Effects of Culture Condition on Solubilization of Coal by Microorganisms (배양 조건의 변화가 미생물에 의한 석탄의 액화에 미치는 영향)

  • Biosolubilization of an Australian lignite was investigated by using Streptomyces viridosporus and Poria cocos. In order to solubilize coals effectively they were pretreated by nitric acid both in surface and liquid cultures. The optimum growth pH was 7.5 for S. viridosporus and 4.5 for P. cocos. The effects of various carbon, nitrogen and metal sources on overall solubilization were also studied. Solubility increased with the addition of urea for S. viridosporus, and peptone and tryptone for P. cocos. However carbon and metal sources had little or negative effects on solubilization. Maximum amount of coal solubilized was 85%(w/w) in a batch fermentation culture. Extracellular materials produced by micro-organism were found to be responsible for the coal solubilization. Approximately 70 to 80% of coal solubilization was determined to be the result of non-enzymatic reactions, and the rest to be the result of enzymatic reactions. Characteristics of the solubilized coal were compared with those of original coal and pretreated coal by the approximate and ultimate composition analysis, and IR-spectrum analysis. The spectroscopic results showed that the mechanism of coal solubilization was caused by continuous oxidation.

Isolation, identification and immobilized-cell characteristics of a bacterium that produces $N_2$ from $NH{_4}{^+}$ under an aerobic condition

  • To treat wastewater efficiently by a one-step process of nitrogen removal, a new strain of $N_2-producing$ bacteria from $NH{_4}{^+}$ under an aerobic condition was isolated and identified. By 16S-rDNA analysis, the isolate was identified as Enterobacter asburiae with 96% similarity. The isolate shows that the capacity of $N_2$ production under an oxic condition was approximately three times higher than that under an anoxic condition. The optimal conditions (pH, temperature and C/N ratio) of the immobilized isolate for $N_2$ production were found to be 7.0, $30^{\circ}C$ and 5, respectively. Under all the optimum reaction conditions, the removal efficiency of $COD_{Cr}$ and TN reached 56.1 and 60.9%, respectively. The removal rates of $COD_{Cr}$ and TN were highest for the first 2.5 hrs (with the removal $COD_{Cr}$ ratios of 32.1), and afterwards the rates decreased as reaction proceeded. For application of the immobilized isolate to a practical process of ammonium removal, a continuous bioreactor system exhibited a satisfactory performance at HRT of 12.1 hr, in which the effluent concentrations of $NH{_4}{^+}-N$ was measured to be 15.4 mg/L with its removal efficiency of 56.0%. The maximum removal rate of $NH{_4}{^+}-N$ reached 1.6 mg $NH{_4}{^+}-N/L/hr$ at HRT of 12.1 hr (with N loading rate of 0.08 $Kg-N/m^3-carrier/d)$. As a result, the application of the immobilized isolate appears a viable alternative to the nitrification-denitrification processes.

  • Currently, there are several newly developed energy resources for the future to replace petroleum resources such as hydrogen fuel cell, solar cell, wind power, and etc. Among them, solar cell has attracted a worldwide concern, because it has an enormous amount of resources. In general, a study of solar cells can be classified in to an area of bulk type and thin-film type. Inorganic solar cells based on silicon have been tremendously developed in technology and efficiency. However, since there are many lithographic steps, high processing temperature approximately $1000^{\circ}C$, and expensive raw materials, a manufacturing cost of device are nearly reaching a limit. Contrary to those disadvantages, organic solar cells can be manufactured at room temperature. Also, it has many advantages such as a low cost, easy fabrication of thin film, and possible manufacture to a large size. Because it can be made to be flexible, research and development on solar cells are actively in progress for the next generation. ever though an efficiency of the organic solar cell is low compared to that of inorganic one, a continuous study is needed. In this paper, we report optimal device structure obtained by a program simulation for design and development of highly efficient organic photovoltaic cells. we have also compared simulated results to experimental ones.

