Apolipoprotein B-100 (Apo-B100) is a major protein component for low density lipoproteins (LDL). A number of mimetic peptides of Apo-B100 were screened from the phase-displayed random peptide library by utilizing monoclonal antibody (B9). Mimetic peptide for B9 epitope against apo B-100 was CRNVPPIFNDVYWIAF (pB1). From the BLAST search, the mimetic peptide pB1 had 40% homology with apo B-100. As a result of the structural determination of this mimotope using homo/hetero nuclear 2D-NMR techniques and NMR-based distance geometry (DG)/molecular dynamic (MD) computations, DG structure had low penalty value of 0.3-0.6 ${\AA}^2$ and the total RMSD was 0.5-1.5 ${\AA}. Moreover, pB1 structure included a weak $3_{10}$-helix from $Ile^7$,/TEX> to $Trp^{13}$.
This study was carried out to examine the association of age, nutrient intake, alcohol drinking and smoking on serum lipid and apolipoprotein levels in 100 healthy adults (54 males and 46 females). The serum total cholesterol, low density lipoprotein cholesterol (LDL-C) (p<0.05), apolipoprotein B (Apo B) levels, LDL-C:high density lipoprotein cholesterol (HDL-C) ratio, Apo B:apolipoprotein(Apo A-I) ratio and atherogenic index (AI) (p<0.001) were significantly higher in males than females, but HDL-cholesterol level was significantly lower (p<0.01) in males than females. The plasma ascorbic acid concentration had no difference in both genders. Most of nutrient intakes except Ca, vitamin A and vitamin $B_2$ were higher than RI (Recommended intake) in both males and females. The meal frequency per day was significantly higher (p<0.01) in males than females, while meal speed, the heaviest meal, meal quantity and meal regularity had no significant differences. The rates of drinking and smoking were significantly higher (p<0.01) in males (59.3%, 37.2%) than females (17.3%, 6.9%). The age was positively correlated with triglyceride level in males (p<0.05), and with total cholesterol (p<0.001), LDL-cholesterol (p<0.001), Apo B (p<0.01), LDL-C:HDL-C ratio (p<0.01), Apo B:Apo A-I ratio (p<0.05) and AI (p<0.01) in females. The levels of serum lipid and apolipoprotein were more correlated with protein intake than other macronutrient intakes in males, while those in females were more correlated energy, carbohydrate and lipid intakes than protein intake. The effect of drinking on serum lipid and apolipoprotein levels was bigger in males than females, while the effect of smoking on those was bigger in females than males. The plasma ascorbic acid concentration was positively correlated with HDL-cholesterol (p<0.05), Apo A-I levels (p<0.001), whereas negatively correlated with Apo B:Apo A-I ratio (p<0.05) in males. And that was positively correlated with HDL-cholesterol (p<0.05) in females. These results suggest that moderate macronutrient intake, less alcohol consumption and non-smoking were necessary to maintain healthy lipid profile with aging in adults.

BACKGROUND/OBJECTIVES: This study was aimed at examining the association between dietary flavanones intake and lipid profiles according to the presence of metabolic syndrome (MetS) in Korean women with type 2 diabetes mellitus (T2DM). SUBJECTS/METHODS: A cross-sectional analysis was performed among 502 female T2DM patients (non-MetS group; n = 129, MetS group; n = 373) who were recruited from the Huh's Diabetes Clinic in Seoul, Korea between 2005 and 2011. The dietary intake was assessed by a validated semi-quantitative food frequency questionnaire (FFQ) and the data was analyzed using the Computer Aided Nutritional Analysis program (CAN-Pro) version 4.0 software. The intake of flavanones was estimated on the basis of the flavonoid database. RESULTS: In the multiple linear regression analysis after adjustment for confounding factors, daily flavanones intake was negatively associated with CVD risk factors such as total cholesterol, LDL-cholesterol, and apoB and apoB/apoA1 ratio only in the MetS group but not in the non-MetS group. Multiple logistic regression analysis revealed that the odds ratio for a higher apoB/apoA1 ratio above the median (${\geq}0.74$) was significantly low in the $4^{th}$ quartile compared to that in the $1^{st}$ quartile of dietary flavanones intake [OR: 0.477, 95% CI: 0.255-0.894, P for trend = 0.0377] in the MetS group. CONCLUSIONS: Dietary flavanones intake was inversely associated with the apoB/apoA1 ratio, suggesting a potential protective effect of flavanones against CVD in T2DM women with MetS.
