Chemical Modification of Rupestonic Acid and Preliminarily In Vitro Antiviral Activity Against Influenza A3 and B Viruses

  • To improve the biological activities of rupestonic acid, 21 new rupestonic acid fatty ester derivatives (2a-2h) and aromatic ester derivatives (2i-2u) were synthesized and preliminarily evaluated for their anti-influenza activity in vitro by the national center for drug screening of China, using the Oseltamivir and Ribavirin as reference drugs. The results showed that 2l ($IC_{50}=0.5{\mu}mol/L$) exhibited potent anti-influenza $A_3$ viral activity among the synthesized compounds and was 10-fold more potent than that of the reference drug Oseltamivir ($IC_{50}=5.1{\mu}mol/L$).

Effects of Elsholtzia splendens and Cirsium japonicum on premenstrual syndrome

  • Premenstrual syndrome is a common chronic disorder in most women of reproductive age. The main symptoms are depression, anxiety, tension, feeling out of control, and mastalgia. In premenstrual syndrome, the effects of aromatic edible Elsholtzia splendens and Cirsium japonicum were investigated for over 3 months in 30 women participants in their twenties. In the Elsholtzia splendens capsule treated group, scores of depression and anxiety were significantly lower than those in the Cirsium japonicum capsule treated group. Moreover, instability of the premenstrual assessment form was significantly decreased in the Elsholtzia splendens capsule treated group. Our results suggest that Elsholtzia splendens could be an effective plant material in relieving symptoms of premenstrual syndrome.

  • There is great demand of nutraceutical secondary metabolites in the world so as to feed the population by improving agricultural production with new varieties of food crops but what is still more important is to add nutrition into the food. Physalis minima, plant of Solanaceae family, contain certain secondary metabolites which enhance the over all metabolic functions of the body. Withasteroids are one of such phytochemicals that are generally regarded as safe. These chemicals are almost monopoly of the plants of Solanaceae. The leaves, stem and roots of P. minima have been examined to yield several steroids, identification of which has been discussed in this paper. These withasteroids contribute to the potential nutraceutical and health function value since incorporation of withanolides in the diet may prevent or decrease the growth of tumors in humans.

  • In order to obtain the basic information for the successful breeding of aromatic tobacco (N. tabacum L.), the combining ability and the mode of inheritance in 28 $F_2$ hybrids by diallel crosses between Korea local and oriental varieties were investigated. Estimates of general combining ability were significant for all characters investigated, and the specific combining ability was significant for stalk height, leaves per plant, leaf length, leaf width and days to flower. The partial dominance was exhibited by all characters investigated as well as additive effects were larger than dominance effects for all characters.

An Efficient In vitro Propagation of Zanthoxylum piperitum DC.

  • A protocol is described for rapid multiplication of Zanthoxylum piperitum DC. (Rutaceae), an important aromatic and medicinal plant, through shoot-tip explant cultures. Murashige and Skoog (MS) medium supplemented with various concentrations of N-6-benzyladenine (BA), N-6-benzylaminopurine (BAP) and thidiazuron (TDZ), in single or in combination with ${\alpha}-naphthaleneacetic$ acid (NAA), was used to determine the rate of shoot proliferation. N-6-benzyladenine (BA) used at 0.5mg/l, was the most effective in initiating multiple shoot proliferation at the rate of 23 microshoots per shoot-tip explants after 40 days of culture. Shoot multiplication increased 1.2-fold in each successive subculture. Induction of rooting (98%) was achieved by transferring the shoots to the same basal medium containing 2 mg/l indole-3-butyric acid (IBA). Plantlets went through a hardening phase in a controlled growth chamber, prior to in vivo transfer. These results represented that possible application for the mass production of plantlets through in vitro culture system of Zanthoxylum piperitum DC.

  • 2-Arylchromones, also known as flavones, are among the most ubiquitous classes of natural products occurring in the plant kingdom. On the other hand, 2-styrylchromones are relatively scarce in nature and only a few compounds has been isolated from the blue-green algae species. Therefore, new 2-arylethenylchromone derivatives ( 4a∼n, 5a∼f) were synthesized by the aldol condensation of 2-methylchromone ( 3) with several aromatic aldehydes in order to evaluate their cytotoxicities using a MTT assay on three tumor cells. 2-Arylethenylchromone derivative 4a showed the significant cytotoxic activities on KB, HL-60 and P-388 cell lines with $IC_{50}$/ values of 25.2, 63.59 and 49.51 $\mu$M, respectively, indicating that 2-arylethenylchromone skeleton has a potential anti-tumor application.

The yield were higher, stalk height, leaf length and leaf width were larger and days to flower was shorter for the Fl hybrids when compared to mid-parent. Estimates of general combining ability were significant for all characters investigated, and specific combining ability were significant for yield, stalk height, leaves Per Plant, leaf width and days to flower. In vr-wr graph, partial dominane was exhibited by all characters investigated.

