마우스 대뇌감염모델을 이용한 Acyclovir의 항Herpes Simplex Virus Type 1 약효평가 (Evaluation of Anti-Herpes Simplex Virus Type 1 Activity of Acyclovir by Using Mouse Intracerebral Infection Model)

  • To establish in vivo antiviral evaluation system by using murine herpesvirus intracerebral infection model, 5-6 female BALB/c mice per group aged 5 weeks were inoculated i.c. into cerebrum with different inocular HSV-1 F. Signs of clinical disease noted everyday for one month. Observed were body weight decrease, neurological signs and death caused by encephalitis. Mice discontinued body weight decrease were recovered from the disease, and keratitis was often observed during recovery. The groups inoculated with higher than 1,000 PFU showed 100% mortaltiy and $LD_{50}$ was <100 PFU/mouse. To study the effect of virus inoculum sizes on antiviral effect of acyclovir (ACV), mice inoculated with different inocula were administered i.p. with different doses of ACV immediately after infection, and twice a day for 5 days. The higher inculum size, the less protective. $ED_{50}$ of ACV was >25, >25, 18.4 and 8.0 mg/kg b.i.d. in the group infected with 1,000,000, 100,000, 10,000 and 1,000 PFU/mouse, respectively. $LD_{50}$ of ACV was 62.5 mg/kg b.i.d. Therapeutic index of ACV was <2.5, <2.5, 3.0 and 7.0 in the groups with inocula 1,000,000, 100,000, 10,000 and 1,000 PFU/mouse, respectively. Inoculum size 1,000 PFU/mouse showing 100% mortaltiy and 5-6 days mean time to death, 5 days drug administration and 14 days observation will be future experimental conditions.

  • 최근 안드로이드 기반의 스마트폰을 대상으로 한 해킹기법중 가장 활발한 것은 스미싱(Smishing)이다. 스미싱은 SMS, MMS등 문자메세지를 이용한 새로운 해킹기법으로 사용자에게 악성코드애플리케이션(이하 악성앱)을 다운로드할 수 있는 주소가 포함된 메세지를 보내 개인정보를 수집하거나 스마트폰을 제어하고, 특히 사용자가 알지 못하게 통신사의 소액결제 서비스를 이용하게 하여 최대 30만원에 달하는 소액결제 사기피해를 유발시킨다. 스미싱은 다양한 유형의 문자메세지를 통해 사용자가 피해를 입을 수 밖에 없게 한다. 본 고는 대표적인 스미싱기법과 그 피해형태에 대해 알아보고, 예방법과 근본적인 해결책을 제시하고자 안드로이드용 스미싱방지 시스템을 설계 및 구현해보았다.

Chitosan Nanoparticles as a New Delivery System for the Anti-HIV Drug Zidovudine

  • Chitosan-based nanoparticles (CSNP) were prepared through ionic cross-linking and gelation of chitosan (CS) by tripolyphosphate (TPP). CS properties such as molecular weight, and preparation conditions were screened and the resulting nanoparticles were examined by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The obtained particles were consistently spherical with an overall diameter of approximately $107{\pm}20$ nm. They were successfully used as a carrier for Zidovudine, an anti-human immunodeficiency virus (HIV) which, to our knowledge, is novel. The encapsulation ability, loading capacity, and controlled release behavior for these CSNP was evaluated. Results indicated that their intrinsic properties were strongly affected by properties inherent to CS such as molecular weight, and by the preparation condition, such as cross-linking density, which depends on the concentration of the cross-linker. In vitro release tests for the entrapped zidovudine showed that the CNNP provided a continuous release that can last upwards 20 h.

바이러스 생활환의 후기 단계에 작용하는 항AIDS제의 탐색을 위한 HIV-1 Complementation System의 응용 (Application of HIV-1 Complementation System to Screen the Anti-AIDS Agents That Targets the Late Stage of HIV-1 Replication Cycle)

  • Continuous efforts are being made to find effective therapeutic agents against HIV-1, the causative agents of AIDS. In this study, we developed a cell-based assay system employing a trans-complementation for production of recombinant viruses which are capable of undergoing one round of replication in CD4+ T cells. This assay system was tested for ability to screen the agents that act at late stage of HIV-1 life cycle. The effect of a protease inhibitor on the trans-complementation assay was assessed. Recombinant HIV-1 viruses were prepared from a trans-complementation in the presence of various concentrations of protease inhibitor. Inhibition of single round infection of these recombinant viruses by protease inhibitor was observed to be a dose-dependent manner. Inhibitory effects of a protease inhibitor on HIV-1 Gag polyprotein processing by HIV-1 protease was detected at concentrations of the protease inhibitor compatible with inhibition of virus infection, confirming that the corresponding step was involved in the inhibitory mechanism of this compound. Together, these results provide evidence that a cell-based assay system established in this study can be used to screen the agents that target the late stage of HIV-1 life cycle.

