Degradation of Phenanthrene and Pyrene by Burkholderia sp. D5 (Burkholderia sp. D5에 의한 phenanthrene과 pyrene 분해)

  • Burkholderia sp. D5, a polyaromatic hydrocarbons(PAHs)-degrading bacterium, was isolated from oil-contaminated soil. The bacterium could utilize phenanthrene (Phe) as a sole carbon source but could not use pyrene (Pyr). However, the strain could degrade Pyr when a cosubstrate such as yeast extract (YE) was supplemented. The PAH degradation rate of the bacterium was enhanced by the addition of other organic materials such as YE, peptone and glucose. YE was a particularly effective additive in stimulating cell growth as well as PAH degradation. When 1 g-YE/L was supplemented into the basal salt medium (BSM) with 215 mg-Phe/L, the specific growth rate (0.28 h-1) and Phe-degrading rate (29.30 μmol/L/h) were enhanced approximately ten and two times more than those obtained in the BSM with 215 mg-Phe/L, respectively. Through kinetic analysis, the maximum specific growth rate (μmax) and PAH degrading rate (Vmax) for Phe were obtained as 0.34/h and 289 ${\mu}mol$/L/h, respectively. Also, μmax and Vmax for Pyr were 0.27 h-1 and 50 ${\mu}mol$/L/h, respectively. The degradation rates for each Phe (2.20 μmol/L/h) and Pyr (2.18 μmol/L/h) were lower in mixture substrates than in a single substrate (29.30 ${\mu}mol$/L/h and 9.58 ${\mu}mol$/L/h, respectively). Burkholderia sp. D5 can degrade Phe and Pyr contained in soil, and the PAH degradation rates in soil were 20.03 ${\mu}mol$/L/h for Phe and 1.09 ${\mu}mol$/L/h for Pyr.

Microbial degradation and other methods for accelerated degradation the Herbicide Imazapyr (제초제 Imazapyr 의 미생물에 의한 분해 및 기타 방법에 의한 분해 촉진)

  • The microbial degradation, photosensitizer-mediated photolysis, and bioceramic- accelerated degradation of the herbicide imazapyr were investigated using four types of soil. 1. Seven strains of microorganisms isolated from the soil A and the active sludge collected from the waste water disposal plant in CheongJu did not give any distinct degradation products in pure culture. When imazapyr (10ppm) was incubated for 14days with each of the 6strains of the known bacteria, they did not produce any noticeable products, either, suggesting that imazapyr was degraded very little by microorganisms in aqueous media. Meanwhile, when 50ppm of imazapyr was incubated in soil A and B for 6months, a degradation product of m/z 279 was detected. It turned out to be 2-[(1-carbamoyl-1,2-dimethylpropyl)carbamoyl]nicotinic acid, which was formed by the hydrolytic cleavage of the imidazolinone ring and by tautomerism. When imazapyr was exposed to sunlight, degradation rates were 14.6% under the control and 66.0, 76.5, 26.7, and 90.0% in the presence of PS-1 (100ppm), PS-1 (200ppm), PS-2(100ppm), and PS-3(100ppm), respectively, and a degradation product of m/z 149 was tentatively identified in the treatment of PS-1. 2. When soil C and D treated with bioceramic were incubated for 7weeks, the $^{14}C$-activities of $^{14}CO_2$ evolved were 2.03 and 1.12% of the originally applied ones, respectively, whereas those in control soils without bioceramic were 1.88 and 0.82% showing no significant defferences.After 5 weeks, however,the differences in the amounts of $^{14}CO_2$ between the two treatments increased gradually, suggesting the bioceramic effect.

Identification of Major Crude Oils Imported into Korea using Molecular and Stable Carbon Isotopic Compositions (분자지표 및 탄소안정동위원소 조성비를 이용한 국내 수입 주요 원유의 식별)

  • Stable carbon isotope ratio of oil components are known to be unaffected by weathering processes and thus has been widely used to determine the origin of spilled oil. In this study, molecular index and composition of stable carbon isotope in 15 crude oils and petroleum product were analyzed and used as oil fingerprints to determine the discriminating power of each fingerprinting method among target crude oils. Through the fingerprints of alkane distribution only Bintulu and B-C(1%) were distinguishable from other crude oils. The pristane/phytane ratio can classify the crude oils into three groups but differentiation of crude oils within a group was impossible using the ratio. The crude oils of A.L., A.S.L., Foroozan and B-C(1%) were differentiated from the other oils using PAH source recognition indexes of C2D/C2P and C3D/C3P. The usage of 4-mD/1-mD and 2/3-mD/1-mD ratio was able to distinguish A.S.L., Bintulu and Oman from the other crude oils. However the PAH source recognition ratios in the other crude oils were similar and thus they were impractical to be used for source identification among the target crude oils. Stable carbon isotope ratios of alkanes were able to uniquely specify each crude oil in the plot of ${\delta}^{13}C_{C21}$ and ${\delta}^{13}C_{C25}$ except A.L., A.M., Qatar-Marine, B-C(1%). The oil fingerprinting method using stable carbon isotope ratios of individual alkane compounds showed more discriminating power among the target crude oils than the conventional source recognition indexes of PAHs or alkanes.

