we examined whether AS treatment would affect small extracellular vesicle (sEV) formation and secretion during CVB3 infection and modulate human monocytic cell (THP-1) response. CVB3-enriched sEVs isolated from HeLa cells were able to infect and replicate THP-1 cells without causing cytotoxicity. Interestingly, sEVs from AS-treated HeLa cells inhibited CVB3 replication in THP-1 cells. These findings suggest that SG formation during CVB3 infection modulates cellular response by inhibiting the release of CVB3-enriched sEVs.

Effect of Vitamin E Supplementation on Hematological and Plasma Biochemical Parameters during Long Term Exposure of Arsenic in Goats

  • The present investigation was designed to determine whether supplementation of different level of vitamin E for 12 months to arsenic exposed goats (50 ppm as sodium arsenite) affords protection against the blood hemato-biochemical parameters caused by the metalloid. A total of 24 crossbred (Alpine${\times}$Beetal) lactating goats were assigned randomly into 4 equal groups (control, $T_1$, $T_2$ and $T_3$) of 6 in each, on the basis of average body weight ($36.10{\pm}0.11$ kg) and milk yield ($1.61{\pm}0.04$ kg/d). The animals in $T_1$, $T_2$ and $T_3$ were given 50 ppm arsenic, while in $T_2$ and $T_3$, additionally; vitamin E at the rate of 100 IU and 150 IU/kg dry matter (DM) respectively was additionally supplemented for the period of 12 months. Hemoglobin (Hb), total leukocyte (TLC) and blood lymphocyte % were decreased (p<0.05) in arsenic fed groups and vitamin E supplementation in the experimental group showed a protective potential. Significant increases (p<0.05) in aspertate transaminase (AST) and alanine transaminase (ALT) activities among arsenic supplemented groups were recorded, however vitamin E supplementation at higher doses showed a protective effect (p<0.05) against AST but in the case of ALT no ameliorating effect was found in either of the doses. Plasma total protein was decreased (p>0.05) but creatinine level was periodically increased in all As supplemented groups and vitamin E supplementation did not produce any protective effect. It can be concluded that arsenic exposure resulted in varying degree of changes in hemato-biochemical parameters and activities of antioxidant enzymes in goats but concomitant treatment with Vitamin E is partially helpful in reducing the burden of arsenic induced effect.

Preliminary Results of Extraction, Separation and Quantitation of Arsenic Species in Food and Dietary Supplements by HPLC-ICP-MS

  • Various extraction procedures were investigated using reference materials and samples to evaluate extraction efficiency and effectiveness. Inductively coupled plasma mass spectrometry (ICP-MS) was used to measure total arsenic and to quantitate arsenic species when coupled to an HPLC (high pressure liquid chromatography). Arsenic species were extracted from rice flour (NIST SRM 1568a) with water/methanol mixtures using accelerated solvent extraction (ASE). Total arsenic extraction efficiency ranged from 42 to 64%, for water and various methanol concentrations. From spinach (NIST SRM 1570), freeze-dried apple, and rice flour (NIST SRM 1568a), arsenic species were extracted with trifluoroacetic acid (TFA) at 100 ${^{\circ}C}$. Total arsenic extraction efficiency was 90% for spinach, 75% for freeze-dried apple, and 83% for rice flour. Enzymatic extraction with alpha-amylase and sonication resulted in extraction efficiency of 104% for rice flour, 98% for freeze-dried apple, and 7% for spinach. Chromatograms of arsenic species extracted by the optimum extraction methods were obtained, and the species were quantified. Arsenite (As(III)), arsenate (As(V)), dimethylarsinic acid (DMA), and monomethylarsonic acid (MMA) were found in the apple sample, and DMA and As(V) in the rice flour sample. As(V) and MMA were found in three herbal dietary supplement samples.

흰쥐 간조직의 세포내 비소결합물질의 특성에 관한 연구 (Studies on the Properties of lntracelluar Arsenic Binding Substances in the Rat Liver)

  • 흰쥐 간세포에 존재하는 비소결합물질의 특성을 조사하기 위하여 4 ppm의 비소를 포함하는 NaASO$_2$수용액의 식수로 15일간 흰쥐에 공급하였다. 비소처리군의 간 cytosol분획의 정상단백 질의 함량은 감소하였으나 8종류의 stress protein의 함량은 증가하였다. Cytosol에 존재하는 비소결합체물질은 한 종류이고,glycine, glutamic acid 및 cysteine의 3종류의 아미노산으로 구성되어있으며 분자량은 500D이었다.Glutathione은 비소와 5:1의 몰비로 결합하였으며 glutathione과 비소의 복합체는 gel filtration chromatography에서 비소결합물질과 같은 이동성을 나타내어 cytosol에 존재하는 미소결합물질은 glutathione으로 추정되었다. Glutathione에 결합한 비소는 미토콘드리아의 호흡, 형태전환 및 팽윤과 수축기능에 있어서 대조군과 별다른 차이를 나타내지 않았다. 이와 같은 결과들로써, 생체에 직접 처리한 비소에 의하여 stress protein합성이 촉진되며, 간 cytosol에 존재하는 비소결합물질은 glutathione으로 추정되고, 비소와 복합체를 형성함으로써 비소의 세포독성에 대한 방어기능을 나타내는 것으로 생각된다.

