Solvent Effects on the Solvolysis of 2-Aryl-1,1-dimethylethyl Bromides

  • Solvolysis rates of substituted 2-aryl-1,1-dimethylethyl bromides (1) were determined in a variety of solvents such as aqueous mixtures of ethanol, acetone, 2,2,2-trifluoroethanol, and also mixtures of ethanol and TFE at 25 ℃, 35 ℃, and 45 ℃. The solvent effects were analyzed in terms of Winstein-Grunwald equation. The solvent effects of 1-4-MeO failed to give a single linear correlation against either Y or YCl (YBr), but exhibited a wide split pattern which could not be related to the solvent nucleophilicity. On the other hand 1-4-CH3 and 1-H gave a fairly good linearity. In the case of 1-4-MeO, a fairly good linearity was observed against YΔ defined from the solvolysis of 4-methoxyneophyl tosylate. It is assumed that resonance interaction between reaction site and aryl-π-system operates to give charge delocalization regardless of the different solvolysis mechanisms. The Hammett-Brown treatment of the solvolytic rate constant of compounds 1 was obtained non-linear two separated lines of - 1.06 to - 1.46, suggesting of mechanistic changeover from kc-ks to kΔ on going from electron-withdrawing to electron-donating substituents as a basis of 4-CH3 group.

멀티 유화 기술 이용 수분산성의 항산화 효능을 함유한 커큐민의 개발 (Development of Curcumin with Anti-Oxidation Effect of Water Dispersibility using Multi-Emulsification Technology)

  • Curcumin is not soluble in water. Therefore, curcumin emulsion that can dissolve well in water were prepared using multi-emulsification technology, and the antioxidant activities and physical properties of emulsion were measured. Although curcumin was not dissolved in water, it was confirmed to be well dispersed in water when prepared in an aqueous dispersion curcumin emulsion. After dissolving curcumin using water and ethanol as solvents, respectively, the DPPH and ABTS radical scavenging abilities of the filtrate and the curcumin emulsion were measured. Because it was not dissolved in water, activities were not shown. However, when curcumin was dissolved in ethanol, the activities increased as the concentration of curcumin increased. On the other hand, when the curcumin emulsion was dissolved in water, it was found to have abilities. The curcumin emulsion was nano-homogenized and the size and distribution of the emulsified spheres were measured. It was confirmed to be nano-sized as it appeared as 9.083 nm/100%. In the results of the DPPH radical and ABTS radical scavenging abilities of curcumin nano-emulsion, it was confirmed that there was no change in the antioxidant abilities. In conclusion, water-dispersible curcumin prepared using multi-emulsification technology, and it was confirmed to exhibit antioxidant activity and emulsion stability.

노루궁뎅이버섯(Hericium erinaceus) 추출공정별 추출물의 대식세포 활성화에 대한 효과 (Effects of the Extracts by Extraction Procedures from Hericium erinaceus on Activation of Macrophage)

  • 다양한 추출공정에 의하여 제조된 노루궁뎅이버섯의 열수 및 50% 에탄올 추출분획이 대식세포의 활성화에 미치는 효과를 마우스의 대식세포주인 RAW264.7 세포를 사용하여 측정하였다. 실험 결과, NO 생산능은 2시간 열수추출한 분획과 60 W에서 50% 에탄올로 3분간 microwave로 추출한 분획이 높았고, 활성산소종(ROS)에 대한 소거활성은 2시간 또는 3시간 열수추출한 분획과 60W에서 50% 에탄올로 3분간 microwave로 추출한 분획 및 0.5% HCl로 추출한 분획에서 높게 나타났다. 50% 에탄올 추출에 있어서 60W, 80W 및 120W에서 3분간 microwave로 추출한 분획들이 RAW264.7 세포의 Candida albicans에 대한 포식활성을 크게 유도한다는 사실을 알았다. 특히 50% 에탄올을 용매로 사용하여 60W에서 3분간 microwave로 추출한 분획은 NO 생산, ROS 소거 및 C. albicans에 대한 포식활성의 전반적인 대식세포의 활성화를 유도하였다. 이 사실은 노루궁뎅이버섯에 있어서 60 W에서 3분간 microwave에 의한 50% 에탄올 추출조건이 함유된 다당류가 관련된 대식세포 활성화 성분의 농화에 유용하게 사용될 수 있음을 보여주었고, 단백다당류가 활성성분으로서 관련되었을 가능성을 시사하였다.