  • Rice quality is much dependent on the pre-and post harvest management. There are many parameters which influence rice or cooked rice qualitys such as cultivars, climate, soil, harvest time, drying, milling, storage, safety, nutritive value, taste, marketing, eating, cooking conditions, and each nations' food culture. Thus, vice evaluation might not be carried out by only some parameters. Physicochemical evaluation of rice deals with amy-lose content, gelatinizing property, and its relation with taste. The amylose content of good vice in Korea is defined at 17 to 20%. Other parameters considered are as follows; ratio of protein body-1 per total protein amount in relation to taste, and oleic/linoleic acid ratio in relation to storage safety. The rice higher Mg/K ratio is considered as high quality. The optimum value is over 1.5 to 1.6. It was reported that the contents of oligosaccharide, glutamic acid or its derivatives and its proportionalities have high corelation with the taste of rice. Major aromatic compounds in rice have been known as hexanal, acetone, pentanal, butanal, octanal, and heptanal. Recently, it was found that muco-polysaccharides are solubilized during cooking. Cooked rice surface is coated by the muco-polysaccharide. The muco-polysaccharide aye contributing to the consistency and collecting free amino acids and vitamins. Thus, these parameters might be regarded as important items for quality and taste evaluation of rice. Ingredients of rice related with the taste are not confined to the total rice grain. In the internal kernel, starch is main component but nitrogen and mineral compounds are localized at the external kernel. The ingredients related with taste are contained in 91 to 86% part of the outside kernel. For safety that is considered an important evaluation item of rice quality, each residual tolerance limit for agricultural chemicals must be adopted in our country. During drying, rice quality can decline by the reasons of high drying temperature, overdrying, and rapid drying. These result in cracked grain or decolored kernel. Intrinsic enzymes react partially during the rice storage. Because of these enzymes, starch, lipid, or protein can be slowly degraded, resulting in the decline of appearance quality, occurrence of aging aroma, and increased hardness of cooked rice. Milling conditions concerned with quality are paddy quality, milling method, and milling machines. To produce high quality rice, head rice must contain over three fourths of the normal rice kernels, and broken, damaged, colored, and immature kernels must be eliminated. In addition to milling equipment, color sorter and length grader must be installed for the production of such rice. Head rice was examined using the 45 brand rices circulating in Korea, Japan, America, Australia, and China. It was found that the head rice rate of brand rice in our country was approximately 57.4% and 80-86% in foreign countries. In order to develop a rice quality evaluation system, evaluation of technics must be further developed : more detailed measure of qualities, search for taste-related components, creation and grade classification of quality evaluation factors at each management stage of treatment after harvest, evaluation of rice as food material as well as for rice cooking, and method development for simple evaluation and establishment of equation for palatability. On policy concerns, the following must be conducted : development of price discrimination in conformity to rice cultivar and grade under the basis of quality evaluation method, fixation of head rice branding, and introduction of low temperature circulation.

Estimation of Physico-Chemical Characteristics of Domestic Aroma Rice and Foreign Aroma Rice (국내 육성 및 도입 향미자원의 이화학적 특성 평가)

  • This study was conducted to provide the fundamental data based on the analysis of phsyco-chemical characteristics of domestic aroma rice and foreign aroma rice for breeding of aroma rice. The average amylose content of domestic aroma rice and widely cultured domestic traditional rice were 18.1% and 19.0% respectively. The Indica and the Japonica types were 25.6% and 25.0%. In the domestic rice of Tongil and Japonica type, the average protein content were 7.6% and 6.4% respectively. And the average protein content of foreign Indica and Japonica type were 7.6% and 7.0% respectively. And the average protein content of widely cultured domestic traditional rice was 5.6%. The Japonica type of rice shown a low protein content compared with Indica type of rice, and the foreign Indica types of rice exhibited a wide range of protein contents. The average alkali digestive value (ADV) of Tongil type of the domestic aroma rice was about 5.0 and the value of Japonica type was about 6.0. The average ADV of foreign Indica and Japonica type were 4.3 and 5.1 respectively. Also the average ADV of widely cultured domestic aroma rice was about 6.0. The foreign aroma rice were distributed a variable range of ADV value and lower than the both domestic aroma rice and widely cultivated domestic rice. In the whole aspect of the amylogram, the highest viscosity, the lowest viscosity and the last viscosity of the total resources, in domestic aroma rice were clearly lower with the next order, Japonica type of foreign aroma rice, Indica type of foreign aroma rice, Japonica type of domestic aroma rice, widely cultured domestic traditional rice, and Tongil type of domestic aroma rice. It shown the difference aspect of amylograms according to the eco-type of the domestic and foreign aroma and the general rice, and it was distinguishable in difference of the aspect of the amylogram of the endosperm of rice as non-glutinous rice, waxy rice, and middle-waxy rice.

