Taurine Possesses In vitro Antimutagenic Activity Comparable to Major Antioxidants

  • Taurine is known to suppress oxidant-induced tissue injury by stabilizing biomembrane and scavanging free radicals. The purpose of this study was to determne the antioxidative and antimutabenic acitvities of taurine, ad to compare those acitivities with major antioxidants. For the measurement of antioxidative capacity, 0.05 , 0.1,0.5 and 1.0mg/ml of taurine, L-Ascorbic acid, alpha-tocopherol, and BHT (dibuty hydroxiy toluene)were prepared and tested for their ability to donate electrons to DPPH (1,1-diphenyl-2-picryl-hydrazyl). Antimutagenic acitivity was examined using the Ames salmonela test system at concentrations of 600, 900 and 1200ug/ml. Results indicated that taurine possesses electron-donating capacity, however, the degree of donation was very weak compared to the major antioxidants tested. However, taurine was evaluated as a potent mutation suppressor. Antimutagenic capacity was in increasing order BHT>taurine>L-ascorbic acid>alpha-tocopherol at concentrations of 600 and 900ug/ml. There was a dose-dependent increase in antimutabenicity of these compounds , however, antimutagenity of the 900ug taurie/plate was not significantly differently from that of 1200ug taurine/plate. These results indicate that taurine effectively suppresses the mutagenicity of AFB1 without noticeable elelctron donating ability.

산벚나무 수피 및 코르크층 추출물의 생리활성 비교 (Comparison of the Physiological Activity of Extracts of Bark and Cork Layer from Prunus sargentii)

  • In this study, we investigated on antioxidative activity, antibacterial activity and tyrosinase inhibitory activity in 75% ethanol extracts of bark and cork layer from Prunus sargentii. The total polyphenol content of cork and bark extract was found to be 217.4 mg/g and 184.3 mg/g. And flavonoid content was found to be 31.9 mg/g and 58.6 mg/g respectively. In DPPH radical scavenging ability, cork extract was exhibited stronger scavenging ability than bark extract. Moreover, cork extract was showed higher inhibitory activity than bark extract in tyrosinase inhibitory activity. In antibacterial activity, bark extract was showed higher growth inhibition effect than cork extract against tested bacterial strains. In measurement of cytotoxicity by MTT assay, bark and cork extract showed fine cell viabilities(95.7~120.9%) against RAW 264.7 cell.

토사자, 복분자 및 작약의 항산화성 및 미백효과에 관한 연구 (Antioxidative Properties and Whitening Effects of the Cuscutae Semen, Rubi Fructus and Paeoniae Radix)

  • In an attempt to find natural sources of antioxidants and whitening agents, comparisons of the antioxidative and tyrosinase inhibitory activities of various ethanol extracts of Cuscutae Semen, Rubi Fructus and Paeoniae Radix were carried out. Comparison of the three ethanol extracts revealed that, Paeoniae Radix had the highest electron-donating ability(79.3%),; however, Rubi Fructus had the highest SOD-like ability(31.1%). The xanthine oxidase experiment exhibited a hindrance effect of 74.3% in Cuscutae Semen, 80.4% in Rubi Fructus and 60.8% in Paeoniae Radix. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 20.1% in the Cuscutae Semen, 54.2% in the Rubi Fructus, 56.3% in the Paeoniae Radix. Based on these results, we suggest that the ethanol extracts of Cuscutae Semen, Rubi Fructus and Paeoniae Radix can be used as food and cosmetic ingredients.

Composition of the Essential Oil of Chrysanthemum sibiricum, and Cytotoxic Properties

  • GC-MS data on the volatile oil (CS-oil) of Chrysanthemum sibiricum herbs led to the identification of 2-methoxythioanisol, (+)-camphor, geraniol, citral, thymol, eugenol, ${\beta}-caryophyllene$ oxide, ${\beta}-caryophyllene$, ${\beta}-eudesmol$, juniper camphor together with an unknown substance using the mass spectral library and literature data. CS-oil exhibited significant cytotoxicities on HL-60 $(IC_{50}\;12.5\;{\mu}g/ml)$ cell and mild on HepG-2 cell $(IC_{50}\;102.4\;{\mu}g/ml)$, though the antioxidant ability was found not to be potent $(IC_{50}\;97.2\;{\mu}g/ml)$. However, the component eugenol showed potent antioxidant ability but mild cytotoxicity. Methyleugenol with no phenolic OH showed less potent cytotoxic and antioxidative properties than eugenol suggesting that phenolic OH plays an important role for the cytotoxic and antioxidant abilities. The oil-pretreatment prevented lipid peroxidation induced by bromobenzene in the rat. Therefore, it was demonstrated that CS-oil could be a cytotoxic agent with antioxidant properties.

