Isolation and Identification of Antimicrobial Substance from Canavalia gladiata

  • Novel antimicrobial substance was isolated from seed coat of Canavalia gladiata by extraction with 75% methanol. Isolation and purification were conducted with solvent fractionation and chromatography on silica gel and sephadex LH-20 columns. Each fraction of antimicrobial activity was tested by paper disc method. Single compound was obtained from the 4th fraction of sephadex LH-20 column chromatography using chloroform/methanol (1:4, v/v), and identified as 3,4,5-trihydroxybenzoic acid methyl ester (methyl gallate) based on HPLC, GC/MS, FT-IR, $^1H$ NMR, and $^{13}C$ NMR analyses. This is the first report describing the presence of methyl gallate in C. gladiata.

항균성을 부여하기 위한 세탁과정에서의 은콜로이드 용액 처리 (Colloidal Silver Treatment of Cotton Fabrics after Washing to Impart Antimicorbial Activity)

  • Underwear is laundered frequently and most of them is made of cotton, but a cotton fiber is more difficult to modify than a synthetic fiber. We have attempted to determine the optimum conditions necessary whereby the lowest concentration of silver solution is needed to produce the greatest antimicrobial properties of cotton fabrics. For this study, colloidal silver was made by electrolysis. The concentration of colloidal silver was increased by increasing the area of the silver plates submerged in the water, the water temperature, the water hardness and the flow time of the water per 1l. However, the colloidal silver concentration was decreased by extending a space between the silver plates and increasing the water velocity. Cotton fabrics treated in the washing machine with 1.3 ppm colloidal silver solution for 10 minutes had effective microbial properties and an unperceivable reduction of reflectance.

  • In this study, a series of 4-amino-5-cyano-3-substituted-2,3-dihydrothiazol-2-thiones (1a-c), as well as their triazolo and triazinopyrimidine derivatives such as 8-substituted-3-benzyl-5-meth-ylthiazolo[5,4-e][1,2,4] triazolo[1,5-c]pyrimidin-2-thiones (4-6, 10) and 3-benzyl-5-methyl thiazolo[5,4-e]pyrimidino[3,4-b][1,2,4]triazin-2-thiones (7a-b) were prepared as potential antimicrobial and antitumor agents. Some of the tested compounds showed promising antimicrobial activity and non of them showed any appreciable antitumor activity.

감귤류 종자 추출물의 향균활성 (Antimicrobial Activity of Extracts from Citrus Seeds)

  • To develope natural food preservatives antimicrobial effect of the natural products against food-related bacteria and yeast The purpose of this study was evaluate antimicrobial effect of the citrus seeds. antimicrobial activities of methanol extracts from the citrus seeds investigate against Escherichia coli O26, Staphylococcus aureus 6358, Saccharomyces cerevisiae IBM 4274, Bacillus licheuiformis 9945a and Alcoligenes faecalis. Citrus seeds is containing to moisture 4∼6.5%, curd protein 11∼15%, curd lipid 32∼46%, curd carbohydrate 22∼45 % and ash 2∼4 %, that is containing to flavornoid 12∼48mg% and phenolic compound 22∼53mg%. Solidity content of the methanol extract from the citrus seeds was 0.8∼1.2%. Almost all of the methanol extracts from citrus seeds exhibited growth inhibiting activities for most of microorganisms tested. The methanol extracts from Citrus grandis, C. sunki, C. sulcata showed the growth inhibitory effects against Escherichia coli O26. The methanol extracts from C. obovoidea, C. sulcata, C. aurantium showed the growth inhibitory effects against Staphylococcus aureus 6358. The methanol extracts from C. obovoidea, C. sulcata, C. tangerina showed the growth inhibitory effects against Saccharomyces cerevisiae IBM 4274. The methanol extracts from C. obovoidea, C sunki, C. sulcata, C. tangerinan, C. natsudaidai, C. iyo, C. aurantium showed the growth inhibitory effects against Bacillus licheuiformis 9945a. The methanol extracts from C. obovoidea, C sunki, C. sulcata, C. aurantium showed the growth inhibitory effects against Alcoligenes faecalis. Among this especially, Showed growth inhibiting activity of the methanol extracts from Citrus sulcata that about microorganisms investigated. If apply searching suitable application method about such the citrus seeds antimicrobial activity, role as good antimicorbial material in storage or cooking of food, processing is expected.