The Solvent Extraction of Univalent Cation Picrates by New Podands (새로운 포단드에 의한 피크린산 일가 양이온 염의 용매추출)

  • Some new podands containing phenyl(B), benzyl(Bz), pyridine(Py), quinoline(Q) and naphthalene(Np) as end-groups, and oxygen(O) and sulfur(S) in ether chains as donor atoms have been synthesized. The univalent cation binding characteristics of these podands have been studied by NMR titration and solvent extraction. By NMR titration we have found that the most of podands form 1:1 complexes with $Ag^+$ ion. Especially, the substituted sulfur atoms in ether chains show the effects to enhance the stabilities. We also carried out the extractions of univalent cation picrates including alkaline metal, $Ag^+$, $Tl^+$ and $NH_4{^-}$ ions from aqueous to chloroform layer by using these podands. We found that the extractabilities of $Ag^+$ ion with the quinoline-containing podands such as, $Q_2O_4$, $Q_2O_5$ and $BQO_5$ were 86.8, 86.6 and 48.0% respectively, but the naphthalene-containing podands such as, $Np_2O_4$ and $Np_2O_5$ extracted quite small amount. Otherwise, in cases of $Bz_2O_3S_2$(89.4%), $B_2O_2S_2$(96.8%), $B_2O_3S_2$(58.9%), $Py_2O_2S_2$(58.8%), $Py_2O_3S_2$(42.1%), and $B_2O_4S$(15.0%), interestingly, $Bz_2O_3S_2$ which have sulfur atoms and benzyl groups showed the highest extraction selectivity for $Ag^+$ ion. This result seems due to not only the strong interaction of $Ag^+$ ion with sulfur donors according to the HSAB theory, but also the effective ${\pi}-{\pi}$ stacking interaction between two aromatic end-groups which is enhanced by the flexible methylene spacing group in benzyl groups instead of phenyl groups. The extraction coefficients gave the similar tendency as the extractabilities and the stabilities. From these results, it could be concluded that the predominant factor affected to extraction coefficients is the stabilities, which are strongly influenced by the structures of podands.

  • For the recent years, considerable efforts have been made to rationalize the techniques of research and monitoring of biological effects of marine pollutants such as heavy metals, polychlorinate biphenyls (PCBs), organochlorine pesticides, polycyclic aromatic hydrocarbons (PAHs), organophosphorus pesticides. This study was designed as a part of efforts to investigate the biochemical pollutant markers for diagnosis of marine pollutions by the changes in lipid components of the flounder (Paralichthys olivaceus) in Yellow Sea of Korea. Homoglobin levels in serum of cultured and wild flounders in Yellow Sea were significantly lower (5 - 15% and 20 -25 % , respectively) than those of wild flounder in Pohang. Triglyceride (TG) contents in serum of cultured flounders in Yellow Sea were 10 - 40% higher than those of wild flounder in Pohang. Total and low density lipoprotein (LDL)-cholesterol contents in serum of flounders in Yellow Sea were significantly 10-15% and 30-35% (cultured), and 10-20% (wild), respectively, higher than those of wild flounder in Pohang. Atherogenic index (AI) and T-Chol/PL rations in serum of cultured and wild flounders in Yellow sea were significantly 20-45% and 10-!5%, 6-35% and 15-35%, respectively, higher than those of wild flounder in pohang. These results suggest that near-coastal waters as well as neritic waters of the Yellow Sea might be affected by pollutant input.