Anti-carcinogenic effects of non-polar components containing licochalcone A in roasted licorice root

  • BACKGROUND/OBJECTIVE: Licorice has been shown to possess cancer chemopreventive effects. However, glycyrrhizin, a major component in licorice, was found to interfere with steroid metabolism and cause edema and hypertension. The roasting process of licorice modifies the chemical composition and converts glycyrrhizin to glycyrrhetinic acid. The purpose of this study was to examine the anti-carcinogenic effects of the ethanol extract of roasted licorice (EERL) and to identify the active compound in EERL. MATERIALS/METHODS: Ethanol and aqueous extracts of roasted and un-roasted licorice were prepared. The active fraction was separated from the methylene chloride (MC)-soluble fraction of EERL and the structure of the purified compound was determined by nuclear magnetic resonance spectroscopy. The anti-carcinogenic effects of licorice extracts and licochalcone A was evaluated using a MTT assay, Western blot, flow cytometry, and two-stage skin carcinogenesis model. RESULTS: EERL was determined to be more potent and efficacious than the ethanol extract of un-roasted licorice in inhibiting the growth of DU145 and MLL prostate cancer cells, as well as HT-29 colon cancer cells. The aqueous extracts of un-roasted and roasted licorice showed minimal effects on cell growth. EERL potently inhibited growth of MCF-7 and MDA-MB-231 breast, B16-F10 melanoma, and A375 and A2058 skin cancer cells, whereas EERL slightly stimulated the growth of normal IEC-6 intestinal epithelial cells and CCD118SK fibroblasts. The MC-soluble fraction was more efficacious than EERL in inhibiting DU145 cell growth. Licochalcone A was isolated from the MC fraction and identified as the active compound of EERL. Both EERL and licochalcone A induced apoptosis of DU145 cells. EERL potently inhibited chemically-induced skin papilloma formation in mice. CONCLUSIONS: Non-polar compounds in EERL exert potent anti-carcinogenic effects, and that roasted rather than un-roasted licorice should be favored as a cancer preventive agent, whether being used as an additive to food or medicine preparations.

투과증발공정을 이용한 물-알코올계에 대한 금속이온이 치환된 이온교환막의 염효과 연구 (Salt Effect of Metal Ion Substituted Membranes for Water-Alcohol Systems Using Pervaporation Processes)

  • PVA/SSA-$H^{+}$으로 제조된 막은 1가 이온 $Li^{+}$, $Na^{+}$, $K^{+}$, 2가 이온 $Mg^{2+}$, $Ca^{2+}$, $Ba^{2+}$, 3가 이온 $Al^{+}$로 치환되었다. 각 금속이온이 치환된 막의 금속이온의 효과를 알아보기 위하여 물-에탄올 및 물-메탄올 혼합용액에 대하여 팽윤도 및 투과증발 실험을 수행하였다. 또한 이들의 치환정도를 알아보기 위해 부분적으로 ESCA 실험을 통하여 알아보았다. 물-에탄올 및 물-메탄올 혼합용액에 대한 팽윤도는 $Li^{+}$> $Na^{+}$>$K^{+}$의 순으로 감소하였는데 이는 금속이온의 `salting-out'효과에 기인하는 것으로 사료되었으며, 2가 및 3가이온의 경우의 팽윤도 결과는 `salting-out' 효과뿐만 아니라 electrostatic 가교 및 금속이온의 반응성에 의해 좌우되는 것으로 사료되었다. 투과증발실험을 통한 투과도면에서는 물-에탄올 용액에 대해 PVA/SSA-$Na^{+}$막에서 최소치를 보였다가 PVA/SSA-$K^{+}$막에서 다시 증가하는 결과를 얻은 반면에 선택도는 반대의 경향을 얻었다. 대표적 분리 결과는 $50^{\circ}C$에서 90%에탄올 수용액에 대하여 투과도는 59 g/$m^{2}$hr, 선택도는 44의 값을 얻었다. 물-메탄올 용액에 대해서는 1가이온의 경우 에탄올 용액과 같은 경향의 결과를 얻었으나 2가 및 3가 이온의 경우 팽윤도 실험 결과와 마찬가지로 `salting-out' 효과 그리고 electrostatic 가교 등에 의해 영향을 받는다고 할 수 있다.

