알로에의 과산화지질 억제효과에 관한 연구 (Studies on the Antilipidperoxidative Effect of Aloe)

  • The antilipidperoxidative and hepatopreventive effects of Aloe water extract (30 mg, 50 mg, 100 mg) were investigated at the levels of liver-total homogenates and the sera of SDrats intoxicated with CCl4 (0.5 cc/100g) and 50% ethanol. We measured MDA (Malondialdehyde) in the liver homogenate, AST (L-Aspartate-2-oxoglutarate aminotransferase) and ALT(L-Alanine-2-oxo-glutraate aminotransferase) in the serum. The analysis of the measurement indicated that Aloe water extract reduced MDA, ALT and AST significantly and their reduction was in relation to dose dependence. In rat liver homogenate intoxicated with ethanol and CCl4, Aloe treatment group markedly inhibited lipidperoxidation by 30%∼70%. In rat serum intoxicated with ethanol and CCl4, Aloe treatment group inhibited AST, ALT by 40%∼90%. In these data Aloe may be used to inhibit or prevent the hapatic toxicity with results from the environmental and alcohlic factors through the further study of its exact antihepatotoxic mechanism.

즙채의 과산화지질 생성 저해효과 (Antilipid Peroxidative Effect of Houttuynia cordata)

  • Houttuynia cordata (Saururaceae) has pungent smell and taste. It has been regarded as detoxicant, antipyretic, antiinflammantory and diuretic agents. In order to evaluate antioxidative and antilipidperoxidative efficacies, its fractions ($H_2O$, 20% MeOH, 40% MeOH, 60% MeOH, 170% MeOH) were measured by DPPH method and TBARS assay on rat liver homogenate. It was revealed that 60% MeOH fractions had potential antioxidative activity and inhibited lipid peroxidation significantly: In active fractions, we isolated rutin, hyperin, quercitrin and quercetin.

천연색소 Brazilin 및 Hematoxylin의 항지질 과산화활성에 관한 연구 (I) (A Study on the Antilipidperoxidative Effects of Brazilin and Hematoxylin(I))

  • 생체내 부정적 영향을 미치는 물질로 알려진 과산화지질의 생성을 억제하는 물질로서 지질대사 개선작용, 간보호작용, 혈소판응집능 억제작용 등 생체내 지질과산화 현상과 밀접한 상관성을 가지고 있을 것으로 사료되는 일련의 생리활성이 확인되었으나 아직 항 과산화지질 활성이 검토 보고된 바 없는 Brazilin 및 그 hydroxy 유도체인 hematoxylin을 대상으로 생체내 지질과산화 유도제로 $CCl_4$와 ethanol을 사용하여 지질과산화 억제효능 및 in vitro에서의 효소적, 비효소적 지질과산화에 대한 억제효능을 검토하였다. Brazilin 및 hematoxylin이 $CCl_4$로 유도된 과산화지질의 억제효과를 보면 간 homogenate에서 각각 90%, 94%, mitochondria 분획에서 각각 68%, 82%, microsome 분획에서 각각 65%, 81%의 억제효과를 보였다. 이에 반해 혈장에서의 억제효과는 각각 37%, 68%로 간에서 낮은 억제효과를 나타내었으나 이 두 물질은 간 뿐만 아니라 혈장에서도 과산화지질 억제효과가 유의성 있게 나타났다. Ethanol로 유도된 과산화지질의 억제효과를 보면 간 homogenate에서 각각 93%, 99.8%, mitochondria 분획에서 각각 74%, 98%, microsome 분획의 경우 각각 70%, 94%의 효과를 보였다. 이에 반해 혈장에서의 억제효과는 Brazilin 및 Hematoxylin이 각각 43%, 80%로 나타났다. 간조직중에서는 Brazilin과 Hematoxylin이 유사한 수준의 지질과산화 억제활성을 나타내고 있으나 혈장 과산화지질치는 현저한 차이를 나타내고 있어 Brazilin과 Hematoxylin은 간으로부터 혈액중으로의 지질의 방출에 상이한 정도로 영향을 미치거나 순환지질 기질의 보호에 의한 지질과산화 억제효과에 차가 있거나 혹은 두 가지 인자의 복합적인 작용에 의한 것으로 추정할 수 있다. 세포하수준에서의 항 지질과산화 활성을 알아보기 위해 실시한 간 misosome 및 mitochondria 분획에 대해 검토해 본 결과 양분획 모두에서 Hematoxylin이 Brazilin보다 강한 항산화 활성을 나타내었고 저지율은 양분획에서 유사한 수준이었다. Brazilin 및 hematoxylin이 효소적 지질과산화 반응인 NADPH 유도 지질과산화 반응에 미치는 영향을 검토한 결과 Brazilin의 경우 88.5%, Hematoxylin의 경우 91.0% 억제효과를 나타내었고 비효소적 지질과산화 반응인 ascorbate 유도 지질과산화 반응에서도 Brazilin이 75.7%, Hematoxylin이 81.9%의 지질과산화 억제효과를 나타내었다. 위의 결과에서 지질과산화 억제효과는 주로 자체의 항산화 작용에 의하나 microsome 약물대사활성에 의한 효소적 과산화 현상에도 억제작용이 있는 것으로 추정된다.

