Scutellaria baicalensis Extracts as Natural Inhibitors of Food Browning (천연 갈변저해제로서 황금 추출물의 효소적 갈변 저해 효과)

  • This study was designed to develop natural browning inhibitors. The anti-browning effects of distilled water (SBD) and 80% ethanol extracts (SBE) of Scutellaria baicalensis Georgi in apple slices were investigated by L and ${\Delta}E$ values. Both SBD and SBE were effective in reducing the browning of apple slices and were successively fractionated into chloroform ($CHCl_3$), ethyl acetate (EtOAc), and water ($H_2O$) fractions. These extracts were measured for total phenolic content, flavonoid content, anti-oxidative activity (through free radical scavenging activity and the FRAP assay), ferrous ion chelation, and the inhibition of PPO (polyphenol oxidase) activity. Overall, fractions of SBE were better than fractions of SBD in all measurements. The highest total phenolic and flavonoid content were measured in the EtOAc and $CHCl_3$ fractions of SBE. EtOAc and $CHCl_3$ fractions also exhibited the highest anti-oxidative activities (in DPPH and ABTS free radical scavenging and the FRAP assay). Unusually, the highest ferrous ion chelating capacity was found in the $H_2O$ fraction of SBD, but the other fractions showed more than triple the ascorbic acid already in use. Also, $CHCl_3$ fractions showed a stronger inhibition of PPO activity than ascorbic acid. All of these results suggest that EtOAc and $CHCl_3$ fractions from Scutellaria baicalensis can be used as natural anti-browning agents.

Occurrence of Skin Browning by Mechanical Injuries on the Fruits of 'Mansu' Pear (만수 품종 배의 과피 갈변 원인 구명)

  • This research was carried out to elucidate the causes of fruit skin browning in 'Mansu' pear for the last 2 years. It was observed that skin browning was induced even by the small mechanical injury produced during grading and packing for the market supplies after harvest on the fruits of 'Mansu' pear. The incidences of fruit skin browning in pears treated with artificial mechanical injuries were investigated between 'Niitaka' and 'Mansu' pears. The results showed that fruits of 'Mansu' are more susceptible to skin browning than those of 'Niitaka', We also found that the epidermis of fruits in 'Mansu' pear was thinner than that of 'Niitaka', and that there was lower incidence of fruit browning in epidermis of pears with high chlorophyll content than those with low chlorophyll content. The skin browning in fruits could be considerably reduced by sorting and grading them wrapped with paper bags for the fruits of 'Mansu' pear.

Efficacy of Sodium Hypochlorite and Acidified Sodium Chlorite in Preventing Browning and Microbial Growth on Fresh-Cut Produce

  • The use of suitable sanitizers can increase the quality of fresh-cut produce and reduce the risk of foodborne illnesses. The objective of this study was to compare the washing effects of 100 mg/L sodium hypochlorite (SH) and 500 mg/L acidified sodium chlorite (ASC) on the prevention of enzymatic browning and the growth of microbial populations, including aerobic plate counts, E. coli, and coliforms, throughout storage at $4^{\circ}C$ and $10^{\circ}C$. Fresh-cut zucchini, cucumbers, green bell peppers, and root vegetables such as potatoes, sweet potatoes, carrots, and radishes were used. Compared to SH washing, ASC washing significantly (p<0.05) reduced microbial contamination on the fresh-cut produce and prevented browning of fresh-cut potatoes and sweet potatoes during storage. More effective inhibition of aerobic plate counts and coliforms growth was observed on fresh-cut produce treated with ASC during storage at $10^{\circ}C$. Polyphenol oxidase (PPO) activity of fresh-cut potatoes and sweet potatoes was more effectively inhibited after washing with ASC. The use of 500 mg/L ASC can provide effective antimicrobial and anti-browning treatments of fresh-cut produce, including processed root vegetables.

  • In the present study, the effects of HML on lipolysis, adipocyte browning, autophagy, and proliferation were investigated. HML affected lipolysis by increasing the protein levels of ATGL and HSL, and phosphorylation levels of HSL and AMPK. Furthermore, HSL decreased the perilipin-1 levels. In addition, free glycerol content was increased by HML treatment. HML affected adipocyte browning by increasing the protein levels of UCP-1, PGC-1α, and PRDM16. In addition, HML affected autophagy by increasing the levels of LC3-I and LC3-II, and decreasing those of SQSTM1/p62. Moreover, HML affected adipocyte proliferation by suppressing the proliferation of 3T3-L1 cells due to arrest of the cell cycle via blocking the expression of β-catenin and cyclin D1. These results suggest that HML induces lipolysis, adipocyte browning, autophagy, and inhibits excessive proliferation of adipocytes.