  • Neural stem cells (NSCs) of the central nervous system (CNS) have raised a great interest not only for their importance in basic neural development but also for their therapeutic potentials in neurologically degenerative diseases such as Parkinson's, Alzheimer and stroke. During the CNS development, two molecular cascades determine specification of midbrain dopamine system. In one pathway, FGF-8, sonic hedgehog and transcription factor Nurr1 specify dopamine neurotransmitter phenotype. In the other, transcription factors $Lm{\times}lb\;and\;Pt{\times}3$ are required for induction of dopaminergic neurons. In Nurr1 knockout mouse, tyrosine hydroxylase (TH) positive cells fail to appear in substantia nigra, indicating that Nurr1 is essential in specification of dopaminergic cell phenotype. In this study, we used the immortalized human NSCs retrovirally transduced with Nurr1 gene to probe the Nurr1 mediated mechanism to induce dopamine phenotype. While Nurr1 over-expression alone did not generate dopamine phenotype in NSCs, applications of retinoid and forskolin induced expression of TH and AADC mRNAs. In addition, co-cultures of Nurr1 expressing NSCs with human astrocytes induced a marked increase of TH expression. In this co-culture system, the addition of retinoid and forskolin dramatically increased expression of TH. These results indicate that the immortalized human NSCs with Nurr1 gene could have a clinical utility for cell replacement for the Parkinson patients.

The Effect of Nebulized Frankincense Essential Oil in an OVA-Induced Allergic Asthma Mouse Model (프랑킨센스 에센셜 오일 흡입이 OVA로 유도된 알러지성 천식 모델 생쥐에 미치는 영향)

  • In this study, we investigated the effects of frankincense essential oil (BSEO) on the immune cell change in the lung, BALF and PBMC using a mouse model of asthma. BALB/c mice after intraperitoneal OVA sensitization (day 1) were challenged intratracheally with OVA on day 14. Then, the asthma was induced by repeated OVA inhalation challenged. The asthma induced mice group inhaled 0.3% BSEO for 30 minutes per trial, three times a week, for 8 weeks using the nebulizer. After 12 weeks from the experiment, the mice was killed and the lung, bronchoalveolar lavage fluid (BALF) and peripheral blood mononuclear cell (PBMC) were obtained. Next, the change of immune cells inside the separated tissues was observed to identity the effects of BSEO on the allergic asthma mice. In conclusion, the hypersensitive reaction of airway to the bronchoconstrictor in the allergic asthma induced mice was effectively suppressed in Frankincense group, in Bermagot, Eucalyptus, Chamomile, Marjoram and Frankincense groups, the natural aromatic essential oil groups. Furthermore, it was also confirmed that the weight of lung, total number of alveolus cells and the number of BALF, MNL and DLN increased after inducing allergic asthma were reduced. BSEO suppressed the percentage of $CD3e^+/CD19^-$, $B220^+/CD23^+$ and $CD11b^+/Gr-1^+$ cells in the lung tissue of allergic asthma mice. Moreover, BSEO also reduced the percentage of $CD4^+/CD8^-$, $B220^+/CD23^+$ and $CD3^+/CCR3^+$ cells in BALF. In addition, the percentage of $CD3e^+/CD19^-$, $CD3^+/CD69^+$ and $B220^+/CD23^+$ cells in PBMC was reduced. The results of this study indicate that BSEO would be effective to treat allergic asthma by the immune control suppressing the activity of immune cells in each tissue.

Effect of Shading Practices on the Chemical Compounds and Antioxidant in Aruncus dioicus (차광재배가 눈개승마(Aruncus dioicus var. kamtschaticus)의 성분 및 항산화 특성에 미치는 영향)

  • This study was conducted to determine chemical compounds and antioxidant in Aruncus dioicus. Further, effects of cultural practices such as the shading conditions on the general compounds, essential oils and antioxidants. The chemical compounds and antioxidant activity of edible extracts on the shading cultivation and harvesting time were measured by crude fiber, pigments, higher fatty acids, essential oils and DPPH free radical scavenging ability and activities of SOD and POD in edible shooting parts of Aruncus dioicus. The contents of crude fiber, total chlorophyll and total carotenoids in extracts of edible shooting by shading cultivation and later harvesting were higher than those of non-shading cultivation and early harvesting. Phenolic compounds such as benzoic acid were identified as the aromatic compounds in the edible extracts of Aruncus dioicus. The contents of azulene, benzaldehyde and linalool among those compounds on the cultivation conditions increased in shading cultivation. $IC_{50}$ values of DPPH free radical scavenging activities were high from 6.644 to $14.499{mu}g/m{\ell}$ in extracts of edible parts and those of edible shooting parts was high such as edible shooting cultivated by 60% shading, 30% shading and non-shading, respectively. The activity of POD and SOD in seeds was lower than that of edible shooting and that by shading cultivation was high in extracts cultivated by non-shading. POD activity of extracts on harvesting time was high in earlier harvesting but SOD activity was low. The numbers of isozyme pattern of POD and SOD in seedling showed 7 bands and 3 bands, respectively, especially, bands of POD and SOD in the first year-growing plant did not show and show a difference according to plant positions, respectively.
