Antibacterial and antifungal effects of Korean propolis against ginseng disease

  • We investigated the anti-microbial activity of propolis against the pathogenic bacteria and fungi on ginseng. We selected six microbials that caused postharvest root rots in ginseng. Propolis extracts were prepared by using the ethanol extraction method. We seeded the bacteria and fungi related to ginseng disease on a specific culture medium, and treated it with propolis extracts by using the paper disc method. Propolis extracts indicate the anti-microbial activity against Paenibacillus polymyxa, Fusarium solani, Rhizoctonia solani AG-1 and Pythium ultimum. However, the anti-fungal activity of propolis is weak on Pseudomonas fluorescens subsp. Cellulosa and Colletotrichum gloeosporioides. As a result, the antimicrobial effects of propolis against microbial that prevent ginseng growth were confirmed. The antimicrobial effects are shown according to the concentration of propolis against root rot. The fungi also showed antibacterial effects in a dose-dependent manner.

저식염 고추장 양조시 항균물질 혼합첨가의 영향 (Effect of Combined Use of various Anti-microbial Materials on Brewing of Low Salted Kochujang)

  • 소금농도를 9%에서 6%로 줄이고 항균물질로 알코올과 겨자, 키토산을 혼합첨가 하여 제조한 저식염 고추장의 숙성중의 효소활성도와 미생물상 및 이화학적 특성을 비교하였다. 숙성 중 수분활성도의 저하는 식염 9%인 대조구에서 심하였고, 숙성 중에 효모수는 증가하였으나 알코올-겨자 혼합 첨가로 효모수를 조절할 수 있었다. 고추장 숙성 중 ${\beta}-amylase$ 활성은 항균물질 첨가시 증가하였으나 ${\alpha}-amylase$와 protease 활성은 조금 감소하였다. 고추장은 숙성이 진행되면서 L차 a-, b-값이 증가하였으며 알코올 첨가 고추장이 $({\Delta}E)$값의 변화가 적었다. 적정산도는 항균물질의 첨가로 숙성 후기에 저하되었으며, 산화환원전위의 저하는 대조구에서 심하였다. 고추장 숙성 중 총당의 감소는 알코올-겨자 혼합 첨가구(EM)에서 적어 환원당량이 많았으며, 숙성 후기의 알코올의 생성은 알코올에 키토산을 혼합 첨가한 고추장(EC)에서 높았다. 고추장 숙성 중기까지 아미노태 질소는 증가하였고, 숙성중 암모니아태 질소의 생성은 알코올-겨자-키토산 혼합 첨가구(EMC)에서 적었다. 12주 숙성된 고추장을 관능 평가한 결과 색과 향기에서 알코올(E) 또는 알코올-겨자 혼합 첨가구(EM)가 좋아 저식염 고추장 제조시 알코올 또는 알코올-겨자를 혼합 첨가하는 것이 전체적인 기호도에서 양호하였다.

천연방부제로서 항균오일의 항균력 비교 (Comparision of Anti-microbial Oils as Natural Preservatives)

  • 티트리오일 2종과 편백나무에서 추출한 천연오일을 합성방부제인 methylparaben과 항균력을 비교하였다. 대상 균주로 그람양성균인 S. aureus (KCTC 1916)와 그람음성균인 E. coli (KCTC 2571)와 P. aeruginosa (KCTC 2513)를 사용하였으며, 진균류인 C. albicans (KCTC 7965)와 A. niger (KCTC 6911), 그리고 여드름균인 P. acnes (KCTC 3314)를 사용하였다. 그람양성균의 항균력실험에서 천연오일은 합성방부제보다 우수한 항균활성을 보였으며, 그람음성균에서는 유사한 정도의 활성을 나타내었다. 특히, 진균과 여드름균에 대해서는 합성물질에 비해 천연오일에서 월등한 활성을 볼 수 있었다. 실험군에서 항균력은 티트리오일 > methylparaben > 편백정유의 순으로 나타났다. 또한, DPPH 법에 의한 항산화실험과 lipoxygenase를 이용한 항염증실험을 통해 천연오일의 항산화 및 항염증 효과를 볼 수 있었다. 실험결과, 천연오일은 널리 사용되고 있는 화학적인 방부제나 항산화제를 충분히 대체할 수 있으리라 기대된다.

