도축 폐혈액 단백질로부터의 Angiotensin I Converting Enzyme 저해 펩타이드의 생산 (Production of Angiotensin I Converting Enzyme Inhibitory Peptides from Bovine Blood Plasma Proteins)

  • 본 연구는 도축 폐기물인 가축혈액을 이용하여 항고혈압 기능성 식품소재로서의 angiotensin I converting enzyme 저해 펩타이드 분획을 생산하기 위한 조건과 가능성을 조사하기 위하여 수행되었다. 산업적으로 이용 가능한 단백분해효소 중 Alcalase가 혈장 원액 및 그로부터 분리된 albumin에 대하여 가장 높은 활성의 가수분해물을 생성하였다. 특히 albumin의 Alcalase 가수분해물과 이를 gel chromatography를 통새 분획하여 얻은 고활성 분획의 $IC_50$값은 각각 0.5 및 0.02 mg/mL로서 지금까지 보고된 식품단백질 유래 펩타이드 혼합물들과 비교할 때 활성이 매우 높은 것에 속함을 알 수 있었다. 또한 이 고활성 펩타이드 분획은 혈장 원액으로부터 단순한 한외여과만을 거쳐도 얻을 수 있음을 확인함으로써 산업적 실용화 가능성이 높은 공정임을 알게 되었다.

Antihypertensive peptides from whey proteins fermented by lactic acid bacteria

  • In this study, whey proteins were fermented with 34 lactic acid bacteria for 48 h at $37^{\circ}C$ and their ability to inhibit angiotensin 1-converting enzyme (ACE) activity were compared. All the lactic acid bacteria displayed varying proteolytic abilities in whey. Their fermentates also displayed varying abilities to inhibit ACE in vitro. Seven fermentates showed strong ACE inhibitory abilities between $84.70{\pm}0.67$ and $52.40{\pm}2.1%$ with $IC_{50}$ values between $19.78{\pm}1.73$ and $2.13{\pm}0.7mg/ml$. Pediococcus acidilactici SDL1414 showed the strongest ACE inhibitory activity of $84.7{\pm}0.67%$ ($IC_{50}=19.78{\pm}1.73{\mu}g/ml$). Mass spectrometry revealed that more than half (57.7%) of the low molecular weight peptides (< 7 kDa) in the P. acidilactici SDL1414 fermented samples were ACE inhibitory peptides. Our results show that P. acidilactici SDL1414 could be used as a starter culture in the dairy industry to develop antihypertensive functional foods for hypertension management.

Biological activity of functional foods and anti-microbial activity of phenolics from Sambucus sieboldiana var. pendula leaves

  • This study aimed to investigate the biological and antibacterial activities of phenolics from Sambucus sieboldiana var. pendula leaf extract. The phenolic contents of the S. sieboldiana var. pendula extracted with water and ethanol was 11.60 and 12.39 mg/g, respectively. The inhibitory effects of the water and ethanol extracts on the angiotensin converting enzyme (ACE) were 71.94-92.08 and 48.42-78.33%, respectively, and on xanthine oxidase (XOase) were 42.11-100 and 35.61-100%, respectively, at a phenolic concentration of 50-200 ㎍/mL. All these effects were found to be concentration-dependent. Additionally, the leaf extracts exhibited excellent antibacterial activities against Streptococcus mutans, Staphylococcus aureus, and Propionibacterium acnes. Hence, S. sieboldiana var. pendula was confirmed to have excellent antihypertensive, antigout, and antimicrobial properties.

Effect of an herbal formulation on DOCA-salt and fructose induced models of hypertension in rats

  • The present study was carried out to investigate the antihypertensive effect of a folklore herbal formulation (HF) (300mg/kg/day; p.o.) in deoxycorticosterone acetate (DOCA)-salt induced and fructose induced hypertensive rats. In DOCA model, DOCA (15 mg/kg, s.c., twice a week) was administered to unilateral nephrectomized rats for 4 weeks. In fructose model, drinking water was replaced with 10% fructose solution for 6 weeks to induce hypertension. Systolic blood pressure (BP) was measured once every week during the treatment schedule. After completion of treatment schedule, BP and vascular reactivity to various agonists like Noradrenaline, Adrenaline, Phenylephrine and Serotonin (5-hydroxytrptamine; 5-HT) were recorded in rats of both models. A cumulative concentration response curve of 5-HT was carried out in isolated rat fundus strip of the DOCA-salt induced and fructose induced hypertensive rats. The results tend to suggest that HF possesses antihypertensive activity.

  • It has been suggested that hyperuricemia is related to the development of essential hypertension. Hypertensive patients with hyperuricemia has decreased glomerular filtration activity as compared to normotensive patients with hyperuricemia. These studies indicates uric acid concentrations in blood is associated with hypertension, Probenecid is an uricosuric agent which decreases uric acid reabsorption at the proximal tubule. Recently, we have shown that probenecid exerts anti-hypertensive action in Spontaneously Hypertensive Rats. Considering these results, I have designed a series of experiments to explore potential mechanism of antihypertensive action, of probenecid. In isolated rat thoracic aorta. probenecid significantly prevented phenylephrine-induced contraction of the blood vessel. When endothelium removed blood vessels were used, probenecid produced same effect as the intact blood vessels, indicating that probenecid directly act through the ${\alpha}$ -adrenergic receptor in vascular smooth muscles rather than through endothelium. These results suggest that one of the mechanism of antihypertensive effects of probenecid is due to the direct inhibition of ${\alpha}$ -adrenergic receptor in blood vessels.