Effect of Anti-Browning Agent Application on the Improvement of Quality Maintenance for Minimally Processed Pear Slices (갈변 방지제 처리가 최소 가공한 배 절편의 저장성 향상에 미치는 영향)

  • This study was conducted to increase the keeping quality of the minimally processed pear slices during the cold storage. Korean pears, 'Shin-go (Niitaka)', 'Chu-hwang' and 'Won-hwang’ were immersed in 1% ascorbic acid, 0.1% calcium propionate, 1% citric acid, 0.2% N-acetyl cysteine and 0.5 M 4-hexylresorcinol solution for 3 three minutes and then stored at $1{\pm}0.5^{\circ}C$ for 10 days. We have also examined into the firmness and the color difference of pears slices as affected by the application of the anti-browning agents. The firmness of pears slices which that were immersed in 0.1% calcium propionate and 0.2% N-acetyl cysteine solution were not significant at did not differ significantly after 10 days after of cold storage. However, the ${\Delta}E^*$ values of 'Shin-go' slices which that were treated with 0.1% calcium propionate and 0.2% N-acetyl cysteine solution and stored for 10 days decreased by 3.18 and 3.83, when compared with that in of the control, which decreased by 6.36. The ${\Delta}E^*$ values of 'Chu-hwang' slices which that were treated with 0.2% N-acetyl cysteine and 0.1% calcium propionate solution had the slight difference by differed by only 2.09 and 2.14, when compared with that in of the control, which differed by 3.04. The ${\Delta}E^*$ values of 'Won-hwang' slices which that were treated with 1% citric acid and 0.5 M 4-hexylresorcinol were 4.49 and 5.83, respectively, while that in of the control decreased by 8.95. It was assumed that the anti-browning agent treatment had the different activities among varieties of the pear varieties slices, however, application of 0.2% N-acetyl cysteine and 0.1% calcium propionate application had greater the higher antibrowning activity.

Establishment of proliferation and regeneration system of PLBs in Phalaenopsis by treatments of a variety of types of medium, sucrose concentrations and anti-browning agents (다양한 배지종류, sucrose 농도 및 갈변억제물질 처리에 의한 팔레놉시스 PLB 증식 및 재분화 체계확립)

  • To establish an efficient proliferation and regeneration of PLBs (protocorm-like bodies) of Phalaenopsis plants, a variety of propagation medium types, various concentraions of sucrose as well as liquid and solid type were tested in this study. Further, activated charcoal, citric acid and ascorbic acid were compared whether these agents are suppose to reduce the browning in culture process using PLBs of Phalanopsis plants. With regard to the proper propagation medium, VW medium showed 1.3 ~ 2 times highr than those of other medium in an index of increasing for fresh weight and 50% higher than those of other medium in the frequency of shoot regeneration. However, regarding liquid and solid types of culture, there were no significant differences in the proliferation of PLBs and regeneration of shoots from PLBs. In the experiment for a variety of sucrose concentrations (0 ~ 50 g/l), 10 g of sucrose showed 30 ~ 50% higher than other concentrations in increasing index and 10 ~ 50% higher in the regeneration of shoots from PLBs. Regarding the reduction of browning in tissue culture via PLBs of Phalaenopsis plants, 1 g of activated charcoal showed only 1.5% browning of PLBs cultured. Whereas, other treatments including citric acid and ascorbic acid showed 6 ~ 16% of browning of PLBs. Therefore, activated charcoal was selected as an efficient anti-browning agents for the culture of PLBs in Phalaenopsis plants. Using above-described results can be contibuted to the establishment of mass propagation system using PLBs of Phalaenopsis plants in the future.

Effect of Postharvest High CO2 Treatment and Anti-moisture Pad on the Quality of Fresh Oyster Mushroom during Export Simulation (모의 수출 조건에서 수확 후 CO2와 부직포 패드 처리가 느타리버섯 품질에 미치는 영향)

  • This study was aimed to find factors responsible for export potential of oyster mushroom through postharvest treatments including high pressure $CO_2$ and anti-moisture pad. Temperature fluctuation during export simulation induced the condensation of excess moisture resulting in the quality loss due to browning and decay. Anti-moisture pad was effective on inhibition of browning occurrence in part. High temperature exposure ($15^{\circ}C$) as retail simulation greatly offset the positive effect of anti-moisture pad. Short term treatment of high pressure $CO_2$ delayed the surface color changes as hunter L, a, and b values. There was no consistent tendency in total sugar and phenolics between treatments. In general, the export potential of small size mush-room (export size) at harvest was higher than large ones (domestic size). The major factors asso-ciated with the quality decrease of oyster mushroom during export were moisture condensation in packages and temperature fluctuation. Further research is required to improve mushroom export procedures.

