Evaluation of Antioxidantive and Antimicrobial Activities of Garlic Stem and Red Cabbage, and Their Application to Pork Patties during Refrigerated Storage (붉은 양배추와 마늘종 추출물의 항산화 및 항균활성 평가와 냉장저장 중 돈육패티에 이용)

  • This study was carried out to investigate antioxidative and antimicrobial activities of ethanol extracted garlic stem and red cabbage, and their application to pork patties during refrigerated storage. Total phenolic compounds of ethanol extracted garlic stem and red cabbage were 3.13 g/100 g and 3.99 g/100 g, respectively. 1,1'-diphenyl-2-picryhdrazyl (DPPH) radical scavenging activity of garlic stem extracts increased up to 0.05%. Iron chelating activities of both garlic stem and red cabbage increased up to 0.5%, and red cabbage up to the concentration of 1% had similar iron chelating activity to those of EDTA. Pork patties containing garlic stem had higher Hunter L and b values, (p<0.05), whereas those with red cabbage had highest Hunter a values, resulting in similar value to those of control (p>0.05). Thiobarbituric acid reactive substances (TBARS) of pork patties with garlic stem were lower than those of the control (p<0.05). In addition, antioxidantive activities of pork patties with red cabbage were higher than those with garlic stem. No differences in microbial contents were observed between the control and treatments. In conclusion, garlic stem and red cabbage extracts might be used as natural antioxidants to be used in the manufacture of functional meat products.

Antioxidant and Antimicrobial Activities of Various Citrus Peels (감귤류 종류에 따른 과피의 산화방지 및 항균 활성)

  • To investigate the functional activity of different citrus fruit peels, antioxidant compounds in 70% ethanol extracts of mandarin, lemon, orange, and grapefruit peel powders were identified, and antioxidant and antibacterial activities were quantitated. Mandarin peel contained the highest content of total phenolic compounds and total flavonoid substances (21.46±0.12 mg GAE/g and 11.57±0.05 mg RE/g, respectively). The total phenolic compound content of the three other citrus fruits was 14.16±0.18-18.44±0.07, and their total flavonoid content was 5.51±0.10-7.46±0.09 mg RE/g. DPPH radical scavenging activity was the highest in lemon peel (87.64±0.21%), and mandarin peel displayed the best antioxidant activity with respective ABTS radical scavenging activity and FRAP measurements of 43.20±0.61% and 78.82±1.06 mM TE/g. Grapefruit peel antimicrobial activity increased with treatment time, and was the most potent among the four tested citrus species, inhibiting Staphylococcus aureus by about 4.05 log cycle. These findings demonstrate that mandarin and grapefruit peel can be used to prevent oxidation, improve food storage capabilities, and potentially preserve food quality.

Antimicrobial Activity of Seeds of Zanthoxylum piperitum against Oral Pathogen Streptococcus mutans (Zanthoxylum piperitum 종자의 치아우식균 Streptococcus mutans에 대한 항균활성)

  • Antimicrobial activity of Zanthoxylum piperitum was investigated against Streptococcus mutans that causes dental caries. Although the methylene chloride extract of seeds exhibited higher antimicrobial activity than other organic solvent extracts, including methanol, ethyl acetate, and hexane extracts of pericarps or seeds of Z. piperitum, essential oils prepared from both seeds and pericarps possessed more potent inhibitory activity than the methylene chloride extract of seeds. The minimal inhibitory concentrations (MICs) of the essential oils of seeds and pericarps were 0.3 mg/ml and 4.0 mg/ml against S. mutans, respectively. When the seed essential oil was further separated into seven fractions (CS-SD-A${\sim}$CS-SD-G) by thin layer chromatography (TLC), all fractions exhibited lower antimicrobial activity than the essential oil. To understand the antimicrobial ingredients of Z. piperitum, seeds the gas chromatography-mass spectrometry (GC-MS) data of the methylene chloride extract of seeds was compared with those of the seed essential oil (CS-SD). Whereas the methylene chloride extract of seeds contained carvacrol (0.24%), ${\beta}$-caryophyllene (1.72%), and ${\alpha}$-humulene (0.88%), which were previously known to inhibit growth of S. mutans, the seed essential oil contained sabinene (1.57%), linalool (1.55%), citronellal (13.67%), terpinene-4-ol (0.45%), citronellol (3.69%), geraniol (0.9%), linalyl acetate (1.35%), ${\beta}$-caryophyllene (1.35%), ${\alpha}$-humulene (0.78%), and ${\delta}$-cadinene (0.67%) in this regard. These results indicate that Z. piperitum seeds possess various inhibitory substances against S. mutans, and an effective method to isolate the active ingredients from the seeds is to prepare the essential oil. These results also suggest that the essential oil of Z. piperitium seeds may be applicable to preventing dental caries.

