Structure-Antifungel Activity Relationships of Cecropin A Hybrid Peptides against Trichoderma sp.

  • The hybrid peptides, CA-ME, CA-MA and CA-BO, with the N-terminal sequence 1-8 of cecropin A and the N-terminal sequences 1-12 of melittin, magainin 2 and bombinin, respectively, have more improved antibacterial activities. CA-MA was found to have stronger antifungal activity against Trichoderma sp than other hybrid peptides and their parental peptides. In order to elucidate the relationships between the peptide structure and antifungal activity, several analogues of CA-MA or CA-BO were also designed and synthesized by the solid phase method. An tifungal activity was measured against T. reesei and T. viride, and hemolytic activity was measured by a solution method against human red blood cells. The residue 16 of CA-MA, Ser, was found to be important for antifungal activity. When the residue was substituted with Leu, showed powerful antifungal activity was dramatically decreased. CA-MA, P1, P4 and P5 designed in this study showed powerful antifungal activity against T. reesei and T. viride with low hemolytic activity against human red blood cells. These hybrid peptides will be potentially useful model to further design peptides with powerful antifungal activity for the effective therepy of fungal infection and understand the mechanisms of antifungal actions of hybrid peptides.

항진균제를 첨가한 수종의 점막 조정제가 Candida albicans 성장 억제에 미치는 효과 (THE INHIBITING EFFECT OF TISSUE CONDITIONER COMBINED WITH ANTIFUNGAL AGENTS ON THE GROWTH OF CANDIDA ALBICANS)

  • Tissue conditioners have been used for treatment of denture stomatitis caused by wearing of dentures. Early studies pointed out Candide albicans (C. albicans) as main etiologic factor, and antifugal agents were added for control of the species. But there is a little information about broad comparison on the effect of tissue conditioners and antifungal agents added. The purpose of the present study was to compare the inhibiting effect of four tissue conditioners and one temporary soft liner on the growth of C. albicans for treatment of denture stomatitis using gel diffusion method by measuring diameter of the zone of growth inhibition. Three antifungal agents were added to each material for evaluation of the effect of added agents. Finally, observation was made to evaluate the effect of the loss of antifungal elements by aging of the specimen. The results of this study were obtained as follows : 1. Tempo had remarkable antifungal effect showing the zone of growth inhibition as 2.35 mm at 1st day, and was most effective on End: 4th and 7th day from incubation (p<0.05). But Coecomfort, Dura conditioner, Visco-gel, Coe-soft had little antifungal effect from the 1st day of incubation. 2. Nystatin was most effective showing 9.60-12.04 mm of zone of inhibition at the 1st day from incubation. The antifungal properties were reduced to amphotericin B, chlorhexidine and materials without agent (p<0.05), and the effect was diminished by time. 3. As pretreatment with amphotericin B, nystatin, chlorhexidine, Tempo was very effective at the 1st day from incubation showing zone of inhibition as 3.65, 12.04, 4.78 mm with addition of each agent. Dura conditioner had strongest antifungal effect at the next day as 2.86, 5.33, 1.29 mm of zone of inhibition, and yielding results of Coe-comfort, Tempo, Coe-soft was shown at 4th and 7th day from incubation (p<0.05). Taken all together, tissue conditioners have little antifungal effect except Tempo. Formation of the zone of growth inhibition was due to agents amphotericin B, nystatin, chlorhexidine and nystatin was most effective. Conclusively, it is advisable to select material which is effective on the growth of C. albican and consider addition of antifungal agents for treatment of denture stomatitis.

끈적긴뿌리버섯(Oudemansiella mucida)의 최적배양조건 및 배양 추출액의 항균작용에 관한 연구 (The Optimal Culture Conditions and Antifungal Activity of Culture Extract from Oudemansiella mucida)

  • 본 연구는 식 의약용버섯으로 알려진 끈적긴뿌리버섯(Oudemansiella mucida) 배양액에서 추출한 물질이 항진균효과를 갖고 있는지 규명하기위한 실험을 수행하였다. 균사체의 배양액에서 항진균 물질을 생산하기 위한 전단계로 끈적긴뿌리버섯의 균사생장에 적합한 온도, pH, 탄소영양원, 질소영양원, 탄소/질소 비율 및 최적의 고체배지 등을 선발하기 위한 실험을 수행하였다. 균사생장에 최적인 온도와 pH는 각각 $25^{\circ}C$와 pH 5이었으며, 최적 탄소영양원과 질소영양원은 각각 maltose와 calcium nitrate였다. 균사생장을 위한 탄소/질소 영양원의 비율이 20 : 1이고 배지의 glucose 농도가 3%일 때, 균사생장이 가장 활발하였으며, 최적의 고체배지에는 Hennerberg 배지가 선정되었다. 균사생장의 실험 결과를 토대로 끈적긴뿌리버섯의 균사체를 배양한 후 그 배양여액을 이용하여 식물병원 곰팡이에 대한 항진균 효과를 규명하였다. 끈적긴뿌리버섯의 균사체 배양액에서 추출한 물질과 시판되고 있는 항진균 농약을 이용하여 Colletotrichum acutatum, Botrytis cinerea 및 Pyricularia oryzae 등 식물병원성 곰팡이를 대상으로 균사생장과 포자발아를 저해하는 효과를 조사한 결과 끈적긴뿌리버섯의 균사체에서 생산된 물질은 식물병원균의 균사생장을 저해하는 효과를 나타내었으나 공시된 항진균 농약에 비해 저해효과는 낮게 나타났다. 공시된 식물병원균 포자의 발아를 억제하는 효과는 folpet을 제외한 모든 공시 농약에 비해 낮게 나타났다. 따라서 끈적긴뿌리버섯의 균사체 배양액에서 추출한 물질에는 항균성을 나타내는 물질을 함유되어 있어서 새로운 항진균 제제로 개발할 수 있는 가능성이 있는 것으로 사료된다.

이탄 토양으로부터 식물생육촉진세균 Pseudomonas sp. SH-26의 분리 및 특성 (Isolation and Characterization of Plant Growth Promoting Bacteria Pseudomonas sp. SH-26 from Peat Soil)

  • 이탄 토양으로부터 분리된 세균의 유기물 분해능, 항균 활성 및 식물생육촉진능을 확인하고 다기능성 유용 미생물을 확보하고자 하였다. 분리된 48 균주를 대상으로 β-glucosidase, cellulase, amylase, protease 생성능을 확인한 결과, SH-23, SH-26, SH-29 그리고 SH-33 균주가 우수 균주로 선발되었다. 선발된 유기물 분해 우수 세균 4 균주는 식물병원성곰팡이(Botrytis cinerea, Rhizoctonia solani, Fusarium oxysporum, Colletotrichum acutatum)에 대한 항진균 활성을 나타내었다. 유기물 분해능이 우수하고 식물병원성 곰팡이에 대해 항진균 활성을 나타낸 SH-26 균주를 최종 우수균주로 선발 하였다. 선발된 SH-26 균주의 16S rRNA 유전자 염기 서열을 결정한 결과, Pseudomonas knackmussii HG322950 B13T, Pseudomonas citronellolis BCZY01000096 NBRC 103043T, 그리고 Pseudomonas delhiensis jgi.1118306 RLD-1T와 100%의 상동성을 나타내었다. Pseudomonas sp. SH-26 균주는 siderophore 생성능, 질소고정능과 Indole-3-acetic acid 생성능이 있는 것으로 확인되었다.
