Enterovirus 71 (EV71) is the predominant cause of hand, foot and mouth disease (HFMD). The antiviral activity of hederasaponin B from Hedera helix against EV71 subgenotypes C3 and C4a was evaluated in vero cells. In the current study, the antiviral activity of hederasaponin B against EV71 C3 and C4a was determined by cytopathic effect (CPE) reduction method and western blot assay. Our results demonstrated that hederasaponin B and 30% ethanol extract of Hedera helix containing hederasaponin B showed significant antiviral activity against EV71 subgenotypes C3 and C4a by reducing the formation of a visible CPE. Hederasaponin B also inhibited the viral VP2 protein expression, suggesting the inhibition of viral capsid protein synthesis.These results suggest that hederasaponin B and Hedera helix extract containing hederasaponin B can be novel drug candidates with broad-spectrum antiviral activity against various subgenotypes of EV71.
Chrysin is a 5,7-dihydroxyflavone and was recently shown to potently inhibit enterovirus 71 (EV71) by suppressing viral 3C protease ($3C^{pro}$ activity. In the current study, we investigated whether chrysin also shows antiviral activity against coxsackievirus B3 (CVB3), which belongs to the same genus (Enterovirus) as EV71, and assessed its ability to prevent the resulting acute pancreatitis and myocarditis. We found that chrysin showed antiviral activity against CVB3 at $10{\mu}M$, but exhibited mild cellular cytotoxicity at $50{\mu}M$, prompting us to synthesize derivatives of chrysin to increase the antiviral activity and reduce its cytotoxicity. Among four 4-substituted benzyl derivatives derived from C(5) benzyl-protected derivatives 7, 9-11 had significant antiviral activity and showed the most potent activity against CVB3 with low cytotoxicity in Vero cells. Intraperitoneal injection of CVB3 in BALB/c mice with $1{\times}10^6TCID_{50}$ (50% tissue culture infective dose) of CVB3 induced acute pancreatitis with ablation of acinar cells and increased serum CXCL1 levels, whereas the daily administration of 9 for 5 days significantly alleviated the pancreatic inflammation and reduced the elevation in serum CXCL1 levels. Collectively, we assessed the anti-CVB3 activities of chrysin and its derivatives, and found that among 4-substituted benzyl derivatives, 9 exhibited the highest activity against CVB3 in vivo, and protected mice from CVB3-induced pancreatic damage, simultaneously lowering serum CXCL1 levels.
Syntheses of new analogues of oxathiin carboxanilide (UC84) and their antiviral activities were described. The heterocyclic carboxylic acids including oxathiins (4), thiazines (9) and dithiins (13) in which the methyl was replaced either by lipophilic trifluoromethyl- or bulky phenylgroup were synthesized starting from $\beta$-keto esters (5). Reaction of 4, 9 and 13 with thionyl chloride followed by treatment of the substituted aniline 22 gave the corresponding carboxanilides (24a~24f). The carboxanilides were subjected to Laweson's reagent the corresponding thiocarboxanilides (24g~24k). The antiviral activities of the synthesized compounds against human immunodeficiency virus type 1 (HIV-1), poliovirus type 1 (PV-1 ), coxsackie B virus type 3 (CoxB-3), vesicular stomatitis virus (VSV) and herpes simplex virus type 1 (HSV-1) were presented. The antiviral activity against HIV-1 of dithiin carboxanilide (24e) was similar with that of UC84 (24a). The corresponding thiocarboxanilides (24g~24k) showed higher inhibitory activity against HIV-1 than the carboxanilides (24a, 24b, 24d, 24e). The compounds in which ether the lipophilic trifluorormethyl substituents (24d, 24f, 24i ,24k) or bulky phenyl substituent is present in the heterocyclic compounds showed lower inhibitory activity than that of the methyl substituents is present in the compounds against the HIV-1. But the trifluoromethylated dithiin (24f) showed higher inhibitory activity against PV-1 and CoxB-3 virus than commercial antiviral agents, ribavirin (RV).
