Antifungal Properties of Rhizopus oligosporus Against Apple Anthracnose Fungi

  • This study was carried out to assess the antifungal potential of R. oligosporus and its ethyl acetate (EtOAc) extract against the fungal pathogens causing anthracnose disease in apple fruits using disc diffusion, antagonistic effect and morphological abnormalities in fungal mycelia. The percentage of inhibition of antifungal effect of the ethyl acetate extract (5 ${\mu}l$ $disc^{-1}$) of the R. oligosporus against C. acutatum KACC 40848, C. gloeosporioides KACC 40897, C. higginsianum KACC 40806, C. orbiculare KACC 40808, C. coccodes KACC 40008, C. musae KACC 40947, C. boninense KACC 40893, C. liliacearum KACC 40981, C. caudatum KACC 41028 and Colletotrichum sp. KACC 40811 was found to be 44.4, 35.5, 40, 31.1, 33.3, 37.7, 40, 51.1, 28.8 and 28.8%, respectively. Also the fungus R. oligosporus showed potential antagonistic effect of antifungal activity against the tested pathogens of Colletotrichum spp. Further, R. oligosporus had a potential detrimental effect on the morphology of the tested fungi of Colletotrichum spp. such as wrinkle abnormalities, abnormal cell formation, lysis of mycelium, empty cell formation, distorted cell formation and breakage of the mycelium. These findings strongly support the role of R. oligosporus to serve as a potential antifungal agent to control plant pathogenic fungi causing anthracnose disease in apple fruits.

  • The fungal isolate Isaria javanica pf185 has potential as a mycopesticide because it demonstrates insecticidal activity against the green peach aphid and antifungal activity against Colletotrichum gloeosporioides. For commercialization of this isolate, determination of the optimal and least expensive culture conditions is required; however, these data are not currently available. This study describes the conditions for optimal development of conidia and production of metabolites for the biocontrol of the fungal pathogen. The optimal culture conditions were examined using cultures on solid agar and liquid media. High growth temperature enhanced spore formation but reduced antifungal activity in both solid and liquid media. The highest spore yield was obtained in a medium containing glucose as a carbon source and yeast extract as a nitrogen source. Soybean powder and wheat bran were effective nitrogen sources that promoted spore production and antifungal activity of the isolate. These results revealed the basic, cost-effective growth media for commercial production of a biopesticide with insecticidal and antifungal properties for use in integrated pest management.

  • AF-001A is a novel antifungal cyclic glyco-peptise isolated from Pseudomonas cepacia AF6008. AF-001A is a mixture of AF-011A1 and AF-001A2. Each compound contains glycine(1), serine(2), asparagine(1), 2,4-diaminobutyric acid(1), beta-hydroxytyrosine(1), xylose(1) and a methylene chain amino acid(1). Additionally, A1 contains one beta-hydroxyasparagine that is replaced with as asparagine in A2. AF-011A showed high in vitro antifungal activity against various animal and plant pathogenic fungi and caused no irritation on the skin of rabbits.

  • 계피의 항진균활성을 검색한 결과 에테르 추출물이 우수한 활성을 나타내는 것으로 평가되어 계피 에테르의 추출물로부터 항진균물질 AF-001을 분리 정제하였으며 이를 동정한 결과 trans-cinnamaldehyde로 확인되었다. AF-001은 피부진균증의 원인 균주인 Tricophyton mentagrophytes, Microsporium gypseum, Epidermophyton floccosum, Cryptococcus neofomans, Aspergillus niger, Candida albicans와 Penicillium avellaneum에 대해 $38{\sim}79\;{\mu}g/ml$의 최소저지농도를 보였으며 imidazole계의 대표적인 항진균제인 Ketoconazole보다 광범위한 항균 스펙트럼을 보였다.

철근콘크리트흄관 라이닝용 니켈계 방균제의 기초적 특성 연구 (A Study on the Base Properties of Nickel Type-Antifungal Agent for Reinforced Concrete Hume Pipe Lining)

