Comparison of Antifungal Activites of Monoterpenes and Sesquiterpenes in Essential Oil from Chamaecyparis obtusa against Dermatophytes (피부사상균에 대한 편백정유의 Mono- 및 Sesquiterpene 항진균 활성 비교)

  • This study was to compare the antifungal activitiy of monoterpenes with that of sesquiterpenes, which were major constituents of antifungal active essential oil of Chamaecyparis obtusa, to investigate which constituents had more effective against dermatophytes, and to evaluate the synergistic effect of combination of the antifungal active constituents. The antifungal activities of seven fractions (A, B, C, D, E, F, and G) from C obtusa essential oil by column chromatography were tested against Trichophyton mentagrophytes. Fraction D, E, F, and G were more active than fraction A, B, and C, and their major compounds were mono- and sesquiterpenes analyzed by GC/MS. Borneol, linalool, and ${\alpha}$-terpineol were selected as monoterpenes, and ${\alpha}$-cedrol, nerolidol, and ${\beta}$-eudesmol as sesquiterpenes, respectively. The antifungal activities of the constituents were respectively examined against Trichophyton mentagrophytes (KCTC6077), Microsporum canis (KCTC6591), and Microsporum gypseum. The constituents of sesquiterpenes were more active than those of monoterpenes. By comparing single and combined sesquiterpenes with fraction F containing the higher ratio of sesquiterpenes, combined sesquiterpenes were the most active. This result indicated that there was the synergistic antifungal effect against dermatophytes when sesquiterpenes were combined together.

Bacillus polyfermenticus CJ9, Isolated from Meju, Showing Antifungal and Antibacterial Activities (메주로부터 분리한 항진균 및 항세균 활성의 Bacillus polyfermenticus CJ9)

  • A CJ9 bacterial strain, which showed antifungal and antibacterial activities, was isolated from meju and identified as Bacillus polyfermenticus based on Gram staining, biochemical properties, as well as its 16S rRNA sequence. B. polyfermenticus CJ9 showed the antimicrobial activity against the various pathogenic molds, yeasts, and bacteria. The antibacterial activity was stable in the pH 5.0~9.0, but the activity was lost at $37^{\circ}C$ for 24 hr. The antifungal activity was stable in the pH range of 3.0~9.0 and reduced at $121^{\circ}C$ for 15 min, but antifungal activity was not completely destroyed. The antibacterial activity was completely inactivated by proteinase K, protease, trypsin, and $\alpha$-chymotrypsin. The antifungal activity was also completely inactivated by protease and $\alpha$-chymotrypsin, and reduced its activity by proteinase which indicated that the antifungal and antibacterial compounds have proteineous nature. The apparent molecular mass of the partially purified antifungal compound, as indicated by using the direct detection method in Tricine-SDS-PAGE, was approximately 1.4 kDa. The molecular mass of the antibacterial compound could not be determined because of its heat-liable characteristic.

Antifungal Activities of Pseudomonas spp. Strains Against Plant Pathogens and Optimization of Culture Conditions (식물병원성 진균에 항균 효과를 지닌 슈도모나스 균주의 항진균 활성 증진을 위한 배양조건의 최적화)

  • To define the optimum conditions for the mass production of four antifungal Pseudomonas spp. isolated from soil, we have investigated culture conditions and effects of various nutrient sources on the bacterial growth and evaluated antagonistic activity against Rhizoctonia solani and Sclerotinia homoeocarpa, plant pathogens. The optimum temperature and pH for the growth of these isolates were determined as pH 7.0 and $20^{\circ}$ or $25^{\circ}C$, respectively. Sucrose, tryptone, and $K_2HPO_4$ generally were more adequate for better growth as carbon, nitrogen and mineral source, respectively. The nutrient sources were also found to be very effective for high antifungal activities against R. solani and S. homoeocarpa. It was elucidated that YUD-F group (P. mandelii and P. fluorescens), which inhabit regions at relatively low temperature, had more broad spectrum and higher antifungal activity than YUD-O group (P. trivialis and P. jessenii) generally against R. solani and S. homoeocarpa. It is thought that the differences of the average temperature in the various habitats of Pseudomonas spp. influence the optimal growth temperature and antifungal activity. Especially, Pseudomonas spp. of YUD-O group showed the better antifungal activity against dollar spot caused by S. homoeocarpa, but showed relatively weaker antifungal activity against brown patch caused by R. solani.

