Clinical Evidence of Effects of Lactobacillus plantarum HY7714 on Skin Aging: A Randomized, Double Blind, Placebo-Controlled Study

  • The beneficial effects of probiotics are now widely reported, although there are only a few studies on their anti-aging effects. We have found that Lactobacillus plantarum HY7714 (HY7714) improves skin hydration and has anti-photoaging effects, and in the present study, we have further evaluated the anti-aging effect of HY7714 via a randomized, double blind, placebo-controlled clinical trial. The trial included 110 volunteers aged 41 and 59 years who have dry skin and wrinkles. Participants took 1 × 1010 CFU/day of HY7714 (probiotic group) or a placebo (placebo group) for 12 weeks. Skin hydration, wrinkles, skin gloss, and skin elasticity were measured every 4 weeks during the study period. There were significant increases in the skin water content in the face (p < 0.01) and hands (p < 0.05) at week 12 in the probiotic group. Transepidermal water loss decreased significantly in both groups at weeks 4, 8, and 12 (p < 0.001 compared with baseline), and was suppressed to a greater extent in the face and forearm in the probiotic group at week 12. Volunteers in the probiotic group had a significant reduction in wrinkle depth at week 12, and skin gloss was also significantly improved by week 12. Finally, skin elasticity in the probiotic group improved by 13.17% (p < 0.05 vs. controls) after 4 weeks and by 21.73% (p < 0.01 vs. controls) after 12 weeks. These findings are preliminary confirmation of the anti-aging benefit to the skin of L. plantarum HY7714 as a nutricosmetic agent.

The Efficacy of Korean Herbal Cosmetics for Facial Skin Elasticity (한방화장품이 안면부의 탄력 개선에 미치는 영향)

  • Objective : The purpose of this study was to evaluate the effect of korean herbal cosmetics for facial skin elasticity. Methods : We recruited patients and divided them to cosmetics group and control group randomly. We provided korean herbal cosmetics to cosmetics group and subjects in cosmetics group applied the provided cosmetics for 4 weeks. Control group waited and applied cosmetics that had previously used for 4 weeks. We assessed all subjects in 0 week, 2 weeks and 4 weeks. Primary assessment instrument was moire topography photo scale. Second assessment instrument was skin moisture, oiliness and elasticity which assessed with skintouch system v1.0, detox/regeneration questionnaire score and self-evaluating facial wrinkle/looseness score. Result : Moire topography photo scale improved in cosmetics group than control group significantly. Skin moisture and elasticity assessed with skintouch system improved in cosmetics group than control group significantly while skin oiliness assessed with skintouch system have not improved significantly. Detox/regeneration questionnaire score and self-evaluating facial wrinkle/looseness score improved in cosmetics group than control group significantly. Conclusion : Korean herbal cosmetics application might be effective method for facial skin elasticity.

Corni Fructus Inhibits Wrinkle Formation by Reduced Advanced glycation end product (AGEs) (산수유의 최종당화산물 억제로 인한 주름 개선효과)

  • Objectives : Corni Fructus (CF) is traditional herbal medicine used on polyuria, low back pain, and tinnitus. This study aimed to evaluate inhibits skin wrinkle formation effect of CF. Methods : To evaluate the produce inhibition effect of CF, SD-rats were distributed into four groups; normal rats (Nor), AGEs (advenced glycation end product)-induced rats (Con), AGEs-induced rats treated with 100mg/kg CF (CF). To induce AGEs, streptozotocin (50mg/kg) was administered intraperitoneally and after 3 days oral administrated 100mM methyl glyoxal for 3 weeks. Results : The oral administration of CF suppressed the reactive oxygen specis (ROS) in serum. The AGEs in skin tissues was significantly reduced through treatment of CF. Furthermore, the expressions of AGEs related proteins such as polyclonal anti-$N^e$-(carboxymethyl)lysine (CML), anti-$N^e$-(carboxyethyl)lysine (CEL), AGE receptors (RAGE) were decreased in CF treated group compared with the control group in skin tissues. Inflammation-related proteins such as Nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kB), monocyte chemoattractant protein-1 (MCP-1), interleukin-6 (IL-6) reduced in CF treatment group than control group. AGE-induced rats exhibited that the significant decreased collagen however, CF treatment (100mg/kg of body weight) up regulated collagen by improved the expression levels of skin fibril-related genes such as Matrix metalloproteinase (MMP-1). Conclusion : Taken together, our study suggests that CF regulates ROS to prevent accumulation of AGEs and inhibits skin wrinkles. Our finding indicate that CF may be an effective agent for inhibits AGEs formation, and improved skin wrinkle.