비수용액 전해질에서 전기도금한 니켈-TiO2 복합 도금층의 표면 및 광분해 특성 연구 (Surface and Photolytic Characteristics of Ni-TiO2 Composite Layer Electro-Plated from Non-Aqueous Electrolyte)

  • Composite plating is a method of co-deposition of plating layer with metallic and/or non-metallic particles to improve the plating layer properties such as high corrosion resistance and photolysis of organic compounds. The properties of nickel-ceramic composite plating are significantly depend on the surface characteristics of co-deposited particles as well as the quantity in electrolyte. In this study, Ni-$TiO_2$ composite coating layer was produced by electrodeposition technique from non-aqueous eletrolyte and its surface characteristics as well as photolytic properties were investigated. The amounts of immobilized $TiO_2$ particles increased with increasing the initial $TiO_2$ particles contents in the bath. Samples electroplated with the current density of $0.5\;A/dm^2$ showed the significantly improved homogeneous $TiO_2$ particles distribution. The corrosion resistance of Ni-$TiO_2$ composite coating layer also improved with increaing the amounts of $TiO_2$ particles. Etched sample showed about 10% increased photolytic rate of organic matter compare to that of the non-etched.

Interpretation of Dispersion Phenomena in Grunwald-Winstein Correlation for Solvolyses of Naphthoyl Chloride

  • Solvolyses rate constant of 1- and 2- naphthoyl chlorides (1 and 2) are reported for aqueous binary mixtures with methanol, ethanol, fluorinated alcohol, acetonitrile and dioxane. Kinetic solvent isotope effects (KISE) in methanol and product selectivities (S) of 2-naphthoyl chloride (2) in alcohol-water are also reported. Dispersions in Grunwald-Winstein correlations $(r{\leq}0.901)$ are discussed by multiple regression analysis incorporating ionizing power $(Y_{Cl})$ scale and rate-rate profiles. Major causes for these phenomena are investigated as an aromatic ring solvation effects, in conjunction with weakly nucleophilic solvation effects ($S_N2$ character), for solvolyses of 1 and for solvolyses of 2, as dual reaction channels, described as $S_N1$-$S_N2$ and $S_AN$-$S_N2$ processes. Distinct border lines between the two pathways are derived from solvolyses rates of 2 in 18 solvent using the results of $log(k/k_o)=mY_{Cl}+lN_T+hI$ plot with values of 1.13 for m, 0.37 for l and 0.15 for h value in 5 aqueous fluorinated alcohol mixtures. Using rate-product correlation, the validity of a third order model based on a general base catalyzed by solvent and contribution from these rate constants, $k_{aa},\;k_{aw}$ and $k_{aw}$, are investigated for $S_AN$-$S_N2$ solvolyses of 2 favored in more rich alcohol media and gradual addition of water to alcohol solvent shows a great shift away from stoichiometric solvation to predominantly medium effects. Rate-rate correlation between solvolyses of 2 and trimethyl acetylchloride (5) with alkyl group in the 29 aqueous solvent mixtures shows appreciable linearity (slope = 0.84, r = 0.987), caused by the same pathway ($S_N1$-$S_N2$ process), even if this correlation coincides with appreciable dispersion (different solvation effect).