옥수수 불검화 추출물(Zea Mays L.)과 후박(Magnoliae cortex) 추출물 혼합물의 치주질환원인균에 대한 항균작용 및 치은섬유아세포 활성도에 미치는 영향 (Antimicorbial effect of Zea Mays L. and Magnoliae cortex extract mixtures on periodontal pathogen and effect on human gingival fibroblast cellular activity)

  • Zea Mays L. has been known to be effective for improving tissue health and Magnoliae cortex to have effective antibacterial and antimicrobial activity against pathogenic microbes. The purpose of this study was to examine the antimicrobial effects of Zea Mays L. and Magnoliae cortex extract mixtures on periodontal pathogens(Prevotella intermedia, Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis, Streptococcus mutans )and to examine the effects on human gingival fibroblast cellular activity. Zea Mays L. and Magnoliae cortex extracts and their mixtures were prepared with various mixing ratios (0.5:1, 1:1, 1.5:1, 2:1). These extracts were loaded to periodontal pathogen cultured petri dish for antimicrobial test and also loaded to cultured human gingival fibroblast for cellular activity test. Each test was repeated 3 times and data were analyzed by one-way ANOVA with 95% confidence level. Mixture of these two extracts showed greater amount of inhibition area on periodontal pathogen and more improved gingival fibroblast activity as Zea Mays L. ratio reduced. So, mixture ratio 0.5:1 (Zea Mays L. : Magnoliae cortex) group showed statistical significance in antimicrobial activity and cellular activity among various mixtures(p < 0.05). In conclusion, 0.5:1 (Zea Mays L. : Magnoliae cortex) mixture possessed best gingival fibroblast cellular activity and antimicrobial activity toward periodontal pathogens.

티트리 (Melaleuca alternifolia) 추출물의 항균 활성 (Antimicrobial Effects of Tea Tree (Melaleuca alternifolia) Extracts)

  • 티트리 (Melaleuca alternifolia, Tea tree)는 terpineol-4, cineol, cymene, sesquiterpenes 등을 함유하여 살균 효과와 피부 보습효과를 지니고 있으며 또한, 여드름 염증 완화, 비듬 치료, 통증 완화, 우울증 해소 등의 특성을 가지고 있다. 본 연구에서는 티트리를 유기용매 (Methanol, Dichloromethane, Ethyl acetate) 및 열수추출법을 이용하여 물질을 추출하였으며, 각 추출액에서 나타나는 항균 및 항진균 활성을 확인하였다. 항균 활성 검증은 9종의 병원성 미생물 (Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Enterococcus faecalis, Staphylococcus aureus, Klebsiella pneumoniae, Escherichia coli, Salmonella typhimurium, Pseudomonas aeruginosa, Vibrio parahaemolyticus)을 대상으로 실시하였으며, 항균 활성 검증에 일반적으로 사용되는 disc diffusion법을 이용하여 추출물의 항균 활성을 확인하였다. 항균활성 검증을 통하여 ethyl acetate 분획추출물과 methanol 분획추출물에서 V. parahemolyticus와, S. aureus에 대한 항균활성을 확인하였다. 또한, 최대 활성을 나타내었던 V. parahaemolyticus를 대상으로 각 분획추출물 및 열수추출물의 항균활성을 확인하였으며, 10 mg/mL 농도로 처리하였을 때 99.9 % 이상의 항균활성을 확인하였다. 항진균활성의 경우, 균종에 따른 차이는 있으나 진균류에 대한 약 45시간 이상의 보존력을 확인하였다. 본 연구를 통해서 티트리 열수추출물과 메탄올 추출물의 V. parahaemolyticus에 대한 항균 활성을 확인하였으며, 추가적인 연구를 통해 항균제 및 항진균제로써의 개발 가능성을 확인하고자 한다.
