Sensory and Antioxidative Characteristics of AF-343 Containing Salt-reduced Dried Bulgogi Bibimbap (AF-343 함유 저염 건조 불고기 비빔밥의 관능적 항산화적 특성)

  • This study investigated the sensory characteristics and antioxidative activity of the reduced salt dried bulgogi bibimbap containing AF-343, a dandelion-derived compound extract. A sample with a 30% reduced salt had a sodium amount of 659.01 mg, which was significantly lower than that of the sample before the reduction (p<0.05). The textural hardness of the SW (Standard sample with the AF-343), showed the lowest value of $1.03N/cm^2$, indicating that the AF-343-containing sample could have a slightly softer texture than the non-containing sample. In the analytical sensory test, the AF-343-containing samples showed a synergistic effect on the aroma and flavor of bulgogi, with values of 5.7 and 8.4, respectively, which were significantly higher than that of the non-containing sample group (4.3 and 4.4, respectively). The hardness by the sensory test was slightly hard with values of 7.2-8.1 in all samples, indicating that textural improvement was needed. In the acceptance test, all the samples scored with slightly lower values of 4-5 points, highlighting the need for future studies to improve the sensory characteristics of dried bibimbap. The flavonoid contents of AF-343-containing SW and RW sample groups were 68.21 and 64.31 mg GAE/100 g, respectively, which were significantly higher than those of the samples without AF-343 (49.06 and 44.82 mg GAE/100 g, respectively) (p<0.05). The ABTS and DPPH radical scavenging activities were similar to those of the flavonoid contents. As a result, in the production of AF-343-containing reduced-salt dried bulgogi bibimbap, more study on the textural improvement will be needed to achieve better palatability.

Antioxidative and Antimicrobial Activities of Eriobotrya japonica Lindl. Leaf Extracts (비파 잎 추출물의 항산화 및 항균활성)

  • Antioxidative, antimicrobial activities and Raw 264.7 cell viability as cytotoxicity of various solvent extracts from leaf of Eriobotrya japonica Lindl. dried by different methods were investigated for processing as functional ingredient. In DPPH radical scavenging activity, RLE (80% EtOH extract of raw leaf) and FLE (80% EtOH extract of freeze-dried leaf) exhibited strong scavenging effect on $300{\mu}M$ DPPH radical solution (1.71 mg/mL and 2.11 mg/mL for RLE $SC_{50}$ and FLE $SC_{50}$). Also in nitric oxide scavenging activity, RLE and FLE showed strong activities (83.9% and 82.2% in 5 mg/mL sample concentration). Total phenolic compound contents of each extracts were found to be $73.7{\sim}215.4$ mg/g and RLE was showed the highest phenolic compound content. Also, total flavonoid contents were found to be $24.85{\sim}110.3$ mg/g and RLE was showed the highest flavonoid content. In antimicrobial activity, RLE was showed higher growth inhibition effect against all microbial strains. RLE, RLW (hot water extract of raw leaf), and FLW (hot water extract of freeze-dried leaf) exhibited strong antimicrobial activities against MRSA and S. aureus. In measurement of cytotoxicity by MTT assay, Raw 264.7 cell viabilities of 80% EtOH extracts showed better effect than water extracts. Especially viability of RLE was found be over 100% in every tested sample concentration.

Antioxidant Effects and Nitrite Scavenging Ability of Extract from Acanthopanax cortex Shoot (오가피순 추출물의 항산화 효과 및 아질산염 소거능)

  • This study was conducted to examine the antioxidative effect and nitrite scavenging ability of extract from Acanthopanax cortex shoot. The total phenolic compound and flavonoids contents of extract from Acanthopanax corex shoot were $116.33{\pm}6.09mg\;GAE/g$ and $65.07{\pm}4.10mg\;RE/g$, respectively. Antioxidative activities were measured by various in vitro models such as DPPH radical scavenging activity, FRAP, reducing power, ABTS radical scavenging activity, ORAC assay. This results showed that the extract of Acanthopanax cortex shoot was effective in scavenging radicals and protecting oxidation when assessed various in vitro systems. Similarly, the nitrite scavenging ability of extract was increased in a dose-dependent manner. Moreover, ORAC value at a concentration of $0.1mg/m{\ell}$ was $103.4{\pm}5.6{\mu}M\;TE/g$. Considering high consumer demand beneficial health effects, Acanthopanax cortex shoot can be utilized to develop functional food health-promoting and natural antioxidant agents.