Characterization of Bacteriocin-Like Substances Produced by Bacillus subtilis MJP1 (Bacillus subtilis MJP1이 생산하는 Bacteriocin-Like Substances)

  • The MJP1 bacterial strain, which possesses antifungal activity, was isolated from meju and identified as Bacillus subtilis based on its morphological and biochemical properties, as well as its 16S rRNA sequence. Antimicrobial activity was found against various species of Gram-positive bacteria, yeasts, and molds, including food-spoilage microorganisms. The antifungal activity was found to be stable after heat and proteolytic enzyme treatment, and in the pH range of $6.0{\sim}10.0$. The antibacterial activity was stable in the pH range of $6.0{\sim}10.0$, but about 50% of the activity was lost after 24 hr at $30^{\circ}C$. The antibacterial compound was also inactivated by proteolytic enzyme treatment, indicating its proteinaceous nature. The apparent molecular masses of the partially purified antifungal and antibacterial compounds, as indicated by using the direct detection method in Tricine-SDS-PAGE, were approximately 2.4 kDa and 4.5 kDa, respectively. These studies suggest that B. subtilis MJP1 produces two bacteriocin-like substances with antifungal and antibacterial activities.

Isolation and Structure Identification of Antibacterial Substances from the Rhizome of Zingiber mioga Roscoe (양하의 근경에서 항균성 물질 분리 및 구조동정)

  • In order to isolate antibacterial substances from the rhizome of Zingiber mioga Roscoe, the ethanol extracts was fractionated according to the activity against Bacillus subtilis, B. cereus and Staphylococcus aureus. Three antibacterial substances were isolated and purified by column chromatography and recrystallization. Compounds I and III showed activity against all the tested bacterias and compound II exhibited the activity against B. subtilis and B. cereus S. aureus. Compound I was examined antimicrobial activity against B. subtilis, B. cereus and S. aureus by optical density using Bioscreen C. Compound I showed strong growth inhibition at 10 ppm on B. subtilis and B. cereus for 72 hrs, and at 25 ppm on S. aureus. On the basis of spectrometric studies including $1^H-NMR$, ${13}^C-NMR$, DEPT, IH-lH COSY, HMQC, HMBC and IR, compounds I, II and III were identified as $(E)-8{\beta}(17)-epoxylabd-12-ene-15,16-dial\;(C_{20}H_{30}O_3,\;MW=318)$, galanolactone $(C_{20}H_{30}O_3\;MW=318)$ and galanal A $(C_{20}H_{30}O_3,\;MW=318)$, respectively. These results are the first reports on the isolation of $(E)-8{\beta}(17)-epoxylabd-12-ene-15,16-dial, galanolactone and galanal A from the rhizome of Zingiber mioga.

Effect of antibacterial substances produced by probiotic lactic acid bacteria on histamine formation in rennet curd (렌넷 커드 내 히스타민 생성에 관한 프로바이오틱 유산균이 생산한 항균 물질의 영향)

  • Purpose of the present study was to investigate the factors affecting the production of antibacterial substances and histamine in rennet curd prepared by inoculation of histamine-producing lactic acid bacteria (LAB) and probiotic LAB. Probiotic Lactobacillus sakei PIL52 and Lactobacillus plantarum FIL20 produced strong antimicrobial agents against histamine-producing bacteria Lactobacillus brevis LAS129, Enterococcus faecium SBP12, and Enterococcus faecalis SBP58. The lactic acid and crude bacteriocin produced from the probiotic LAB inhibited histamine-producing bacteria in a concentration-dependent manner. As the number of probiotic LAB inoculated for the production of rennet curd increased, the antibacterial activity against histamine-producing bacteria was elevated due to the increased amount of lactic acid and crude bacteriocin in the sample. The growth of probiotic LAB as well as histamine-producing bacteria was inhibited by addition of 10% NaCl, thus the antibacterial substances and histamine contents in rennet curd were significantly lower than those of the control (P < 0.05). Meanwhile, the histamine content was not significantly increased when the rennet curd prepared by mixing probiotic LAB and histamine-producing bacteria was stored at $25^{\circ}C$ for 5 days. However, the amount of histamine detected in the rennet curd was significantly (P < 0.05) increased because the antibacterial activity of the bacteriocin produced by the probiotic LAB was decreased at $20^{\circ}C$ for 20 days.

