This study assessed the contamination level of food poisoning bacteria on handles of food service equipment in childcare centers to prevent food poisoning and analyzed toxin genes and antibiotic resistance of isolated strains. The isolates used in this study were collected from 101 childcare centers in Jeollanam-do. Four strains of Bacillus cereus and two strains of Staphylococcus aureus were isolated on the handles of food service equipment (refrigerators and freezers). The toxin genes of B. cereus were detected as nheA, nheB, nheC, entFM, and cytK. No toxin genes of S. aureus were detected. B. cereus showed resistance to β-lactam antibiotics, such as ampicillin and cefepime. S. aureus also showed resistance to antibiotics such as ampicillin and cefepime. Therefore, microbial safety and hygiene management, such as periodic sterilization of handles, should be strengthened to prevent food poisoning caused by cross-contamination of food service equipment handles in childcare centers.

Frequent isolation of Enterococcus faecalis from root canal treated teeth with apical periodontitis, has proposed the role of this organism in endodontic treatment failures. Different factors have been suggested in the pathogenicity of this organism. In this study, 22 E. faecalis isolates from canals of root-filled teeth were identified, and phenotypic and genotypic characteristics were investigated. No resistance to vancomycin and gentamicin was noted, and most isolates (91%) were susceptible to ampicillin. Biofilm formation was detected in 73% of the isolates and may be considered as the most important virulence factor involved in the pathogenesis of these isolates.
This study was conducted to investigate the characteristics of antibiotic resistant E. coli. its antibiotic susceptibility and pathogenicity were analyzed via molecular convergence technique, for the relationship of antibiotic susceptibility and pathogenicity. The 60 isolated strains consisted of ESBL(+)(8) and ESBL(-)(52) strains. The ESBL(+)(8) strains consisted of 2 strains without a pathogenic gene, stb(3), flich7(1), and flich7-eae(2). The ESBL(-)(52) strains consisted of 26 strains without a pathogenic gene, stx1(3), stb(10), flich7(2), eae(2), stx1-flich7(2), stx1-stb(4), flich7-stb(2), and flich7-stb-eae(1). In conclusion, antibiotic resistance is increasingly, Focused on molecular convergence, showed the correlation of pathogenicity with antibiotic resistance was poor. However, It will be able to find the exact pathogenic factor in the future through convergence technique including the analysis of virulence genes.
This study aimed at determining the genetic and virulence characteristics of the Listeria monocytogenes from smoked ducks. L. monocytogenes was isolated by plating, and the isolated colonies were identified by PCR. All the obtained seven L. monocytogenes isolates possessed the virulence genes (inlA, inlB, plcB, and hlyA) and a 385 bp actA amplicon. The L. monocytogenes isolates (SMFM2018 SD 1-1, SMFM 2018 SD 4-1, SMFM 2018 SD 4-2, SMFM 2018 SD 5-2, SMFM 2018 SD 5-3, SMFM 2018 SD 6-2, and SMFM 2018 SD 7-1) were inoculated in tryptic soy broth (TSB) containing 0.6% yeast extract at 60℃, followed by cell counting on tryptic soy agar (TSA) containing 0.6% yeast extract at 0, 2, 5, 8, and 10 min. We identified five heat resistant isolates compared to the standard strain (L. monocytogenes ATCC13932), among which three exhibited the serotype 1/2b and D-values of 5.41, 6.48, and 6.71, respectively at 60℃. The optical densities of the cultures were regulated to a 0.5 McFarland standard to assess resistance against nine antibiotics after an incubation at 30℃ for 24 h. All isolates were penicillin G resistant, possessing the virulence genes (inlA, inlB, plcB, and hlyA) and the 385-bp actA amplicon, moreover, three isolates showed clindamycin resistance. In conclusion, this study allowed us to characterize L. monocytogenes isolates from smoked ducks, exhibiting clindamycin and penicillin G resistance, along with the 385-bp actA amplicon, representing higher invasion efficiency than the 268-bp actA, and the higher heat resistance serotype 1/2b.
Recently, a number of countries are now considering the reuse of effluents from wastewater treatment for various water applications. To improve the reuse of wastewater effluent, the development of appropriate micro-pollutant removal technology is necessary. Although several researche have been studied for removing micro-pollutants in water, little study has been conducted for the removal of emerging contaminant such as antibiotic resistant genes (ARGs) by disinfection processes. Therefore, the aim of this study is to compare the capacity of disinfection technologies such as chlorination, ozone, and electron beam, for removing antibiotic resistant bacteria (ARB) and ARGs. Based on this study, better ARG removal can be achieved by ozonation and electron beam. Relatively, high CT values of chlorination or ozonation are needed to remove ARB and ARG compared to conventional pathogens.