Extraction Characteristics and Antioxidative Activity of Cassia tora L. Extracts (결명자 추출물의 추출특성 및 항산화 효과)

  • The present study was conducted to investigate extraction characteristics and antioxidative activity of Cassia tora L. extracts. Cassia tora L. was extracted by reflux extraction under different extraction conditions including solvent. The solid yield, turbidity, color value, titratable acidity, free sugar contents, electron donating ability and superoxide dismutase-like ability of Cassia tora L. extracts were determined. The highest solid yield value was obtained with water of 10 fold. No significant difference in turbidity and color value were found among the extracts prepared with various extraction solvents, 75% ethanol, 50% ethanol and water. The highest titratable acidity was obtained with 50% ethanol of Cassia tora L.. The free sugar contents of Cassia tora L. extracted with water showed the highest value. Cassia tora L. extracts with water included higher contents of free sugar compared with then of the other solvent extracts, 50% ethanol and 75% ethanol extracts. The total polyphenol compound content of Cassia tora L. extracted with 50% ethanol showed the highest value. Cassia tora L. extracts with 50% ethanol included higher contents of total polyphenol compound compared with those of the other solvent extracts, water and 75% ethanol extracts.

  • The present study was conducted to investigate extraction characteristics and antioxidative activity of Schiznadra chinensis extracts. Schiznadra chinensis was extracted by reflux extraction(RE) under different extraction conditions including solvent. The solid yield, turbidity, color value, titratable acidity, free sugar contents, electron donating ability(EDA) and superoxide dismutase(SOD)-like ability of Schiznadra chinensis extracts were determined. The total polyphenol compound content of Schiznadra chinensis extracted with 50% ethanol showed the highest value. Schiznadra chinensis extracts with 50% ethanol included higher contents of total polyphenol compound compared with those of the other solvent extracts, water and 75% ethanol extracts. The electron donating ability of extracts were 60.87% in water, 57.24% in 50% ethanol, and 55.61% in 75% ethanol.

  • The purpose of this study was to evaluated the antioxidative constituents and their activities of the 80% methanolic extracts from fruit of Sorbaria sorbifolia var. stellipila MAX. The isolation of active compound was performed in three steps: solvent partition, open column chromatography, and high-performance liquid chromatography (HPLC). The solvent fractions were tested for their antioxidant activities by oxygen radical absorbance capacity (ORAC). The antioxidant activity of 80% methanolic extracts by various solvent partitions was in the order of 80% MeOH (1.68 ${\pm}$ 0.027), n-hexane (1.02 ${\pm}$ 0.036), $CH_2Cl_2$ (0.95 ${\pm}$ 0.025), EtOAc (1.98 ${\pm}$ 0.065), n-BuOH (1.94 ${\pm}$ 0.054) and Water (1.28 ${\pm}$ 0.032). Therefore, the results indicated that the potential antioxidant activities and functional values were observed significantly at EtOAc fraction from fruit of S. sorbifolia, flavonoid compound isolated.

Validation of Asiaticoside as Marker Compound of Centella asiatica Juice and Extract, and Its Antioxidant Activity (병풀(Centella asiatica) 착즙액과 추출물의 Asiaticoside 분석법 검증 및 항산화 활성)

  • Centella asiatica (C. asiatica) has been widely used in food, cosmetics, and pharmaceutical industry as a functional material. In a previous study, we have investigated not only pharmacological effects such as antioxidative and anti-inflammatory effects, but also analyzed various functional ingredients. In this study, triterpenoids were analyzed using HPLC-DAD to determine marker compounds among functional ingredients. When triterpenoids were analyzed, asiaticoside from C. asiatica was determined as an optimal marker compound. Next, specificity, linearity, limited of detection (LOD), limited of quantification (LOQ), precision, accuracy, and range were evaluated using HPLC-DAD to determine asiaticoside contents in C. asiatica juice and extracts. The specificity was elucidated by chromatogram and retention time using an established analytical method. The coefficient of correlation obtained was 0.9996. LOD was 4.99 ㎍/mL and LOQ was 15.12 ㎍/mL. Intra- and inter-day precision of asiaticoside were determined to be 0.48~1.68% and 0.08~1.09%, respectively. Furthermore, the recovery rate of asiaticoside was 98.88% and the analytical range of Field-70E was determined to be 0.625~10 mg/mL. As a results of evaluating ABTS, DPPH, and FRAP antioxidative effect, Field-70E showed potent antioxidant activities. Results of this study could be used as basic data for quality standardization of C. astiatica juice and extracts.