MONNTORING AIR QUALITY AND ACIDDEPOSITION IN SOUTHERN U.S.

  • Atmospheric monitoring capabilities were established in 1988 by the University of Florida at Duke forest, near Durham. NC: Cary forest, near Gainesville, FL: and Austin forest, near Nacogdoches, TX. Continuous (hourly averaged) measurements of air quality (ozone, nitrogen oxides and sulfur dioxide) and meteorological variables were made at these three low elevation (< 200 meters), rural locations in the southeastern U.S. for more than three years. During the same period at these sites wet and dry acid deposition samples were collected and analyzed on an event and weekly basis, respectively The monitoring locations were selected to determine actual atmospheric exposure indices for southern pine species in support of on-site surrogate exposure chamber studies conducted by Southern Commercial Forest Research Cooperative (SCFRC) investigators. Daily and quarterly averaged ozone maxima were higher (55 ppb) at the northernmost site in the network (Duke forest) in the second and third quarters (spring and summer seasons) and lower (35 ppb) in the first and fourth quarters (winter and fall seasons), when compared to ozone levels at the two southernmost sites (Cary and Austin forests). Seasonal ozone levels at the latter two sites were similar Nitrogen oxieds and sulfur dioxide levels were insignificant (< 5 ppb) most of the time at all sites, although soil emissions of NO at two sites were found to influence nighttime ozone concentrations. Typical maximum quarterly and annual aggregate ozone exposure indices were significantly higher at Duke forest (92.5/259 ppm-hr) than those values observed at the two southern sites (65.6/210 ppm-hr). Acid deposition (wet and dry) components concentrations and deposition fluxes observed at the Duke forest, NC piedmont site, were generally greater, dependent on site and season, than corresponding variables measured at either of the two southern coastal plain sites (Cary and Austin forests). Acid deposition variables observed at the latter two sites were remarkably similar, both qualitatively and quantitatively, although the sites were located 1300 km apart. A comparison of deposition fluxes of elemental nitrogen (NO3, NH4') and sulfur (5042-, SO3) components in wet and dry forms indicated that wet deposition accounts for approximately 70% of the total nitrogen and 73% of the total sulfur input on an annual equivalent basis at all sites.

  • 4D CT is a dynamic volume imaging system of moving organs with an image quality comparable to conventional CT, and is realized with continuous and high-speed cone-beam CT. In order to realize 4D CT, we have developed a novel 2D detector on the basis of the present CT technology, and mounted it on the gantry frame of the state-of-the-art CT-scanner. In the present report we describe the design of the first model of 4D CT-scanner as well as the early results of performance test. The x-ray detector for the 4D CT-scanner is a discrete pixel detector in which pixel data are measured by an independent detector element. The numbers of elements are 912 (channels) ${\times}$ 256 (segments) and the element size is approximately 1mm ${\times}$ 1mm. Data sampling rate is 900views(frames)/sec, and dynamic range of A/D converter is 16bits. The rotation speed of the gantry is l.0sec/rotation. Data transfer system between rotating and stationary parts in the gantry consists of laser diode and photodiode pairs, and achieves net transfer speed of 5Gbps. Volume data of 512${\times}$512${\times}$256 voxels are reconstructed with FDK algorithm by parallel use of 128 microprocessors. Normal volunteers and several phantoms were scanned with the scanner to demonstrate high image quality.

