Oxyresveratrol의 기원, 생합성, 생물학적 활성 및 약물동력학 (Source, Biosynthesis, Biological Activities and Pharmacokinetics of Oxyresveratrol)

  • Oxyresveratrol (trans-2,3',4,5'-tetrahydroxystilbene) has been receiving increasing attention because of its astonishing biological activities, including antihyperlipidemic, neuroprotection, antidiabetic, anticancer, antiinflammation, immunomodulation, antiaging, and antioxidant activities. Oxyresveratrol is a stilbenoid, a type of natural phenol and a phytoalexin produced in the roots, stems, leaves, and fruits of several plants. It was first isolated from the heartwood of Artocarpus lakoocha, and has also been found in various plants, including Smilax china, Morus alba, Varatrum nigrum, Scirpus maritinus, and Maclura pomifera. Oxyresveratrol, an aglycone of mulberroside A, has been produced by microbial biotransformation or enzymatic hydrolysis of a glycosylated stilbene mulberroside A, which is one of the major compounds of the roots of M. alba. Oxyresveratrol shows less cytotoxicity, better antioxidant activity and polarity, and higher cell permeability and bioavailability than resveratrol (trans-3,5,4'-trihydroxystilbene), a well-known antioxidant, suggesting that oxyresveratrol might be a potential candidate for use in health functional food and medicine. This review focuses on the plant sources, chemical characteristics, analysis, biosynthesis, and biological activities of oxyresveratrol as well as describes the perspectives on further exploration of oxyresveratrol.

Bifidus Fermentation Increases Hypolipidemic and Hypoglycemic Effects of Red Ginseng

  • Antihyperlipidemic and antihyperglycemic effects of Red Ginseng (RG, steamed and dried root of Panax ginseng C.A.Meyer, family Araliaceae), major component of which is ginsenoside Rg3, and Bifidodoterium-fermented RG (FRG), major component of which is ginsenoside Rh2, were investigated. Orally administered RG and FRG potently reduced the serum triglyceride levels in com-oil-induced hypertriglycemidemic mice as well as total cholesterol and triglyceride levels in Triton WR-1339-induced hyperlipidemic mice. Of the saponin and polysaccharide fractions of RG and FRG, the polysaccharide fraction inhibited postprandial blood glucose elevation of maltose- or starch-loaded mice and reduced the blood triglyceride levels in com-oil-induced hypertriglycemidemic mice. The saponin fraction and its ginsenosides Rg3 and Rh2 reduced blood triglyceride and total cholesterol levels in Triton WR1339-induced hyperlipidemic mice. The inhibitory effect of FRG and its main constituents against hyperlipidemia and hyperglycemia in mice were more potent than those of RG. These findings suggest that hypolipidemic and hypoglycemic effects of RG can be enforced by Bifidus fermentation and FRG may improve hyperlipidemia and hyperglycemia.

고지방식이로 유도된 비만 Mice에서 쇠비름 에탄올 추출물의 고지혈증 개선효과 (Antihyperlipidemic Effect of Diet Containing Portulaca oleracea L. Ethanol Extract in High Fat Diet-Induced Obese Mice)

  • 쇠비름 에탄올 추출물의 고지혈증 개선 효과를 알아보기 위하여 C57BL/6J mice에게 9주 동안 고지방식이를 섭취시켰다. 마지막 4주 동안은 고지방식이에 쇠비름 에탄올 저농도투여군(HFD+POE 75) 및 쇠비름 에탄올 고농도투여군(HFD+POE 125)으로 나누어 하루에 한 번씩 경구투여 하였으며 고지방식이군(HFD)은 증류수를 투여하였다. HFD+POE 75군 및 HFD+POE 125군의 체중변화량은 유의적인 차이를 나타나지는 않았으나 식이효율은 HFD군에 비해 유의적으로 감소하였다. 혈중 ALT 함량은 HFD군에 비해 HFD+POE 125군에서 유의적으로 감소하였으나 AST 함량은 그룹 간 차이를 나타내지 않았다. 실험식이 4주 때에 혈청 중 총콜레스테롤, 중성지질 및 LDL-콜레스테롤 함량은 쇠비름 에탄올 추출물 경구투여로 유의적으로 감소하였으나 HDL-콜레스테롤 함량은 유의적 차이를 나타내지 않았다. 심순환기 질환의 위험성을 나타내는 지표로 알려진 동맥경화지수와 심혈관위험지수는 고농도의 쇠비름 에탄올 추출물 경구투여로 HFD군에 비해 유의적으로 감소하였다. 이상의 결과 고지방식이에 쇠비름 에탄올 추출물 경구투여로 혈중 및 간장 중의 총콜레스테롤 및 LDL-콜레스테롤, 중성지질이 저하되었는데 이는 쇠비름 에탄올 추출물이 고지혈증 개선 효과가 있는 것으로 사료되나 쇠비름 에탄올 추출물의 어떤 성분에 의한 지질 저하효과를 나타내는지와 어떠한 기전에 의해 지질 저하효과를 나타내는지 보다 자세한 연구가 요구된다.