  • Recently plant extracts have obtained increased attention because of their health-related beneficial aspects. In this study, we evaluated anti-oxidation, whitening and anti-wrinkle of four different extracts obtained from Rubus coreanus (ethanol extracts of unripe, ripe fruits, leaves/stems, and roots). The ethanol extract of unripe fruits of Rubus coreanus generally showed stronger whitening and the anti-wrinkle activities than other extracts of Rubus coreanus. As a results, we knew that the ethanol extract of unripe Rubus coreanus fruit had mote possibility as a whitening and anti-wrinkle ingredient for skin care products.

Enhancement of Skin Whitening and Anti-wrinkle Activities of the Co-culture of Lactobacillus rhamnosus and Lactobacillus paracasei (Lactobacillus rhamnosus와 Lactobacillus paracasei 복합 배양의 피부 미백 및 주름 개선 활성 증진)

  • In this study, cosmeceutical activities of the co-cultures of both lactic acid bacteria isolated from the hands of women, Lactobacillus rhamnosus (L. rhamnosus) and Lactobacillus paracasei (L. paracasei) were first reported: For whitening and anti-wrinkle activities, the co-culture showed the highest tyrosinase inhibition of 20.68%, compared L. paracasei and L. rhamnosus. The co-culture also showed the highest inhibition of melanin synthesis as 63.7%. In observing the anti-wrinkle activities of the co-culture, it generated only 3726.3 pg/mL of matrixmetalloproteinase-1 (MMP-1) production when 13613.5 pg/mL and 13012.0 pg/mL of MMP-1 production were estimated from L. rhamnosus and L. paracasei. Besides these, the extract from the co-culture yielded higher collagen production as 380.7 ng/mL, compared to 323.4 ng/mL and 304.1 ng/mL from L. paracasei and L. rhamnosus. These results indicate that the co-culture of both lactic acid could improve its cosmetic activities. This hypothesis was also confirmed that the co-culture of both bacteria showed strong antioxidant activities of DPPH free radical scavenging while the extract of L. rhamnosus and L. paracasei.

  • Vaccinium uliginosum L. (VU) is a flowering plant in the genus Vaccinium has berry fruit. This study was performed to investigate the effect of water extract of Vaccinium uliginosum L. and fractions on inhibition of melanogenesis and wrinkle formation. One hundred grams of the Vaccinium uliginosum L. was extracted with 2,000 mL of water ($90^{\circ}C$, 16h, 2 times). The water extracts were lyophilized and stored at $4^{\circ}C$ until used. Vaccinium uliginosum L. extracts showed scavenger activities on DPPH radical, superoxide anion radical, hydroxyl radical, hydrogen peroxide and singlet oxygen radical, dose dependently. And VU extract and fractions reduced melanin contents on B16F10 melanoma and inhibited the expression of melanogenesis-related proteins, tyrosinase, tyrosinase-related protein (TRP-1) and dapachrometa utomerase (Dct, TRP-2). Moreover VU extract and fractions stimulated procollagen production and inhibited MMP-1 production in human fibroblast. And it decreased degree of wrinkle formation in hairless mouse skin that induced by DVB irradiation for 9 weeks. From the above results, it is possible that Vaccinium uliginosum L. may be developed to be the health functional food and functional cosmetics that have anti-melanogenesis and anti-wrinkle effect.

Experimental studies about the inhibitory effect on tyrosinase and elastase activities by various herb medicines (수종(數種)의 한약재의 Tyrosinase와 Elastase 활성 억제 효과에 대한 실험적 연구)

  • Objectives : The purpose of this study is to investigate whitening effects and anti-wrinkle effects of a few 80% ethanol extracted herbal medicines. Methods : In the first study, a few 80% ethanol extracted herbal medicines were screened for their inhibitory activities against the tyrosinase. In the second study, a few 80% ethanol extracted herbal medicines were screened for their inhibitory activities against elastase. Results : 1. We showed 28%, 27% and 19% inhibitions of mushroom tyrosinase at 500 $\mu$g/ml concentration of ASR, AIF and ABR extracts and they were showed higher anti-tyrosinase activity than arbutin's. We also could observe that the decreased mushroom tyrosinase activities in RR, CML, LR, AGR and TH extracts. 2. RR, AF and ABR (final concentrstion 1 mg/ml) were appeared 60%, 98%, 83% of inhibitions of elastase activity, and they were showed higher anti-elastase activity than that of ursolic acid. We also could observe that the decreased elastase activities in AIF, AR, LR and CML extracts. Conclusions : These results suggest that ASR, AIF and ABR extracts contribute to the anti-melanin activities and represent potential sources of whitening agent, and RR, AF and ABR extracts contribute to the anti-elastase activities and represent potential sources of anti-wrinkle agent. These results suggest that some herbal medicines could be strong potential sources of inhibition about anti-aging and whitening effects for the skin.