Conservation of Thymus pallidus Cosson ex Batt. by shoot tip and axillary bud in vitro culture

  • Here, we describe an efficient and rapid protocol for the micropropagation of Thymus pallidus Cosson ex Batt., a very rare medicinal and aromatic plant in Morocco. After seed germination, we tested the effect of different macronutrients, cytokinins alone or in combination with gibberellic acid (GA3) or auxins, on T. pallidus plantlet growth. We found that Margara macronutrients (N30K) had the best effect on the in vitro development of the plantlets. The addition of 0.93 μM/L 1,3-diphenylurea (DPU), 0.46 μM/L adenine (Ad), and 0.46 and 0.93 μM/L kinetin (Kin) resulted in the best shoot multiplication and elongation. In addition, the combination of 0.46 μM/L Kin, DPU, or Ad with gibberellic acid, in particular, 0.46 μM/L Ad + 0.58 μM/L GA3 and 0.46 μM/L Kin + 1.15 μM/L GA3, led to better bud and shoot multiplication. Moreover, the integration of the combinations of 0.46 μM/L Kin and auxins, namely 0.46 μM/L Kin + 2.85 μM/L indole-3-acetic acid (IAA), 0.46 μM/L Kin + 2.85 or 5.71 μM/L indole-3-butyric acid (IBA), and 0.46 μM/L Kin + 0.3 or 0.57 μM/L 1-naphthaleneacetic acid (NAA), in the culture medium led to better root development and optimized aerial growth. Finally, the in vitro plants from the medium containing N30K + 0.46 μM/L Kin + 2.85 μM/L IAA were successfully acclimatized; these plants served as a source for repeating in vitro culture.

한국 야생더덕 수집종의 노지 재배시 생육 특성과 향기성분 조성 (Agronomic Characteristics and Aromatic Compositions of Korean Wild Codonopsis lanceolata Collections Cultivated in Field)

  • 한국산 야생더덕 수집종 자생지의 생육환경과 동위산소 패턴 및 향기성분 조성을 조사하여 얻어진 결과를 요약하면 다음과 같다. 1. 야생더덕 군락지역은 표고 450∼800m에 걸쳐 자생하였고 표고 450m 이하와 800 m이상에서는 발견할 수 없었다. 2. 야생더덕 자생지의 조도량은 1,970∼8,400 Lux로 노지 재배지인 안동의 약 8.3%에 불과한 약광상태였다. 3. 자생지 토양의 이화학적 특성은 pH은 4.8∼6.4정도로 산성을 띠는 토양이 많았으며 유효인산함량은 9∼23ppm으로 매우 낮았고 유기물 함량은 4.0∼32.1%로 일반 밭 토양 2.8%보다 훨씬 높았다. 4. 자생지의 식생군락은 천남성, 곰취, 잔대, 쥐오줌풀 등의 초본류와 단풍나무, 생강나무, 조팝나무, 참나무 등의 목본류가 주로 생육하는 개엽수림하에서 자생하고 있었다. 5. 야생더덕 수집종의 안동 재배지에서 지상부 생육은 개화기가 8월 5일에서 8월 16일로 소백산더덕이 가장 빠른 개화특성을 보였고, 특히 자방수는 일월산 더덕이 평균 3.2개로 다른 지역수집종과 뚜렷한 차이를 나타내었고 종자수는 주왕산 더덕이 평균 121개로 가장 많은 경향을 보였다. 6. 야생더덕 수집종간의 단백질 밴드 및 동위효소 패턴은 뚜렷한 차이가 인정되지 않았으나 동위효소인 peroxidase의 경우 잎 조직은 덕유산 더덕, 뿌리 조직은 학가산 더덕, 덕유산 더덕, 일본산 더덕의 차이가 다소 인정되었고, esterase의 경우 잎 조직에서는 가야산 더덕, 오대산 더덕, 청량산 더덕이 차이를 보였고, 뿌리조직에서는 덕유산 더덕에서 차이가 있었다. 7. 수집종간의 식물정유에 대한 수율은 평균 0.007%인 것에 비하여 덕유산 더덕이 0.009%로 가장 많이 추출되었고, 소백산 더덕을 비롯한 10개 지역의 더덕은 0.004∼0.007% 정도를 보였다. 8. 향기성분 조성에 있어서는 수집종간에 뚜렷한 차이를 보이지 않았으나 지리산 더덕에서는 식품분야에서 식용유지나 가공식품의 산패를 방지하는 등 장기보존용의 물질로 사용되는 저산화 물질인 BHT 성분이 동정되었고 주된 성분은 지방족 알콜류가 11종으로 대부분을 차지하였으며 trans-2-hexanol, cis-3-hexanol, 1-hexanol등의 향기성분은 모든 수집종에서 높은피이크 면적(%)을 나타내어 금후 고방향성 더덕 품종육성을 위한 육종의 기초자료로 유망시되었다.

Pharmacophore Modeling and Molecular Dynamics Simulation to Find the Potent Leads for Aurora Kinase B

  • Identification of the selective chemical features for Aurora-B inhibitors gained much attraction in drug discovery for the treatment of cancer. Hence to identify the Aurora-B critical features various techniques were utilized such as pharmacophore generation, virtual screening, homology modeling, molecular dynamics, and docking. Top ten hypotheses were generated for Aurora-B and Aurora-A. Among ten hypotheses, HypoB1 and HypoA1 were selected as a best hypothesis for Aurora-B and Aurora-A based on cluster analysis and ranking score, respectively. Test set result revealed that ring aromatic (RA) group in HypoB1 plays an essential role in differentiates Aurora-B from Aurora-A inhibitors. Hence, HypoB1 used as 3D query in virtual screening of databases and the hits were sorted out by applying drug-like properties and molecular docking. The molecular docking result revealed that 15 hits have shown strong hydrogen bond interactions with Ala157, Glu155, and Lys106. Hence, we proposed that HypoB1 might be a reasonable hypothesis to retrieve the structurally diverse and selective leads from various databases to inhibit Aurora-B.