  • Recently, with the development of the ICT environment, the use of the software is growing rapidly. And the number of the web server software used with a variety of users is also growing. However, There are also various damage cases increased due to a software security vulnerability as software usage is increasing. Especially web shell hacking which abuses software vulnerabilities accounts for a very high percentage. These web server environment damage can induce primary damage such like homepage modification for malware spreading and secondary damage such like privacy. Source code weaknesses checking system is needed during software development stage and operation stage in real-time to prevent software vulnerabilities. Also the system which can detect and determine web shell from checked code in real time is needed. Therefore, in this paper, we propose the system improving security for web server by detecting web shell attacks which are invisible to existing detection method such as Firewall, IDS/IPS, Web Firewall, Anti-Virus, etc. while satisfying existing secure coding guidelines from development stage to operation stage.

색채치료에 이용되는 가시광선 제시 시스템 개발 (Development of visible light stimulation system for color therapy)

  • 본 연구는 가시광선영역에서 각각의 색을 구현하여 색채치료에 이용하기 위한 시스템을 개발하는데 있다. 사용된 광원은 에너지 밀도가 전파장에 걸쳐서 일정한 Dichroic reflector hallogen lamp를 사용하였고, 광원에서 발생하는 적외선을 차단하기 위하여 IR filter를 사용하였다. 색채치료에 사용 가능한 순수한 파장의 색을 분리하기 위하여 long pass filter와 short pass filter로 구성되는 color filter set를 사용하여 구성하였다. 개발된 가시광선 제시 시스템은 빨강, 파랑, 노란 광을 낼 수 있도록 되어 있으며 각각의 광도는 2390 lx, 1020 lx, 17400 lx이다. 개발된 시스템의 객관적 효과를 검증하기 위하여 피부 서식균으로 항균 실험을 하나 결과 노란 광이 빨강이나 파란 광에 비해 세포성장 억제가 컸다. 대식세포와 피부암세포로 실시한 항염 실험은 각각의 색광에 대한 영향이 크지 않았다.

HIV Integrase Inhibitory Activity of Agastache regosa

  • We have been screening anti-HIV integrase compounds from Korean medicinal plants by using an in vitro assay system which is mainly composed of recombinant human immunodeficency virus type 1 integrase and radiolabeled oligonucleotides. From the above screening, the aqueous methanolic extract of the roots of Agastache rugosa exhibited a significant activity. Bioactivity-guided chromatographic fractionation of the methanolic extract resulted in the isolation of rosmarinic acid. The structure of the compound was determined by spectroscopic data and by the comparison with the reported values. The $IC_{50}$ of the rosmarinic acid was approximately $10{\mu}g/ml$ against HIV integrase.

안드로이드 앱 추천 시스템을 위한 Sybil공격과 Malware의 관계 분석 (Relationship Analysis between Malware and Sybil for Android Apps Recommender System)