Two molecular species of apolipophorin-III (spoLp-III) were purified from the last instar larval hemolymph of Galleria mellonella by gel permeation chromatography (Sephadex G-100), ion exchange chromatography (DE-52), heat treatment (90C for 30 min) and Mono S FPLC, and were named apoLp-III-a and apoLp-lll-b, respectively. They were indistinguishable by SDS-PAGE but could be separated by native PAGE. The molecular mass of apoLp-III determined by SDS-PAGE was approximately 18 kDa. The N-terminal amino acid sequence of apoLp-III-b revealed high similarities with the apoLp-III from Manduca sexta.
The very low density apolipoprotein-II (apoVLDL-II) gene is closely related with the constitution of the lipoprotein in various tissues. The apoVLDL-II gene have main functions for reducing fat elements from tissues and muscles. Previous results indicated that the polymorphisms in apoVLDL-II gene were positively related with growth and body composition traits in chicken. In this study, we analyzed previously identified apoVLDL-II gene polymorphisms using the PCR-RFLP method and investigated allele and genotype frequencies in three chicken breeds. Data indicated that Korean native chicken and Korean Oge chicken have similar B and F gene frequencies, indicating that this marker can be used for the improvement of growth and body composition traits in those breeds and can be used as marker assisted selection with further verifications.
Apolipoprotein A-I (apoA-I) has anti-inflammatory and anti-oxidative properties. This study was designed to investigate whether apoA-I affects apoptosis and cytokine production of human blood neutrophils in an in vitro culture system. Spontaneous apoptosis of neutrophils was significantly delayed by apoA-I. In addition, high density lipoprotein containing apoA-I also delayed apoptosis of neutrophils. Apoptosis of neutrophils was inhibited by anti-scavenger receptor type B-I antibodies. The amounts of interleukin-8, interferon (IFN)-inducible protein 10 (IP-10), and tumor necrosis factor-$\alpha$ (TNF-$\alpha$) in the supernatants of cultured neutrophils treated with apoA-I were significantly increased. Combined treatment of neutrophils with IFN-$\gamma$ and apoA-I produced higher amounts of IP-10 and TNF-$\alpha$ than did treatment with IFN-$\gamma$ or apoA-I alone. The present study reveals that apoA-I activates neutrophils to produce cytokines and delays spontaneous apoptosis of neutrophils. These findings suggest that apoA-I, although a well-known negative acute-phase protein, has a pro-inflammatory effect in neutrophils.
The purpose of this research was to examine the relationship between the plasma LDL particle size and blood lipid profile, dietary factors and anthropometric values (body mass index, waist circumference and waist/hip ratio). The subjects were 173 adults aged 23 to 81 years, selected from the Outpatient Clinic and Cardiovascular Department of the Seoul Municipal Hospital. Dietary data were obtained using a 3-day food record and analyzed using Korean and US nutrient databases. The subjects were divided into three groups by LDL particle size : type A (large buoyant LDL, > 25.5 nm, n=96), type I (Intermediate LDL,$25.2\leq-\leq25.5$ nm, n=18), and type B (small dense LDL, < 25.2 nm, n=59) groups. The type B group had higher age, waist circumference, and waist/hip ratio (WHR) than the type A and type I groups. Serum concentration of triglyceride, Apo B, LDL/HDL cholesterol ratio and atherogenic index were significantly higher in the type B group as compared to those in the other two groups. HDL cholesterol level and Apo A-I/Apo B ratio were significantly lower in the type B group than the other two groups. The plasma LDL particle size was highly correlated with triglyceride (r= -0.450), Apo B (r= -0.402) and HDL cholesterol (r= 0.418). However, there was no correlation between plasma LDL particle size and dietary intakes. This study showed that small dense LDL was an important biochemical risk factor that was associated with other risk factors.