AIIoxan 유도 당뇨병상태에서 과산화지질생성에 미치는 Brazilin의 효과 (Antilipidperoxidative Effects of Brazilin in Alloxan-induced Diabetic Mice)

  • Brazilin was tested for its ability to inhibit alloxan induced lipidperoxidation. Lipid peroxide contents in liver, kidney and serum were measured by the TBA method. ICR mice receiving alloxan at a dose of 43mg/kg via the tail vein after a 24 hrs starvation showed significantly increased lipid peroxide contents as compared to untreated control. Lipid peroxide contents in liver, kidney and serum of alloxan-induced diabetic mice were dosedependently decreased by the treatment of brazilin at a dose of 10mg/kg, 50mg/kg, 100 mg/kg for 5 days.

  • Morus alba extract(MAE) was tested for its ability to inhibit alloxan induced lipidperoxidation. Lipid peroxide contents in serum, liver, kidney and heart were measured by the TBA method. ICR mice receiving alloxan at a dose of 6mg/kg intraperitoneally after a 24hrs starvation showed significantly increased lipid peroxide contents as compared to untreated control. Lipid peroxide contents in serum, liver, kidney of alloxan-diabetic mice were decreased by the treatment of MAE at the dose of 50mg/kg, 100mg/kg for 7 days.

호장근의 사염화탄소로 유도된 지질과산화 저해활성 (Antilipoperoxidant Activity of the Root Polygonum cuspidatum on Carbon Tetrachloride Induced Hepatotoxicity)

  • The root of Polygonum cuspidatum (Polygonaceae) has been used as treatments of hyperlipidemia, dermatitis, gonorrhea, favus athlete's foot, inflammation in traditional medicine. In order to examine anti-lipidperoxidation activity, hexane, EtOAc, BuOH and water fractions of its methanol extract were administered to carbon tetrachloride intoxicated rats. Ethylacetate fraction exhibited antilipidperoxidative effect on liver lipid homogenate and the radical scavenging effect on DPPH. Serum transaminase, AlP, triglyceride and total cholesterol contents significantly decreased by administrations of ethylacetate fraction.

알콜유도 지질과산화에 미치는 어성초 영향 (Effect of Houttuynia cordata on EtOH-induced Lipidperoxidation in Rat)

  • The antilipidperoxidative and hepatopreventive effects of Houttuynia cordata extract(HCE) were investigated at the level of liver homogenates and serum of SD rats intoxicated EtOH(3g/kg for 3days). We measured MDA(malondialdehyde) in the live. homogenate, AST(1-aspartate-2-oxoglutarate aminotransferase) and ALT( 1-alanine-2-oxoglutarate aminotransferse) in the serum. The analysis of the measurement indicated that HCE reduced MDA, ALT and AST significantly and their reduction was in relation to dose dependence. In rat liver homogenate intoxicated with EtOH, HCE treatment group inhibited lipidperoxidation by 50-75%. In rat serum intoxicated EtOH, HCE inhibited AST and ALT by 40-70%.

천연색소 Brazilin의 항지질 과산화 활성에 관한 연구(II) (A Study on the Antilipidperoxidative Effects of Brazilin(II))

  • Alloxan으로 유도된 당뇨병상태에서 Brazilin 투여시 mitochondira 분획 및 microsome 분획에서 용량의존적으로 억제효과가 증가하였으며 Brazilin 100mg/kg을 투여시는 mitochondria 분획에서 56.7% 억제를 , microsome 분획에서는 68.4%의 억제효과를 보였다. 또한 Brazilin 100mg/kg을 시간별 투여시 시간의존적으로 억제효과가 증가하였으며 30일간 투여시에 mitochondria 분획에서 66.2%, microsome 분획에서 74.3%의 억제효과를 보았다.

황기의 지질과산화 억제작용 (Antilipidperoxidative activity of Astragalus membranaceus)

  • The root of Astragalus membranaceus (Leguminosae) has been used in traditional chinese prescriptions for strengthning the superficial resistance, promoting pus discharge and tissue regeneration, diuretis and alleviating edema. Lipid peroxidation has been suggested as a major cause of atherosclerosis, cancer, liver disease, and the aging process. In order to investigate the anti-lipid peroxidation activity, Astragali Radix was extracted with $80\%$ MeOH and fractionated with hexane, $CH_{2}Cl_2$, EtOAc, BuOH and Water. The antilipid peroxidation activities of them were determined by human erythrocyte ghost and $CCl_4$-induced lipid peroxidation. $CH_{2}Cl_2$ and EtOAc fractions, especially, isoflavonoids, 7,2'-dihydroxy-3',4'-dimethoxyisoflavan-7-O-${\beta}$-D-glucoside and calyco sin-7-O-${\beta}$-D-glucoside from EtOAc fraction showed anti-lipid peroxidation activities.