Effects of Plant Growth Regulators and Anti-oxidants on Rapid Multiplication of Cymbidium kanran (한란의 급속증식을 위한 생장조절물질과 항산화제 처리효과)

  • Effects of plant growth regulators and anti-oxidants for rapid multiplication of Cymbidium kanran were investigated. The best gelling agent was 2.5 g/1 gelrite which needed less quantity (about 28%) and half price than 9 g/1 chemi-cal agar. Undefined edible agar was only a little bit worse than chemical agar in growth, but the price was half as much as the latter. The higher concentration of BA and NAA, the deeper browning of medium that prevented from performing its functions of plant growth regulators. Polyvinylpyrrolidone (M.W. 40,000) was the most effective anti-oxidant other than ascorbic acid, aspartic acid, and rutin in protecting the browning of medium, enhancing the effect of plant growth regulators, and thus prolonging the subculture cycle. Vigorous seedlings were obtained by 0.1∼1.0 mg/1 BA,0.1 mg/1 NAA and 1 g/1 polyvinylpyrrolidone treatments. Therefore, the best result for growth and econo-mic aspects in rhizome culture of Cymbidium kanran were obtained by using MS basal medium with 2.5 g/1 gelrite, 1 g/1 polyvinylpyrrolidone, 0.1∼1.0 mg/1 BA and 0.1 mg/1 NAA.

Easy production techniques for clear pear juice and its antioxidant activities of 'Chuwhangbae' pear (추황배 청징배즙의 제조 및 항산화 활성)

  • This study was carried out to optimize the filtration, clarification, anti-browning processing conditions of clear pear juice and to investigate changes in antioxidant activity of pear juice produced through different heating treatment. For the filtration with cheese cloth, filter paper, or centrifugation (10 min at 3,000 rpm), the pear juice was most efficiently filtered with centrifugation because it showed the highest lightness (L value) and lowest yellowness (a value). Among various clarifying agents, 1% of gelatin or bentonite clarified effectively pear juice but tannin or egg albumin did not. Among anti-browning agents (0.1%) like L-ascorbic acid, NaCl or citric acid, L-ascorbic acid prevented the browning of pear juice with the lowest browning index value (2.62), compared to that of NaCl (2.74), or citric acid (2.87). Fructose, sucrose, glucose and sorbitol were present in the pear juice, the fructose and glucose contents increased but that of sucrose decreased in the heated pear juice. The total polyphenol content of the heated pear juice significantly increase, and did the total flavonoid contents in the clear and heated pear juice. The DPPH radical scavenging activity and nitrate scavenging activity were higher in the clear and heated pear juice than in the fruit crush.

Quality Changes in Fresh-Cut Potato (Solanum tuberosum var. Romano) after Low-Temperature Blanching and Treatment with Anti-Browning Agents (저온 blanching 및 갈변저해물질의 처리에 따른 최소가공 감자의 저장 중 품질변화)

  • Quality changes in fresh-cut potatoes during storage at $4^{\circ}C$ after treatment with low-temperature blanching and antibrowning agents were studied. Fresh-cut potatoes were treated by dipping for 1.5 min in a browning inhibitor solution containing 0.5% (w/v) ascorbic acid, 0.5% (w/v) citric acid, 0.5% (w/v) sodium chloride, 0.1% (w/v) trehalose, and 0.005% (w/v) biotin, at $60^{\circ}C$, with subsequent cooling for 1.5 min and storage at $4^{\circ}C$. The browning properties of fresh-cut potatoes were examined by measurement of polyphenol oxidase (PPO) activity and total phenolic content. Changes in quality attributes over a 14-day period were assessed in terms of titratable acidity, pH, water-soluble solid level, and gas analysis at $4^{\circ}C$. During storage, PPO activity increased, with the lowest activity seen after about 7 days of storage. Treatment with antibrowning solution at $4^{\circ}C$ increased visual sensory attributes during storage. Low-temperature blanching in distilled water more effectively inhibited browning compared with exposure to browning inhibitor solution, as assessed after 7 days of storage. Fresh-cut potatoes respired aerobically after different treatments during storage at $4^{\circ}C$.