Quantification of Pre-parturition Restlessness in Crated Sows Using Ultrasonic Measurement

  • This study presents a non-video, non-invasive, automatic, on-site monitoring system the system employs ultrasonic transducers to detect behavior in sows before, during and after parturition. An ultrasonic transmitting/receiving (T/R) circuit of 40 kHz was mounted above a conventional parturition bed. The T/R units use ultrasonic time-of-flight (TOF) ranging technology to measure the height of the confined sows at eight predetermined locations. From this data, three momentary postures of the sow are determined, characterized as standing-posture (SP), lateral-lying-posture (LLP) and sitting posture (STP). By examining the frequencies of position switch Stand-Up-Sequence (SUS) between standing-posture (SP), lateral-lying-posture (LLP) and sitting-posture (STP) rate can be determined for the duration of the sow' confinement. Three experimental pureblooded Landrace sows undergoing normal gestation were monitored for the duration of confinement. In agreement with common observation, the sows exhibited increased restlessness as parturition approached. Analysis of the data collected in our study showed a distinct peak in Stand-Up-Sequence (SUS, i.e. the transition from lying laterally to standing up ) and sitting-posture (STP) rate approximately 12 h prior to parturition, the observed peak being 5 to 10 times higher than observed on any other measurement day. It is concluded that the presented methodology is a robust, low-cost, lowlabor method for the continuous remote monitoring of sows and similar large animals for parturition and other behavior. It is suggested that the system could be applied to automatic prediction of sow parturition, with automatic notification of remote management personnel so human attendance at birth could reduce rates of sow and piglet mortality. The results of this study provide a good basis for enhancing automation and reducing costs in large-scale sow husbandry and have applications in the testing of various large mammals for the effects of medications, diets, genetic modifications and environmental factors.

Potential Soil Loss Prediction for Land Resource Management in the Nakdong River Basin (토지자원관리를 위한 낙동강 유역의 잠재적 토양유실량 산정)

  • The purpose of this study is to analyze the potential soil loss and hazard zone by the Revised Universal Soil Loss Equation(RUSLE) for preservation and management of land resources which is the base of ecosystem, and to grasp the relationship between RUSLE factors in the Nakdong River Basin. All thematic maps used in RUSLE are constructed through GIS and spatial analysis method derived from digital topographic maps, detailed soil maps, land-cover maps, and mean annual precipitation of 30 years collected respectively from National Geographic Information Institute, National Institute of Agricultural Science and Technology, and Ministry of Environment. The slope length of LS-factor that takes much times by the study area's wideness was calculated automatically through AML(Arc Macro Language) program developed by Van Remortel et al.(2001, 2003). The results are as follows; First, according to the soil loss estimation by the RUSLE, it shows that approximately 82% of the study area have relatively lower possibility of soil loss which is the 1 ton/ha in annual soil loss. While, 9.4% ($2,228km^2$) needed intensive and continuous management for soil loss. Because the amount of their annual soil loss was greater than 10 ton/ha that is optimum level suggested by Morgan(1995). For these areas, the author believe that a new approach which can minimize environmental impacts from soil loss through improvement of cultivation process and buffer forest zone should be applied. Second, according to the relationship between the RUSLE factors, topographical(LS-factor) and cover management(C-factor) conditions have a lot of influence on soil loss in case of the Nakdong River Basin. However, because of RUSLE factor's influence that affect to soil loss might be different based on the variety of spatial hierarchy and extent, it is necessary to analyze and evaluate factor's relationship in terms of spatial hierarchy and extent through field observations and further studies.

Field and Culture Studies on the Growth and Reproduction of Campylaephora hypnaeoides (석묵 (Campylaephora hypnaeoides)의 생장과 성숙에 대한 야외 및 배양 연구)