  • 본 연구는 철근콘크리트 흄관의 내구성을 개선시킬 목적으로 흄관 내부에 폴리머 시멘트 모르타르로 라이닝 처리한 부분에 황산니켈 6수화물을 혼입시켜, 하수용으로 사용되는 철근콘크리트 흄관의 열화에 크게 영향을 미치는 황산화 세균의 번식을 억제하기 위하여 실시하였다. 본 연구에서는 황산화세균의 증식을 억제시키기 위하여 황산니켈 6수화물의 농도에 따른 성능을 평가하였으며, 방균제에 함유된 황산이온이 시멘트 모르타르의 팽창요인 되어 역학적성질에 영향을 미칠 수 있기 때문에 팽창성 시험과, 니켈의 중금속의 용출로 환경오염은 물론 흄관의 내구성에 영향을 미치기 때문에 용출시험도 실시하였다. 본 연구 결과, 니켈계 방균제가 시멘트 콘크리트의 부식균인 Thiobacillus novellus의 증식을 억제하기 위해서는 20mM 이상을 사용하여야 효과가 있는 것으로 평가되었으며, 방균제를 혼입한 시멘트 모르타르의 압축강도 및 휨강도는 방균제의 혼입여부에 따라 크게 영향을 받지 않았다. 또한 방균제 혼입에 따른 특별한 팽창현상은 발견되지 않았으며, 모든 종류의 시멘트 모르타르에서 니켈성분이 전혀 용출되지 않았다. 본 연구를 통하여 철근콘크리트 흄관의 내구성 개선을 위하여, 흄관 내부의 라이닝과 함께 방균제를 혼입한다면, 두가지 방식재료의 성능이 서로 보완효과를 일으킬 수 있을 것으로 판단되었다.

Identification and Characterization of an Antifungal Protein, AfAFPR9, Produced by Marine-Derived Aspergillus fumigatus R9

  • A fungal strain, R9, was isolated from the South Atlantic sediment sample and identified as Aspergillus fumigatus. An antifungal protein, AfAFPR9, was purified from the culture supernatant of Aspergillus fumigatus R9. AfAFPR9 was identified to be restrictocin, which is a member of the ribosome-inactivating proteins (RIPs), by MALDI-TOF-TOF-MS. AfAFPR9 displayed antifungal activity against plant pathogenic Fusarium oxysporum, Alternaria longipes, Colletotrichum gloeosporioides, Paecilomyces variotii, and Trichoderma viride at minimum inhibitory concentrations of 0.6, 0.6, 1.2, 1.2, and 2.4 μg/disc, respectively. Moreover, AfAFPR9 exhibited a certain extent of thermostability, and metal ion and denaturant tolerance. The iodoacetamide assay showed that the disulfide bridge in AfAFPR9 was indispensable for its antifungal action. The cDNA encoding for AfAFPR9 was cloned from A. fumigatus R9 by RT-PCR and heterologously expressed in E. coli. The recombinant AfAFPR9 protein exhibited obvious antifungal activity against C. gloeosporioides, T. viride, and A. longipes. These results reveal the antifungal properties of a RIP member (AfAFPR9) from marine-derived Aspergillus fumigatus and indicated its potential application in controlling plant pathogenic fungi.

Streptomyces sp. A252의 배양적 특성 및 항진균활성 (Antifungal Activity and Cultural Characteristics of the Streptomyces sp. A252)

  • A252 균주의 항균성물질의 생성 및 균주의 배양적 특성을 조사한 결과 균체의 생장은 TSB와 malt extract-yeast extract (ISP-2) 배양기가 좋았으며, 배양여액의 항균활성은 TSB와 nutrient broth에서 효과적이였다. TSB 배지에서 균사생장 및 배양여액의 항균활성은 $25^{\circ}C$와 pH 6.5에서 최적화 되었으며, 2$\%$ TSB 농도에서는 1$\%$에 비해 생장은 좋았으나 배양여액의 항균활성에는 차이가 없었다. A252 균주의 균사 생장은 접종 72시간 후에 최대치에 달하였으며 반면 배양액의 항균력은 접종 60시간 후인 idiophase에 가장 높은 수준을 보였지만 96시간 후에는 약간 감소하는 경향을 나타냈다. 배양여액의 항균활성은 $121^{\circ}C$ 고온에서도 안정하였으며, pH에 대한 안정성은 4.0 - 9.0의 범위였다. A252 균주의 생리적 특성 및 분생자경의 형태 등을 관찰한 결과 Streptomyces sp.로 동정되었다.