금속매염제인 초산구리의 세포독성에 대한 자귀나무잎 추출물의 보호 효과 (Protective Effect of Albizzia julibrissin Leaf Extract on the Cytotoxicity Induced by Cupric Acetate Metallic Mordant)

  • Objectives: This study assessed the cytotoxicity of the metallic mordant cupric acetate (CA) and the protective effect of Albizzia julibrissin (AJ) leaf extract on CA-induced cytotoxicity in NIH3T3 fibroblasts. Methods: For this study, cell viability and antioxidative effects such as the inhibitory activity of lipid peroxidation (LP) and superoxide anion-radical (SAR) scavenging activity were assessed. Results: CA significantly decreased cell viability in a dose-dependent manner, and the $XTT_{50}$ value was measured as $55.0{\mu}M$ of CA. The cytotoxicity of CA was determined as highly toxic by Borenfreund and Puerner's toxic criteria. The catalase antioxidant significantly increased cell viability diminished by CA-induced cytotoxicity in these cultures. Regarding the protective effect of AJ leaf extract on CA-induced cytotoxicity, AJ leaf extract remarkably increased the SAR scavenging ability and the inhibitory ability of LP. From these findings, it is suggested that oxidative stress is involved in the cytotoxicity of CA, and AJ leaf extract effectively protected CA-induced cytotoxicity via antioxidative effects. Conclusions: Natural resources like AJ leaf extract may be a putative therapeutic agent for treatment or alleviation of the toxicity induced by CA metallic mordant.

Protective Effect of Aster tataricus L. Extract on the Dermal Cytotoxicity Induced by Sodium Bromate, Oxidant of Hair Dye

  • This study evaluated the dermal cytotoxicity of sodium bromate (NaBrO3) and the protective effect of Aster tataricus L. (AT) extract against NaBrO3-induced cytotoxicity in the cultured NIH3T3 fibroblasts. For this study, it was done the antioxidative effects such as electron donating (ED) activity and lipid peroxidation (LP) activity as well as cell viability. NaBrO3 significantly decreased cell viability in a dose-dependent manner and its XTT50 value was measured at a concentration of 54.4 μM in these cultures. The cytotoxicity of NaBrO3 was determined as highly-toxic by Borenfreund and Puerner's toxic criteria. The quercetin, antioxidant significantly increased cell viability against NaBrO3-induced cytotoxicity. Regarding the protective effect of Aster tataricus (AT) L. extract on NaBrO3-induced cytotoxicity, AT extract significantly increased the cell viability, the ED ability and the inhibitory ability of LP. From these findings, it suggested that the oxidative stress is involved in the cytotoxicity of NaBrO3, and AT extract effectively protected NaBrO3-induced cytotoxicity by antioxidative effects. Conclusively, the natural component like AT extract may be a putative therapeutic agent for the diminution or treatment of the cytotoxicity correlated with oxidative stress like hair dye component, NaBrO3.

Amino-carbonyl 반응에 의한 glucose-poly-${\gamma}$-glutamate (Glu-PGA) 갈변 반응물질의 항산화적 특성 연구 (Antioxidative Characteristics of Browning Reaction Products of Glucose-Poly-${\gamma}$-Glutamate (GIu-PGA) obtained from Amino-carbonyl Reaction)

  • 청국장 점질물인 PGA의 갈변 및 갈변 반응물질의 분획에 따른 갈변도와 전자 공여능에 따른 항산화력을 알아보았다. PGA의 갈변은 자체적으로는 갈변이 잘 일어나지 않고, 염기적 조건 하에서 glucose을 첨가하고 가열하면 갈변이 급격하게 일어나며 전자 공여능 또한 높게 나타났다. 염기적 조건 하에서 얻은 PGA 갈변 반응 물질(Glu-PGA, pH 8.2)을 Sephadex G-50을 이용하여 분획하고, 분획에 따른 갈변도 및 DPPH를 이용한 전자 공여능을 측정하였다. 분획된 갈변 반응 물질들은 비교적 광범위한 갈변도 및 전자 공여능을 보여 주었다. 이들 중 갈변도 및 전자 공여능이 상대적으로 높은 F-7과 F-20의 UV-VIS 흡수대에 있어서는 F-20은 멜라노이딘의 흡수대인 260-320nm에 걸쳐 완만한 흡수곡선을 나타내었고, F-7은 270nm 부근에서 특징적인 최대 흡수대를 나타내었다. 두 분획 물질의 분자량은 SDS-PAGE로 분석한 결과 F-7은 대략 35kDa 이상으로 나타났고, F-20은 SDS-PAGE상에서 뚜렷한 크기는 알 수 없었으나, F-7 보다는 비교적 저분자량을 갖는 물질이라는 것을 알 수 있었다.