노로바이러스는 인간에게 급성위장염을 일으키는 식중독 바이러스로, 최근 전세계적으로 노로바이러스 식중독 사고가 증가하고 있다. 따라서 본 연구에서는 다양한 생리활성 작용이 있다고 알려진 향신료, 차(茶), 식물성 한약재등 11종의 천연물질을 대상으로 노로바이러스의 대체모델인 feline calicivirus (FCV 에 대한 항바이러스 활성을 조사하였다. 각 추출물에 대한 항바이러스 활성은 end point dilution assay 방법인 50% tissue culture infectious dose ($TCID_{50}$)으로 바이러스 감염가를 측정하였다. 녹차(Camellia sinensis L.) 추출물의 경우 3.13 mg/ml의 농도에서 1 시간만에 FCV가 완전히 불활성화 되었으며, 홍화씨 (Carthamus tinctorius L.) 추출물은 6.25 mg/ml의 농도에서 5시간 만에 바이러스가 불활성화 되어 녹차추출물이 FCV에 대한 항바이러스 활성이 가장 뛰어난 것으로 조사되었으며, 농도 및 반응시간에 유의적으로 항바이러스 활성을 나타내는 것으로 관찰되었다. 녹차 및 홍화씨 추출물 등과 같은 천연식물 자원을 이용하여 FCV에 대한 항바이러스 활성을 조사한 결과는 궁극적으로는 노로바이러스에 의한 식중독 저감을 위한 기초자료에 이용될 수 있을 것으로 기대된다.
We aimed to investigate the antiviral activity of Zanthoxylum species against influenza virus A/WS/33, A/PR/8 and B/Lee/40 used by sulforhodamine B (SRB) assay and the action of leaves extracts of Zanthoxylum piperitum on life cycle of influenza virus A/WS/33. Among the twelve extracts, only the leaf extract of Z. piperitum exhibited strong antiviral activity at low concentration of less than 10${\mu}g/m{\ell}$ with no citotoxicity (50${\mu}g/m{\ell}$) against all of three viruses. In addition, only oseltamivir showed antiviral activity with $IC_{50}$ of 65.3${\mu}g/m{\ell}$ against influenza A/WS/33 among the viruses. Furthermore, the leaf extract of Z. piperitum suppressed infection of influenza virus A/WS/33, when added just prior (-1 hr) or after virus inoculation (0 hr). Leaf extract of Z. piperitum directly affect the infectivity of influenza virus A/WS/33 particles. Therefore, Leaf extract of Z. piperitum exhibited higher antiviral activity against three influenza viruses than that of the oseltamivir, which directly interacts with influenza A/WS/33 particles, affecting the initial stages of infection such as receptor binding and virus entry.
Influenza viruses cause significant morbidity and mortality in humans through epidemics or pandemics. Currently, two classes of anti-influenza virus drugs, M2 ion-channel inhibitors (amantadin and rimantadine) and neuraminidase inhibitors (oseltamivir and zanamivir), have been used for the treatment of the influenza virus infection. Since the resistance to these drugs has been reported, the development of a new antiviral agent is necessary. In this study, we examined the antiviral efficacy of the plant extracts against the influenza A/PR/8/34 infection. In vitro, the antiviral activities of the plant extracts were investigated using the cell-based screening. Three plant extracts, Thuja orientalis, Aster spathulifolius, and Pinus thunbergii, were shown to induce a high cell viability rate after the infection with the influenza A/PR/8/34 virus. The antiviral activity of the plant extracts also increased as a function of the concentration of the extracts and these extracts significantly reduced the visible cytopathic effect caused by virus infections. Furthermore, the treatment with T. orientalis was shown to have a stronger inhibitory effect than that with A. spathulifolius or P. thunbergii. These results may suggest that T. orientalis has anti-influenza A/PR/8/34 activity.