알코올성 간독성 흰쥐의 혈청 지질 및 조직 과산화지질 농도에 미치는 발효당귀의 영향 (Effects of Monascus-fermented Angelica gigas Nakai on the Contents of Serum Lipid and Tissue Lipid Peroxidation in Alcohol Feeding Rats)

  • Monascus purpureus 균주에 의해 발효된 당귀분말을 식이 중에 5% 수준으로 첨가하여 30% 알코올과 함께 4주간 흰쥐에 급여한 후 혈중 지질 농도 및 조직 내 산화 스트레스에 미치는 영향에 대하여 검토하였다. 혈중 중성지질 농도 및 유리 지방산 농도는 알코올 대조군(Alc)에서의 증가가 알코올 실험군에서 모두 감소하였고, 특히 발효당귀군(AFAG)에서 낮은 감소수치를 보였다. 혈중 총 콜레스테롤 농도는 전반적으로 유의적인 차이는 없었으나, HDL-cholesterol 농도에서 발효당귀군(AFAG)이 증가수치를 나타내었다. 간장, 심장, 비장,신장, 고환 및 혈청에서 생체막 과산화지질 생성 정도는 정상군(N)에 비해 알코올 대조군(Alc)에서 유의적으로 증가하였고, 이러한 증가는 발효당귀군(AFAG)과 양성 대조군(ASM)에서 정상 수준으로 회복되었다. 간 조직 내 내인성 항산화 물질인 glutathione 농도는 알코올 대조군(Alc)에서 유의적으로 감소하였고, 발효당귀군(AFAG)에서 유의적으로 증가하여, 과산화 지질 생성정도와 glutathione 농도는 유의적인 차이가 인정되었다. 이상의 실험결과 발효당귀는 알코올 투여 동물실험에서 혈중 지질 개선효과는 물론 조직 내 항산화 활성증가로 산화스트레스를 경감시키는 효능이 있는 것으로 사료되며, 향후 간 독성 개선에 효능을 가지는 건강식품 개발 가능성이 높은 소재로 판단되어진다.

탱자 열매 추출액의 항산화 및 지질저하효과 (Antihyperlipidemic and Antioxidant Effects of Poncirus trifoliata)

  • 탱자 추출액이 비만을 유도한 흰쥐의 지질강하 및 항산화효과에 미치는 영향을 검토했다. 혈장유리지방산 및 triglyceride 량은 대조군과 비교하여 탱자 처리군이 유의하게 낮은 값을 보였다. 그러나 정상군의 수준까지 나타내지는 않았다. 총콜레스테롤 및 LDL-cho.농도는 탱자 처리군 모두가 대조군보다 현저하게 하락한 값을 나타내었다. HDL-cho.은 대조군을 비롯하여 전 처리군에서 유의한 차이를 나타내지 않았다. 간장내 총콜레스테롤과 triglyceride량은 정자 처리군이 대조군 보다 현저하게 낮은 값을 보였다. 혈장 및 간장의 TBARS 농도는 탱자처리량이 증가함에 따라 감소하였다. GSH-Px, SOD 및 CAT 활성치는 탱자 처리군에서 높은 활성치를 보였다. 그러나 SOD의 활성치는 대조군과 유의한 차이를 나타내지는 않았다.

고지혈증 랫트를 이용한 수종 전통 한약제의 항고지혈 효과 (Antihyperlipidemic Activity of Several Traditional Herbal Medicines on Experimental Hyperlipidemia in Rats)

  • Three herbal medicine recipes were tried on the animal model of diet induced hypercholesterolemia to screen for the lipid-lowering effect. The recipes adopted were based on the prescription for atherosclerosis-related symptoms by folk-medicine practioners. Hyperlipidemia was induced in rats by giving the high lipid/cholesterol diet for 7 days. Then, the recipes started with the normal diet. Blood was sampled at 1,2,3 and 5 week-points of the treatment, and levels of total cholesterol, triglyceride, high density lipoprotein and lower density lipoprotein were measured. Dae-shiho-tang decreased total cholesterol level significantly. Hwangryun-haedok-tang and Samhwang-sasim-tang slightly decreased total cholesterol level, although it was not statistically significant.

산마늘 기능성 활성물질 탐색 및 자원화 (Effect of Allium victorialis var.platyphyllum Leaves on Triton WR-1339-induced and Poloxamer-407-induced hyperlipidemic Rats and on Diet-Induced Obesity Rats)