Detection of Endolichenic Fungi Producing Antifungal Compound (항진균성 물질을 생산하는 지의류 내생 곰팡이의 탐색)

  • To isolate a novel antifungal compound, we obtained 100 kinds of endolichenic fungi from Korean Lichen & Allied Bioresources Center and examined their antifungal capability. Three fungi Usnea rigidula (2326), Parmotrema pseudotinctorum (2202) and Myelochroa sp. (2292) showed high antifungal activity against Candida albicans when they grew in both liquid and solid media. We extracted the culture supernatants of these three fungi with chloroform and then with ethyl acetate. Chloroform fraction exhibited the highest antifungal activities when those fractions were examined for the growth inhibition of Candida albicans with disc diffusion method. The chloroform faction was on further analysis with $C_{18}$ column chromatography to see whether the inhibitors are already known or not. Two peak fractions were collected from 4-day culture extract for Usnea rigidula and from 6-day culture extract for Parmotrema pseudotinctorum on the HPLC. A peak fraction from chloroform extracts of 4-day culture filtrate of Parmotrema pseudotinctorum showed higher antifungal activities against C. albicans and C. glabrata than another peak fraction. It appears that the antifungal materials are relatively nonpolar as usnic acid often found in lichenic fungi.

Antimicrobial and Antifungal Activities of Lisianthus (Eustoma grandiflorum) Essential Oil (리시안셔스 유래 에센셜 오일의 항세균 및 항진균 효과)

  • Essential oils are fragrant oils extracted from the leaves, stems, peels, petals and roots of aromatic plants cultivated by natural means or using organic agricultural techniques. Essential oils have commonly been used as antibacterial and antifungal agents. In the present study, essential oil was extracted from lisianthus (Eustoma grandiflorum [Raf.] Shinn.) and tested for antifungal activities against three eumycetes (Penicillium pinophilum, Chaetomium glogosum and Aspergillus niger). Lisianthus essential oil showed high antifungal activities against three eumycetes, especially against Aspergillus niger, for which the resulting minimum inhibitory concentration (MIC) was 0.005 mg/ml. In addition, the extracted essential oil was shown to have antimicrobial activity against ten intestinal pathogenic bacteria (Escherichia coli, Salmonella typhimurium, Klebsiella pneumonia, Staphylococcus aureus, Bacillus cereus, Listeria monocytogenes, Pseudomonas aeruginosa, Bacillus subtilis, Enterococcus faecalis and Vibrio parahaemolyticus) according to the disc diffusion method and was also shown to exhibit strong antibacterial activity against an additional three pathogenic bacteria (Bacillus subtilis, Listeria monocytogenes and Vibrio parahaemolyticus). These results indicate that lisianthus essential oil could be used as an antibiotic against harmful bacteria that produce intestinal illnesses. From the present study, we suggest that lisianthus extracts can be utilized as potential antifungal and antibacterial agents and for the development of pharmaceutical and cosmetic products.

Seed Disinfectant Effect of Pleurotus ostreatus (Heuktari) Extract on Fusarium fujikuroi Nirenberg (흑타리버섯 추출물의 벼 키다리병에 대한 종자소독 효과)

  • This study was carried out to investigate antimicrobial activities of water extracted Pleurotus ostreatus var Heuktari (PO), and their application to F. fujikuroi Nirenberg growth inhibition material. Various organic solvents (chloroform, hexane, ethyl acetate, ethanol) were tested to investigate the antimicrobial activities of Pleurotus ostreatus var Heuktari against F. fujikuroi Nirenberg. Chloroform, hexane, ethyl acetate and hot water extracts had no antimicrobial activities, but butanol extract showed 2.5% strong activities in order of F. fujikuroi Nirenberg disk diffusion test. Then we observe Antifungal activity using green house. Bed soil and mushroom extract mixed at 10%, 7.5%, 5%, 2.5%, 1% total bed soil mass. Screening of Antifungal activity was tested two periode 18day and 25day. The Antifungal activity rate of each period and extract density was 7.5% mixed bedsoil has investigated 80% Antifungal activity rate. The result indicates that the Pleurotus ostreatus var Heuktari extract using F. fujikuroi Nirenberg registance, which shows the development of organic agriculture seed disinfectant.