Background: Canine parvoviral enteritis (CPE) is a fatal disease worldwide. The treatment of CPE is based mainly on supportive and symptomatic treatment. Antiviral addition to the treatment may result in a higher survival. Objectives: This study evaluated the effects of antiviral treatments with a standardized treatment (ST) on the clinical and inflammatory response of dogs with naturally occurring CPE. Methods: Twenty-eight dogs with CPE caused by canine parvovirus type 2 were divided randomly into treatment groups. The ST group received fluid, antibiotic, antiemetic, and deworming treatments. The antiviral treatment groups received the same ST with an additional antiviral drug, recombinant feline interferon omega (rFeIFN-ω), oseltamivir (OSEL) or famciclovir (FAM). Results: Compared to the healthy control, the tumor necrosis factor-α, interleukin-1β, interferon (IFN)-α, IFN-γ, haptoglobin, and C-reactive protein values were high (p < 0.05) on day zero. At presentation, mild lymphopenia, neutropenia, and a high neutrophil to lymphocyte (LYM) ratio (NLR) were also observed. Adding rFeIFN-ω to the ST produced the best improvement in the clinical score with a decreased NLR, while leucocytes remained low and inflammatory markers stayed high on day three. The survival rates of the groups were 85.7% in ST+IFN, 71.4% in ST+OSEL, 71.4% in ST+FAM, and 57.1% in ST groups on day seven. Conclusions: Antiviral drugs may be valuable in treating CPE to improve the clinical signs and survival. In addition, the decrease in NLR in favor of LYM may be an indicator of the early prognosis before the improvement of leukocytes, cytokines, and acute phase proteins in CPE.
인수공통 호흡기바이러스인 인플루엔자바이러스 감염으로 인해 공중보건과 가축산업에 심각한 피해가 지속적으로 발생하고 있다. 인플루엔자 백신 접종을 통해 항원형이 일치하는 바이러스 감염에 대해 우수한 방어면역을 제공하고 있으나, 효과적인 바이러스 감염 제어에는 여전히 큰 공백이 존재하고 있다. 다양한 항원형을 갖는 바이러스에 동시방어가 가능한 범용인플루엔자백신 개발과 함께 바이러스 치료효과를 제공하는 항바이러스제의 개발도 중요한 접근법으로 고려되고 있다. 현재 널리 사용되고 있는 인플루엔자 항바이러스제의 불완전한 치료효과와 내성바이러스의 출현 등의 문제들로 인해 식물 유래 천연물의 항바이러스 활성에 대한 관심이 증가하고 있다. 특히, 현재 진행 중인 코로나-19 팬데믹은 범용적인 항바이러스 활성을 갖는 안전하고 효과적인 항바이러스제 개발의 필요성을 뚜렷이 보여준다. 본 리뷰는 현재까지 보고된 천연물의 항인플루엔자바이러스 활성을 요약하였다. 또한, 항바이러스 활성을 갖는 천연물의 바이러스 사멸활성과 면역증강활성을 이용하는 신규 백신개발과 면역증강제 개발 가능성에 대해서도 분석하였다.

본 연구에서는 다중 코팅 폴리에스터(PET) 섬유 여재의 항바이러스 소재 응용 가능성을 고찰하기 위해 금속산화물, 키토산, 및 구리이온의 첨착 조건에 따른 PET의 항균 및 항바이러스 성능을 평가하였다. 항균 물질이 단독으로 코팅된 PET 대비 다중 코팅 PET의 경우 첨착량을 감소시킬 수 있을 뿐만 아니라 배양 후 박테리아가 육안상으로 검출되지 않는데(< 10 CFU/mL) 필요한 균 접촉시간을 단축할 수 있음을 확인하였다. 금속산화물은 촉매반응에 의해 라디칼 등의 산소 활성종을 생성하며, 구리이온은 접촉 살균 효과를 가지면서 산소 활성종에 의한 박테리아와 바이러스의 손상에 기여한다. 키토산은 구리이온의 배위결합을 통한 고정화뿐만 아니라 아민기에 의한 살균 효과로 코팅 PET의 항균 성능 향상 효과를 보였다. 다중 코팅 PET는 대장균과 황색포도상구균에 대한 항균 효과 외에 인플루엔자 A (H1N1)와 SARS-CoV-2에 대해 99.9% 이상의 항바이러스 효과가 있음을 확인하였다. 대면적 roll-to-roll 공정을 통해서도 다중 코팅 PET 섬유 여재의 제조가 가능하고 높은 항바이러스 성능이 유지됨을 보였으며, 이는 공기정화용 필터, 마스크, 및 개인 보호용구과 같은 항바이러스 직물 소재로서 관련 산업에 응용될 수 있음을 시사한다.