  • A 63 year old male had suffered from hypertension and angina pectoris for 4 years, On physical examination, blood pressure was 150/110 mmHg with medication of antihypertensive drugs. Aortogram showed the stenosis of the left renal artery, the complete occlusion of the right renal artery, and atherosclerotic change of abdominal aorta. Blood urea nitrogen was 25 mg/dl, serum creatinine was 1.2 mg/dl, and renin activity in peripheral blood was 8.7 ng /ml /hour, The stenosis of left renal artery and the complete occlusion of right renal artery should have produced the renovascular hypertension Bilateral aorto-renal bypasses with saphenous grafts were done for treatment of ren-ovascular hypertension Postoperatively, blood pressure was normalized with only small dosage of antihypertensive drugs.

  • Symplocarpus renifolius, a folk medicinal herb has been used for treatment of hypertension, rheumatis, tetanus, ringworm, cough and epilepsy in north and middle part of Korea. The extracts from the root of S. renifolius were evaluated for antibacterial, antihypertensive and analgesic activities. The crude extract of the root of S. renifolius showed antibacterial activity against Gram(+) bacteria and dose dependantly decrease the blood pressure of SHR. The chloroform extract of the roots of S. renifolius was also exibits analgesic action in mice.

  • This study was conducted to compare and analyze the changes in the biochemical characteristics and biological activity of peptide extracts derived from Chickso, Hanwoo, and Wagyu beef during digestion. The results of the in vitro digestion analysis revealed that the digestion rate, total free amino acid content, and antioxidant and antihypertensive activities of Chickso loin and shank myofibrillar proteins were significantly higher (p<0.05) than those of Hanwoo and Wagyu loin and shank myofibrillar proteins. Particularly, the peptide extracts of Chickso loin and shank had a high angiotensin-converting enzyme inhibitory activity. In mice in vivo digestion experiment, the blood serum of mice fed with Chickso loin peptide extract (<10 kDa) showed the highest antioxidant enzyme activity. Thus, Chickso peptide extracts were deemed to be similar or more bioactive than Hanwoo and Wagyu peptide extracts, and can be used as bioactive materials.

쉐리단 포도주의 항고혈압성과 품질에 머루첨가의 영향 (Effects of Vitis coignetiae on the Quality and Antihypertension of Vitis hybrid Red Wine)

  • 항고혈압활성을 가진 고품질 국산 포도주를 개발하기 위하여 쉐리단 포도주 제조과정에서 다양한 약용식물과 머루 등을 첨가하여 이들이 쉐리단 포도주의 품질과 생리기능성에 미치는 영향을 조사하였다. 쉐리단 포도 주스에 머루를 첨가하여 발효시킨 쉐리단-머루 포도주가 에탄올 함량과 기호도가 가장 높았고 68.5%의 높은 항고혈압성 안지오텐신 전환효소(ACE) 저해활성을 보였다. 쉐리단-머루 포도주는 60일간 후발효 시켰을 때 ACE 저해활성은 80.7%($IC_{50}$: 28 mg/mL)로 상승하였고 기호성도 좋았다. 또한 60일 후발효 시킨 쉐리단-머루 포도주의 $C_{18}$ solid phase 추출물은 본태성 고혈압쥐 (SHR)에서 항고혈압성이 확인되었다.

항고혈압 활성을 가진 식물유래 젖산균의 생균제 특성 (Probiotic Potential of Plant-Derived Lactic Acid Bacteria with Antihypertensive Activity)

  • Lactic acid bacteria (LAB) are industrially important microorganisms for probiotics. The recent widespread application of LAB for preparation of functional food is attributable to the accumulating scientific evidence showing their beneficial effects on human health. In this study, we isolated and characterized plant-derived LAB that show angiotensin-converting enzyme (ACE) inhibitory and antioxidant activities. The selected strain K2 was isolated from Kimchi, and identified as Lactobacillus plantarum by 16S rRNA gene analysis. The strain grew under static and shaking culture systems. They were also able to grow in different culture conditions like $25^{\circ}C{\sim}37^{\circ}C$ temperature, 4~10 pH range and ~6% NaCl concentration. L. plantarum K2 was highly resistant to acid stress; survival rate of the strain at pH 2.5 and 3 were 80% and 91.6%, respectively. The strain K2 also showed high bile resistance to 0.3% bile bovine and 0.3% bile extract with more than 74% of survival rate. The cell grown on MRS agar plate containing bile extract formed opaque precipitate zones around the colonies, indicating they have bile salt hydrolase activity. The strain showed an inhibitory activity against pathogenic bacteria such as Escherichia coli, Staphylococcus aureus and Listeria monocytogenes; antibacterial activity was probably due to the lactic acid. The K2 strain showed relatively higher autoaggregation values, antihypertensive and antioxidant activities. These results suggest that L. plantarum K2 could be not only applied as a pharmabiotic for human health but also is also starter culture applicable to fermentative products.