메주로부터 분리한 Bacillus polyfermenticus CJ6의 항진균 활성 (Antifungal Activity of Bacillus polyfermenticus CJ6 Isolated from Meju)

  • 메주로부터 곰팡이 및 세균 등에 생육 저해활성을 나타내는 균주 B. polyfermenticus CJ6을 분리 동정하였다. 분리 균주 B. polyfermenticus CJ6는 2단 대수기를 나타내는 생육 곡선상 특이점을 나타내었으며 배양 30시간 이후부터 최대 활성을 나타내었고 사멸기 이후 활성이 다소 감소되었으나 120시간까지 활성을 유지하였다. B. polyfermenticus CJ6의 항진균 활성 물질은 $70^{\circ}C$ 이상에서 활성이 감소되었으나 $121^{\circ}C$에서 15분간 열처리 시 역가가 완전히 소실되지 않았다. pH 안정성 실험에서는 pH $3.0{\sim}9.0$ 구간에서 안정한 활성을 나타내었으며, 각종 효소에 대한 영향에서 항진균 활성물질은 proteinase K, protease, ${\alpha}$-chymotrypsin 등의 단백분해효소 처리로 역가를 상실하거나 일부 감소되어 단백질성 물질임을 추정하였다. 균주의 항진균 활성 물질을 $C_{18}$ Sep-Pak column으로 부분 정제한 후 Tricine-SDS-PAGE 및 direct detection 실험을 통하여 분자량이 약 1.4 kDa의 물질임을 확인하였다. B. polyfermenticus CJ6가 생산하는 항진균 활성 물질은 기존에 거의 보고되지 않은 B. polyfermenticus 유래의 단백질성 항진균 활성 물질로서 천연보존제 및 천연항균제재로 사용이 기대되며, 이를 위하여 항진균 활성 물질들의 정제 및 구조분석 등의 연구가 필요하다.

목재문화재 보존을 위한 수목추출물의 부위별 생리활성 탐색 (Studies of biological activities of tree extracts for conservation of wooden cultural properties)

  • Wooden artifacts are biologically damaged by animals, insects, and micro-organisms. We focused on the evaluation of the antifungal and insecticidal activities of tree extracts to control deterioration of wooden cultural properties. 12 kinds of methanol extracts from 9 species of tree were investigated for their biocidal activities against two micro-organisms and two spices of insect. Most tree extracts showed fungicidal activities to Tyromyces palustris and Trametes versicolor. Among 12 different tree extracts, the strongest antifungal activity against T.palustris and T.versicolor was observed from the extracts of Pinus densiflora stem-bark extract and Pinus koraiensis leaf extracts. However, in the insecticidal activity test, Pinus densiflora stem-bark extracts against termite was relatively effective. Chemical compositions of methanol extracts were analyzed by gas chromatography/mass spectrometry. As a result, it was confirmed that development of biocide based on the natural extracts can be expected to conservate of wooden cultural properties.

항진균제를 첨가한 수종의 점막 조정제가 Candida albicans 성장 억제에 미치는 효과 (THE INHIBITING EFFECT OF TISSUE CONDITIONER COMBINED WITH ANTIFUNGAL AGENTS ON THE GROWTH OF CANDIDA ALBICANS)

  • Tissue conditioners have been used for treatment of denture stomatitis caused by wearing of dentures. Early studies pointed out Candide albicans (C. albicans) as main etiologic factor, and antifugal agents were added for control of the species. But there is a little information about broad comparison on the effect of tissue conditioners and antifungal agents added. The purpose of the present study was to compare the inhibiting effect of four tissue conditioners and one temporary soft liner on the growth of C. albicans for treatment of denture stomatitis using gel diffusion method by measuring diameter of the zone of growth inhibition. Three antifungal agents were added to each material for evaluation of the effect of added agents. Finally, observation was made to evaluate the effect of the loss of antifungal elements by aging of the specimen. The results of this study were obtained as follows : 1. Tempo had remarkable antifungal effect showing the zone of growth inhibition as 2.35 mm at 1st day, and was most effective on End: 4th and 7th day from incubation (p<0.05). But Coecomfort, Dura conditioner, Visco-gel, Coe-soft had little antifungal effect from the 1st day of incubation. 2. Nystatin was most effective showing 9.60-12.04 mm of zone of inhibition at the 1st day from incubation. The antifungal properties were reduced to amphotericin B, chlorhexidine and materials without agent (p<0.05), and the effect was diminished by time. 3. As pretreatment with amphotericin B, nystatin, chlorhexidine, Tempo was very effective at the 1st day from incubation showing zone of inhibition as 3.65, 12.04, 4.78 mm with addition of each agent. Dura conditioner had strongest antifungal effect at the next day as 2.86, 5.33, 1.29 mm of zone of inhibition, and yielding results of Coe-comfort, Tempo, Coe-soft was shown at 4th and 7th day from incubation (p<0.05). Taken all together, tissue conditioners have little antifungal effect except Tempo. Formation of the zone of growth inhibition was due to agents amphotericin B, nystatin, chlorhexidine and nystatin was most effective. Conclusively, it is advisable to select material which is effective on the growth of C. albican and consider addition of antifungal agents for treatment of denture stomatitis.