ApolipophorinIII (apoLp-III) is a protypical exchangeable apolipoprotein that is abundant in hemolymph of many insect species. Its function lies in the stabilization of low-density lipophorin particles (LDLp) crossing the hemocoel in phases of high energy consumption to deliver lipids from the fat body to the flight muscle cells. But, recent studies with naive Galleria mellonella-apoLp-III gave first indication of an unexpected role of that protein in insect immune activation. In this research, we cloned a cDNA encoding putative apoLp-III from the silkworm, Bombyx mori injected with E. coli and characterized its role. We constructed a cDNA library using whole bodies of B. mori larvae injected with E. coli, carried out the differential screening, and selected the up-regulated clones. Among these clones, we focused on a cDNA showing a high sequence similarity to the apolipophorinIII from other insects and analyzed the nucleotide and deduced amino acid sequences. The pupative B. mori Jam123 apoLp-III cDNA contained 1,131 bp encoding 186 amino acid residues. Phylogenetic analysis revealed that the nucleotide and amino acid sequences of the B. mori apoLp-III cDNA formed a highly inclusive subgroup with Bombycidae. But, it was interesting that B. mori Jam123 is closer to B. mandarina than B. mori P50 and B. mori N4. Northern blot analysis showed a signal in the fat body, posterior silkgland and midgut.
This study describes a stable and simple method for the measurement of cholesteryl ester transfer protein(CETP) activities using reconstituted HDL and LDL as substrates. Apolipoproteins (apo) A-I and -B were purified from hog plasma by a new strategy without ultracentrifugation and delipidation. a simple two-step column chromatography was administered. In the first step of phenyl-sepharose CL-4B column chro-matography, hydrophobic plasma proteins were isolated. The most hydrophobic proteins bound to the column appeared to be A-I and apo-B. Contaminat proteins were efficiently eliminated from the sample by washing the column with 0.3M NaCI containing buffer after loading the plasma on the column. Two pure proteins showing each single band on SDS-PSGE of apo A-I and apo-B were individually obtained by a subsequent gel filtration column chromatography(Sephadex G-200). This two-step purification was simple and inexpensive compared to the ultracentrifugation and/or delipidation method that are most commonly used. Reconstituted hight-density lipoproteins(HDL) and low-density lipoproteins(LDL) were prepared using the purified apo A-I and-B, respectively. When these artificially prepared HDL and LDL were used in the assays for CETP as the cholesteryl ester(CE) donor and acceptor respectively, the specific transfer of CE increased up to two fold compared to that used the native HSL and LDL.
To investigate effects of Ixeris dentata EtOH ext. on lowering lipid levels and anti-oxidation activity, hyperlipidemic rats were treated with Ixeris dentata EtOH ext. and lipid levels and anti-oxdation activity were measured. The Ixeris dentata EtOH ext. groups showed low concentration of plasma FFA, plasma triglyceride, plasma total cholesterol, and plasma LDL-cholesterol compared to control group. However, concentration of plasma HDLcholesterol was not significantly different among all the treatment groups. The Ixeris dentata EtOH ext. groups showed lower level of liver total cholesterol, liver triglyceride, plasma TBARS, and liver TBARS than those of control group. The Ixeris dentata EtOH ext. groups also showed higher level of GSH-Px activity, SOD activity, and CAT activity than those of control group. Moreover, the Ext. showed lower level of TNF-${\alpha}$, Apo-B, Apo-E, and leptin expression than those of control group. The results in this study shows that the Ixeris dentata EtOH ext. have positive effect in lowering lipid level, and anti-oxidative activity.