The separation of arsenic metabolites in urine by high performance liquid chromatography-inductively coupled plasma-mass spectrometry

  • Objectives The purpose of this study was to determine a separation method for each arsenic metabolite in urine by using a high performance liquid chromatography (HPLC)-inductively coupled plasma-mass spectrometer (ICP-MS). Methods Separation of the arsenic metabolites was conducted in urine by using a polymeric anion-exchange (Hamilton PRP X-100, $4.6mm{\times}150mm$, $5{\mu}m$) column on Agilent Technologies 1260 Infinity LC system coupled to Agilent Technologies 7700 series ICP/MS equipment using argon as the plasma gas. Results All five important arsenic metabolites in urine were separated within 16 minutes in the order of arsenobetaine, arsenite, dimethylarsinate, monomethylarsonate and arsenate with detection limits ranging from 0.15 to $0.27{\mu}g/L$ ($40{\mu}L$ injection). We used G-EQUAS No. 52, the German external quality assessment scheme and standard reference material 2669, National Institute of Standard and Technology, to validate our analyses. Conclusions The method for separation of arsenic metabolites in urine was established by using HPLC-ICP-MS. This method contributes to the evaluation of arsenic exposure, health effect assessment and other bio-monitoring studies for arsenic exposure in South Korea.

철 및 망간 산화물로 코팅된 입자활성탄을 이용한 수용액 중 As(III) 및 As(V)의 제거 (Removal of As(III) and As(V) in Aqueous Phases by Fe and Mn Oxides Coated Granular Activated Carbon)

  • 본 연구에서는 수용액상의 비소를 산화 및 흡착기작을 이용하여 제거하기 위해서 철과 망간 산화물로 코팅된 입자 활성탄 (FMOCG)을 제조하고, 이의 표면특성 및 회분식 실험을 통하여 비소제거 특성을 규명하였다. 회분식 실험에서는 네 가지 코팅매질의 비소 산화 및 흡착 동역학, pH 영향, 등온흡착실험을 실시하였다. 코팅매질의 철과 망간 함량은 FMOCG-1(178.12 Fe mg/g, 11.25 Mn mg/g)가 비교적 많은 것으로 나타났다. 비소흡착 동역학을 통하여 As(III)의 경우 산화 및 흡착을 통하여 제거됨을 확인하였다. pH 영향실험 결과 pH가 낮을수록 비소의 제거율이 높아지는 것으로 나타났다. 등온흡착실험 결과는 Langmuir isotherm에 잘 적용되었으며 As(III)보다 As(V)의 흡착량이 약간 높았으며, 최대 흡착량은 1.38~8.44 mg As(III)/g과 2.91~9.63 mg As(V)/g이었다.

Lime based stabilization/solidification (S/S) of arsenic contaminated soils

  • Lime based stabilization/solidification (S/S) can be an effective remediation alternative for the immobilization of arsenic (As) in contaminated soils and sludges. However, the exact immobilization mechanism has not been well established, Based on previous research, As immobilization could be attributed to sorption and/or inclusion in pozzolanic reaction products and/or the formation of calcium-arsenic (Ca-As) precipitates. In this study, suspensions of lime-As were studied in an attempt to elucidate the controlling mechanism of As immobilization in lime treated soils. Aqueous lime-As suspensions (slurries) with varying Ca/As molar ratios (1:1, 1.5:1, 2:1, 2.5:1 and 4:1) were prepared and soluble As concentrations were determined. X-ray diffraction (XRD) analyses were used to establish the resulting mineralogy of crystalline precipitate formation. Depending on the redox state of the As source, different As precipitates were identified. When As (III) was used, the main precipitate formation was Ca-As-O. With As(V) as the source, Ca4(OH)2(AsO4)2${\cdot}$4H2O formed at Ca/As molar ratios greater than 1:1. A significant increase in As (III) immobilization was observed at Ca/As molar ratios greater than 1:1. Similarly, a substantial increase in As (V) immobilization was noted at Ca/As molar ratios greater than or equal to 2.5: 1. This observation was also confirmed by XRD. The effectiveness of both As (III) and As(V) immobilization in these slurries appeared to increase with increasing Ca/As molar ratios.