Detection of residual antibiotic and sulfonamide agents in emergency slaughtered cattle (절박 도축우의 항생제 및 설파제 잔류 조사)

  • This survey was carried out to detect the residual antibiotics and sulfonamides in emergency slaughtered cattle(n=265) from slaughter houses in Kyeonggi province by EEC 4-plates method, Charm II and HPLC. The results were summarized as follows ; 1. Antimicrobial substances were detected from 24 samples(9.1%) by EEC 4-plates method and the detection ratio were highest in summer(13.8%). 2. Twenty-two of the 24 positive samples were classified as sulfonamide(34.4%), TCs(31.3%), $\beta$-lactam(23.5%) and aminoglycoside(9.3%) by Charm II test. 13(59.1%) of the 22 samples contained single agent and 9 samples(40.9%) contained 2 or more agents. 3. Oxytetracycline(27.3%), penicillin G (27.3%) and sulfathiazole(18.2%) were detected in 20 from 22 samples by the HPLC and Charm II test. 4. The residual concentration of oxytetracycline, penicillin G and sulfonamide were 0.29~9.30 ppm, 0.05~9.58 ppm and 0.04~7.59 ppm, respectively and 19 samples(7.2%) were exceeded tolerance levels.

Manufacturing and Antibacterial Characteristics of Functional Non-woven Fabrics Including Nano-silver Particles (은 나노 입자를 함유한 기능성 부직포의 제조와 항균특성)

  • The functional non-woven fabrics have been applied in various industry fields, such as clothing, hygiene, environment, medical and so forth. The functional non-woven fabrics were manufactured by meltblown and finishing processes. These functional non-woven fabrics were based on 5wt% masterbatch using 2,000ppm nano-silver resin composite. Silver is one of the most universal antimicrobial substances. Nano-technology enables us to expand the surface area of silver particles markedly. Silver nano particles were successfully produced less than 50nm in size. The functional non-woven fabrics including nano-silver particles showed excellent antibacterial activities against Staphylococcus aureus (ATCC 6538) and Klebsieila pneumoniae (ATCC 4352). From the results, functional non-woven fabrics including silver nano particels probably will be available as a good and safe antibiotic alternative, such as mask medium filter, water purifier filter, hygiene wet tissues, marine products pad and so forth.

Construction of Probability Identification Matrix and Selective Medium for Acidophilic Actinomycetes Using Numerical Classification Data

  • A probability identification matrix of acidophilic Streptomyces was constructed. The phenetic data of the strains were derived from numerical classification described by Seong et al. The minimum number of diagnostic characters was determined using computer programs for calculation of different separation indices. The resulting matrix consisted of 25 clusters versus 53 characters. Theoretical evaluation of this matrix was achieved by estimating the chuster overlap and the identification scores for the Hypothetical Median Organisms (HMO) and for the representatives of each cluster. Cluster overlap was found to be relatively small. Identification scores for the HMO and the randomly selected representatives of each cluster were satisfactory. The matrix was assessed practically by applying the matrix to the identification of unknown isolates. Of the unknown isolates, 71.9% were clearly identified to one of eight clusters. The numerical classification data was also used to design a selective isolation medium for antibiotic-producing organisms. Four chemical substances including 2 antibiotics were determined by the DLACHAR program as diagnostic for the isolation of target organisms which have antimicrobial activity against Micrococcus luteus. It was possible to detect the increased rate of selective isolation on the synthesized medium. Theresults show that the numerical phenetic data can be applied to a variety of purposes, such as construction of identification matrix and selective isolation medium for acidophilic antinomycetes.

Purification and Characterization of Antioxidant Substance from the Stem Bark of Rhus verniciflua (옻나무 껍질에서 항산화물질의 정제와 특성)

  • In order to isolate antioxidant substances from Rhus verniciflua (RV) , the dried stem bark was extracted with water. The crude water extracts was purified by using HPLC method with a DEAE (anionic type) , CN and ODS column. The purified compound remained stable at pH 3.0∼6.0. but unstable above pH 6.5. It was stable at 100$^{\circ}C$ for 4 hours, but still had about 80% of residual activity after treatment at 100$^{\circ}C$ for 5 hours. In antimicrobial test, no inhibition was observed against Gram-positive and negative bacteria. This compound was stronger than that of commercial antioxidant showed that DPPH test, such as BHT, BHC at the same concentration (20 $\mu\textrm{g}$/ml) .
