Characterization of Bacteriocin produced Lactobacillus bulgaricus acting on bovine mastitis pathogens

  • The antimicrobial substance produced by Lactobacillus bulgaricus was inactivated by pretense, which confirmed it as a bacteriocin and referred to 'bulgaricin HJ'. The bulgaricin HJ showed the inhibitory activity against mastitis pathogens, gram-positive and gram-negative bacteria. The optimal conditions for the production of bulgaricin HJ were at the temperature of $30^{\circ}C$ and 10 h after cultivation of L. bulgaricus. Staph. and Strep. agalactiae, common bovine mastitis pathogens, were treated with bulgaricin HJ by the agar well diffusion method and showed antimirobial activities to the bovine mastitis pathogens. The activity of the bulgaricin HJ was maintained at pH 6-7 and $100^{\circ}C$ for 60 min against the mastitis pathogens. The bulgaricin HJ was determined as class IV bacteriocin by various enzyme treatments. Colony forming units analysis with indicator strains by the treatments of bulgaricin HJ indicates that the mode of bacteriocin action was bactericidal rather than bacteriostatic.

  • Several extracts of Chinese herbs are known to have antimicrobial efface on Helicobacter pylori. The inhibition action is believed to be anti-urease activity of the substance in the herbs. In this study ethanol (70%) extracts of Mume Fructus showed anti-urease activity far better than previously reported substance such as Caesalpinia sappan L. and Forsythiae suspensa VAHL.

  • We compared in vitro antibacterial activity of DWC-751, a new parenteral cephalosporin antibiotic, with those of cefpirome (CPR), cefotaxime (CTX) and ceftazidime (CAZ). DWC-751 showed a broad antimicrobial spectrum against Gram-positive and negative bacteria. The antibacterial activity of DWC-751 against Stapylococcus aureus was equal to that of CPR and superior to those of CTX and CAZ. The activity of it against Excherichia coli was more potent than those of CPR, CTX and CAZ. Against Pseudomonas aeruginosa, DWC-751 was slightly inferior to that of CAZ and superior to those of CPR and CTX. The antibacterial activity of DWC-751 was superior to those of CPR, CTX and CAZ against clinical isolates and ofloxacin resistant strains. DWC-751 showed bactericidal action against Escherichia coli at concentrations close to the MIC and induced the formation of filament and burge and lysis of Escherichia coli in a microscopic examination.

  • A new bacteriocin, named lacticin 10790, was purified from Lactococcus lactis subsp. cremoris KFCC 10790 by sequential adsorption, immobilized metal-affinity, cation-exchange, and $C_{18}$ reverse-phase chromatographies. The molecular mass of the bacteriocin was estimated to be between 3,000 and 3,500 Da. Lacticin 10790 showed a broad antimicrobial spectrum against many gram-positive bacteria. The bacteriocin was stable to heat and in the pH range between 2 and 6. Lacticin 10790 was destroyed by digestion with proteases and exhibited a bactericidal mode of action. An amino acid composition analysis of purified lacticin 10790 revealed a high concentration of hydrophobic amino acids. The N terminus of the bacteriocin was found to be blocked, upon analysis by Edman degradation. The results suggest that lacticin 10790 is a class I bacteriocin.

Partial Characterization and Purification of Enterocin K25 Linked to the Plasmid in Enterococcus sp. K25

  • The antimicrobial activity of partially purified enterocin K25, produced by Enterococcus sp. K25, was abolished by proteases such as pepsin and proteinase K. The bacteriocin was resistant to heat treatment at $75^{\circ}C$ for 15 min and lost 75% of its activity at $100^{\circ}C$ for 30 min. Enterocin K25 showed bactericidal mode of action against an indicator strain, Lactobacillus plantarum NCDO 955. Enterocin K25 was purified to 112.6-fold purity via conventional steps of ammonium sulfate precipitation, ion exchange chromatography, and reversed phase high performance liquid chromatography (RP-HPLC). The molecular mass of the purified enterocin K25 was estimated as 4.3 kDa on an electrophoresis gel. Plasmid (${\sim}6.5\;kb$) linkage of production of enterocin K25 was confirmed by plasmid curing.

  • Chitin, a poly $\beta$-(1longrightarrow14)-N-acetyl-D-glucosamine, is best known as a cell wall component of fungi and as a skeletal materials of invertebrates. Chitosan is derived from chitin by deacetylation in the presence of alkali. Chitosan has been developed as new physiological materials since it possesses antibacterial activity, hypocholesterolemic activity and antihypertensive action. However, the actions of chitosan in vivo still remain ambiguous as the physiological functional properties because most animal intestines, especially the human gastrointestinal tract, do not possess enzyme such as chitosanase which directly degrade the $\beta$-glucosidic linkage in chitosan, and consequently the unbroken polymers may be poorly absorbed into the human intestine. Therefore, recent studies as chitosan have attracted interest for chitosan oligosaccharides, because the oligosaccharides process not only water-soluble property but also versatile functional properties such as antitumor activity, immune-enhancing effects, enhancement of protective effects against infection with some pathogens in mice and antimicrobial activity (Kingsnorth et al., 1983, Mori et al., 1997).   • 초임계추출법과 수증기증류법을 이용하여 황련해독탕의 정유성분을 추출하여 성분 패턴을 비교하였다. 이때 초임 계추출조건은 압력 200 atm, $45^{\circ}C$ 그리고 추출시간 25분 이었다. 이때의 추출된 성분의 93.9%인 37개의 성분을 가스크로마토그래피/질량분석기로 확인하였다. 주요성분으로는 tetradecenoic acid (11.7%), Vanillin (5.9%), dl-Limonene (5.5%) 및 Eicosane (4.6%)으로 나타났다. 수증기증류법으로 추출한 정유에서는 34개 성분을 확인할수 있었다. 주요성분으로는 tetradecenoic acid (8.9%), Vanillin (5.8%)및 Eicosane (4.7%)를 확인 할 수 있었다. 또한 구강내의 12균주를 이용하여 항균효과를 측정하여 최소엑제농도(MIC)와 사멸농도(MBC) 0.025 - 12.8 mg/ml와 0.05 - 12.8 mg/ml 각각 나타났다.

  • Chitin, a linked polysaccharide composed of 2-acetamido-2-deoxy-$\beta$-D-glucopytanose residues, is distributed widely in nature. It has been utilized on various application field due to the development of chitin derivatives such as chitosan, partial deacetylated chitin, carboxylmethyl chitin, sulfated chitin, and so on. Chitin and chitosan have been recently interested in antitumor and antimicrobial activities, because of a powerful tumor inhibitory effect against experimental mouse tumors. Especially, the oligosaccharides obtained by partial degradation of them exhibited a remarkable antitumor effect against sarcoma 180, MM 48 and Meth Asolid tumors and antimetastatic effect against Lewis lung carcinoma in mice. This review describes on antitumor effects of chitin, chitosan and their oligosaccharides by their mechanism of action involving enhancement of immunological system.

  • Silybin has a potent antibacterial activity, more potent than silymarin II, against gram-positive bacteria without hemolytic activity, whereas it has no antimicrobial activity against gram-negative bacteria or fungi. The mode of action of silybin against the gram-positive bacterial cell was examined by investigating the change in plasma membrane dynamics of bacterial cells using 1 ,6-diphenyl-1,3,5-hextriene (DPH) as a membrane probe and by assessing the inhibition of macromolecular synthesis using radiolabeled incorporation assay. The results showed that silybin inhibited RNA and protein synthesis on gram-positive bacteria.