Tetracycline (TC) is one of the antibiotics used for treatment of bacterial infection in Korea. Inadequate usage and abuse cause the resistance to antibiotics, like Tetracycline, Erythromycin, and Fluoroquinolone. It can also affect severe economic loss in aquaculture field in Korea. We isolated 101 bacterial samples from diseased fish at aquaculture sites in Gyeonggi-do during 2015~2018. Minimum inhibitory concentration (MIC) method has been used to determine distribution and to identify bacterial isolates resistant to antibiotics including Oxytetracycline (OTC), Ampicillin (AMP), Clindamycin (CLI), Enrofloxacin (ENRO), Gentamycin (GEN). TC resistant isolates were confirmed antibiotic resistant genes by conventional PCR. Bacterial isolates were identified as Aeromonas spp. (43.5%), Pseudomonas spp. (4.0%) and Vibrio spp. (5.0%). It was confirmed that multi-resistant isolates (77.2%) were predominant over single-resistant one (22.8%). TC resistant genes like tet(A), tet(D), tet(E), tet(G), tet(M), and tet(S) were detected and tet(A) was the most prevalent. Aeromonas spp. is a dominant strain in bacterial infections in fishes of aquaculture sites, and further investigation on various antibiotic resistance genes will be needed for clear understanding of aquaculture sites in Gyeonggi-do.
The occurrence of tetQ and aacC2 genes encoding tetracycline and gentamicin resistance determinant, respectively, was assessed in total bacterial community DNA isolated from Dongchon stream of Sunchon area. To examine the resistance potential of bacteria that were not cultured, total DNA from 1 liter of stream water was extracted by freeze-thaw method. The PCR technique was employed to determine the abundance of the target genes. The highest frequency of tetQ gene was obtained from site 1, located near the animal farms area, whereas the incidence of aacC2 was highest in site 5, the downstream area. These results showed that the occurrence of antibiotic resistance gene may be used as a convenient marker of water quality related to source.

The purpose of this study was to investigate the ability to treat and prevent infection by multiple Gram-negative bacterial pathogens as a last choice option in the treatment of serious infections in clinical settings. The global spread of extended-spectrum 𝛽-lactamases (ESBLs) and/or carbapenemases in microorganisms are of enormous concern to health services because they are often associated with multi-drug resistance which significantly restricts the antibiotic treatment options. In this study, the antimicrobial resistance profiles of bacteria isolated from South Korean market-derived meat samples were determined by the disc diffusion method. PCR was used to detect the presence of antibiotic resistance genes and ESBL producing genes. In total, we tested 181 isolated colonies from 36 market-derived meat samples. Single PCR and DNA sequencing results revealed that genes blaVIM, blaBIC, blaKPC, and blaSIM were present in the bacteria isolated from retail meat. The bacteria in the meat were separately sequenced and based on alignment, four different bacteria were identified. These findings suggest that bacteria found in retail meats are a reservoir for the spreading of ESBL blaVIM, blaBIC, blaKPC, and blaSIM resistance genes and bacteria strains.
Two macrolide-lincosamide-streptogramin B (MLS) antibiotic resistance genes, one expressed inducibly and the other expressed constitutively were recognized from a single Staphylococcus aureus DH1 strain. The inducible MLS resistance gene was isolated and cloned from the R-plasmid pDE1(7.4kb) and the constitutive gene was from chromosomal DNA. Base sequence of the inducible MLS resistance gene (1.2kb) was determined and found as same that of pE194. The restriction map of the cloned constitutive MLS resistance gene was compared with that of the inducible gene. Two genes have same restriction map except leader region. In the constitutive gene there is no leader region which is doing major role in inducible expression.
Decreased porin-mediated outer membrane penetration of hydrophilic antibiotics is a common mechanism of antibiotic resistance in Gram-negative bacteria. This study was undertaken to determine whether a null mutation in Pseudomonas putida would suppress porin synthesis, and therefore reduce the susceptibility of the organism to streptomycin, norfloxacin, and tetracycline. Inverse PCR amplification and double-stranded DNA sequencing were used to identify chromosomal genes carrying TnphoA'-1 inserts. Genome database available was used to identify putative homologue genes, one of which encodes protein with homology to domains of the MutS of P. putida, suggesting a crucial role in the multidrug resistance. Increased resistance to streptomycin, norfloxacin, and tetracycline might be due to accumulation of compensatory mutations. Either no growth or slow growth was observed in P. putida KH1027 when grown in minimal medium containing gluconate, glucose, or citrate; however, it is not clear whether the growth patterns contributed to the multidrug resistance.