  • 스마트 폰에서 활용할 수 있는 다양한 앱 (Apps)들의 개수가 기하급수적으로 증가함에 따라 개인 맞춤형 앱들을 추천해주는 시스템이 각광받고 있다. 하지만, 다양한 목적으로 악성 앱 (Malware)을 제작하여 구글 플레이(GooglePlay) 사이트에 등록 후 배포하는 경우가 동시에 증가함에 따라 사용자들은 만족도 하강의 단순 피해부터 개인정보 노출 및 금전 탈취 등 심각한 수준의 많은 피해까지 겪고 있다. 또한, 소셜 네트워크가 발전함에 따라 물리적인한 사용자가 많은 거짓 계정들을 만들어서 구글 플레이 사이트의 각 앱의 평점 (rating)들을 조작하는 시빌 공격(Sybil)도 존재할 수 있다. 이때까지 악성 앱과 시빌 공격 연구는 독립적으로 진행되어 왔다. 하지만 실시간으로 발전하고 있는 지능화된 공격 종류들을 고려했을 때 악성 앱 제작자가 구글 플레이 사이트에 노출 된 평점까지 조작 후 인지도를 높여서 결국 악성 앱을 다운받도록 유도하는 지능화된 공격의 유무를 판단하는 것이 중요하다. 따라서, 본 논문에서는 구글 플레이어 사이트를 직접 크롤링하고 시빌 공격과 악성 앱의 상관관계를 실험적으로 밝힌다. 실험결과, 구글 플레이어 사이트에서는 아직 시빌과 악성 앱의 상관관계가 낮음을 알 수 있었다. 이는 악성 앱 배포자가 인지도 및 평점까지 다수 조작하여 많은 사람들에게 노출되면 다양한 Anti-Virus (AV) 벤더들에게 오히려 더 빨리 탐지되어 목적을 달성할 수 없기 때문에 이를 고려하지 않았거나, 악성 앱 배포자가 악성 앱을 만들고 배포하는 것에만 초점을 두고 사이트 인지도 및 평점 조작까지는 아직 동시에 고려하지 않음으로 해석될 수 있다.

Insect Cell Surface Expression of Hemagglutinin (HA) of Egyptian H5N1 Avian Influenza Virus Under Transcriptional Control of Whispovirus Immediate Early-1 Promoter

  • In the present study, whispovirus immediate early 1 promoter (ie-1) was used to initiate surface expression of the hemagglutinin (HA) protein of Egyptian H5N1 avian influenza virus (AIV) by using the baculovirus expression vector system. The HA gene and whispovirus ie-1 promoter sequence were synthesized as a fused expression cassette (ie1-HA) and successfully cloned into the pFastBac-1 transfer vector. The recombinant vector was transformed into DH10Bac competent cells, and the recombinant bacmid was generated via site-specific transposition. The recombinant bacmid was used for transfection of Spodoptera frugiperda (Sf-9) insect cells to construct the recombinant baculovirus and to induce expression of the HA protein of H5N1 AIV. The recombinant glycoprotein expressed in Sf-9 cells showed hemadsorption activity. Hemagglutination activity was also detected in both extra- and intracellular recombinant HAs. Both the HA and hemadsorption activities were inhibited by reference polyclonal anti-H5 sera. Significant expression of the recombinant protein was observed on the surface of infected insect cells by using immunofluorescence. SDS-PAGE analysis of the expressed protein revealed the presence of a visually distinguishable band of ~63 kDa in size, which was absent in the non-infected cell control. Western blot analysis confirmed that the distinct 63 kDa band corresponded to the recombinant HA glycoprotein of H5N1 AIV. This study reports the successful expression of the HA protein of H5N1 AIV. The expressed protein was displayed on the plasma membrane of infected insect cells under the control of whispovirus ie-1 promoter by using the baculovirus expression vector system.

노로바이러스 검출을 위한 측면유동면역분석법 기반의 바이오리셉터 선별기법 개발 (Norovirus Targeted Bioreceptor Screening Method based on Lateral Flow Immunoassay (LFIA))

  • Later flow immunoassay (LFIA) is a protein analytical method based on immunoreaction. On the LFIA based protein analytical method, bioreceptor molecule plays a key role, and so a system that evaluates and manages the binding affinity of bioreceptor is needed to secure detection reliability. In this study, Lateral Flow Immunoassay based rapid Bioreceptor Screening Method (rBSM) is presented that provide a simple and quick evaluating method for the binding affinity to the target protein of the antibody as model bioreceptor. To verify this evaluation method, Virus-like particles (VLP) and anti-VLP antibodies are selected as a model norovirus, which is target protein, and the candidate bioreceptors respectively. Among the 5 different candidate antibodies, appropriate antibody could be sorted out within 30 minutes through rBSM. In addition, selected antibodies were applied to two representative LFIA based techniques, sandwich assay and competitive assay. Among these methods, sandwich assay showed more effective VLP detection method. Through applying selected antibodies and techniques to the commercialized mass production lines, an VLP detecting LFIA kit was developed with a detection limit of 1012 copies/g of VLPs in real samples. Since this proposed method in this study could be easily transformable into other combinations with bioreceptors, it is expected that this technique would be applied to LFIA kit development system and bioreceptor quality management.