가시상추 유래 생리활성물질의 분리 및 특성 규명 (Isolation and Characterization of Bio-active Materials from Prickly Lettuce (Lactuca serriola))

  • 가시상추(Prickly lettuce : Lactuca serriola)로부터 여러 가지 추출용매를 사용하여 생리활성물질을 분리하였다. 가시상추의 일반성분, vitamin, polyphenol성 물질 그리고 flavonoid 성분의 함량을 분석하였다. 또한, MeOH 조추출물에 의한 아질산염 제거 활성, 항산화 활성, 항균 활성을 측정하였다. MeOH 조추출물의 EtOAc 분획에서 가장 높은 아질산염 제거 활성, 항산화 활성 그리고 항균 활성을 보여주었다. 그리고 polyphenol성 물질과 flavonoid의 함량도 가장 높았다. EtOAc 분획 중 subfraction 2 (EtOAc:MeOH=3:1)에서 가장 높은 항산화 활성을 보여주었다. Subfraction 2는 GC-MS에 의해 ${\rho}$-coumaric acid와 caffeic acid로 동정되었다. 가시상추 추출물의 용매 분획별 항균 활성을 검색한 결과, EtOAc 분획에서 가장 높은 항균 활성을 보여주었다. 이러한 연구 결과는 가시상추 유래의 생리활성물질의 생물학적 활성을 이해하는데 필요한 기본 데이터를 제공해 줄 것이다.

  • Bamboo salt is a special processed salt by Korean traditional recipe. Recent study results showed that bamboo salt or bamboo salt with some other materials like herbal extracts have the anti-microbial activity, inhibition effects of dental plaque and gingival inflammation. Bamboo salt also showed anti-cariogenic effects; remineralization and acid resistance. Compare to fluoride toothpaste, bomboo salt toothpaste with fluoride showed the more effective remineralization on inner part of the early dental caries lesion. It increased the surface hardness and decreased lesion depth of early dental caries lesion. Thus, it is suggested thai bamboo salt could be used as a anti-microbial, anti-plaque, anti-inflammatory and anti-cariogenic material for oral disease prevention. Especially, bamboo salt dentifrice with fluoride can be recommanded as a useful remineralizing agent.

천연소재의 화장품약리활성에 관한 연구 (Study on Cosmeceutical Acitivies of Natural Material)

  • The most extensively used synthetic antioxidants are butylated hydroxyanisole (BHA), butylated hydroxy toluene (BHT) and tert-butylhydroquinone (TBHQ). However BHT and BRA have been suspented of being responsible for liver damage andcar cinogenesis. Therefore, the importance of the search and exploitation of natural antioxidant, especially of plant origin, has greatly increased in recent years. Plant contain a wide variety of chemicals that have potent biological effects. As a result, there has been a growing interest in the use of herbs as a source of therapeutic drugs. The aim of this study was to assess the antioxidant and cosmeceutical of natural materials. The antioxidant and cosmeceutical activity of natural materials were investigated by hydroxyl radical scavenging, superoxide dismutase (SOD) -like, xanthine oxidase inhibition, tyrosinase inhibition, anti-microbial and astringent.

Comparative study on bone regeneration between silk mat incorporated 4-hexylresorcinol and collagen membrane

  • The purpose of this study was (1) to demonstrate the anti-microbial properties of 4-hexylresorcinol (4HR) loaded silk mat and (2) comparison of bone formation between 4HR incorporated silk mat and collagen membrane. Anti-microbial properties of 4HR incorporated silk mat was done after sterilization of silk mat (autoclaving and ethylene oxide gas). For the evaluation of bone formation, bilateral bony defects (size: 8 mm) were prepared in the parietal bone of the rabbits (n=10). 4HR incorporated silk mat (size: $10{\times}10mm$) was applied on the right defect. For the comparative purpose, the same size of commercial collagen membrane was applied on the left defect. The anti-microbial properties of 4HR incorporated silk mat were maintained after sterilization process. When compared bone mineral density and bone volume, there was no statistically significant difference between groups at 4 weeks and 8 weeks after operation (p>0.05). In conclusion, 4HR incorporated silk mat could be autoclaved without concern of anti-microbial properties loss. In addition, 4HR incorporated silk mat showed similar bone regeneration to collagen membrane. Therefore, 4HR incorporated silk mat might be considered for the application of open membrane technique.