  • The herb of Allium victorialis var. platyphyllum (Liliaceae) has been used as an edible wild herb and to treat heart failure and gastritis. We have already reported antihyperlipidemic anti-tumor effects of this plant. To enlarge the commercial availability of this food, it was investigated whether the extracts of A. victorialis var.platyphyllum reduce hyperlipidemia and obesity or not. The plants tested in this experiment were collected from two eco-types of IS. Ullung and Mt. Odae cultivated at Pyongchang. Extracts were prepared by extracting the fresh leaves and those dried at $36^{\circ}C$ and $90^{\circ}C$, respectively. Pretreatment with the ethanolic extracts for two weeks (p.o.) reduced serum triglyceride-, total cholesterol- and LDL-cholesterol contents in rats induced by Triton WR-1339, respectively. Furthermore, oral administration of the extracts also inhibited the hyperlipidemia induced with oral diet of 30% corn oil. In the other attempt to find to alleviate the obesity, the model rats with obesity were induced by the high fat-diet for six weeks. Post-treatment with the extracts for two weeks significantly reduced the hyperlipidemia. Retroperitoneal-, epidymal- and total abdominal fat pad weights were considerably reduced at 100 mg/kg oral administration of the extracts. Increased feces lipid contents were also found in the rat treated with the extracts. The extract of Mt. Odae eco-type showed more potent activity than that of Is. Ullung one. These results suggest that use of the fresh leaves may lead to the higher activity in treatment of hyperlipidemia and obesity than of the dried one.

Anti-hyperlipidemic Effect of Insamsansa-eum in Mice

  • Hypolipidemic effect of Insamsansa-eum (ISE, Ren-Shen-Shan-Zah-Yin in Chinese) consisted of Red ginseng (RG; the steamed root of Panax ginseng C.A. Meyer) and Crataegii fructus (CF, the fruit of Crataegus pinnatifida BGE) is used frequently in China and Korea as a herbal medicine to treat arteriosclerosis, was investigated. Treatment of ISE significantly reduced blood triglyceride (TG) and total cholesterol (TC) levels in Triton WR-1339-induced hyperlipidemic mice and serum TG levels in corn oil-induced hypertriglyceridemic mice. ISE significantly lowered the high blood TG and TC levels as well as body and epididymal mass weights in hyperlipidemic mice induced by long-term feeding of a high-fat diet and increased blood HDL cholesterol level. ISE and its ingredients, RG and CF, inhibited pancreatic lipase and HMG-CoA reductase activities. Of its ingredients, RG reduced the blood TC level and HMG-CoA reductase activity more than CF. CF reduced blood TG level and pancreatic lipase activity more than RG. These finding suggest that the antihyperlipidemic effect of ISE may be due to synergistic inhibition of HMG-CoA reductase and pancreatic lipase by RG and CF, and that ISE may be effective hypolipidemic agents.

Sprague-Dawley계 흰쥐에서 현지초 부탄올 분획물의 항비만 및 항고지혈증 효과 (Anti-obesity and Anti-hyperlipidemic Effects of Butanol Soluble Fraction from Methanol Extract of Geranium thunbergii in Sprague-Dawley Rats)

  • The purpose of this study was to investigate anti-obesity and anti-hyperlipidemic effects of extracts from Geranium thunbergii (GT) in high-fat-diet-induced obese rats. Animals were randomly divided into four groups [normal diet, high fat diet, MeOH extract of GT (GTM), and BuOH fraction of GT (GTB)] and GT samples were treated with dose of 100 mg/kg for 8 weeks. It was observed that GTB-treated group significantly reduced body weight gain, food intake, epididymal fat weight, and triglyceride level in serum and liver compared to control group. The rats fed GTB also decreased contents of thiobarbituric acid-reactive substances (TBARS), hydroxyl radical, and xanthin oxidase (XO) increased by high fat diet. Furthermore, the anti-oxidant enzymes such as superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase were increased by GTB treatment. The experimental results indicate that GTB has anti-obesity and anti-hyperlipidemic effects, as well as radical scavenging activity.

Antidiabetic activity of Argyreia speciosa (sweet) (Burm.f.)Boj. in normoglycemic and Streptozotocin-induced diabetic rats

  • Effect of ethanol (ASE) and water (ASW) extracts of Argyreia speciosa on blood glucose and lipid profile was investigated in normoglycemic and Streptozotocin (STZ)-induced diabetic animals. In oral glucose and sucrose tolerance test, treatment with ASE and ASW (100 and 200 mg/kg) and Glidenclamide (10 mg/kg) significantly improved the glucose and sucrose tolerance in normal animals. In addition, respective treatment for fifteen-day resulted in significant percentage reduction in serum glucose (SG) ie., 30.39% (lower dose of ASE) and 33.21% (higher dose of ASW). In standardized STZ (50 mg/kg, iv)-induced diabetic rats, a single dose of ASE and ASW treatment exhibited reduction in SG levels at different time intervals compared to basal levels. Administration of both the doses of ASE and ASW for fifteen-day days exhibited greater percentage reduction in glycemia (24.6%, 24.7%, 23.9% and 21.9% respectively) and also ameliorated restored to near normal value of all tested lipid parameters. Further, treatment also exhibited significantly improved glucose tolerance over the period of 120 min compared to diabetic control group. Eventhough treatment failed to increase serum insulin levels significantly but peripheral utilization of insulin was increased as evident by insulin tolerance test. Taken together, present study supports the traditional usage of title plant in the treatment of diabetes mellitus.