찰옥수수 $F_1$의 지방산 조성과 항산화 활성 (Fatty Acid Composition and Antioxidative Activity in Waxy Corn (Zea mays L.) $F_1's$)

  • 우리나라 찰옥수수의 품질을 향상시키는 성분육종의 기초자료를 제공하고자, 310개 $F_1$의 지방산 조성과 항산화 활성, 페놀화합물, tocopherols, carotenoids의 함량을 분석한 결과는 다음과 같다. Palmitic acid와 stearic acid, linoleic acid, 불포화지방산은 각각 $14.6{\pm}2.5%,\;2.5{\pm}1.0%,\;24.8{\pm}4.2%,\;55.6{\pm}5.4%,\;81.8{\pm}3.1%$였고, 항산화 활성과 페놀화합물, tocopherols, carotenoids의 함량은 각각 $22.8{\pm}5.9%,\;36.2{\pm}11.5\;{\mu}g/mL,\;120.3{\pm}27.7\;{\mu}g/mL,\;0.4{\pm}0.6\;{\mu}g/mL$였다. Linoleic acid와 oleic acid 간에는 고도의 부의 상관관계가 있었다. 항산화활성은 페놀화합물과 tocopherols 함량과는 고도의 상관관계가 있었으나, carotenoids의 함량과는 유의성이 없었다

Antioxidative Action of Corni Fructus Aqueous Extract on Kidneys of Diabetic Mice

  • This study investigated the antioxidative action of Corni Fructus aqueous extract on kidneys of diabetic mice. The electron donating abilities of Corni Fructus aqueous extract and its antioxidant activities (XO, SOD, CAT, GST, eNOS) in kidneys of C57BL/6 or db/db mice were evaluated. For in vivo study, seven week-old male mice were divided into normal control group (NC, C57BL/6 mice), diabetic control group (DC, db/db mice) and Corni Fructus (500 mg/kg/day for 8 weeks) treated diabetic group (DCF, db/db mice). The electron donating abilities of Corni Fructus aqueous extract exhibited 7%, 24.4%, and 42.7% at concentrations of 100, 500, and $1000\;{\mu}g/ml$, respectively. The activity of XO in the DCF group was significantly lower than the DC group by 35% (p < 0.05). The SOD activity was significantly higher in the DCF group than the DC group by 26% (p < 0.05). The activities of CAT and GST were lowered in the DCF group than the DC group by 26% (p < 0.05) and 7.6%, respectively. The mRNA expression of eNOS in kidneys was lower in the DCF group than the DC group by 24%. These results indicate that Corni Fructus reduced oxidation stress as evidenced by the restoration of the enzymatic antioxidative defense system in renal tissues of db/db mice. It is suggested that these antioxidative actions of Corni Fructus on renal tissues in db/db mice could contribute to its renoprotective effects on diabetic nephropathy.

Antioxidative and Antiaging Activities and Component Analysis of Lespedeza cuneata G. Don Extracts Fermented with Lactobacillus pentosus

  • Lespedeza cuneata G. Don is a traditional herb that has been associated with multiple biological activities. In this study, we investigated the antioxidative/antiaging activities and performed an active component analysis of the non-fermented and fermented (using Lactobacillus pentosus) extracts of Lespedeza cuneata G. Don. The antioxidative activities of the fermented extract were higher than those of non-fermented extracts. The elastase inhibitory activity, inhibitory effects on UV-induced MMP-1 expression, and ability to promote type I procollagen synthesis were investigated in Hs68 human fibroblasts cells. These tests also revealed that the fermented extract had increased antiaging activities compared with the non-fermented extract. A component analysis of the ethyl acetate fractions of non-fermented and fermented extracts was performed using TLC, HPLC, and LC/ESI-MS/MS to observe changes in the components before and after fermentation. Six components that were different before and after fermentation were investigated. It was thought that kaempferol and quercetin were converted from kaempferol glucosides and quercetin glucosides, respectively, via bioconversion with the fermentation strain. These results indicate that the fermented extract of L. cuneata G. Don has potential for use as a natural cosmetic material with antioxidative and antiaging effects.