  • The relationship between chemical structure and antifungal activity of benzene ring substituents of 3-phenyl-5,5-dimethyloxazolidine-2,4-diones is studied by testing of model compounds and use of conformational data. The analysis revealed a number of structural features as essential for the antifungal effect: (1) the presence of an intact -NCO group and benzene moiety; (2) the distance between para substituents and the N atom within the 5.32∼5.37${\AA}$ range; (3) the distance between substituents X3 and X4, in the interval 5.59 ∼ 5.77 ${\AA}$ for the active analogues.

  • The purpose of this study was to investigate the functional properties of natural dyed Hanji for cultural properties conservation and storage. To estimate functional properties of Hanji, antimicrobial, insect repellent test, and Oddy test were executed. Hanji dyed with 10% Golden thread (Coptis chinensis), a natural dye and silver nitrate mordanting showed the most excellent antifungal activities against A. niger, P. chrysogenum, and Trichoderma sp. before and after aging. The insects repellent test followed didn't indicate significant effects on treated Hanji against Lasioderma serricorne, Sitophilus zeamais, and Tribolium castaneum. According to Oddy test result, Hanji dyed with Golden thread and silver nitrate caused no significant corrosions on copper, silver, and lead after aging 28 days.

  • 6-[(N-3,4-Dibromophenyl)amino]-7-chloro-5,8-quinolinedione(FCK13) was tested for antifungal activities. The MIC values were determined by the two-fold dilution method. The therapeutic potential of RCK13 had been assessed in comparison with ketoconazole and fluconazole against systemic infections with candida albicans in normal mice. RCK13 had ED50,0.80$\pm$0.21 mg/kg but ketoconazole had ED50, 8.00$\pm$0.73 mg/kg respectively. And administered RCK13 at the ED50 for 14 days improved survival rates as well as ketoconazole. Acute oral toxicity studies of RCK13 were carried out in ICR mice of both sexes. These acute oral toxicities of RCK13 were low and LD50 values were over 2,850 mg/kg in ICR mice. The genotoxicities of RCK13 had been evaluated. RCK13 was negative in Ames test with Salmonella typhimurium and chromosomal aberration test in CHL cells. The clastogenicity was tested on the RCK13 with in vivo mouse micronucleus assay. RCK13 did not show any clastogenic effect in mouse peripheral blood and was negative in mouse micronucleus assay. These results indicate that RCK13 has no genotoxic potential under these experimental conditions.

Study of antibacterial and antifungal activity of traditional Cedrus deodara and Pinus roxburghii Sarg

  • In the present study, the volatile oil, chloroform extract and methanol extract of the woods of the plants Cedrus deodara and Pinus roxburghii were screened for their antibacterial and antifungal activities against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus subtilis, Streptococcus pyogenes, Candida albicans, Aspergillus niger, and Aspergillus clavatus using the Agar diffusion method. The susceptibilities of the microorganisms to the extracts were compared with each other and with a selected standard antibiotic. It was observed that the volatile oil and chloroform extracts showed the significant antibacterial activities while the least antibacterial activity was recorded with the methanolic extracts. The higher Minimum Inhibitory Concentration value of the extracts and oils against fungus suggested that the plants may possess less antifungal activity. Phytochemical analysis and thin-layer chromatography profiling revealed the presence of flavonoids and terpenoids in the oil and chloroform extracts, which could explain the antimicrobial activity. The findings suggest that the Cedrus deodara and Pinus roxburghii have antimicrobial properties and they can be used in the treatment of infectious diseases. However, further work is required in order to isolate the active constituents of the plants responsible for the antibacterial activity.