  • 아세톤, 에탄올, 메탄올 이성분 혼합수용액에서 파라치환된 cinnamoyl chloride의 가용매분해반응을 25$^{\circ}C$에서 수행하였다. 이들 속도자료로부터 Grunwald-Winstein관계식, Hammett 식, PES 모형과 QM모형을 이용하여 해석 하였다. 파라 치환된 cinnamoyl chloride의 일차반응속도상수를 이용하여 Grunwald-Winstein 도시를 한 결과 세 개의 분리된 곡선으로 분산 현상을 보였으며 알코올 수용액에서 큰 m 값을 나타내었다. PES 모형과 QM 모형에 의한 전이상태 변화에 대한 예측으로부터 본 연구에서의 반응은 결합 이탈이 많이 진전된 $S_N1$$S_N2$ 반응으로 진행되고 있음을 알 수 있었다.

약용식물 추출물의 에탄올대사 효소활성에 미치는 영향 (Effect of Medicinal Plant Extracts on the Ethanol-Metabolizing Enzyme Activities)

  • 약용식물의 열수 추출물이 in vitro에서 alcohol dehydrogenase (ADH)와 aldehyde dehydrogenase (ALDH)의 활성에 미치는 영향을 확인하였다. 약용식물에 20배의 증류수를 넣고 $80^{\circ}C$에서 8시간 추출하여 얻어진 추출액을 시료로 사용하였다. 50종의 약용식물 중에서 마늘과 육계 추출물이 숙취해소 천연소재로서의 활용 가능성이 가장 높게 나타났다. 마늘 추출물은 ADH에 비해 ALDH의 활성을 2배 이상 촉진시킴으로써 acetaldehyde의 분해가 잘 되게 하였다. 육계 추출물은 ALDH의 활성에 비해 ADH의 활성을 획기적으로 저해함으로써 acetaldehyde의 생성을 크게 억제하였다. 육계 추출물은 농도에 비례하여 ADH와 ALDH의 활성을 저해하였으며, $45.33{\mu}g/mL$의 농도에서 ADH의 활성을 52.8% 저해하였고 ALDH의 활성을 11.0% 저해하였다.

우리나라 유산양 알코올 양성유의 특징에 관한 연구 (Characteristics of dairy goat milk positive reaction of the alcohol precipitation test in Korea)

  • The objective of the present study was to determine the minimum alcohol (ethanol) concentration that gives rise to the coagulation of goat milk for the alcohol precipitation test, and to evaluate the physical parameters of goat milk which include alcohol and heat stability. A total of 1,295 udder-half milk samples from 648 lactating dairy goats were collected from seven farms in Jeonnam province, Republic of Korea, to determine the alcohol and heat stability. The majority (99.6%) of the samples were coagulated when 70% ethanol was added to the milk, while only 11.0% of the samples were precipitated by the addition of an equal volume of 45% ethanol. With the concentration of 65%, 60%, 55% and 50% aqueous ethanol, 99.2%, 96.8%, 81.0% and 52.8% of the milk samples were coagulated, respectively. Of 1,295 dairy goat milk samples tested for heat stability, 127 (9.8%) were coagulated by boiling. Among the 143 alcohol test-positive udder-half milk samples, 52 (4.0%) were unstable by heat test, while 1,032 (79.7%) of the 1,152 alcohol test-negative milk samples were stable by heat test. According to the results of boiling test, sensitivity and specificity of 45% alcohol precipitation test were 0.3023 (95% CI: 0.2346~0.3772) and 0.9190 (95% CI: 0.9017~0.9344), respectively. The contents of protein and the specific gravity were higher in the milk samples of 45% alcohol test-positive than in those of 45% alcohol test-negative. However, lower levels of lactose and milk urea nitrogen were observed in the milk samples of 45% alcohol test-positive compared to the alcohol test-negative milk samples. The lowest pH values ($6.73{\pm}0.20$) were shown in the 45% alcohol test-negative and heat-unstable milk samples, while the lowest values of somatic cell counts and bacterial counts were shown in the 45% alcohol test-negative and heat-stable milk samples. Results of this study suggest that the alcohol precipitation for dairy goat milk may have to be tested with ethanol concentration less than 45% for the determination of freshness and heat-stability.