In vivo와 In vitro 평가모델을 利用한 韓藥抽出物의 毛髮成長 및 促進에 미치는 實驗的 硏究 (Extracts for the Hair Growth Stimulation using In vivo and In vitro Test Models)

  • To screen the effective materials for hair loss treatment, several natural extracts were tested using in vivo and in vitro test models. Firstly, all test materials were applicated onto the back skin of C57BL/6 mouse and then hair growth promoting effect was measured using hair growth index. As a result, Prunus mume, black bean, Brassica campestris subsp. black sesame and Rubi Fructus showed potent hair growth promoting effect, ranking as 1.5-2.0 of hair growth index. However, there were no plant extracts, which have remarkable potential of growth promotion of human hair dermal papilla cells cultured in vitro. In the experiments of 5${\alpha}$-reductase type Ⅱ inhibition assay, Prunus mume, Eriobotryae Folium showed effective potential to inhibit the activity of 5${\alpha}$-reductase type Ⅱ. To investigate the possible involvement of the effect of several plant extracts on the gene expression of growth factors in human hair dermal papilla cells, RT - PCR analyses were performed. However, there were no plant extracts, which have profound effect on the gene expression of several growth factors such as IGF-I, KGF, HGF and VEGF in the dermal papilla cells. Another tests for inhibition of microbial such as P. acne were also carried out to find whether these plant extracts have anti -microbial activities. Rubi Fructus showed anti -microbial effects on Propionibacterium acnes, which is believed as a pathogen of acne. Together, these results showed several plant extracts can be used for hair growth promotion.

To investigate the possible involvement of effects of several plant extracts on the gene expression of growth factors in human hair dermal papilla cells, RT-PCR analyses were performed. As a consequences, Mentha haplocalyx Briq., Cimicifuga foetida L., Eclipta prostrata (L.) L., Pinus densiflora S. et. Z, and Polygonum muitifiorum Thunb revealed the regulatory roles on the expression of growth factors such as IGF-I, KGF, HGF and VEGF in the dermal papilla cells. Another test for inhibition of microbial such as P. acne and P. ovale were also carried out to find whether these plant extracts have anti-microbial activities. Morus alba L. and Chaenomelis Fructus showed anti-microbial effects on Propionibacterium acnes, which is believed as a pathogen of acne. Together, these results showed several plant extracts can be used for hair growth promotion.

Achyranthes japonica Nakai Water Extract Suppresses Binding of IgE Antibody to Cell Surface FcεRI.

  • Achyranthes japonica Nakai (AJN) water extract has a variety of physiological properties, including anti-diabetic, anti-cancer, anti-inflammatory, anti-microbial, and anti-oxidative activities. In the present study, the inhibitory effects of AJN extract were investigated in high affinity immunoglobulin E receptor ($Fc{\varepsilon}RI$)-mediated KU812F cells activation. AJN extract showed suppressive effects on histamine release and intracellular calcium [$Ca^{2+}$]i elevation from anti$Fc{\varepsilon}RI$ antibody (CRA-1)-stimulated cells in a dose-dependent manner. Flow cytometric analysis showed that AJN extract treatment caused a dose-dependent decrease in the cell surface $Fc{\varepsilon}RI$ expression and the binding between the cell surface $Fc{\varepsilon}RI$ and the IgE antibody. Moreover, reverse transcription-polymerase chain reaction analysis showed that levels of the mRNA for the $Fc{\varepsilon}RI$ ${\alpha}$ chain was decreased by treatment with AJN extract. These results indicate that AJN extract may exert anti-allergic effects via the inhibition of calcium influx and histamine release, which occurs as a result from the downregulation of the binding of IgE antibody to cell surface $Fc{\varepsilon}RI$. This mechanism may occur through $Fc{\varepsilon}RI$ expression inhibition.