Chemical Compositions and Antioxidative Activity of Leek (Allium tuberusum) Seeds (부추(Allium tuberusum) 씨의 이화학적 특성과 항산화 활성)

  • The chemical compositions as amino acids, minerals, fatty acids, and total polyphenolic compounds of the seeds of leek (Allium tuberusum) were analyzed. The antioxidative activity of water soluble extract from leek seeds was also tested in DPPH ($\alpha$, $\alpha$ - diphenyl-$\beta$ -picrylhydrazyl) method. The chemical compositions of leek seeds were moisture 4.4%, curde protein 25.7%, crude fat 16.6%, and crude ash 2.9%. Major amino acid compositions were proline 11 g, glutamic acid 4.9 g, arginine 2.1g, aspartic acid 1.6g, leucine 1.3g, valine 1.2 g, and methionine 1.1 g as per 100g. Mineral contents were K 215 ppm, Ca 142 ppm, Fe 124 ppm, and Mg 100 ppm. Major fatty acid compositions were linoleic acid 71.9%, oleic acid 12.7%, palmitic acid 8.6%, and stearic acid 1.4%. The changes of contents in polyphenolic compound from leek seeds caused by heat treatment were also listed in the following order; $20^{\circ}C$(364mg/100g), $40^{\circ}C$(462 mg/100g), and $60^{\circ}C$(551 mg/100g). Antioxidative activity as electron donating ability showed in the following order; 0.05% BHT(butylated hydroxytoluene)(45.6%)>0.05% water-extract(31.3%)>0.1% water extract(30.3%). On the basis of chemical analysis, the leek seedsshowed to have relatively high contents of nutrients as amino acids, minerals, fatty acids.

Antioxidative and Antiaging Effects of Sorbus commixta Twig Extracts (마가목 가지 추출물의 항산화 및 항노화에 관한 연구)

  • In this study, the antioxidative effects and inhibitory effects on tyrosinase and elastase of Sorbus commixta (S. commixta) twig extracts were investigated. The aglycone fraction of S. commixta twig extract showed the prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity($FSC_{50}$, $13{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities of S. commixta twig extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated by the luminol-dependent chemiluminescence assay. The 50 % ethanol extract among extracts showed the most prominent ROS scavenging activity ($OSC_{50}$, $0.189{\mu}g/mL$). The cellular protective effects of extract/fractions of S. commixta twig on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The 50 % ethanol extract and ethyl acetate fraction showed the cellular protective effects against ROS in a concentration dependent manner ($5{\sim}50{\mu}g/mL$). The inhibitory effect of S. commixta twig extract on tyrosinase was investigated to assess the whitening efficacy. The ethyl acetate ($IC_{50}$, $113.2{\mu}g/mL$) and aglycone fraction($IC_{50}$, $105.3{\mu}g/mL$) on tyrosinase showed more remarkable inhibitory effect than arbutin($IC_{50}$, $226.88{\mu}g/mL$), known as the whitening agent. The inhibitory effect of aglycone fraction ($IC_{50}$, $6.9{\mu}g/mL$) on elastase was simillar to quercetin($IC_{50}$, $6.1{\mu}g/mL$), flavonoid known as reference compound. These results indicate that extract/fractions of S. commixta twig can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. S. commixta twig extracts can be applicable to new functional cosmetics for anti-aging products.

Physiological Activities of Dried Persimmon, Fresh Persimmon and Persimmon Leaves (곶감, 생감 및 감잎 추출물의 생리활성 효과)

  • Antioxidative, antidiabetes, antibacterial, anticancer and angiotensin-converting enzyme (ACE) inhibitory activities of methanol extracts of dried persimmon, fresh persimmon and persimmon leaves were investigated. Total polyphenol content of dried persimmon, fresh persimmon and persimmon leaves were 147.79, 301.45 and $315.90\;{\mu}g/mg$, respectively, of which fresh persimmon and persimmon leaves had significantly higher total polyphenol than dried persimmon. Activities of DPPH radical scavenging, lipid peroxidation inhibition and salivary $\alpha$-amylase inhibition were increased in persimmon leaves related to total polyphenol contents. Anticancer activities against AGS of fresh persimmon and persimmon leaves were $65{\sim}70%$; however, there were no significant differences between dried persimmon and fresh persimmon on free radical scavenging activity and inhibitory activity of salivary $\alpha$-amylase. Also, extracts of dried persimmon, fresh persimmon and persimmon leaves showed good ACE inhibitory activities. Dried persimmon and fresh persimmon showed antibacterial activities on E.coli O157:H7. Therefore, there are many difference activities by dried and parts of persimmon. From this result, it is suggested that persimmon leaves is believed to have possible antioxidative, antidiabetes and anticancer capacities by polyphenol, but further studies on the identification of the active compound(s) as antioxidant, antidiabetic, antihypertensive and antibacterial materials will be needed to develop a better understanding of its potency on persimmons.