  • The phenology of Campylaephora hypnaeoides J. Agardh and optimal conditions for carpospore release, growth and reproduction were examined in the field and in the laboratory from January to December 2007. In the field population of C. hypnaeoides, approximately 50% of the plants were vegetative during the study period. Additionally, the percentages of carposporophytes and tetrasporophytes were maximal in April (37%) and June (57%), respectively. Maximum growth in plant length, dry weight, and hook number coincided with the tetrasporophyte reproductive peak in the field. In culture, carpospore release, sporeling growth and reproduction were affected by environmental factors such as daylength, temperature, and salinity. The liberation of carpospores was maximum under continuous light and at a combination of $15^{\circ}C$ and $10\;{\mu}mol$ photons $m^{-2}\;s^{-1}$. Maximum growth of tetrasporophyte sporelings occurred at a combination of $20\;{\mu}mol$ photons $m^{-2}\;s^{-1}$ of constant light and $25^{\circ}C$. However, the growth of gametophyte sporelings was maximal under $40\;{\mu}mol$ photons $m^{-2}\;s^{-1}$ of constant light and in a combination of $20^{\circ}C$ and 35 psu. The tetrasporophyte sporelings were grew faster than gametophytes, indicating that gametophyte- and tetrasporophyte-sporelings have different physiological responses to irradiance and temperature. Tetrasporangial branches and cystocarps of C. hypnaeoides were produced from carpospores and tetraspores within 1 month, and they were stimulated at high temperature and irradiance levels. In conclusion, C. hypnaeoides should be seeded using carpospores during early winter (November-December) because cystocarps are easily identified by fishermen, and tetrasporophytes grow faster than gametophytes.

Change of Temperature using the Twentieth Century Reanalysis Data (20CR) on Antarctica (20세기 재분석 자료(20CR)를 이용한 남극대륙의 기온 변화)

  • Antarctica is very sensitive to climate change but the number of stations is not sufficient to accurately analyze climate change in this regoin. Model reanalysis data supplements the lack of observation and can be used as long term data to verify climate change. In this study, the 20CR (Twentieth Century Reanalysis) Project data from NCEP/NCAR and monthly mean data (temperature, solar radiation and longwave radiation) from 1871 to 2008, was used to analyze the temperature trend and change in radiation. The 20CR data was used to validate the observation data from Antarctica since 1950 and the correlation coefficients between these data were determined to be over 0.95 at all stations. The temperature increased by approximately $0.23^{\circ}C$/decade during the study period and over $0.20^{\circ}C$/decade over all of the months. This increasing trend was observed throughout the Antarctica and a slight increase was observed in the Antarctic Peninsula. In addition, solar radiation (surface) and longwave radiation (surface and top of atmosphere) trends correlated with the increase in temperature. As a result, outgoing longwave radiation at the surface is attenuated by atmospheric water vapor or clouds and radiation at the top of the atmosphere was reduced. In addition, the absorbed energy in the atmosphere increases the temperature of the atmosphere and surface, and then the heated surface emits more longwave radiation. Eventually these processes are repeated in a positive feedback loop, which results in a continuous rise in temperature.

EXAMINATION OF CALCULATION METHOD FOR THE FLEXURAL RIGIDITY OF CROP STALKS

  • Calculation of the flexural rigidity value (EI) is indispensable for prescription of deflection characteristics of crop stalks in harvesting□Conventionally□EI has been determined by either average EI of the whole stalk or average EI of each stems divided into node through the calculation method of cantilever with homogeneous section□However□deflection characteristics of crop stalks caused by mechanical operation such as combine harvester were not exactly presumed by these conventional EI through the experiment by authors. Further, actual EI of a stalk changes in company with a change of moisture contents as time passes during the experiment. Finally, efficient calculation method for determining EI is needed in order to improve these problems. In this study, mechanical model based on actual structure of the crop stalk with variety sectional area was proposed. This mechanical model is calculated by the theory of cantilever with continuous stages. Therefore, improvement of both calculating accuracy on EI and efficiency of measuring system was tried. At first, this calculation method was applied to piano wire of which EI was recognized in advance. As a result, EI calculated from this new method coincided approximately with piano wire's EI. Next, applying to crop stalks as same as piano wire, relationship between loads acting on crop stalks and deflection values calculated by EI using this new calculation method was exactly presumed in comparison with conventional method. Further, measuring time of deflection test was greatly reduced. Finally, new calculation method of EI will be available for estimating mechanical characteristics of so many kinds of crop stalks in harvesting operation. Further, in this study, new deflection test using image-processing apparatus by computer will be introduced.