Effects of Sibseonsan as an Anti-Inflammatory, Anti-Wrinkle, and Skin Whitening Treatment

  • Background: The purpose of this study was to investigate whether Sibseonsan (SSS) is an effective antiinflammatory, anti-wrinkling, and whitening agent. Methods: To determine whether SSS had an anti-inflammatory effect, a murine macrophage cell line was used (RAW 264.7) and production of DPPH, NO, TNF-α, and PGE2 were measured. To ascertain potential anti-wrinkle effects of SSS in these cells, collagenase and elastase production were measured. To verify whether SSS had a whitening effect, tyrosinase activity and DOPA staining were performed using a melanoma cell line (B16/F10). Results: There was no significant reduction in survival of SSS-treated RAW 264.7 cells, up to 400 ㎍/mL. Free radical scavenging (23.96 ± 1.85%) was observed in RAW 264.7 cells treated with SSS at a concentration of 400 ㎍/mL. The SSS treatment group (400 ㎍/mL) significantly inhibited NO production compared with the LPS stimulated treatment group. The SSS treatment of macrophage cells appeared to reduce production of TNF-α in a concentration dependent manner. There was a significant reduction in the concentration of PGE2 by about 25% in the SSS treatment (400 ㎍/mL) group (p = 0.05). Compared with the control, the production of collagenase and elastase in B16/F10 cells treated with SSS (400 ㎍/mL) was greater by 26.37% and 45.71%, respectively. The SSS treatment (400 ㎍/mL) group showed a significant reduction by about 17% in tyrosinase production in B16/F10 cells. The SSS treatment group showed little change in DOPA staining. Conclusion: SSS extract may be useful for the treatment and prevention of inflammatory diseases and may have anti-wrinkle and whitening effects. These results may support the use of SSS in clinical practice.

  • The human skin is constantly exposed to environmental irritants such as ultraviolet, smoke and chemicals. Free radicals and reactive oxygen species caused by them play critical roles in cellular damage. They not only injure the skin structure but also participate in the immensely complex inflammatory reaction. Anti-wrinkle effects of the Oriental herb extracts(OHE) were evaluated by the determination of anti-oxidation, collagenase inhibition and collagen synthesis in normal human fibroblast. OHE showed antioxidative activity as high as vitamin C, trolox and DL-penicillamine. Also OHE showed promotive effect on collagen synthesis and inhibitory effect on collagenase activity. These results demonstrated that OHE could be useful as an anti-wrinkle cosmetic ingredient.

In vitro anti-skin-aging effects of dried pomegranate concentrated powder

  • Purpose : In this study, we intended to observe the anti-wrinkle and moisturizing effects of dried pomegranate juice concentration powder (PCP) using in vitro test. Materials and methods : Antioxidant effects of PCP were determined by free radical scavenging capacity (DPPH assay) and the cytotoxicity of PCP was examined in human keratinocyte (HaCaT) and human primary dermal fibroblast-neonatal (HDF) cells. To investigate the moisturizing effect of PCP, hyaluronan synthesis was examined in HaCaT cells. Activity of procollagen production were assessed in HDF cells and elastase inhibition properties of PCP were evaluated in cell free condition, to determine their anti-wrinkle effects. Metalloproteinase 1 (MMP-1) activity was also assessed following UVB irradiation, in the current in vitro experiment. Results : No PCP treatment related significant cytotoxic effects were demonstrated against to the both HDF and HaCaT cells. PCP showed favorable free radical scavenging activities in dose-dependent manner. In PCP-treated HaCaT cells, hyaluronan synthesis was non-significantly but markedly increased, and pro-collagen productions were significantly increased in HDF cells, at all three different concentrations (0.25, 0.75 and 1 mg/ml), and elastase inhibitory activities were observed by PCP treatment. A significant decrease in UVB-induced MMP-1 activity was also observed in 1 mg/ml PCP-treated HDF cells as compared to those of UVB-exposed cells. Conclusions : Taken together, these results suggest that PCP has favorable antioxidant, anti-wrinkle and moisturizing effects without meaningful cytotoxicity on HDF and HaCaT cell lines.
